    Background: Bacterial resistance is a fundamental aspect of One Health, which is defined as the inseparable unity of animal, human, and environmental health. Epidemiological surveillance on the spread of bacterial resistance in animals and their derived products is essential given that meat, milk, and dairy products can carry resistant microorganisms that may reach humans through the food chain either by direct consumption or by handling the product. To eliminate the scarcity of information, it is necessary to characterize the epidemiological situation in terms of bacterial resistance in dairy production in northeastern Brazil. Thus, the objective of this study was to determine the frequency and antimicrobial susceptibility profile of bacteria isolated from goat milk samples from some municipalities in the Brazilian state of Sergipe.Materials, Methods & Results: The study included 28 goat farms in 4 municipalities of the Semiarid region of the State of Sergipe in Northeastern Brazil, namely CanindĂ© de SĂŁo Francisco (n = 11), Nossa Senhora da GlĂłria (n = 6), Poço Verde (n = 6), and Porto da Folha (n = 5). All lactating does of each herd (n = 263) aged >1 year were, sampled randomly by non-probabilistic convenience sampling. Milk samples were collected from both teats, resulting in 526 samples in total.Bacterial culturing and isolation were performed, followed by antimicrobial susceptibility profile analysis to the following active principles: amoxicillin with and without clavulanic acid, amikacin, ampicillin with sulbactam, ciprofloxacin, cefalexin, cefalotin, ceftriaxone, chloramphenicol, doxycycline, enrofloxacin, gentamicin, levofloxacin, ofloxacin, penicillin G, and tetracycline. A survey form was used to obtain zootechnical information for each farm. Data are described as absolute and relative frequencies. The significance assessment of the differences between herd characteristics and bacterial isolation was performed using Pearson's chi-squared test. Bacterial isolation occurred in 15.4% (81/526) of the samples from 23.2% (61/263) of the goats. Escherichia coli (45.9% = 28/61), Staphylococcus caprae (16.4% = 10/61) and Enterococcus faecalis (11.5% = 7/61), were the most frequently isolated species. Bacterial isolations were predominant in dairy herds with up to 50 animals, production of 20 to 50 L/day and in the municipality of Porto da Folha. In terms of antimicrobial susceptibility, most isolates demonstrated resistance to penicillin and amoxicillin (88.5%), followed by ceftriaxone (23%), ofloxacin (23%), tetracycline (23%), doxycycline (19.7%), chloramphenicol (11.5%), levofloxacin (11.5%), ampicillin/sulbactam (8.2%), amikacin (6.6%), cephalothin (4.9%), cephalexin (3.3%) and gentamicin (3.3%). Approximately 20% of the isolates were multidrug resistant, especially E. coli (50%) and S. aureus (16.7%).Discussion: E. coli was the most frequently isolated species from the samples. It is considered an environmental pathogen, and its high frequency in different herds indicates poor milking hygiene. E. coli also stood out as the species presenting the most multidrug-resistant (MDR) isolates (50%), with strains resistant to beta-lactams, aminoglycosides, quinolones, tetracycline, and chloramphenicol. Coagulase-negative staphylococci are recognized as a public health problem as they areetiological agents of various diseases and can easily acquire antimicrobial resistance genes. Although it was not the most frequently isolated species, S. aureus was the species with the second-highest frequency of MDR strains. The presence of MDR species is relevant and indicates the need for urgent action to reduce the dissemination of antimicrobial resistance.     Enhanced mitochondrial superoxide scavenging does not Improve muscle insulin action in the high fat-fed mouse

