Serum levels and removal by haemodialysis and haemodiafiltration of tryptophan-derived uremic toxins in ESKD patients
Tryptophan is an essential dietary amino acid that originates uremic toxins that contribute to end-stage kidney disease (ESKD) patient outcomes. We evaluated serum levels and removal during haemodialysis and haemodiafiltration of tryptophan and tryptophan-derived uremic toxins, indoxyl sulfate (IS) and indole acetic acid (IAA), in ESKD patients in different dialysis treatment settings. This prospective multicentre study in four European dialysis centres enrolled 78 patients with ESKD. Blood and spent dialysate samples obtained during dialysis were analysed with high-performance liquid chromatography to assess uremic solutes, their reduction ratio (RR) and total removed solute (TRS). Mean free serum tryptophan and IS concentrations increased, and concentration of IAA decreased over pre-dialysis levels (67%, 49%, -0.8%, respectively) during the first hour of dialysis. While mean serum total urea, IS and IAA concentrations decreased during dialysis (-72%, -39%, -43%, respectively), serum tryptophan levels increased, resulting in negative RR (-8%) towards the end of the dialysis session (p < 0.001), despite remarkable Trp losses in dialysate. RR and TRS values based on serum (total, free) and dialysate solute concentrations were lower for conventional low-flux dialysis (p < 0.001). High-efficiency haemodiafiltration resulted in 80% higher Trp losses than conventional low-flux dialysis, despite similar neutral Trp RR values. In conclusion, serum Trp concentrations and RR behave differently from uremic solutes IS, IAA and urea and Trp RR did not reflect dialysis Trp losses. Conventional low-flux dialysis may not adequately clear Trp-related uremic toxins while high efficiency haemodiafiltration increased Trp losses
Tropical larval and juvenile fish critical swimming speed (U-crit) and morphology data
Fish swimming capacity is a key life history trait critical to many aspects of their ecology. U-crit (critical) swimming speeds provide a robust, repeatable relative measure of swimming speed that can serve as a useful surrogate for other measures of swimming performance. Here we collate and make available one the most comprehensive datasets on U-crit swimming abilities of tropical marine fish larvae and pelagic juveniles, most of which are reef associated as adults. The dataset includes U-crit speed measurements for settlement stage fishes across a large range of species and families obtained mostly from field specimens collected in light traps and crest nets; and the development of swimming abilities throughout ontogeny for a range of species using reared larvae. In nearly all instances, the size of the individual was available, and in many cases, data include other morphological measurements (e.g. âpropulsive areaâ) useful for predicting swimming capacity. We hope these data prove useful for further studies of larval swimming performance and other broader syntheses

A simple device for multiplex ELISA made from melt-extruded plastic microcapillary film
We present a simple device for multiplex quantitative enzyme-linked immunosorbant assays (ELISA) made from a novel melt-extruded microcapillary film (MCF) containing a parallel array of 200”m capillaries along its length. To make ELISA devices different protein antigens or antibodies were immobilised inside individual microcapillaries within long reels of MCF extruded from fluorinated ethylene propylene (FEP). Short pieces of coated film were cut and interfaced with a pipette, allowing sequential uptake of samples and detection solutions into all capillaries from a reagent well. As well as being simple to produce, these FEP MCF devices have excellent light transmittance allowing direct optical interrogation of the capillaries for simple signal quantification. Proof of concept experiments demonstrate both quantitative and multiplex assays in FEP MCF devices using a standard direct ELISA procedure and read using a flatbed scanner. This new multiplex immunoassay platform should find applications ranging from lab detection to point-of-care and field diagnostics
Inverse problem for wave equation with sources and observations on disjoint sets
We consider an inverse problem for a hyperbolic partial differential equation
on a compact Riemannian manifold. Assuming that and are
two disjoint open subsets of the boundary of the manifold we define the
restricted Dirichlet-to-Neumann operator . This
operator corresponds the boundary measurements when we have smooth sources
supported on and the fields produced by these sources are observed
on . We show that when and are disjoint but
their closures intersect at least at one point, then the restricted
Dirichlet-to-Neumann operator determines the
Riemannian manifold and the metric on it up to an isometry. In the Euclidian
space, the result yields that an anisotropic wave speed inside a compact body
is determined, up to a natural coordinate transformations, by measurements on
the boundary of the body even when wave sources are kept away from receivers.
Moreover, we show that if we have three arbitrary non-empty open subsets

