    Pollen Ultrastructure of Genus Dendrobium Orchids as a Learning Resource

    Anggrek genus Dendrobium merupakan salah satu genus dari divisi spermatophyta yang merupakan kelompok tumbuhan yang berkembang dengan menggunakan biji. Tumbuhan berbiji tentu memiliki bunga sebagai alat perkembangan generatifnya. Perkembangan generatif pada bunga artinya pertemuan antara sel gamet jantan dan sel gamet betina. Sel gamet betina pada tumbuhan dihasilkan oleh putik, sedangkan sel gamet jantan disebut serbuk sari atau pollen. Genus Dendrobium merupakan salah satu kekayaan alam Indonesia, jumlahnya diperkirakan mencapai 275 spesies. Penelitian ini bertujuan untuk mengetahui bagaimana ultrastruktur pollen anggrek genus Dendrobium dari sepuluh spesies yang diamati menggunakan SEM. Jenis penelitian ini adalah deskriptif kualitatif. Hasil penelitian menunjukkan unit pollen untuk semua spesies yaitu kategori pollinia, sedangkan untuk polaritas pollen adalah apolar. Jenis aperture kesepuluh spesies yang diamati mempunyai aperture dengan pola yang tidak beraturan dan lebih dari enam yang disebut colpate. Bentuk pollen dari semua spesies yang diteliti bentuknya adalah subprolate hingga prolate dan ukuran pollen termasuk dalam kategori minuta hingga media. Ornamentasi pollen atau skluptur pollen tidak teridentifikasi dikarenakan ukuran pollen yang terlalu kecil, permukaan pollen terlihat kurang jelas. Sumber belajar yang digunakan adalah atlas

    DETERMINANTS OF FARMERS' DECISIONS IN THE TAKING OF AGRICULTURAL SECTOR CREDIT: A STUDY IN LAMAKNEN SUBDISTRICT OF BELU REGENCY, EAST NUSATENGGARA PROVINCE, INDONESIA

    The development of the agricultural sector has limitations in increasing production. At this time, the productivity of the agricultural sector is still low; one of them is caused by capital constraints, both internal and external. This study aims to examine the influence of land ownership area (X1), education (X2), income (X3), and counseling service (X4) to the decision-making of agricultural credit (Y) in Lamaknen Subdistrict Belu Regency East Nusa Tenggara Province. This study uses a quantitative method with the population namely all farmers in Lamaknen Sub-district consisting of 10 village units. Furthermore, the samples studied namely all farmers in Makir Village. The sampling technique used is probability sampling. Primary data was obtained using a questionnaire and then tested with the Logistic Regression analysis method. The results of the analysis show a significance value < (0,05) with the Negelkerke R Square value equal to 0,617. Thus, it can be concluded that the area of land ownership (X1), education (X2), income (X3), and counseling service (X4) had a significant influence on the decision making of agricultural credit (Y) with an influence of 61,7%. The right credit decision making also contributes to an increase in agricultural production, so that needed cooperation between village government and credit institutions to provide socialization to farmers related to agricultural business credit

    Positive association between physical outcomes and patient-reported outcomes in late-onset Pompe disease: a cross sectional study

    BACKGROUND: Pompe disease is a rare, progressive metabolic myopathy. The aim of this study is to investigate the associations of physical outcomes with patient-reported outcome measures (PROMs) in late-onset Pompe disease. METHODS: We included 121 Dutch adult patients with Pompe disease. Physical outcomes comprised muscle strength (manual muscle testing using Medical Research Council [MRC] grading, hand-held dynamometry [HHD]), walking ability (6-min walk test [6MWT]), and pulmonary function (forced vital capacity [FVC] in upright and supine positions). PROMs comprised quality of life (Short Form 36 health survey [SF-36]), participation (Rotterdam Handicap Scale [RHS]) and daily-life activities (Rasch-Built Pompe-Specific Activity [R-PAct] Scale). Analyses were cross-sectional: the time-point before, and closest to, start of Enzyme Replacement Therapy was chosen. Associations between PROMs and physical outcomes were investigated using linear regression models. RESULTS: RHS and R-PAct scores were better in patients with higher FVC supine and upright, HHD, MRC and 6MWT scores, accounting for the effect of sex, disease duration, use of wheelchair and ventilator support. While the SF-36 Physical Component Summary (PCS) was correlated positively with FVC upright, HHD, MRC and 6MWT scores, there was no significant relationship between the SF-36 Mental Component Summary (MCS) and any of the physical outcomes. CONCLUSIONS: Participation, daily-life activities, and the physical component of quality of life of adult Pompe patients are positively correlated to physical outcomes. This work serves as a first step towards assessing how changes over time in physical outcomes are related to changes in PROMs, and to define the minimal change in physical outcomes required to make an important difference for the patient

