    International audienceInterleukin-1β (IL-1β), a key-cytokine in osteoarthritis, impairs TGFβ signaling through TβRII down-regulation by increasing its degradation. Here, we investigated the molecular mechanism that controls TßRII fate in IL-1ß treated cells. Chondrocytes were treated with IL-1ß in the presence of different inhibitors. TßRII and Cav-1 expression were assayed by Western blot and RT-PCR. We showed that IL-1ß-induced degradation of TßRII is dependent on proteasome and on its internalization in caveolae. In addition, IL-1ß enhances Cav-1 expression, a major constituent of lipid raft. In conclusion, we enlighten a new mechanism by which IL-1ß antagonizes TGFß pathway and propose a model of TßRII turnover regulation upon IL-1ß treatment

    Removal of the adhesive gum layer surrounding naturally fertilised ballan wrasse (Labrus bergylta) eggs

    Commercial production of ballan wrasse (Labrus bergylta) as a cleaner fish for the removal of sea lice (Lepeophtheirus salmonis) from farmed salmonids (Salmo salar) has increased due to its proven efficiency. One bottleneck in commercial hatchery production is working with the benthic adhesive eggs, which makes disinfection and incubation of eggs challenging; therefore, this study aimed to find a chemical or enzymatic treatment and process to remove the adhesive gum layer. Naturally spawned eggs were collected from artificial spawning substrates up to 24h post spawning from wild caught broodstock kept in captivity at the Marine Harvest, Machrihanish facility. Four treatments were tested: tannic acid (0.2, 0.1, and 0.05%), sodium sulfite (2, 1, and 0.5%),l-cysteine (2, and 1%), and enzyme alcalase® (4.0, 3.0, 2.0, 1.0, and 0.5%)in vitro. Eggs were exposed for 25min while being continually agitated, and the proportion of “degummed” eggs was counted at the end of each time period. Enzyme alcalase® was the only treatment that proved successful in degumming eggs, with the time to complete degumming (≥96%) inversely related to enzyme concentration. Complete degumming occurred between 15 and 30min for all enzyme alcalase® dose rates. Mean hatch rates for eggs treated with enzyme alcalase ® were not compromised by the treatment and in the highest dose tested were actually found to be higher in treated eggs (78.9±2.4%) than controls (71.3±3.3%). The use of enzyme alcalase® has proven effective in degumming ballan wrasse eggs without affecting hatch rates. However, translation of this method toin situdegumming and thus removal of eggs from spawning substrate on farm remains to be standardised

    Unravelling travellers' route choice behaviour at full-scale urban network by focusing on representative OD pairs in computer experiments

    In a city-scale network, trips are made in thousands of origin-destination (OD) pairs connected by multiple routes, resulting in a large number of alternatives with diverse characteristics that influence the route choice behaviour of the travellers. As a consequence, to accurately predict user choices at full network scale, a route choice model should be scalable to suit all possible configurations that may be encountered. In this article, a new methodology to obtain such a model is proposed. The main idea is to use clustering analysis to obtain a small set of representative OD pairs and routes that can be investigated in detail through computer route choice experiments to collect observations on travellers behaviour. The results are then scaled-up to all other OD pairs in the network. It was found that 9 OD pair configurations are sufficient to represent the network of Lyon, France, composed of 96,096 OD pairs and 559,423 routes. The observations, collected over these nine representative OD pair configurations, were used to estimate three mixed logit models. The predictive accuracy of the three models was tested against the predictive accuracy of the same models (with the same specification), but estimated over randomly selected OD pair configurations. The obtained results show that the models estimated with the representative OD pairs are superior in predictive accuracy, thus suggesting the scaling-up to the entire network of the choices of the participants over the representative OD pair configurations, and validating the methodology in this study

    Salirasib inhibits the growth of hepatocarcinoma cell lines in vitro and tumor growth in vivo through ras and mTOR inhibition

    <p>Abstract</p> <p>Background</p> <p>Dysregulation of epidermal growth factor and insulin-like growth factor signaling play important roles in human hepatocellular carcinoma (HCC), leading to frequent activation of their downstream targets, the ras/raf/extracellular signal-regulated kinase (ERK) and the phosphoinositide 3-kinase (PI3K)/Akt/mammalian Target of Rapamycin (mTOR) pathways. Salirasib is an S-prenyl-cysteine analog that has been shown to block ras and/or mTOR activation in several non hepatic tumor cell lines. We investigated <it>in vitro </it>the effect of salirasib on cell growth as well as its mechanism of action in human hepatoma cell lines (HepG2, Huh7, and Hep3B) and its <it>in vivo </it>effect in a subcutaneous xenograft model with HepG2 cells.</p> <p>Results</p> <p>Salirasib induced a time and dose dependent growth inhibition in hepatocarcinoma cells through inhibition of proliferation and partially through induction of apoptosis. A 50 percent reduction in cell growth was obtained in all three cell lines at a dose of 150 μM when they were cultured with serum. By contrast, salirasib was more potent at reducing cell growth after stimulation with EGF or IGF2 under serum-free conditions, with an IC<sub>50 </sub>ranging from 60 μM to 85 μM. The drug-induced anti-proliferative effect was associated with downregulation of cyclin A and to a lesser extent of cyclin D1, and upregulation of p21 and p27. Apoptosis induction was related to a global pro-apoptotic balance with caspase 3 activation, cytochrome c release, death receptor upregulation, and a reduced mRNA expression of the apoptosis inhibitors cFLIP and survivin. These effects were associated with ras downregulation and mTOR inhibition, without reduction of ERK and Akt activation. <it>In vivo</it>, salirasib reduced tumour growth from day 5 onwards. After 12 days of treatment, mean tumor weight was diminished by 56 percent in the treated animals.</p> <p>Conclusions</p> <p>Our results show for the first time that salirasib inhibits the growth of human hepatoma cell lines through inhibition of proliferation and induction of apoptosis, which is associated with ras and mTOR inhibition. The therapeutic potential of salirasib in human HCC was further confirmed in a subcutaneous xenograft model.</p

