    The Effects of Cannabidiol Supplementation on Measures of Performance and Fatigue Following Eccentric Exercise

    Following intense exercise, there is a period of time where performance is decreased. This period of reduced performance is characterized by several factors including myofibrillar disruption, reduced range-of-motion, inflammation, and an influx of enzymes and proteins. Cannabidiol (CBD) has been marketed as a recovery supplement capable of reducing markers of fatigue and inflammation following exercise, yet this claim has not been investigated. PURPOSE: The purpose of this study was to determine if CBD supplementation limits fatigue and expedites a return to performance following intense eccentric exercise. METHODS: A double-blind, crossover design with repeated measures was used. Twenty-four NCAA female athletes (age = 21.2 ± 1.8 yrs., height = 166.4 ± 8 cm, weight = 64.9 ± 9.1 kg) were randomized to either receive 5 mg/kg of CBD in pill form (Cannabidiol Life, Longwood, FL) or a matched weight placebo. Treatments were consumed two hours prior to, immediately following, and ten hours following muscle damage sessions. All participants consumed both treatments, with each separated by approximately 28 days to control for the menstrual cycle. To induce muscle damage, participants completed 10 sets of 10 repetitions of unilateral eccentric leg extension at 60°/sec on an isokinetic dynamometer (Biodex Medical Systems Inc., Shirley, NY). Concentrations of a blood marker indicative of muscle damage (myoglobin), in addition to measures of fatigue (visual analogue fatigue scale [VAFS]) and performance (vertical jump, peak dynamic knee extensor torque at 60, 180, and 300°/sec, and peak isometric knee extensor torque), were collected before and 4, 24, and 48 hours following muscle damaging sessions. A repeated measures MANOVA was conducted to analyze the performance measures, and separate repeated measures ANOVAs were conducted to analyze myoglobin concentrations and results from the VAFS with a significance level of 0.05. RESULTS: A significant increase (p = 0.002) in myoglobin levels was observed for both treatments 4 hours following the muscle damaging session but no significant differences (p \u3e 0.05) were observed between the CBD and placebo groups at any of the 4 measured time points. Peak torque at 60°/sec (p = 0.001) and peak isometric torque (p = 0.02) were significantly lower 24 hours following muscle damage, but none of the 5 measured performance variables were significantly different (p \u3e 0.05 for all) between the CBD and placebo treatment at any time point. Subjective fatigue as measured by the VAFS was not significantly different (p \u3e 0.05) between the CBD and placebo treatments at any measured time point. CONCLUSION: Cannabidiol supplementation was unable to reduce fatigue and restore performance when compared to a placebo in well-trained female participants. It does not appear that CBD supplementation is of beneficial use as a recovery supplement following intense exercise in athletes

    Revising upper-ocean sulfur dynamics near Bermuda : new lessons from 3 years of concentration and rate measurements

The definitive version was published in Environmental Chemistry 13 (2016): 302-313, doi:10.1071/EN15045.Oceanic biogeochemical cycling of dimethylsulfide (DMS), and its precursor dimethylsulfoniopropionate (DMSP), has gained considerable attention over the past three decades because of the potential role of DMS in climate mediation. Here we report 3 years of monthly vertical profiles of organic sulfur cycle concentrations (DMS, particulate DMSP (DMSPp) and dissolved DMSP (DMSPd)) and rates (DMSPd consumption, biological DMS consumption and DMS photolysis) from the Bermuda Atlantic Time-series Study (BATS) site taken between 2005 and 2008. Concentrations confirm the summer paradox with mixed layer DMS peaking ~90 days after peak DMSPp and ~50 days after peak DMSPp : Chl. A small decline in mixed layer DMS was observed relative to those measured during a previous study at BATS (1992–1994), potentially driven by long-term climate shifts at the site. On average, DMS cycling occurred on longer timescales than DMSPd (0.43 ± 0.35 v. 1.39 ± 0.76 day–1) with DMSPd consumption rates remaining elevated throughout the year despite significant seasonal variability in the bacterial DMSP degrader community. DMSPp was estimated to account for 4–5 % of mixed layer primary production and turned over at a significantly slower rate (~0.2 day–1). Photolysis drove DMS loss in the mixed layer during the summer, whereas biological consumption of DMS was the dominant loss process in the winter and at depth. These findings offer new insight into the underlying mechanisms driving DMS(P) cycling in the oligotrophic ocean, provide an extended dataset for future model evaluation and hypothesis testing and highlight the need for a reexamination of past modelling results and conclusions drawn from data collected with old methodologies.The authors acknowledge funding from the National Science Foundation (NSF) (OCE-0425166) and the Center for Microbial Oceanography Research and Education (CMORE) a NSF Science and Technology Center (EF-0424599)

