    Bipyridinium-carboxylate ligands towards photo- and thermo-chromic porous coordination polymers

    Porous coordination polymers (PCP) or metal−organic frameworks (MOF) have focused attention going from synthetic strategy to applications in heterogeneous catalysis, molecular recognition and gas storage. Up to now the main strategy to increase the sorption abilities has been to introduce coordinatively unsaturated metal centres. Only few examples have been reported based on the assembly of cationic bridging ligands. Our original approach consists in combining the coordination versatility of carboxylate functions, commonly used in this field, to electro-active viologen derivatives in order to synthesize new porous coordination polymers for gas storage and redox mediators with optical and magnetic properties. The designed ligands are based on N-substituted-4,4’-bipyridinium monocation and N,N’-disubstituted-4,4’-bipyridinium dication bearing one or several carboxylate groups. In addition to act as charge-separated organic linkers, these pyridinium-carboxylate ligands will i) afford a cationic surface to enhance the interactions with guests and ii) exhibit reversible redox states involving monocationic radicals with good stability and large absorptions coefficients in the visible range. Some of our new results will be presented here and a particular attention will be dedicated to compounds that also exhibit thermo-, solvato- and photo-chromic behavior.

    A new scintillating fiber dosimeter using a single optical fiber and a CCD camera

    Radiotherapy treatments become more and more accurate, using very small irradiation fields and complex dose depositions. So small dosimeters for real time and in vivo dosimetry, suitable for photons as well as for electrons beams are highly desired. In this context, a scintillating fiber dosimeter (SFD) has been developed by the Laboratoire de Physique Corpusculaire de Caen (LPC Caen), France, in collaboration with one of the French regional center for cancer treatment Centre Regional de lutte contre le cancer F. Baclesse (CRLCC F. Baclesse), Caen, France, and the ELDIM Company, Herouville, France. This plastic dosimeter is water equivalent, and it is suitable for photons as well as for electrons beams without correction. It is a real time dosimeter, with an excellent signal to noise ratio, and a spatial resolution of about a few millimeters. The aim of this study was to reduce the size of the scintillator in order to improve the spatial resolution of this dosimeter. So, a new light collection device has been developed to reduce the length of the scintillator from 1 cm to 1 mm without loss in the signal to noise ratio. The accuracy of this improved prototype has been tested by comparison with standard ionization chambers and the difference between the two devices never exceeded one percent for photon and for electron irradiation beams. A first set of commercial SFD is under completion at ELDIM and it will be soon clinically tested in several French centers for cancer treatment

    Coupled networks of permanent protected areas and dynamic conservation areas for biodiversity conservation under climate change

    The complexity of climate change impacts on ecological processes necessitates flexible and adaptive conservation strategies that cross traditional disciplines. Current strategies involving protected areas are predominantly fixed in space, and may on their own be inadequate under climate change. Here, we propose a novel approach to climate adaptation that combines permanent protected areas with temporary conservation areas to create flexible networks. Previous work has tended to consider permanent and dynamic protection as separate actions, but their integration could draw on the strengths of both approaches to improve biodiversity conservation and help manage for ecological uncertainty in the coming decades. As there are often time lags in the establishment of new permanent protected areas, the inclusion of dynamic conservation areas within permanent networks could provide critical transient protection to mitigate land-use changes and biodiversity redistributions. This integrated approach may be particularly useful in highly human-modified and fragmented landscapes where areas of conservation value are limited and long-term place-based protection is unfeasible. To determine when such an approach may be feasible, we propose the use of a decision framework. Under certain scenarios, these coupled networks have the potential to increase spatio-temporal network connectivity and help maintain biodiversity and ecological processes under climate change. Implementing these networks would require multidisciplinary scientific evidence, new policies, creative funding solutions, and broader acceptance of a dynamic approach to biodiversity conservation

    Mendelian randomisation study of height and body mass index as modifiers of ovarian cancer risk in 22,588 BRCA1 and BRCA2 mutation carriers