also for the finite-time boundary measurements when the hyperbolic equation

A rigorous analysis of high order electromagnetic invisibility cloaks
There is currently a great deal of interest in the invisibility cloaks
recently proposed by Pendry et al. that are based in the transformation
approach. They obtained their results using first order transformations. In
recent papers Hendi et al. and Cai et al. considered invisibility cloaks with
high order transformations. In this paper we study high order electromagnetic
invisibility cloaks in transformation media obtained by high order
transformations from general anisotropic media. We consider the case where
there is a finite number of spherical cloaks located in different points in
space. We prove that for any incident plane wave, at any frequency, the
scattered wave is identically zero. We also consider the scattering of finite
energy wave packets. We prove that the scattering matrix is the identity, i.e.,
that for any incoming wave packet the outgoing wave packet is the same as the
incoming one. This proves that the invisibility cloaks can not be detected in
any scattering experiment with electromagnetic waves in high order
transformation media, and in particular in the first order transformation media
of Pendry et al. We also prove that the high order invisibility cloaks, as well
as the first order ones, cloak passive and active devices. The cloaked objects
completely decouple from the exterior. Actually, the cloaking outside is
independent of what is inside the cloaked objects. The electromagnetic waves
inside the cloaked objects can not leave the concealed regions and viceversa,
the electromagnetic waves outside the cloaked objects can not go inside the
concealed regions. As we prove our results for media that are obtained by
transformation from general anisotropic materials, we prove that it is possible

Lack of correlation of stem cell markers in breast cancer stem cells
BACKGROUND: Various markers are used to identify the unique sub-population of breast cancer cells with stem cell properties. Whether these markers are expressed in all breast cancers, identify the same population of cells, or equate to therapeutic response is controversial. METHODS: We investigated the expression of multiple cancer stem cell markers in human breast cancer samples and cell lines in vitro and in vivo, comparing across and within samples and relating expression with growth and therapeutic response to doxorubicin, docetaxol and radiotherapy. RESULTS: CD24, CD44, ALDH and SOX2 expression, the ability to form mammospheres and side-population cells are variably present in human cancers and cell lines. Each marker identifies a unique rather than common population of cancer cells. In vivo, cells expressing these markers are not specifically localized to the presumptive stem cell niche at the tumour/stroma interface. Repeated therapy does not consistently enrich cells expressing these markers, although ER-negative cells accumulate. CONCLUSIONS: Commonly employed methods identify different cancer cell sub-populations with no consistent therapeutic implications, rather than a single population of cells. The relationships of breast cancer stem cells to clinical parameters will require identification of specific markers or panels for the individual cancer
Prepubertal Children With Metabolically Healthy Obesity or Overweight Are More Active Than Their Metabolically Unhealthy Peers Irrespective of Weight Status: GENOBOX Study
Background and Aim: The association of a metabolically healthy status with the practice of physical activity (PA) remains unclear. Sedentarism and low PA have been linked to increased cardiometabolic risk. The aim of this study was to evaluate the PA levels in metabolically healthy (MH) or unhealthy (MU) prepubertal children with or without overweight/obesity. Methods: A total 275 children (144 boys) with 9 ± 2 years old were selected for the GENOBOX study. PA times and intensities were evaluated by accelerometry, and anthropometry, blood pressure, and blood biochemical markers were analyzed. Children were considered to have normal weight or obesity, and further classified as MH or MU upon fulfillment of the considered metabolic criteria. Results: Classification resulted in 119 MH children (21% with overweight/obesity, referred to as MHO) and 156 MU children (47% with overweight/obesity, referred to as MUO). Regarding metabolic profile, MHO showed lower blood pressure levels, both systolic and diastolic and biochemical markers levels, such as glucose, Homeostatic Model Assessment of Insulin Resistance, triglycerides and higher HDL-c levels than MUO (P < 0.001). In addition, MHO children spent more time in PA of moderate intensity compared with MUO children. In relation to vigorous PA, MH normal weight (MHN) children showed higher levels than MUO children. Considering sex, boys spent more time engaged in moderate, vigorous, and moderateâvigorous (MV) PA than girls, and the number of boys in the MH group was also higher. Conclusion: Prepubertal MHO children are less sedentary, more active, and have better metabolic profiles than their MUO peers. However, all children, especially girls, should increase their PA engagement, both in terms of time and intensity because PA appears to be beneficial for metabolic health status itself. Does fish larval dispersal differ between high and low latitudes?

connectivity of marine animals differs with latitude. We examine this expectation for
demersal shorefishes, including relevant mechanisms, assumptions, and evidence.
We explore latitudinal differences in: 1) biological (e.g., species composition,
spawning mode, pelagic larval duration (PLD)), 2) physical (e.g., water movement,
habitat fragmentation), and 3) biophysical factors (primarily temperature, which could
strongly affect development, swimming ability, or feeding). Latitudinal differences
exist in taxonomic composition, habitat fragmentation, temperature, and larval
swimming, and each could influence larval dispersal. Nevertheless, clear evidence
for latitudinal differences in larval dispersal at the level of broad faunas is lacking.
For example, PLD is strongly influenced by taxon, habitat, and geographic region,
but no independent latitudinal trend is present in published PLD values. Any trends
in larval dispersal may be obscured by a lack of appropriate information, or use of
âoff the shelfâ information that is biased with regard to the species assemblages in
areas of concern. Biases may also be introduced from latitudinal differences in taxa
or spawning modes, as well as limited latitudinal sampling. We suggest research to