    The Effects of Equine-Assisted Therapy on Gait in Adults with Parkinson’s Disease: A Preliminary Analysis

    Parkinson’s disease (PD) is a chronic, progressive, neurological disorder caused by the destruction of dopamine-producing neurons in the substantia nigra of the brain. To treat motor symptoms of PD, individuals may choose alternative modes of exercise, such as equine-assisted therapy (EAT), to improve physiological health. PURPOSE: To determine changes in gait following 8 weeks of Equine Assisted Therapy (EAT) in older men with PD. METHODS: Six older adults (age = 68.0 ± 8.6 yrs; height = 178.6 ± 8.3 cm; weight = 93.4 ± 16.3 kg; Hoehn and Yahr classification = 2.8 ± 0.4; time since diagnosis = 7.3 ± 5.0 yrs) performed two, 60-minute riding sessions weekly for 8 weeks. Before, midway, immediately after, and at two points following the EAT program (at 8 weeks and 16 weeks), spatiotemporal parameters of gait were measured using motion capture with infrared markers strategically placed on lower-body anatomical landmarks. Participants walked at a self-selected speed without the use of an assistive device in the capture space for 15 consecutive strides, turned around, and walked back across the space. Gait velocity, step length, time spent in stance phase, time spent in swing phase, toe clearance were collected. Data were analyzed using a repeated-measures analysis-of-variance and a Bonferroni correction with a significance level of 0.05. RESULTS: Gait velocity (101.8 ± 29.1 vs. 109.2 ± 34.9 vs. 116.3 ± 32.8 vs. 105.7 ± 26.2 vs. 108.0 ± 25.4 cm/s for pre-EAT, midway through EAT, immediately post-EAT, 8 weeks after EAT concluded, and 16 weeks after EAT concluded, respectively) was significant across all time points (p = 0.03). All other variables were statistically similar (p \u3e 0.05) across all time points. CONCLUSION: There is an observable trend towards significance in each gait variable immediately after 8 weeks of EAT. The study is ongoing and will further explore these differences

    Population dynamics of rhesus macaques and associated foamy virus in Bangladesh.

    Foamy viruses are complex retroviruses that have been shown to be transmitted from nonhuman primates to humans. In Bangladesh, infection with simian foamy virus (SFV) is ubiquitous among rhesus macaques, which come into contact with humans in diverse locations and contexts throughout the country. We analyzed microsatellite DNA from 126 macaques at six sites in Bangladesh in order to characterize geographic patterns of macaque population structure. We also included in this study 38 macaques owned by nomadic people who train them to perform for audiences. PCR was used to analyze a portion of the proviral gag gene from all SFV-positive macaques, and multiple clones were sequenced. Phylogenetic analysis was used to infer long-term patterns of viral transmission. Analyses of SFV gag gene sequences indicated that macaque populations from different areas harbor genetically distinct strains of SFV, suggesting that geographic features such as forest cover play a role in determining the dispersal of macaques and SFV. We also found evidence suggesting that humans traveling the region with performing macaques likely play a role in the translocation of macaques and SFV. Our studies found that individual animals can harbor more than one strain of SFV and that presence of more than one SFV strain is more common among older animals. Some macaques are infected with SFV that appears to be recombinant. These findings paint a more detailed picture of how geographic and sociocultural factors influence the spectrum of simian-borne retroviruses

    PatientExploreR: an extensible application for dynamic visualization of patient clinical history from electronic health records in the OMOP common data model.

