    Chromium Speciation in Wastewater and Sewage by Solid-Phase Extraction Using a New Diphenylcarbazone-Incorporated Resin

    A new procedure for the determination of chromium species in polluted environmental samples by flame atomic absorption spectrometry was developed in this work. A new material containing 1,5-diphenylcarbazone included in a polymeric matrix was prepared and employed as a solid-phase extraction material for selective separation of Cr(III) ions under dynamic conditions. Chromium(III) ions were retained on this sorbent with high efficiency and repeatability (95 %, RSD = 1 %) from solutions with pH 9.0. The quantitative recovery of analyte was obtained with 0.1 mol L−1 EDTA. The concentration of Cr(VI) ions was calculated from the difference between the concentration of total chromium and Cr(III) ions. The prepared sorbent exhibits good chemical and mechanical stability, sorption capacity and selectivity towards Cr(III) ions in the presence of Cu(II), Ni(II), Mn(II) and Ca(II) ions. The accuracy of the separation method was proved by analysis of reference material of wastewater RES 10.2.     Preferred analysis methods for single genomic regions in RNA sequencing revealed by processing the shape of coverage

    The informational content of RNA sequencing is currently far from being completely explored. Most of the analyses focus on processing tables of counts or finding isoform deconvolution via exon junctions. This article presents a comparison of several techniques that can be used to estimate differential expression of exons or small genomic regions of expression, based on their coverage function shapes. The problem is defined as finding the differentially expressed exons between two samples using local expression profile normalization and statistical measures to spot the differences between two profile shapes. Initial experiments have been done using synthetic data, and real data modified with synthetically created differential patterns. Then, 160 pipelines (5 types of generator × 4 normalizations × 8 difference measures) are compared. As a result, the best analysis pipelines are selected based on linearity of the differential expression estimation and the area under the ROC curve. These platform-independent techniques have been implemented in the Bioconductor package rnaSeqMap. They point out the exons with differential expression or internal splicing, even if the counts of reads may not show this. The areas of application include significant difference searches, splicing identification algorithms and finding suitable regions for QPCR primer

    Analiza profilu demograficznego pacjentów leczonych z powodu niepłodności metodami rozrodu wspomaganego w latach 2005-2010

    Objectives: analysis of the demographic profile of patients, causes for infertility and effectiveness of infertility treatment methods in the years 2005-2010. Material and methods: Retrospective research was conducted to analyze data of 1705 randomly selected couples who underwent in vitro fertilization procedure at the Department of Reproduction and Gynecological Endocrinology, Medical University of Bialystok, between 2005 and 2010. The analyzed data included mainly causes for infertility, age of the female and male subjects, place of residence and final treatment results. Results: The percentage of pregnancy rate increased significantly to approximately 40% in 2007. The contribution of male and female infertility factors remained at a similar level, but the idiopathic factor continued to steadily increase (to 20% in the last years of the study). We observed a greater prevalence of the male factor among couples living in cities compared to inhabitants of rural areas (42.3% vs. 34.3%, p=0.004), whereas the tubal factor dominated among couples living in the countryside when compared to city dwellers (29.7% vs. 21.6%, p=0.001). The average age of women entering treatment was significantly higher in cities than the countryside (pCel pracy: Analiza profilu demograficznego pacjentów, przyczyn niepłodności oraz skuteczności leczenia metodami rozrodu wspomaganego w latach 2005-2010. Materiał i metody: W badaniu retrospektywnym przeanalizowano dane dotyczące 1705 losowo wybranych cykli leczenia niepłodności metodami rozrodu wspomaganego, przeprowadzonych w latach 2005-2010 w Klinice Rozrodczości i Endokrynologii Ginekologicznej Uniwersytetu Medycznego w Białymstoku. Wyniki: Odsetek uzyskanych ciąż wyraźnie wzrósł w 2007 roku do poziomu około 40%. Udział męskich i żeńskich czynników niepłodności kształtuje się na porównywalnym poziomie, stale wzrasta udział czynnika niewyjaśnionego pochodzenia (do około 20% w ostatnich latach badania). Wśród par z miasta stwierdza się większy niż na wsi odsetek występowania czynnika męskiego (42,3% vs. 34,3%, p=0,004), natomiast u par ze wsi – większy niż w miastach udział czynnika jajowodowego (29,7% vs. 21,6%, p=0,001).     Successes and pitfalls of chronic peritoneal dialysis in infants – a Polish nationwide outcome study