    Strong obstruction of the Berends-Burgers-van Dam spin-3 vertex

    In the eighties, Berends, Burgers and van Dam (BBvD) found a nonabelian cubic vertex for self-interacting massless fields of spin three in flat spacetime. However, they also found that this deformation is inconsistent at higher order for any multiplet of spin-three fields. For arbitrary symmetric gauge fields, we severely constrain the possible nonabelian deformations of the gauge algebra and, using these results, prove that the BBvD obstruction cannot be cured by any means, even by introducing fields of spin higher (or lower) than three.Comment: 19 pages, no figur

    Seasonal changes in broodstock spawning performance and egg quality in ballan wrasse (Labrus bergylta)

    Sea lice continue to be one of the largest issues for the salmon farming industry and the use of ballan wrasse (Labrus bergylta) as a biological control is considered to be one of the most sustainable solutions in development. Broodstock management has proved challenging in the initial phases due to the significant lack of understanding of basic reproductive physiology and behaviour in the species. The aim of the study was to monitor captive breeding populations throughout a spawning season to examine timing and duration of spawning, quantify egg production, and look at seasonal changes in egg quality parameters as well as investigate the parental contribution to spawning events. A clear spawning rhythm was shown with 3&ndash;5 spawning periods inclusive of spawning windows lasting 1&ndash;9days followed by interspawning intervals of 8&ndash;12days. Fertilization rate remained consistently high (&gt;87.5%) over the spawning season and did not differ significantly between spawning populations. Hatch rate was variable (0&ndash;97.5%), but peaked in the middle of the spawning season. Mean oocyte diameter and gum layer thickness decreased slightly over the spawning season with no significant differences between spawning populations. Fatty acid (FA) profile of eggs remained consistent throughout the season and with the exception of high levels of ARA (3.8&plusmn;0.5% of total FA) the FA profile was similar to that observed in other marine fish species. Parental contribution analysis showed 3 out of 6 spawning events to be single paired mating while the remaining 3 had contributions from multiple parents. Furthermore, the proposed multiple batch spawning nature of this species was confirmed with proof of a single female contributing to two separate spawning events. Overall this work represents the first comprehensive dataset of spawning activity of captive ballan wrasse, and as such and will be helpful in formulating sustainable broodstock management plans for the species

    Consistent couplings between spin-2 and spin-3 massless fields

    We solve the problem of constructing consistent first-order cross-interactions between spin-2 and spin-3 massless fields in flat spacetime of arbitrary dimension n > 3 and in such a way that the deformed gauge algebra is non-Abelian. No assumptions are made on the number of derivatives involved in the Lagrangian, except that it should be finite.     AsrR Is an Oxidative Stress Sensing Regulator Modulating Enterococcus faecium Opportunistic Traits, Antimicrobial Resistance, and Pathogenicity

    Oxidative stress serves as an important host/environmental signal that triggers a wide range of responses in microorganisms. Here, we identified an oxidative stress sensor and response regulator in the important multidrug-resistant nosocomial pathogen Enterococcus faecium belonging to the MarR family and called AsrR (antibiotic and stress response regulator). The AsrR regulator used cysteine oxidation to sense the hydrogen peroxide which results in its dissociation to promoter DNA. Transcriptome analysis showed that the AsrR regulon was composed of 181 genes, including representing functionally diverse groups involved in pathogenesis, antibiotic and antimicrobial peptide resistance, oxidative stress, and adaptive responses. Consistent with the upregulated expression of the pbp5 gene, encoding a low-affinity penicillin-binding protein, the asrR null mutant was found to be more resistant to \u3b2-lactam antibiotics. Deletion of asrR markedly decreased the bactericidal activity of ampicillin and vancomycin, which are both commonly used to treat infections due to enterococci, and also led to over-expression of two major adhesins, acm and ecbA, which resulted in enhanced in vitro adhesion to human intestinal cells. Additional pathogenic traits were also reinforced in the asrR null mutant including greater capacity than the parental strain to form biofilm in vitro and greater persistance in Galleria mellonella colonization and mouse systemic infection models. Despite overexpression of oxidative stress-response genes, deletion of asrR was associated with a decreased oxidative stress resistance in vitro, which correlated with a reduced resistance to phagocytic killing by murine macrophages. Interestingly, both strains showed similar amounts of intracellular reactive oxygen species. Finally, we observed a mutator phenotype and enhanced DNA transfer frequencies in the asrR deleted strain. These data indicate that AsrR plays a major role in antimicrobial resistance and adaptation for survival within the host, thereby contributes importantly to the opportunistic traits of E. faecium