    MotivationElectronic health records (EHRs) are quickly becoming omnipresent in healthcare, but interoperability issues and technical demands limit their use for biomedical and clinical research. Interactive and flexible software that interfaces directly with EHR data structured around a common data model (CDM) could accelerate more EHR-based research by making the data more accessible to researchers who lack computational expertise and/or domain knowledge.ResultsWe present PatientExploreR, an extensible application built on the R/Shiny framework that interfaces with a relational database of EHR data in the Observational Medical Outcomes Partnership CDM format. PatientExploreR produces patient-level interactive and dynamic reports and facilitates visualization of clinical data without any programming required. It allows researchers to easily construct and export patient cohorts from the EHR for analysis with other software. This application could enable easier exploration of patient-level data for physicians and researchers. PatientExploreR can incorporate EHR data from any institution that employs the CDM for users with approved access. The software code is free and open source under the MIT license, enabling institutions to install and users to expand and modify the application for their own purposes.Availability and implementationPatientExploreR can be freely obtained from GitHub: https://github.com/BenGlicksberg/PatientExploreR.     Improvement of neuropathology and transcriptional deficits in CAG 140 knock-in mice supports a beneficial effect of dietary curcumin in Huntington's disease

    <p>Abstract</p> <p>Backgound</p> <p>No disease modifying treatment currently exists for Huntington's disease (HD), a fatal neurodegenerative disorder characterized by the formation of amyloid-like aggregates of the mutated huntingtin protein. Curcumin is a naturally occurring polyphenolic compound with Congo red-like amyloid binding properties and the ability to cross the blood brain barrier. CAG140 mice, a knock-in (KI) mouse model of HD, display abnormal aggregates of mutant huntingtin and striatal transcriptional deficits, as well as early motor, cognitive and affective abnormalities, many months prior to exhibiting spontaneous gait deficits, decreased striatal volume, and neuronal loss. We have examined the ability of life-long dietary curcumin to improve the early pathological phenotype of CAG140 mice.</p> <p>Results</p> <p>KI mice fed a curcumin-containing diet since conception showed decreased huntingtin aggregates and increased striatal DARPP-32 and D1 receptor mRNAs, as well as an amelioration of rearing deficits. However, similar to other antioxidants, curcumin impaired rotarod behavior in both WT and KI mice and climbing in WT mice. These behavioral effects were also noted in WT C57Bl/6 J mice exposed to the same curcumin regime as adults. However, neither locomotor function, behavioral despair, muscle strength or food utilization were affected by curcumin in this latter study. The clinical significance of curcumin's impairment of motor performance in mice remains unclear because curcumin has an excellent blood chemistry and adverse event safety profile, even in the elderly and in patients with Alzheimer's disease.</p> <p>Conclusion</p> <p>Together with this clinical experience, the improvement in several transgene-dependent parameters by curcumin in our study supports a net beneficial effect of dietary curcumin in HD.</p

    Optimal model complexity for terrestrial carbon cycle prediction

    The terrestrial carbon cycle plays a critical role in modulating the interactions of climate with the Earth system, but different models often make vastly different predictions of its behavior. Efforts to reduce model uncertainty have commonly focused on model structure, namely by introducing additional processes and increasing structural complexity. However, the extent to which increased structural complexity can directly improve predictive skill is unclear. While adding processes may improve realism, the resulting models are often encumbered by a greater number of poorly determined or over-generalized parameters. To guide efficient model development, here we map the theoretical relationship between model complexity and predictive skill. To do so, we developed 16 structurally distinct carbon cycle models spanning an axis of complexity and incorporated them into a model–data fusion system. We calibrated each model at six globally distributed eddy covariance sites with long observation time series and under 42 data scenarios that resulted in different degrees of parameter uncertainty. For each combination of site, data scenario, and model, we then predicted net ecosystem exchange (NEE) and leaf area index (LAI) for validation against independent local site data. Though the maximum model complexity we evaluated is lower than most traditional terrestrial biosphere models, the complexity range we explored provides universal insight into the inter-relationship between structural uncertainty, parametric uncertainty, and model forecast skill. Specifically, increased complexity only improves forecast skill if parameters are adequately informed (e.g., when NEE observations are used for calibration). Otherwise, increased complexity can degrade skill and an intermediate-complexity model is optimal. This finding remains consistent regardless of whether NEE or LAI is predicted. Our COMPLexity EXperiment (COMPLEX) highlights the importance of robust observation-based parameterization for land surface modeling and suggests that data characterizing net carbon fluxes will be key to improving decadal predictions of high-dimensional terrestrial biosphere models.</p

