Official travel is a trip carried out by employees in an institution/company related to official duties. Official travel occurs in each institution, for instance, at the Office of Investment, Energy and Mineral Resources, and Transmigration in Gorontalo Province. Yet, there are several problems encountered in the official travel system. The problem is that institution’s data recap regarding official travel will be lost every year. Another problem is in the process of report filling, where the report filling can only be performed by the treasurer. As a result, the treasurer has difficulties since s/he must input various reports, and it took a long time. Equally important,as long as the report has not been filled in, the employees cannot perform official travel. Based on the problem above, the proposed solution is creating an information system that can facilitate the treasurer in managing the data of employees and searching the annual reports. In order to improve the welfare conditions for animals, here with focus on elephants, in zoological facilities, scientific data on their ethology are needed. The Zoo Heidelberg is the first German facility, which keeps a bachelor herd of young elephant bulls. This is due to support the European breeding program (EEP) for Elephas maximus. The separation of young males from their natal family group is a behavioral strategy to avoid inbreeding. After leaving the family group male elephants form loose all-male groups. This juvenile phase is an important time in the life of nearly every social living mammal. During that time they learn relevant social behavior. The aim of this study was to assess the social structure and the group dynamics within the bachelor group of young elephant bulls. Therefore, the social behavior was recorded over a time period of twenty weeks by using the focal animal sampling. In a group of young bulls a lot of motion is expected. To analyze their movements, all four animals where outfitted with a GPS device in an anklet. Within the elephant group a linear rank order was observed. By means of the social behavior different associations and individual characteristics were identified. On the basis of the GPS measurements important information about the elephants’ amount of movement in the Zoo Heidelberg was obtained. These findings conform to the walking rates of their wild conspecifics

Skingineering II: transplantation of large-scale laboratory-grown skin analogues in a new pig model

Background: Tissue engineering of skin with near-normal anatomy is an intriguing novel strategy to attack the still unsolved problem of how to ideally cover massive full-thickness skin defects. After successful production of large, pig cell-derived skin analogues, we now aim at developing an appropriate large animal model for transplantation studies. Materials and methods: In four adult Swiss pigs, full-thickness skin defects, measuring 7.5×7.5cm, were surgically created and then shielded against the surrounding skin by a new, self-designed silicone chamber. In two animals each, Integra dermal regeneration templates or cultured autologous skin analogues, respectively, were applied onto the wound bed. A sophisticated shock-absorbing dressing was applied for the ensuing 3weeks. Results were documented photographically and histologically. Results: All animals survived uneventfully. Integra healed in perfectly, while the dermo-epidermal skin analogues showed complete take of the dermal compartment but spots of missing epidermis. The chamber proved effective in precluding ("false positive”) healing from the wound edges and the special dressing efficiently kept the operation site intact and clean for the planned 3weeks. Conclusion: We present a novel and valid pig model permitting both transplantation of large autologous, laboratory-engineered skin analogues and also keeping the site of intervention undisturbed for at least three postoperative weeks. Hence, the model will be used for experiments testing whether such large skin analogues can restore near-normal skin, particularly in the long term. If so, clinical application can be envisione

Chicken γδ T cells proliferate upon IL-2 and IL-12 treatment and show a restricted receptor repertoire in cell culture

In chickens, γδ T cells represent a large fraction of peripheral T cells; however, their function remains largely unknown. Here, we describe the selective in vitro expansion of γδ T cells from total splenocytes by stimulation with the cytokines IL-2 and IL-12. Under these conditions, γδ T cells proliferated preferentially and reached frequencies of >95% within three weeks. Although IL-2 alone also triggered proliferation, an increased proliferation rate was observed in combination with IL-12. Most of the expanded cells were γδ TCR and CD8 double-positive. Splenocytes sorted into TCR1+CD8+, TCR1highCD8−, and TCR1lowCD8− subsets proliferated well upon dual stimulation with IL-2/IL-12, indicating that none of the three γδ T cell subsets require bystander activation for proliferation. TCR1+CD8+ cells maintained CD8 surface expression during stimulation, whereas CD8− subpopulations showed varied levels of CD8 upregulation, with the highest upregulation observed in the TCR1high subset. Changes in the γδ T-cell receptor repertoire during cell culture from day 0 to day 21 were analyzed by next-generation sequencing of the γδ variable regions. Overall, long-term culture led to a restricted γ and δ chain repertoire, characterized by a reduced number of unique variable region clonotypes, and specific V genes were enriched at day 21. On day 0, the δ chain repertoire was highly diverse, and the predominant clonotypes differed between animals, while the most frequent γ-chain clonotypes were shared between animals. However, on day 21, the most frequent clonotypes in both the γ and δ chain repertoires were different between animals, indicating that selective expansion of dominant clonotypes during stimulation seems to be an individual outcome. In conclusion, IL-2 and IL-12 were sufficient to stimulate the in vitro outgrowth of γδ T cells. Analyses of the TCR repertoire indicate that the culture leads to an expansion of individual T cell clones, which may reflect previous in vivo activation. This system will be instrumental in studying γδ T cell function

Diboryldiborenes: pi-conjugated B4 chains isoelectronic to the butadiene dication

sp2–sp3 diborane species based on bis(catecholato)diboron and N-heterocyclic carbenes (NHCs) are subjected to catechol/bromide exchange selectively at the sp3 boron atom. The reduction of the resulting 1,1-dibromodiborane adducts led to reductive coupling and isolation of doubly NHC-stabilized 1,2-diboryldiborenes. These compounds are the first examples of molecules exhibiting pi-electron delocalization over an all-boron chain

    A. S. is grateful to the Australian Research Council for support and a fellowship. C. A. O. thanks the Australian Research Council for Discovery Project grants DP11015530 and DP130100483.We have synthesized and characterized the monomeric diiminophosphinate-stabilized group 13 metal(I) complexes [DipLE:], DipL = Ph2P(NDip)2, Dip = 2,6-iPr2C6H3; E = Ga ( 1 ), In ( 2 ) and Tl ( 3 ). In addition, we structurally characterized the dimeric complex [(DipLGa)2], 12 . Similar synthetic attempts using MesL = Ph2P(NMes)2, Mes = 2,4,6-Me3C6H2 afforded product mixtures from which the mixed oxidation state species [(MesL)3Ga4I3] 4 was isolated. [DipLGa:] 1 is converted with dry air to the gallium(III) oxide species [(DipLGaO)2] 5 . Density Functional Theory studies on [DipLE:] and [(DipLE)2], E = Al-Tl, shed light on the bonding in these compounds and show that the newly formed E-E bonding interactions can be described as weak single σ-bond with no significant π-bonding contribution for E = Al, Ga. A large contribution to the dimer binding enthalpies results from London dispersion forces.PostprintPeer reviewe
