    Spindle waves occur during the early stage of slow wave sleep and are thought to arise in the thalamic reticular nucleus (TRN), causing inhibitory postsynaptic potential spindle-like oscillations in the dorsal thalamus that are propagated to the cortex. We have found that thalamocortical neurons exhibit membrane oscillations that have spindle frequencies, consist of excitatory postsynaptic potentials, and co-occur with electroencephalographic spindles. TRN lesioning prolonged oscillations in the medial geniculate body (MGB) and auditory cortex (AC). Injection of GABA~A~ antagonist into the MGB decreased oscillation frequency, while injection of GABA~B~ antagonist increased spindle oscillations in the MGB and cortex. Thus, spindles originate in the dorsal thalamus and TRN inhibitory inputs modulate this process, with fast inhibition facilitating the internal frequency and slow inhibition limiting spindle occurrence

    Nuances are the Key: Unlocking ChatGPT to Find Failure-Inducing Tests with Differential Prompting

    Automatically detecting software failures is an important task and a longstanding challenge. It requires finding failure-inducing test cases whose test input can trigger the software's fault, and constructing an automated oracle to detect the software's incorrect behaviors. Recent advancement of large language models (LLMs) motivates us to study how far this challenge can be addressed by ChatGPT, a state-of-the-art LLM. Unfortunately, our study shows that ChatGPT has a low probability (28.8%) of finding correct failure-inducing test cases for buggy programs. A possible reason is that finding failure-inducing test cases requires analyzing the subtle code differences between a buggy program and its correct version. When these two versions have similar syntax, ChatGPT is weak at recognizing subtle code differences. Our insight is that ChatGPT's performance can be substantially enhanced when ChatGPT is guided to focus on the subtle code difference. We have an interesting observation that ChatGPT is effective in inferring the intended behaviors of a buggy program. The intended behavior can be leveraged to synthesize programs, in order to make the subtle code difference between a buggy program and its correct version (i.e., the synthesized program) explicit. Driven by this observation, we propose a novel approach that synergistically combines ChatGPT and differential testing to find failure-inducing test cases. We evaluate our approach on Quixbugs (a benchmark of buggy programs), and compare it with state-of-the-art baselines, including direct use of ChatGPT and Pynguin. The experimental result shows that our approach has a much higher probability (77.8%) of finding correct failure-inducing test cases, 2.7X as the best baseline.Comment: Accepted to the 38th IEEE/ACM International Conference on Automated Software Engineering (ASE 2023

    Knowledge, practices, compliance and beliefs of university nursing students’ toward hand hygiene: A cross-sectional survey

    Nursing students can contribute to healthcareassociated infections if having inadequate knowledge and noncompliance of hand hygiene. Effective hand hygiene is considered the most effective measure for preventing healthcare-associated infections. This study investigated the knowledge, practices, compliance, and beliefs of university nursing students toward hand hygiene. By using a well-validated, self-report hand hygiene questionnaire, a cross-sectional survey was performed. In the convenience sampling, 421 out of 506 questionnaires were returned (83.2%). There were 169 (40.14%), 170 (40.38%) and 82 (19.48%) nursing students from Years 1, 2 and 3, respectively. The respondents’ overall mean score for hand hygiene knowledge was 7.33/10 (SD = 1.52) with an increasing trend of knowledge performance along study year was observed. The hand hygiene practice inventory was 4.76/5 (SD = 0.303). The mean scores for respondents from Years 1, 2 and 3 were 4.75 (SD = 0.367), 4.82 (SD = 0.20) and 4.69 (SD = 0.290), respectively. The self-reported hand hygiene compliance rate was 88.17% (SD = 11.922) and the mean score on the health beliefs scale was 4.03/5 (SD = 0.34). The university nursing students demonstrated moderate knowledge, good practices, a high compliance rate and positive beliefs toward hand hygiene. More effort on educating healthcare professionals about alcohol-based hand rubs and the promotion of hand cream usage to prevent hand-hygiene-induced skin irritations are suggested. Regular training workshops or seminars could be arranged to maintain knowledge levels and cultivate a positive attitude toward hand hygiene

    Isolation and characterization of ZK002, a novel dual function snake venom protein from Deinagkistrodon acutus with anti-angiogenic and anti-inflammatory properties