    Neurocognitive function in HIV infected patients on antiretroviral therapy

    OBJECTIVE To describe factors associated with neurocognitive (NC) function in HIV-positive patients on stable combination antiretroviral therapy. DESIGN We undertook a cross-sectional analysis assessing NC data obtained at baseline in patients entering the Protease-Inhibitor-Monotherapy-Versus-Ongoing-Triple therapy (PIVOT) trial. MAIN OUTCOME MEASURE NC testing comprised of 5 domains. Raw results were z-transformed using standard and demographically adjusted normative datasets (ND). Global z-scores (NPZ-5) were derived from averaging the 5 domains and percentage of subjects with test scores >1 standard deviation (SD) below population means in at least two domains (abnormal Frascati score) calculated. Patient characteristics associated with NC results were assessed using multivariable linear regression. RESULTS Of the 587 patients in PIVOT, 557 had full NC results and were included. 77% were male, 68% Caucasian and 28% of Black ethnicity. Mean (SD) baseline and nadir CD4+ lymphocyte counts were 553(217) and 177(117) cells/µL, respectively, and HIV RNA was <50 copies/mL in all. Median (IQR) NPZ-5 score was -0.5 (-1.2/-0) overall, and -0.3 (-0.7/0.1) and -1.4 (-2/-0.8) in subjects of Caucasian and Black ethnicity, respectively. Abnormal Frascati scores using the standard-ND were observed in 51%, 38%, and 81%, respectively, of subjects overall, Caucasian and Black ethnicity (p<0.001), but in 62% and 69% of Caucasian and Black subjects using demographically adjusted-ND (p = 0.20). In the multivariate analysis, only Black ethnicity was associated with poorer NPZ-5 scores (P<0.001). CONCLUSIONS In this large group of HIV-infected subjects with viral load suppression, ethnicity but not HIV-disease factors is closely associated with NC results.     Compact HI clouds from the GALFA-HI survey

    The Galactic Arecibo L-band Feed Array HI (GALFA-HI) survey is mapping the entire Arecibo sky at 21-cm, over a velocity range of -700 to +700 km/s (LSR), at a velocity resolution of 0.18 km/s and a spatial resolution of 3.5 arcmin. The unprecedented resolution and sensitivity of the GALFA-HI survey have resulted in the detection of numerous isolated, very compact HI clouds at low Galactic velocities, which are distinctly separated from the HI disk emission. In the limited area of ~4600 deg2^2 surveyed so far, we have detected 96 of such compact clouds. The detected clouds are cold with a median Tk,max_{k,max} (the kinetic temperature in the case in which there is no non-thermal broadening) of 300 K. Moreover, these clouds are quite compact and faint, with median values of 5 arcmin in angular size, 0.75 K in peak brightness temperature, and 5×10185 \times 10^{18} cm2^{-2} in HI column density. Most of the clouds deviate from Galactic rotation at the 20-30 km/s level, and a significant fraction show evidence for a multiphase medium and velocity gradients. No counterparts for these clouds were found in other wavebands. From the modeling of spatial and velocity distributions of the whole compact cloud population, we find that the bulk of the compact clouds are related to the Galactic disk, and their distances are likely to be in the range of 0.1 to a few kpc. We discuss various possible scenarios for the formation and maintenance of this cloud population and its significance for Galactic ISM studies.Comment: Accepted for publication in the Astrophysical Journa