    Improving mitochondrial oxidant scavenging may be a viable strategy for the treatment of insulin resistance and diabetes. Mice overexpressing the mitochondrial matrix isoform of superoxide dismutase (sod2(tg) mice) and/or transgenically expressing catalase within the mitochondrial matrix (mcat(tg) mice) have increased scavenging of O2(˙-) and H2O2, respectively. Furthermore, muscle insulin action is partially preserved in high fat (HF)-fed mcat(tg) mice. The goal of the current study was to test the hypothesis that increased O2(˙-) scavenging alone or in combination with increased H2O2 scavenging (mtAO mice) enhances in vivo muscle insulin action in the HF-fed mouse. Insulin action was examined in conscious, unrestrained and unstressed wild type (WT), sod2(tg), mcat(tg) and mtAO mice using hyperinsulinemic-euglycemic clamps (insulin clamps) combined with radioactive glucose tracers following sixteen weeks of normal chow or HF (60% calories from fat) feeding. Glucose infusion rates, whole body glucose disappearance, and muscle glucose uptake during the insulin clamp were similar in chow- and HF-fed WT and sod2(tg) mice. Consistent with our previous work, HF-fed mcat(tg) mice had improved muscle insulin action, however, an additive effect was not seen in mtAO mice. Insulin-stimulated Akt phosphorylation in muscle from clamped mice was consistent with glucose flux measurements. These results demonstrate that increased O2(˙-) scavenging does not improve muscle insulin action in the HF-fed mouse alone or when coupled to increased H2O2 scavenging

    Validation of a digital photographic method for assessment of dietary quality of school lunch sandwiches brought from home.

    Background: It is a challenge to assess children's dietary intake. The digital photographic method (DPM) may be an objective method that can overcome some of these challenges. Objective: The aim of this study was to evaluate the validity and reliability of a DPM to assess the quality of dietary intake from school lunch sandwiches brought from home among children aged 7–13 years. Design: School lunch sandwiches (n=191) were prepared to represent randomly selected school lunch sandwiches from a large database. All components were weighed to provide an objective measure of the composition. The lunches were photographed using a standardised DPM. From the digital images, the dietary components were estimated by a trained image analyst using weights or household measures and the dietary quality was assessed using a validated Meal Index of Dietary Quality (Meal IQ). The dietary components and the Meal IQ obtained from the digital images were validated against the objective weighed foods of the school lunch sandwiches. To determine interrater reliability, the digital images were evaluated by a second image analyst. Results: Correlation coefficients between the DPM and the weighed foods ranged from 0.89 to 0.97. The proportion of meals classified in the same or an adjacent quartile ranged from 98% (starch) to 100% (fruits, vegetables, fish, whole grain, and Meal IQ). There was no statistical difference between fish, fat, starch, whole grains, and Meal IQ using the two methods. Differences were found for fruits and vegetables; Bland–Altman analyses showed a tendency to underestimate high amounts of these variables using the DPM. For interrater reliability, kappa statistics ranged from 0.59 to 0.82 across the dietary components and Meal IQ. Conclusions: The standardised DPM is a valid and reliable method for assessing the dietary quality of school lunch sandwiches brought from home

    Uses Genres and Media Ensembles : A Conceptual Roadmap for Research of Convergent Audiences

    This chapter tackles one of the main methodological and conceptual challenges to current audience research: fragmentation of viewers’ practices of reception. The use of digital and networked media and the consequent multiplication of screens, distribution channels and content sources have further complicated the notion of “watching television” and, along with that, academic and applied audience research. The chapter reintroduces Maria Bakardjieva’s concept of uses genres and connects it with the concept of media ensemble, suggesting that for research on the domestic consumption of films and TV series, the application of these concepts in qualitative (ethnographic) research and in audience surveys comes with strong advantages. Firstly, the concepts help to identify distinct types of consumption practices linked with specific technological objects, with specific audiovisual content and with typical everyday situations, and they enable us to analyze consumption explicitly within the contexts of the spatiotemporal and social organization of everyday life. Secondly, in cases of small- and peripheral-market audiences, the concepts enable us to identify specifics in audiences’ practices linked with the characteristics of these markets (e.g., with localized and non-localized content, with domestic and global production, etc.). And thirdly, the concepts explicitly acknowledge power both involved in and shaping the analyzed practices by emphasizing the “generative process of technology,” i.e., the transformative role of users’ practices in shaping technological and economic systems

    Aquatic community response to volcanic eruptions on the Ecuadorian Andean flank: evidence from the palaeoecological record

