    Alguns dos requisitos do projeto sísmico de edifícios de madeira são compatíveis com as recomendações para o dimensionamento tendo em conta a robustez estrutural. Neste estudo é analisado o efeito do dimensionamento sísmico na robustez de pórticos de madeira.     The Farther the Better: Effects of Multiple Environmental Variables on Reef Fish Assemblages along a Distance Gradient from River Influences

    The conservation and management of site-attached assemblages of coastal reefs are particularly challenging because of the tremendous environmental variation that exists at small spatial scales. In this sense, understanding the primary sources of variation in spatial patterns of the biota is fundamental for designing effective conservation policies. We investigated spatial variation in fish assemblages around the windward and leeward sides of coastal islands situated across a gradient of riverine influence (13 km in length). Specifically, relationships between rocky reef fish assemblages and benthic, topographic and physical predictors were assessed. We hypothesized that river induced disturbances may overcome local habitat features in modeling spatial patterns of fish distribution. Fish assemblages varied primarily due to the strong directional gradient of riverine influence (22.6% of the estimated components of variation), followed by topographic complexity (15%), wave exposure (9.9%), and benthic cover (8%). The trophic structure of fish assemblages changed from having a high abundance of invertebrate feeders in macroalgae-dominated reefs close to river mouths to a high proportion of herbivores, planktivores and invertebrate feeder species in reefs with large boulders covered by epilithic algal matrices, as the distance from rivers increased. This gradient led to an increase of 4.5-fold in fish richness and fish trophic group diversity, 11-fold in fish biomass and 10-fold in fish abundance. Our results have implications for the conservation and monitoring of assemblages patchily distributed at small spatial scales.     Fragility analysis of a mass-timber frame structure with ring-doweled moment-resisting connections

    © WCTE 2018 Committee. The nonlinear behaviour of connections between structural elements is critical to the performance of mass-timber structures under seismic loads. However, limited work has been developed in nonlinear modelling and fragility assessment of mass-timber structures. To improve the accuracy of this approach, in particular when considering structures with ring-doweled moment-resisting connections, a nonlinear modelling approach and fragility assessment are proposed and a prototype example of a three-story building is analysed herein as a case study. For the case study, connections and members were designed following the prescriptions in Eurocode 5 and Eurocode 8, considering a high ductility structure. The mechanical properties of the structure are modelled as random variables to evaluate the impact of uncertainty on the prediction of the structural performance, in particular, on the probability of occurrence of ductile and brittle failure modes. The structure is studied under both nonlinear static analysis and multi-record incremental dynamic analysis. From these, fragility curves for different damage levels are computed and a q-factor is proposed. Results indicate that the requirements of Eurocode 5 and Eurocode 8 are sufficient to guarantee adequate performance for this type of structure, albeit these may be overconservative. Moreover, it is shown that uncertainties in material properties have a significant impact on the collapse capacity of these structures.This work had financial support of the Portuguese Science Foundation (FCT), through PhD grant PD/BD/113679/2015 included in InfraRisk-PhD program.     Compartmentalized PDE4A5 signaling impairs hippocampal synaptic plasticity and long-term memory

    Alterations in cAMP signaling are thought to contribute to neurocognitive and neuropsychiatric disorders. Members of the cAMP-specific phosphodiesterase 4 (PDE4) family, which contains >25 different isoforms, play a key role in determining spatial cAMP degradation so as to orchestrate compartmentalized cAMP signaling in cells. Each isoform binds to a different set of protein complexes through its unique N-terminal domain, thereby leading to targeted degradation of cAMP in specific intracellular compartments. However, the functional role of specific compartmentalized PDE4 isoforms has not been examined in vivo. Here, we show that increasing protein levels of the PDE4A5 isoform in mouse hippocampal excitatory neurons impairs a long-lasting form of hippocampal synaptic plasticity and attenuates hippocampus-dependent long-term memories without affecting anxiety. In contrast, viral expression of a truncated version of PDE4A5, which lacks the unique N-terminal targeting domain, does not affect long-term memory. Further, overexpression of the PDE4A1 isoform, which targets a different subset of signalosomes, leaves memory undisturbed. Fluorescence resonance energy transfer sensor-based cAMP measurements reveal that the full-length PDE4A5, in contrast to the truncated form, hampers forskolin-mediated increases in neuronal cAMP levels. Our study indicates that the unique N-terminal localization domain of PDE4A5 is essential for the targeting of specific cAMP-dependent signaling underlying synaptic plasticity and memory. The development of compounds to disrupt the compartmentalization of individual PDE4 isoforms by targeting their unique N-terminal domains may provide a fruitful approach to prevent cognitive deficits in neuropsychiatric and neurocognitive disorders that are associated with alterations in cAMP signaling

    A Phage-Like Plasmid Carrying blaKPC-2 Gene in Carbapenem-Resistant Pseudomonas aeruginosa