    An integrated cell atlas of the lung in health and disease

    Single-cell technologies have transformed our understanding of human tissues. Yet, studies typically capture only a limited number of donors and disagree on cell type definitions. Integrating many single-cell datasets can address these limitations of individual studies and capture the variability present in the population. Here we present the integrated Human Lung Cell Atlas (HLCA), combining 49 datasets of the human respiratory system into a single atlas spanning over 2.4 million cells from 486 individuals. The HLCA presents a consensus cell type re-annotation with matching marker genes, including annotations of rare and previously undescribed cell types. Leveraging the number and diversity of individuals in the HLCA, we identify gene modules that are associated with demographic covariates such as age, sex and body mass index, as well as gene modules changing expression along the proximal-to-distal axis of the bronchial tree. Mapping new data to the HLCA enables rapid data annotation and interpretation. Using the HLCA as a reference for the study of disease, we identify shared cell states across multiple lung diseases, including SPP1 + profibrotic monocyte-derived macrophages in COVID-19, pulmonary fibrosis and lung carcinoma. Overall, the HLCA serves as an example for the development and use of large-scale, cross-dataset organ atlases within the Human Cell Atlas. © 2023, The Author(s)

    We present an approach to determining the speed of wave-front solutions to reaction-transport processes. This method is more accurate than previous ones. This is explicitly shown for several cases of practical interest: (i) the anomalous diffusion reaction, (ii) reaction diffusion in an advective field, and (iii) time-delayed reaction diffusion. There is good agreement with the results of numerical simulations

    Predicting climate change impacts on polar bear litter size

    Predicting the ecological impacts of climate warming is critical for species conservation. Incorporating future warming into population models, however, is challenging because reproduction and survival cannot be measured for yet unobserved environmental conditions. In this study, we use mechanistic energy budget models and data obtainable under current conditions to predict polar bear litter size under future conditions. In western Hudson Bay, we predict climate warming-induced litter size declines that jeopardize population viability: ∼28% of pregnant females failed to reproduce for energetic reasons during the early 1990s, but 40–73% could fail if spring sea ice break-up occurs 1 month earlier than during the 1990s, and 55–100% if break-up occurs 2 months earlier. Simultaneously, mean litter size would decrease by 22–67% and 44–100%, respectively. The expected timeline for these declines varies with climate-model-specific sea ice predictions. Similar litter size declines may occur in over one-third of the global polar bear population

    Increasing Costs Due to Ocean Acidification Drives Phytoplankton to Be More Heavily Calcified: Optimal Growth Strategy of Coccolithophores

    Ocean acidification is potentially one of the greatest threats to marine ecosystems and global carbon cycling. Amongst calcifying organisms, coccolithophores have received special attention because their calcite precipitation plays a significant role in alkalinity flux to the deep ocean (i.e., inorganic carbon pump). Currently, empirical effort is devoted to evaluating the plastic responses to acidification, but evolutionary considerations are missing from this approach. We thus constructed an optimality model to evaluate the evolutionary response of coccolithophorid life history, assuming that their exoskeleton (coccolith) serves to reduce the instantaneous mortality rates. Our model predicted that natural selection favors constructing more heavily calcified exoskeleton in response to increased acidification-driven costs. This counter-intuitive response occurs because the fitness benefit of choosing a better-defended, slower growth strategy in more acidic conditions, outweighs that of accelerating the cell cycle, as this occurs by producing less calcified exoskeleton. Contrary to the widely held belief, the evolutionarily optimized population can precipitate larger amounts of CaCO3 during the bloom in more acidified seawater, depending on parameter values. These findings suggest that ocean acidification may enhance the calcification rates of marine organisms as an adaptive response, possibly accompanied by higher carbon fixation ability. Our theory also provides a compelling explanation for the multispecific fossil time-series record from ∼200 years ago to present, in which mean coccolith size has increased along with rising atmospheric CO2 concentration

    Decoupling genetics, lineages, and microenvironment in IDH-mutant gliomas by single-cell RNA-seq

    Tumor subclasses differ according to the genotypes and phenotypes of malignant cells as well as the composition of the tumor microenvironment (TME).We dissected these influences in isocitrate dehydrogenase (IDH)-mutant gliomas by combining 14,226 single-cell RNA sequencing (RNA-seq) profiles from 16 patient samples with bulk RNA-seq profiles from 165 patient samples. Differences in bulk profiles between IDH-mutant astrocytoma and oligodendroglioma can be primarily explained by distinct TME and signature genetic events, whereas both tumor types share similar developmental hierarchies and lineages of glial differentiation. As tumor grade increases, we find enhanced proliferation of malignant cells, larger pools of undifferentiated glioma cells, and an increase in macrophage over microglia expression programs in TME. Our work provides a unifying model for IDH-mutant gliomas and a general framework for dissecting the differences among human tumor subclasses.National Cancer Institute (U.S.) (Grant P30-CA14051