    Aquatic ecosystems in the tropical Andes are under increasing pressure from human modification of the landscape (deforestation and dams) and climatic change (increase of extreme events and 1.5 °C on average temperatures are projected for AD 2100). However, the resilience of these ecosystems to perturbations is poorly understood. Here we use a multi-proxy palaeoecological approach to assess the response of aquatic ecosystems to a major mechanism for natural disturbance, volcanic ash deposition. Specifically, we present data from two Neotropical lakes located on the eastern Andean flank of Ecuador. Laguna Pindo (1°27.132â€ČS–78°04.847â€ČW) is a tectonically formed closed basin surrounded by a dense mid-elevation forest, whereas Laguna Baños (0°19.328â€ČS–78°09.175â€ČW) is a glacially formed lake with an inflow and outflow in high Andean PĂĄramo grasslands. In each lake we examined the dynamics of chironomids and other aquatic and semi-aquatic organisms to explore the effect of thick (> 5 cm) volcanic deposits on the aquatic communities in these two systems with different catchment features. In both lakes past volcanic ash deposition was evident from four large tephras dated to c.850 cal year BP (Pindo), and 4600, 3600 and 1500 cal year BP (Baños). Examination of the chironomid and aquatic assemblages before and after the ash depositions revealed no shift in composition at Pindo, but a major change at Baños occurred after the last event around 1500 cal year BP. Chironomids at Baños changed from an assemblage dominated by Pseudochironomus and Polypedilum nubifer-type to Cricotopus/Paratrichocladius type-II, and such a dominance lasted for approximately 380 years. We suggest that, despite potential changes in the water chemistry, the major effect on the chironomid community resulted from the thickness of the tephra being deposited, which acted to shallow the water body beyond a depth threshold. Changes in the aquatic flora and fauna at the base of the trophic chain can promote cascade effects that may deteriorate the ecosystem, especially when already influenced by human activities, such as deforestation and dams, which is frequent in the high Andes

    Safer topical treatment for inflammation using 5α-tetrahydrocorticosterone in mouse models

    Use of topical glucocorticoid for inflammatory skin conditions is limited by systemic and local side-effects. This investigation addressed the hypothesis that topical 5α-tetrahydrocorticosterone (5αTHB, a corticosterone metabolite) inhibits dermal inflammation without affecting processes responsible for skin thinning and impaired wound healing. The topical anti-inflammatory properties of 5αTHB were compared with those of corticosterone in C57Bl/6 male mice with irritant dermatitis induced by croton oil, whereas its effects on angiogenesis, inflammation, and collagen deposition were investigated by subcutaneous sponge implantation. 5αTHB decreased dermal swelling and total cell infiltration associated with dermatitis similarly to corticosterone after 24 h, although at a five fold higher dose, but in contrast did not have any effects after 6 h. Pre-treatment with the glucocorticoid receptor antagonist RU486 attenuated the effect of corticosterone on swelling at 24 h, but not that of 5αTHB. After 24 h 5αTHB reduced myeloperoxidase activity (representative of neutrophil infiltration) to a greater extent than corticosterone. At equipotent anti-inflammatory doses 5αTHB suppressed angiogenesis to a limited extent, unlike corticosterone which substantially decreased angiogenesis compared to vehicle. Furthermore, 5αTHB reduced only endothelial cell recruitment in sponges whereas corticosterone also inhibited smooth muscle cell recruitment and decreased transcripts of angiogenic and inflammatory genes. Strikingly, corticosterone, but not 5αTHB, reduced collagen deposition. However, both 5αTHB and corticosterone attenuated macrophage infiltration into sponges. In conclusion, 5αTHB displays the profile of a safer topical anti-inflammatory compound. With limited effects on angiogenesis and extracellular matrix, it is less likely to impair wound healing or cause skin thinning

    Sociodemographic correlates of food habits among school adolescents (12–15 year) in north Gaza Strip