    Introduction: Peritoneal dialysis (PD) is a preferred method of renal replacement therapy for end-stage renal disease in children. Recent advances have allowed chronic PD to be provided to children of all ages and sizes. Material and methods: The study was designed as a national (10 dialysis centres), multicentre retrospective analysis of the medical history of 33 children who started chronic peritoneal dialysis in their infancy between 1993 and 2005, with a follow-up period of at least 24 months. Results: The nutritional status of the infants was unsatisfactory. The mean SDS of body weight at the start was -2.0, at 1 year of age -1.7. Only 40% of infants were adequately nourished at 1 year of age. Long-term follow-up analysis showed that 12 children received a kidney transplant, 13 were still on dialysis (4 changed method) and 6 died (mortality rate in the first year of life of 9%). In 2 children we observed an improvement of renal function. We observed a relatively high (1/8.8 patient-months) peritonitis rate in the analysed children when compared to 1 : 22 patient-months in all children undergoing PD in Poland.     W niniejszej publikacji przedstawiono zasady nowego modelu wspomagania pracy szkół, które od stycznia 2016 roku będą obowiązywać placówki doskonalenia nauczycieli, poradnie psychologiczno-pedagogiczne oraz biblioteki pedagogiczne.Celem tego opracowania jest pokazanie roli szkoły – dyrektora i nauczycieli – oraz zadań stojących przed instytucjami wspierającymi szkołę w pracy. W opracowaniu publikacji uczestniczył zespół pracowników ośrodków doskonalenia nauczycieli, poradni psychologiczno-pedagogicznych oraz bibliotek pedagogicznych, który wykorzystał różnorodne źródła – opracowania teoretyczne, wyniki badań, opisy dobrych praktyk, a przede wszystkim własne doświadczenia we współpracy ze szkołami uczestniczącymi w pilotażu nowego systemu wspomagania

    rnaSeqMap: a Bioconductor package for RNA sequencing data exploration

    BACKGROUND: The throughput of commercially available sequencers has recently significantly increased. It has reached the point where measuring the RNA expression by the depth of coverage has become feasible even for largest genomes. The development of software tools is constantly following the progress of biological hardware. In particular, as RNA sequencing software can be regarded genome browsers, exon junction tools and statistical tools operating on counts of reads in predefined regions. The library rnaSeqMap, freely available via Bioconductor, is an RNA sequencing software which is independent of any biological hardware platform. It is based upon standard Bioconductor infrastructure for sequencing data and includes several novel features focused on deeper understanding of coverage expression profiles and discovery of novel transcription regions. RESULTS: rnaSeqMap is a toolbox for analyses that may be performed with the use of gene annotations or alternatively, in an unsupervised mode, on any genomic region to find novel or non-standard transcripts. The data back-end may be a MySQL database or a set of files in standard BAM format. The processing in R can be run on a machine without any particular hardware requirements, and scales linearly with the number of genomic loci and number of samples analyzed. The main features of rnaSeqMap include coverage operations, discovering irreducible regions of high expression, significance search and splicing analyses with nucleotide granularity. CONCLUSIONS: This software may be used for a range of applications related to RNA sequencing by building customized analysis pipelines. The applicability and precision is expected to increase in parallel with the progress of the genome coverage in sequencers

    Zorian Dołęga Chodakowski (born as Adam Czarnocki, 1784–1825) is mainly known as a collector of Slavic antiquities and the author of O Sławiańszczyźnie przed chrześcijaństwem [On the Slavic Lands before Christianity], a work marking the need to study the peasants in order to extract from their culture the relics of ancient ‘Slavicness’. The first descriptions of the researcher clearly emphasized the aspect of his frequent personal and scientific travels. This wandering way of living was interpreted as an experience constituting a unique personality type of a rebellious romantic, which significantly influenced the further reception of Chodakowski’s work. Through the analysis of the construction of these biographies and selected texts of the researcher, the author presents the elements that may have contributed to the creation of the image of Chodakowski as a romantic wanderer.Zorian Dołęga Chodakowski (wcześniej: Adam Czarnocki, 1784–1825) znany jest polskiej nauce jako zbieracz słowiańskich starożytności i autor O Sławiańszczyźnie przed chrześcijaństwem, tekstu zaznaczającego potrzebę pochylenia się nad warstwą chłopską w celu wydobycia z jej wytworów pozostałości dawnej „sławiańskości”.
