    A comparison between faecal sterols and coliform counts in the investigation of sewage contamination in sediments

    Em setembro de 2002, foram coletadas nove amostras de sedimento superficial (0-2 cm) na Enseada de Botafogo (sudoeste da Baía de Guanabara/RJ), a fim de comparar o uso de marcadores químicos (coprostanol) e biológicos (E. coli e coliformes totais) na identificação da contaminação fecal da região. Os resultados obtidos (carbono orgânico - 6,0 to 64,8 mg g-1; coprostanol - 1,4 to 105 µg g-1; E. coli - < 30 to 2400 NMP/10g e coliformes totais - 40 to 9300 NMP/10g) foram iguais ou maiores aos observados em outras áreas contaminadas da Baía de Guanabara. Nas estações próximas à linha de costa, as concentrações de coprostanol e as contagens das bactérias confirmaram que o esgoto doméstico se acumulou no sedimento da enseada. As concentrações de coprostanol se mantiveram altas nas estações distantes das fontes de contaminação, porém o material fecal representou uma menor fração do carbono orgânico. Nessas mesmas estações, redução na contagem de colimetria foi proporcionalmente mais alta, provavelmente pela baixa sobrevivência das bactérias devido ao efeito de luz, salinidade e temperatura. durante o transporte e deposição das partículas de esgoto. Portanto, baseando-se nos resultados obtidos, o coprostanol foi um indicador mais adequado da contaminação fecal nos sedimentos da Enseada de Botafogo.In September 2002, nine sediment samples (0-2 cm) were collected from Botafogo Cove (southwestern part of Guanabara Bay) in order to compare the use of chemical (coprostanol) and biological markers (E. coli and total coliforms) in identifying faecal contamination. The values found (organic carbon - 6.0 to 64.8 mg g-1; coprostanol - 1.4 to 105 µg g-1; E. coli - < 30 to 2400 NMP/10g and total coliforms - 40 to 9300 NMP/10g) were similar to or even higher than those observed in other contaminated areas of Guanabara Bay. In stations close to the shoreline, both coprostanol concentration and bacteria count confirmed that domestic sewage had accumulated in the cove. Coprostanol concentrations were still relatively high in the stations furthest from contamination sources, although faecal material represented a smaller fraction of the total organic carbon originated by dilution in waters containing algae. Reduction was proportionately higher by virtue of the use of the colimetric assessment method. This result may be associated with the effect that environmental variables (light, salinity, temperature) may have on the survival of bacteria during transport and deposition of sewage particles. Consequently, based on these results, coprostanol may be considered the better indicator of the faecal contamination level in Botafogo Cove sediments

    Multi-pathology detection and lesion localization in WCE videos by using the instance segmentation approach

    The majority of current systems for automatic diagnosis considers the detection of a unique and previously known pathology. Considering specifically the diagnosis of lesions in the small bowel using endoscopic capsule images, very few consider the possible existence of more than one pathology and when they do, they are mainly detection based systems therefore unable to localize the suspected lesions. Such systems do not fully satisfy the medical community, that in fact needs a system that detects any pathology and eventually more than one, when they coexist. In addition, besides the diagnostic capability of these systems, localizing the lesions in the image has been of great interest to the medical community, mainly for training medical personnel purposes. So, nowadays, the inclusion of the lesion location in automatic diagnostic systems is practically mandatory. Multi-pathology detection can be seen as a multi-object detection task and as each frame can contain different instances of the same lesion, instance segmentation seems to be appropriate for the purpose. Consequently, we argue that a multi-pathology system benefits from using the instance segmentation approach, since classification and segmentation modules are both required complementing each other in lesion detection and localization. According to our best knowledge such a system does not yet exist for the detection of WCE pathologies. This paper proposes a multi-pathology system that can be applied to WCE images, which uses the Mask Improved RCNN (MI-RCNN), a new mask subnet scheme which has shown to significantly improve mask predictions of the high performing state-of-the-art Mask-RCNN and PANet systems. A novel training strategy based on the second momentum is also proposed for the first time for training Mask-RCNN and PANet based systems. These approaches were tested using the public database KID, and the included pathologies were bleeding, angioectasias, polyps and inflammatory lesions. Experimental results show significant improvements for the prFCT national funds, under the national support to R&D units grant, through the reference project UIDB/04436/2020 and UIDP/04436/2020 and through the PhD Grants with the references SFRH/BD/92143/2013 and SFRH/BD/139061/201