    Background: Lateral gene transfer plays a central role in the dissemination of carbapenem resistance in bacterial pathogens associated with nosocomial infections, mainly Enterobacteriaceae and Pseudomonas aeruginosa. Despite their clinical significance, there is little information regarding the mobile genetic elements and mechanism of acquisition and propagation of lateral genes in P. aeruginosa, and they remain largely unknown.Objectives: The present study characterized the genetic context of blaKPC-2 in carbapenem-resistant P. aeruginosa strain BH9.Methods:Pseudomonas aeruginosa BH9 sequencing was performed using the long-read PacBio SMRT platform and the Ion Proton System. De novo assembly was carried out using the SMRT pipeline and Canu, and gene prediction and annotation were performed using Prokka and RAST.Results:Pseudomonas aeruginosa BH9 exhibited a 7.1 Mb circular chromosome. However, the blaKPC-2 gene is located in an additional contig composed by a small plasmid pBH6 from P. aeruginosa strain BH6 and several phage-related genes. Further analysis revealed that the beginning and end of the contig contain identical sequences, supporting a circular plasmid structure. This structure spans 41,087 bp, exhibiting all the Mu-like phage landmarks. In addition, 5-bp direct repeats (GGATG) flanking the pBH6 ends were found, strongly indicating integration of the Mu-like phage into the pBH6 plasmid. Mu phages are commonly found in P. aeruginosa. However, for the first time showing a potential impact in shaping the vehicles of the dissemination of antimicrobial (e.g., plasmid pBH6) resistance genes in the Pseudomonas genus.Conclusion: pBH6 captured the Mu-like Phage BH9, creating a co-integrate pBH6::Phage BH9, and this phage-plasmid complex may represent novel case of a phage-like plasmid

    Optimized generation of spatial qudits by using a pure phase spatial light modulator

    We present a method for preparing arbitrary pure states of spatial qudits, namely, D-dimensional (D ≥ 2) quantum systems carrying information in the transverse momentum and position of single photons. For this purpose, a set of D slits with complex transmission are displayed on a spatial light modulator (SLM). In a recent work we have shown a method that requires a single phase-only SLM to control independently the complex coefficients which define the quantum state of dimension D. The amplitude information was codified by introducing phase gratings inside each slit and the phase value of the complex transmission was added to the phase gratings. After a spatial filtering process we obtained in the image plane the desired qudit state. Although this method has proven to be a good alternative to compact the previously reported architectures, it presents some features that could be improved. In this paper we present an alternative scheme to codify the required phase values that minimizes the effects of temporal phase fluctuations associated to the SLM where the codification is carried on. In this scheme the amplitudes are set by appropriate phase gratings addressed at the SLM while the relative phases are obtained by a lateral displacement of these phase gratings. We show that this method improves the quality of the prepared state and provides very high fidelities of preparation for any state. An additional advantage of this scheme is that a complete 2\pi modulation is obtained by shifting the grating by one period, and hence the encoding is not limited by the phase modulation range achieved by the SLM. Numerical simulations, that take into account the phase fluctuations, show high fidelities for thousands of qubit states covering the whole Bloch sphere surface.     Preparing arbitrary pure states of spatial qudits with a single phase-only spatial light modulator

    Spatial qudits are -dimensional (≥2) quantum systems carrying information encoded in the discretized transverse momentum and position of single photons. We present a proof-of-principle demonstration of a method for preparing arbitrary pure states of such systems by using a single phase-only spatial light modulator (SLM). The method relies on the encoding of the complex transmission function corresponding to a given spatial qudit state onto a preset diffraction order of a phase-only grating function addressed at the SLM. Fidelities of preparation above 94% were obtained with this method, which is simpler, less costly, and more efficient than those that require two SLMs for the same purpose.Instituto de Física La Plat

    Alouatta guariba is endemic to the Atlantic Forest in eastern Brazil and northeast Argentina. In the south, its distribution is limited by the Camaquã river basin (Printes et al. 2001) and to the north by the Paraguaçu river in the state of Bahia (Gregorin 2006; Neves et al. 2018). The western boundary is marked by the limits of the Atlantic Forest. It occurs in the Brazilian states of Bahia, Espírito Santo, Rio de Janeiro, Minas Gerais, São Paulo, Paraná, Santa Catarina and Rio Grande do Sul (Mendes et al. 2008, Bicca-Marques et al. 2018).     Numerical modeling of CLT diaphragms tested on a shake-table experiment

    Current standards and existing literature provide very limited information regarding the design of cross-laminated timber (CLT) floor diaphragms. In addition, limited procedures exist to develop analytical models to estimate the deformation response of CLT floor diaphragms. This paper presents a modelling approach that captures the response of CLT timber diaphragms, with a special focus on CLT spline panel-to-panel connections. The modeling approach is validated through the comparison of the results of the computation model with experimental data obtained from a series of shake-tables test performed on a two-story full-scale building tested in the summer of 2017 at UC San Diego Large High Performance Outdoor Shake Table. The two-story building included two diaphragm designs at each floor level. The first solution consists of CLT panels connected with plywood surface splines that are fastened using self-tapping screws, while the second consists of a CLT-concrete composite floor solution. Results from the nonlinear pushover analysis describe accurately the experimental data obtained.(undefined
