    Neurofilaments form flexible bundles during neuritogenesis in culture and in mature axons in situ

    Neurofilaments (NFs) undergo cation-dependent phospho-mediated associations with each other and other cytoskeletal elements that support axonal outgrowth. Progressive NF-NF associations generate a resident, bundled population that undergoes exchange with transporting NFs. We examined the properties of bundled NFs. Bundles did not always display a fully linear profile but curved and twisted at various points along the neurite length. Bundles retracted faster than neurites and retracted bundles did not expand following extraction with Triton, indicating that they coiled passively rather than due to pressure from the cell. Bundles consisted of helically wound NFs, which may provide flexibility necessary for turning of growing axons during pathfinding. Interactions between NFs and other cytoskeletal elements may be disrupted en masse during neurite retraction or regionally during remodeling. It is suggested that bundles within long axons that cannot be fully retracted into the soma could provide maintain proximal support yet still allow more distal flexibility for remodeling and changing direction during pathfinding

    Eddy Impacts on the Florida Current

    The Gulf Stream in the Atlantic carries warm water northwards and forms both the return closure of the subtropical gyre as well as the upper limb of the meridional overturning circulation. Recent time series recorded east of the Bahamas at 26°N indicate that from May 2009 to April 2011, in contrast with past observations, the northward flowing Antilles Current covaried with the Gulf Stream in the Florida Straits—the Florida Current—even though the Florida and Antilles Currents are separated by banks and islands spanning 150?km. The peak-to-trough amplitude of transport variations during this period was 15?×?106?m3?s?1 for the Florida Current and 12?×?106?m3?s?1 for the Antilles Current, at time scales of 50?days to a year. From satellite observations, we show that the fluctuations in both the Florida and Antilles Currents between May 2009 and April 2011 are driven by eddy activity east of the Bahamas. Since the Florida Current time series is a critical time series for the state of the oceans, and often compared to climate models, this newly identified source of variability needs careful consideration when attributing the variability of the Florida Current to changes in the larger-scale circulations (e.g., gyre and overturning) or wind forcing.<br/

    Equivalence of operator-splitting schemes for the integration of the Langevin equation

    We investigate the equivalence of different operator-splitting schemes for the integration of the Langevin equation. We consider a specific problem, so called the directed percolation process, which can be extended to a wider class of problems. We first give a compact mathematical description of the operator-splitting method and introduce two typical splitting schemes that will be useful in numerical studies. We show that the two schemes are essentially equivalent through the map that turns out to be an automorphism. An associated equivalent class of operator-splitting integrations is also defined by generalizing the specified equivalence.Comment: 4 page

    Multiplex giant magnetoresistive biosensor microarrays identify interferon-associated autoantibodies in systemic lupus erythematosus.

    High titer, class-switched autoantibodies are a hallmark of systemic lupus erythematosus (SLE). Dysregulation of the interferon (IFN) pathway is observed in individuals with active SLE, although the association of specific autoantibodies with chemokine score, a combined measurement of three IFN-regulated chemokines, is not known. To identify autoantibodies associated with chemokine score, we developed giant magnetoresistive (GMR) biosensor microarrays, which allow the parallel measurement of multiple serum antibodies to autoantigens and peptides. We used the microarrays to analyze serum samples from SLE patients and found individuals with high chemokine scores had significantly greater reactivity to 13 autoantigens than individuals with low chemokine scores. Our findings demonstrate that multiple autoantibodies, including antibodies to U1-70K and modified histone H2B tails, are associated with IFN dysregulation in SLE. Further, they show the microarrays are capable of identifying autoantibodies associated with relevant clinical manifestations of SLE, with potential for use as biomarkers in clinical practice

    Winter Wheat Quality Responses to Water, Environment, and Nitrogen Fertilization

    Decreasing carbon (C) footprints by reducing nitrogen (N) and water inputs has been speculated to have negative impacts on wheat grain yield and flour processing quality. The objective of this study was to determine the impact of N and water stress on winter wheat grain yield, protein composition, and dough quality. Wheat fertilized at two N rates (unfertilized and recommended) was grown under water-stressed and well-watered environments. Nitrogen and water stress were measured using the 13C isotopic approach. Research showed that (1) N fertilizer and the water-management environment produced similar impacts on wheat quality and yield loss due to N stress and yield loss due to water stress (YLWS); (2) N fertilizer increased flour protein, dough stability, and relative concentration of glutenin (%Glu), unextractable polymeric protein (UPP), and relative amount of high-molecular-weight glutenin subunits (HMW-GS/LMW-GS); (3) the well-watered environment reduced protein contents when N mineralization was low, whereas it did not influence protein content when mineralization was high; and (4) the %Glu was negatively correlated with yield loss due to N stress (YLNS) and positively correlated with stability. This study showed that a clear understanding of the complex relationship between soil variability and climatic conditions should make it possible to develop adaptive management practices, increase profitability, and improve quality

    R-parity violation in superstring derived models

    The ALEPH collaboration has recently reported a significant excess of four-jet events at the LEP 1.5 and LEP 2. While not yet confirmed by the other collaborations, it was recently proposed that this excess may be explained by certain R-parity violating operators in supersymmetric models. R-parity violating operators introduce the danger of inducing rapid proton decay. I discuss how the operators required to explain the ALEPH four jet events may arise from superstring derived models without inducing rapid proton decay.Comment: 11 pages. Standard Latex.     Anomalies in field theories with extra dimensions

    We give an overview of the issue of anomalies in field theories with extra dimensions. We start by reviewing in a pedagogical way the computation of the standard perturbative gauge and gravitational anomalies on non-compact spaces, using Fujikawa's approach and functional integral methods, and discuss the available mechanisms for their cancellation. We then generalize these analyses to the case of orbifold field theories with compact internal dimensions, emphasizing the new aspects related to the presence of orbifold singularities and discrete Wilson lines, and the new cancellation mechanisms that are becoming available.