    Morphology and biometry of the thymus gland in macaw of Ara and Anodorhynchus genera

    Foram utilizados 12 exemplares de Ara ararauna – seis fêmeas e seis machos –, cinco exemplares de Ara chloropterus (uma fêmea e quatro machos) e dois exemplares de Anodorhynchus hyacinthinus – uma fêmea e um macho –, todos adultos, doados por criadouro particular, após óbito natural. Os lobos foram dissecados e medidos com paquímetro – comprimento x largura x espessura – e analisados quanto ao peso, à topografia e à morfologia individual. Independentemente do gênero, foram identificados, em 17 casos (89,5%), lobos tímicos nos antímeros cervicais esquerdo e direito, e em oito casos (42,1%), lobos na cavidade celomática. Os lobos apresentaram formatos alongados – 52,6% –, arredondados – 21,1% – ou ovalados – 15,8% –, posicionados preferencialmente ventromedialmente ao longo do plexo vasculoneural do pescoço, com número médio de cinco lobos por antímero, tamanho médio de 0,49cm de comprimento, 0,12cm de largura e 0,05cm de espessura e peso médio de 0,076g.Twelve samples of Ara ararauna – six females and six males –, five samples of Ara chloropterus (one female and four males) and two samples of Anodorhynchus hyacinthinus (one female and one male), all adults from a Breeding Park, were used after natural death. The lobes were dissected and measured with electronic calliper (length x width x thickness) and analyzed taking their weight, topography and individual morphology into account. Regardless of gender, 89.5% of the cases presented timic lobes in the left and right cervical antimere, and 42.1% of the cases presented lobes in the celomatic cavity. The lobes were shown in shapes – long (52.6%), round (21.1%) or oval (15.8%), positioned mostly ventromedially, along the neurovascular plexus of the neck, with an average of five lobes per antimere, and an average size of 0.49cm length, 0.12cm width, and 0.05cm thick and average weight of 0.076g

    Sacrificial Adhesive Bonding: A Powerful Method For Fabrication Of Glass Microchips

Initially, the method involves the use of an uncured SU-8 intermediate to seal two glass slides irreversibly as in conventional adhesive bonding-based approaches. Subsequently, an additional step removes the adhesive layer from the channels. This step relies on a selective development to remove the SU-8 only inside the microchannel, generating glass-like surface properties as demonstrated by specific tests. Named sacrificial adhesive layer (SAB), the protocol meets the requirements of an ideal microfabrication technique such as throughput, relatively low cost, feasibility for ultra largescale integration (ULSI), and high adhesion strength, supporting pressures on the order of 5 MPa. Furthermore, SAB eliminates the use of high temperature, pressure, or potential, enabling the deposition of thin films for electrical or electrochemical experiments. Finally, the SAB protocol is an improvement on SU-8-based bondings described in the literature.     Arterial vascularization of the thoracic lobes of the thymus in stillborns of the lineage C40