    Introduction: Pathological angiogenesis, the abnormal or excessive generation of blood vessels, plays an important role in many diseases including cancer, diabetic retinopathy, psoriasis, and arthritis. Additionally, increasing evidence supports the close linkage between angiogenesis and inflammation. Snake venoms are a rich natural source of biologically active molecules and carry rich potential for the discovery of anti-angiogenic and anti-inflammatory modulators.Methods: Here, we isolated and purified a novel protein, ZK002, from the venom of the snake Deinagkistrodon acutus, and investigated its anti-angiogenic and anti-inflammatory activities and mechanisms.Results: ZK002 was identified as a 30 kDa heterodimeric protein of α and ÎČ chains, which exhibited anti-angiogenic activity in various in vitro assays. Mechanistically, ZK002 inhibited activation of VEGF signaling and related mediators including eNOS, p38, LIMK, and HSP27. ZK002 also upregulated the metalloproteinase inhibitor TIMP3 and inhibited components of the VEGF-induced signaling cascade, PPP3R2 and SH2D2A. The anti-angiogenic activity of ZK002 was confirmed in multiple in vivo models. ZK002 could also inhibit the in vitro expression of pro-inflammatory cytokines, as well as in vivo inflammation in the carrageenin-induced edema rat model.Conclusion: Our findings highlight the potential for further development of ZK002 as a dual function therapeutic against diseases with involvement of pathogenic angiogenesis and chronic inflammation

    Cladielloides A and B: New Eunicellin-Type Diterpenoids from an Indonesian Octocoral Cladiella sp

    Two new eunicellin-type diterpenoids, cladielloides A (1) and B (2), which were found to possess a 2-hydroxybutyroxy group in their structures, were isolated from an Indonesian octocoral identified as Cladiella sp. The structures of eunicellins 1 and 2 were elucidated by spectroscopic methods. Cladielloide B (2) exhibited moderate cytotoxicity toward CCRF-CEM tumor cells and this compound displayed significant inhibitory effects on superoxide anion generation and elastase release by human neutrophils

    ALMA Survey of Orion Planck Galactic Cold Clumps (ALMASOP) : How Do Dense Core Properties Affect the Multiplicity of Protostars?

    During the transition phase from a prestellar to a protostellar cloud core, one or several protostars can form within a single gas core. The detailed physical processes of this transition, however, remain unclear. We present 1.3 mm dust continuum and molecular line observations with the Atacama Large Millimeter/submillimeter Array toward 43 protostellar cores in the Orion molecular cloud complex (lambda Orionis, Orion B, and Orion A) with an angular resolution of similar to 0.'' 35 (similar to 140 au). In total, we detect 13 binary/multiple systems. We derive an overall multiplicity frequency (MF) of 28% +/- 4% and a companion star fraction (CSF) of 51% +/- 6%, over a separation range of 300-8900 au. The median separation of companions is about 2100 au. The occurrence of stellar multiplicity may depend on the physical characteristics of the dense cores. Notably, those containing binary/multiple systems tend to show a higher gas density and Mach number than cores forming single stars. The integral-shaped filament of the Orion A giant molecular cloud (GMC), which has the highest gas density and hosts high-mass star formation in its central region (the Orion Nebula cluster), shows the highest MF and CSF among the Orion GMCs. In contrast, the lambda Orionis GMC has a lower MF and CSF than the Orion B and Orion A GMCs, indicating that feedback from H ii regions may suppress the formation of multiple systems. We also find that the protostars comprising a binary/multiple system are usually at different evolutionary stages.Peer reviewe

    ALMA Survey of Orion Planck Galactic Cold Clumps (ALMASOP) : Detection of Extremely High-density Compact Structure of Prestellar Cores and Multiple Substructures Within

    Prestellar cores are self-gravitating dense and cold structures within molecular clouds where future stars are born. They are expected, at the stage of transitioning to the protostellar phase, to harbor centrally concentrated dense (sub)structures that will seed the formation of a new star or the binary/multiple stellar systems. Characterizing this critical stage of evolution is key to our understanding of star formation. In this work, we report the detection of high-density (sub)structures on the thousand-astronomical-unit (au) scale in a sample of dense prestellar cores. Through our recent ALMA observations toward the Orion Planck Galactic Cold Clumps, we have found five extremely dense prestellar cores, which have centrally concentrated regions of similar to 2000 au in size, and several 10(7) cm(-3) in average density. Masses of these centrally dense regions are in the range of 0.30 to 6.89 M. For the first time, our higher resolution observations (0.8 '' similar to 320 au) further reveal that one of the cores shows clear signatures of fragmentation; such individual substructures/fragments have sizes of 800-1700 au, masses of 0.08 to 0.84 M, densities of 2 - 8 x 10(7) cm(-3), and separations of similar to 1200 au. The substructures are massive enough (greater than or similar to 0.1 M) to form young stellar objects and are likely examples of the earliest stage of stellar embryos that can lead to widely (similar to 1200 au) separated multiple systems.Peer reviewe
