    What we need to know about exercise, neurodegenerative disease and ageing? A Systematic Review.

    As doenças neurode-generativas são caracterizadas por um conjunto de patologias crónicas e irreversíveis do sistema nervoso. Até ao momento não foram realizadas revisões de literatura que relacionem o exercício, as doença neurodegenerativas e o envelhecimento. Assim, o objetivo desta revisão sistemática foi analisar a literatura actual sobre os efeitos e associações entre o exercício físico em quatro doenças neurodegenerativas: Alzheimer, Parkinson, Huntington e Demência Vascular.Neurodegenerative diseases are characterized by a set of chronic and irreversible pathologies of the nervous system.     The Yeast PNC1 Longevity Gene Is Up-Regulated by mRNA Mistranslation

    Translation fidelity is critical for protein synthesis and to ensure correct cell functioning. Mutations in the protein synthesis machinery or environmental factors that increase synthesis of mistranslated proteins result in cell death and degeneration and are associated with neurodegenerative diseases, cancer and with an increasing number of mitochondrial disorders. Remarkably, mRNA mistranslation plays critical roles in the evolution of the genetic code, can be beneficial under stress conditions in yeast and in Escherichia coli and is an important source of peptides for MHC class I complex in dendritic cells. Despite this, its biology has been overlooked over the years due to technical difficulties in its detection and quantification. In order to shed new light on the biological relevance of mistranslation we have generated codon misreading in Saccharomyces cerevisiae using drugs and tRNA engineering methodologies. Surprisingly, such mistranslation up-regulated the longevity gene PNC1. Similar results were also obtained in cells grown in the presence of amino acid analogues that promote protein misfolding. The overall data showed that PNC1 is a biomarker of mRNA mistranslation and protein misfolding and that PNC1-GFP fusions can be used to monitor these two important biological phenomena in vivo in an easy manner, thus opening new avenues to understand their biological relevance

    Adesão ao tratamento de diabetes Mellitus e relação com a assistência na atenção primária

    RESUMO OBJETIVO: verificar a associação entre a adesão ao tratamento medicamentoso e não medicamentoso e as práticas assistenciais prestadas às pessoas com diabetes Mellitus tipo 2 (DM2) pelas equipes da Estratégia Saúde da Família (ESF). MÉTODO: estudo transversal, do tipo inquérito domiciliar, realizado com pessoas com DM2 cadastradas nas 65 equipes urbanas da ESF, selecionadas aleatoriamente e de forma estratificada por equipe. Os dados foram coletados no primeiro semestre 2014 mediante entrevista estruturada e na análise, usando-se regressão logística, considerando-se associação significativa quando p≤0,05. RESULTADOS: as 408 pessoas participantes tinham idade média de 66,5 anos, 84,1% relataram aderir ao tratamento medicamentoso, 29,4% realizavam atividade física regularmente e 24% tinham alimentação adequada. Após ajustes, as variáveis que permaneceram associadas ao tratamento medicamentoso foram: não participação em atividade de educação em saúde (p=0,012) e ser atendido pelo mesmo enfermeiro (p=0,048). Em relação ao medicamentoso, a adoção de alimentação adequada apresentou associação com verificação trimestral da glicemia capilar (p=0,011) e ser questionado, durante o atendimento, sobre a prática de atividade física (p=0,012) e a prática de atividade física regular com participação em atividades de educação em saúde (p=0,031), estar satisfeito com a assistência (p=0,04), ser atendido no mesmo dia em que procurou a UBS (p=0,017) e os profissionais perguntarem sobre sua saúde (p=0,011). CONCLUSÃO: as pessoas com DM2 apresentaram boa adesão ao tratamento medicamentoso e baixa adesão ao não medicamentoso, indicando que as equipes da ESF precisam ampliar a implementação de ações de promoção da saúde, prevenção e controle da doença e suas complicações

    A Roadmap for HEP Software and Computing R&D for the 2020s

    Particle physics has an ambitious and broad experimental programme for the coming decades. This programme requires large investments in detector hardware, either to build new facilities and experiments, or to upgrade existing ones. Similarly, it requires commensurate investment in the R&D of software to acquire, manage, process, and analyse the shear amounts of data to be recorded. In planning for the HL-LHC in particular, it is critical that all of the collaborating stakeholders agree on the software goals and priorities, and that the efforts complement each other. In this spirit, this white paper describes the R&D activities required to prepare for this software upgrade.Peer reviewe

    Lifelong robbery victimisation and mental disorders at age 18 years: Brazilian population-based study

    Purpose: Urban violence is a major problem in Brazil and may contribute to mental disorders among victims. The aim of this study was to assess the association between robbery victimisation and mental health disorders in late adolescence. Methods: At age 18 years, 4106 participants in the 1993 Pelotas Birth Cohort Study were assessed. A questionnaire about history of robbery victimisation was administered, the Self-Report Questionnaire was used to screen for common mental disorders, and the Mini International Neuropsychiatric Interview was used to assess major depressive disorder and generalised anxiety disorder. Cross-sectional prevalence ratios between lifetime robbery victimisation and mental disorders were estimated using Poisson regression with robust standard errors, adjusting for socioeconomic variables measured at birth and violence in the home and maltreatment measured at age 15. Results: There was a dose–response relationship between frequency of lifetime robberies and risk of mental disorders. Adolescents who had been robbed three or more times had twice the risk (PR 2.04; 95% CI 1.64–2.56) for common mental disorders, over four times the risk for depression (PR 4.59; 95% CI 2.60–8.12), and twice the risk for anxiety (PR 1.93; 95% CI 1.06–3.50), compared with non-victims, adjusting for covariates. Experiencing frequent robberies had greater impact on common mental disorders than experiencing an armed robbery. Population attributable fractions with regard to robbery were 9% for common mental disorders, 13% for depression, and 8% for anxiety. Conclusions: Robberies are associated with common mental disorders in late adolescence, independently of violence between family members. Reducing urban violence could significantly help in preventing common mental illnesses

    Antifungal and Antivirulence Activities of Hydroalcoholic Extract and Fractions of Platonia insignis Leaves against Vaginal Isolates of Candida Species

    Vulvovaginal candidiasis is a common fungal infection in women. In this study, Platonia insignis hydroalcoholic extract (PiHE) and its fractions were evaluated for antifungal and antivirulence activities against vaginal Candida species. Dichloromethane (DCMF) and ethyl acetate fractions (EAF) obtained from PiHE effectively inhibited the pathogen. Electrospray ionization mass spectrometry was used for identifying the main compounds in extracts. Minimal inhibitory and fungicidal concentrations (MIC and MFC, respectively) were determined by a broth microdilution assay. Furthermore, we evaluated the effect of the extract and fractions on the virulence properties of Candida albicans, and their cytotoxicity effect was determined on RAW 264.7 cells. Compounds found in extracts were flavonoid glycosides, mainly derivatives of quercetin and myricetin. Extracts showed antifungal potential, with the lowest MIC value for EAF (1.3 mg/mL) and inhibited Candida adherence and biofilm formation. EAF disrupted 48 h biofilms with an inhibition rate of more than 90%. The extract and its fractions exhibited no cytotoxicity. The antifungal effects were attributed to the ability of these extracts to alter the mitochondrial membrane potential for the release of pro-apoptotic factors in the cytosol. In conclusion, our data suggest that PiHE and EAF could act as novel candidates for the development of new therapeutic treatments against fungal infections