    As origens e distribuições das artérias que vascularizaram os lobos torácicos do timo foram estudadas em fetos de 30 suínos da linhagem C40, sendo 12 machos e 18 fêmeas. Os exemplares tiveram o sistema arterial preenchido com solução aquosa a 50% de Neoprene Látex corado e, em seguida, foram submetidos à fixação em solução aquosa a 10% de formaldeído. Os lobos torácicos do timo foram vascularizados por ramos diretos das artérias torácica interna direita (63,33%) e esquerda (53,33%), subclávia esquerda (3,33%), vertebral esquerda (3,33%), cervical superficial direita (3,33%) e esquerda (3,33%), carótida comum esquerda (3,33%), coronária direita (3,33%) e pelos troncos braquiocefálico (33,33%) e costocervical (3,33%). Observaram-se ainda os ramos indiretos das artérias torácica interna direita (70%) e esquerda (76,67%), subclávia esquerda (23,33%), cervical superficial esquerda (3,33%) e do arco aórtico (6,67%).The origins and distributions of the arteries that vascularization the thoracic lobes of the thymus, right and left, were studied in 30 swine of the lineage C40, fetus, being 12 males and 18 females. The animals had the arterial system filled with aqueous colored solution in a 50% of Neoprene Latex, and than were fixed in 10% aqueous solution formaldehyde. The thoracic lobes of thymus were vascularization by direct branches of the right (53.33%) and left (63.33%) internal thoracic arteries, left subclavian (3.33%), left vertebral (3.33%), right (3.33%) and left (3.33%) superficial cervical, left common carotid (3.33%), right coronary (3.33%) and brachiocephalic trunk (33.33%) and costocervical trunk (3.33%). The right internal thoracic arteries (70%) and left internal thoracic (76.67%), left subclavian (23.33%), left superficial cervical (3.33%) of the aortic arch (6.67%) provided indirect branches for the correspondent thoracic lobes of the thymus in each side

    Nematodes Affecting Potato and Sustainable Practices for Their Management

    Plant-parasitic nematodes are a significant factor limiting potato production and tuber quality in several regions where potato is produced. Overall, parasitic nematodes alone cause an estimated annual crop loss of $ 78 billion worldwide and an average crop yield loss of 10–15%. As a result, sustainable food production and food security are directly impacted by pests and diseases. Degrading land use with monocultures and unsustainable cropping practices have intensified problems associated with plant pathogens. Proper identification of nematode species and isolates is crucial to choose effective and sustainable management strategies for nematode infection. Several nematode species have been reported associated with potato. Among those, the potato cyst nematodes Globodera rostochiensis and G. pallida, the root-knot nematode Meloidogyne spp., the root lesion nematode Pratylenchus spp., the potato rot nematode Ditylenchus destructor and the false root-knot nematode Nacobbus aberrans are major species limiting potato yield and leading to poor tuber quality.     Safety analysis of sacubitril/valsartan in patients with heart failure in Vitória, Espírito Santo

    Background: Sacubitril/valsartan has proven its efficacy to reduce cardiovascular mortality, all-cause mortality and sudden death in heart failure with reduced ejection fraction (HFrEF). Thus, it becomes important to evaluate the safety profile of the medication in clinical practice. Objectives: This study aimed to assess safety outcomes on the use of sacubitril/valsartan in patients with HFrEF attended in a Brazilian specialized service. Methods: Prospective observational study that included patients with HFrEF from a specialized ambulatory service, in functional class II-IV, initiated on sacubitril/valsartan as per clinical indication, with a four-month follow-up. Primary outcomes were the occurrence of symptomatic arterial hypotension, hyperkalemia and reduction of renal function. Serum potassium values, blood pressure and creatinine clearance were analyzed at inclusion and at the end of follow-up. A 5% significance level was considered for comparisons. Results: Twenty-six patients were analyzed, 57.7% male, mean age 57.8 ± 10 years, average left ventricle ejection fraction 29.9 ± 7.7%. Symptomatic hypotension occurred in 53.8%, hyperkalemia in 19.2% and reduction of renal function in 6.7%. There was significant difference from initial to final systolic (122 ± 24mmHg versus 109 ± 15mmHg; p=0.024) and diastolic (76 ± 18mmHg versus 66 ± 12mmHg; p=0.022) blood pressure, but no difference in serum potassium (4.8 ± 0.4mEq/L versus 5.0 ± 0.3mEq/L; p=0.07) and creatinine clearance (65 ± 23mL/min/1.73m² versus 66 ± 29mL/min/1.73m²; p=0.89). Conclusions: Symptomatic hypotension was the most frequent side-effect of sacubitril/valsartan. Reduction of blood pressure was observed at the end of follow-up, but no reduction of renal function or significant increase of serum potassium