    Background: There is little information about meal patterns and food consumption of adolescents in Palestine. The objective of this study was to describe the association between sociodemographic factors and food intake, and meal patterns among Palestinian school adolescents (12–15 year) in North Gaza Strip. Methods: A cross-sectional study was conducted in 2002 comprising 944 subjects in 10 schools in Gaza city, Jabalia village and Jabalia refugee camp. Self-administered questionnaires were filled in by students and parents to obtain data on frequency of meals, food intake and sociodemographic characteristics. Results: High household socioeconomic status (SES) was associated with the increased number of meals and the increased intakes of many nutritious foods such as; animal food items, fruits and vegetables and dairy foods. The percentage of adolescents having breakfast daily of high and low SES was 74.5% vs 55% in boys and 65.6% vs 45% in girls. The percentage of girls with refugee status who had lunch was higher (90.2%) compared to the local citizen girls (83.9%), (p = 0.03). Girls were less likely to skip daily lunch (OR = 0.55, 95% CI = 0.36–0.87, p = 0.01) compared to boys. Risk of skipping lunch was three times higher among adolescents living in the village compared to Gaza well-off area (OR = 3.3, 95% CI = 1.72–6.31, p < 0.001). Adolescents who were having lunch daily were less likely to skip breakfast or dinner. Only 11.6% of boys and 16.2% of girls consumed fruits daily. In multivariate analysis, SES was positively associated with food frequency intake scores in both genders. Boys from the refugee camp and the village had a significant higher consumption of fruits and vegetables than boys from high and low income area in Gaza City, while it was the opposite in girls. Conclusion: Meal skipping is common, particularly among those of low SES and the intakes of many nutritious foods such as animal food items, fruits and vegetables and dairy foods seem to be low among adolescents of low SES.     A blended knowledge translation initiative to improve colorectal cancer staging [ISRCTN56824239]

    BACKGROUND: A significant gap has been documented between best practice and the actual practice of surgery. Our group identified that colorectal cancer staging in Ontario was suboptimal and subsequently developed a knowledge translation strategy using the principles of social marketing and the influence of expert and local opinion leaders for colorectal cancer. METHODS/DESIGN: Opinion leaders were identified using the Hiss methodology. Hospitals in Ontario were cluster-randomized to one of two intervention arms. Both groups were exposed to a formal continuing medical education session given by the expert opinion leader for colorectal cancer. In the treatment group the local Opinion Leader for colorectal cancer was detailed by the expert opinion leader for colorectal cancer and received a toolkit. Forty-two centres agreed to have the expert opinion leader for colorectal cancer come and give a formal continuing medical education session that lasted between 50 minutes and 4 hours. No centres refused the intervention. These sessions were generally well attended by most surgeons, pathologists and other health care professionals at each centre. In addition all but one of the local opinion leaders for colorectal cancer met with the expert opinion leader for colorectal cancer for the academic detailing session that lasted between 15 and 30 minutes. DISCUSSION: We have enacted a unique study that has attempted to induce practice change among surgeons and pathologists using an adapted social marketing model that utilized the influence of both expert and local opinion leaders for colorectal cancer in a large geographic area with diverse practice settings

    Human place and response learning: navigation strategy selection, pupil size and gaze behavior.

    In this study, we examined the cognitive processes and ocular behavior associated with on-going navigation strategy choice using a route learning paradigm that distinguishes between three different wayfinding strategies: an allocentric place strategy, and the egocentric associative cue and beacon response strategies. Participants approached intersections of a known route from a variety of directions, and were asked to indicate the direction in which the original route continued. Their responses in a subset of these test trials allowed the assessment of strategy choice over the course of six experimental blocks. The behavioral data revealed an initial maladaptive bias for a beacon response strategy, with shifts in favor of the optimal configuration place strategy occurring over the course of the experiment. Response time analysis suggests that the configuration strategy relied on spatial transformations applied to a viewpoint-dependent spatial representation, rather than direct access to an allocentric representation. Furthermore, pupillary measures reflected the employment of place and response strategies throughout the experiment, with increasing use of the more cognitively demanding configuration strategy associated with increases in pupil dilation. During test trials in which known intersections were approached from different directions, visual attention was directed to the landmark encoded during learning as well as the intended movement direction. Interestingly, the encoded landmark did not differ between the three navigation strategies, which is discussed in the context of initial strategy choice and the parallel acquisition of place and response knowledge