    Item does not contain fulltextBACKGROUND: Height and body mass index (BMI) are associated with higher ovarian cancer risk in the general population, but whether such associations exist among BRCA1/2 mutation carriers is unknown. METHODS: We applied a Mendelian randomisation approach to examine height/BMI with ovarian cancer risk using the Consortium of Investigators for the Modifiers of BRCA1/2 (CIMBA) data set, comprising 14,676 BRCA1 and 7912 BRCA2 mutation carriers, with 2923 ovarian cancer cases. We created a height genetic score (height-GS) using 586 height-associated variants and a BMI genetic score (BMI-GS) using 93 BMI-associated variants. Associations were assessed using weighted Cox models. RESULTS: Observed height was not associated with ovarian cancer risk (hazard ratio [HR]: 1.07 per 10-cm increase in height, 95% confidence interval [CI]: 0.94-1.23). Height-GS showed similar results (HR = 1.02, 95% CI: 0.85-1.23). Higher BMI was significantly associated with increased risk in premenopausal women with HR = 1.25 (95% CI: 1.06-1.48) and HR = 1.59 (95% CI: 1.08-2.33) per 5-kg/m(2) increase in observed and genetically determined BMI, respectively. No association was found for postmenopausal women. Interaction between menopausal status and BMI was significant (Pinteraction < 0.05). CONCLUSION: Our observation of a positive association between BMI and ovarian cancer risk in premenopausal BRCA1/2 mutation carriers is consistent with findings in the general population

    Avaliação da qualidade de vida, da dor nas costas, da funcionalidade e de alterações da coluna vertebral de estudantes de fisioterapia

    O objetivo deste estudo foi avaliar a qualidade de vida, dor nas costas, funcionalidade e as alterações da coluna vertebral de estudantes de fisioterapia. Participaram 42 universitários, sendo avaliada a coluna vertebral por meio do arcômetro, a dor nas costas por meio de um questionário multidimensional de dor, a qualidade de vida por meio do questionário SF-36 e funcionalidade a partir do Roland-Morris. Foi realizada estatística descritiva e teste qui-quadrado (α=0,05). Os resultados demonstraram: prevalência de alterações nas curvaturas da coluna vertebral de 38,1%; (n=16); alta prevalência de dor nas costas (69%; n=29); baixa prevalência de comprometimento da funcionalidade (3,4%; n=1); que não há associação entre as alterações nas curvaturas da coluna vertebral e funcionalidade e dor nas costas; e que os escores dos domínios do SF-36 foram maiores que 45, exceto o domínio "dor", cujo escore aproximado foi de 35. Conclui-se que quanto menor os níveis de intensidade da dor melhor o nível de qualidade de vida

    Functional mechanisms underlying pleiotropic risk alleles at the 19p13.1 breast-ovarian cancer susceptibility locus

    A locus at 19p13 is associated with breast cancer (BC) and ovarian cancer (OC) risk. Here we analyse 438 SNPs in this region in 46,451 BC and 15,438 OC cases, 15,252 BRCA1 mutation carriers and 73,444 controls and identify 13 candidate causal SNPs associated with serous OC (P=9.2 × 10-20), ER-negative BC (P=1.1 × 10-13), BRCA1-associated BC (P=7.7 × 10-16) and triple negative BC (P-diff=2 × 10-5). Genotype-gene expression associations are identified for candidate target genes ANKLE1 (P=2 × 10-3) and ABHD8 (P<2 × 10-3). Chromosome conformation capture identifies interactions between four candidate SNPs and ABHD8, and luciferase assays indicate six risk alleles increased transactivation of the ADHD8 promoter. Targeted deletion of a region containing risk SNP rs56069439 in a putative enhancer induces ANKLE1 downregulation; and mRNA stability assays indicate functional effects for an ANKLE1 3′-UTR SNP. Altogether, these data suggest that multiple SNPs at 19p13 regulate ABHD8 and perhaps ANKLE1 expression, and indicate common mechanisms underlying breast and ovarian cancer risk

    Genome-Wide Association Study in BRCA1 Mutation Carriers Identifies Novel Loci Associated with Breast and Ovarian Cancer Risk

    BRCA1-associated breast and ovarian cancer risks can be modified by common genetic variants. To identify further cancer risk-modifying loci, we performed a multi-stage GWAS of 11,705 BRCA1 carriers (of whom 5,920 were diagnosed with breast and 1,839 were diagnosed with ovarian cancer), with a further replication in an additional sample of 2,646 BRCA1 carriers. We identified a novel breast cancer risk modifier locus at 1q32 for BRCA1 carriers (rs2290854, P = 2.7×10-8, HR = 1.14, 95% CI: 1.09-1.20). In addition, we identified two novel ovarian cancer risk modifier loci: 17q21.31 (rs17631303, P = 1.4×10-8, HR = 1.27, 95% CI: 1.17-1.38) and 4q32.3 (rs4691139, P = 3.4×10-8, HR = 1.20, 95% CI: 1.17-1.38).
