    Testing convex hypotheses on the mean of a Gaussian vector. Application to testing qualitative hypotheses on a regression function

    In this paper we propose a general methodology, based on multiple testing, for testing that the mean of a Gaussian vector in R^n belongs to a convex set. We show that the test achieves its nominal level, and characterize a class of vectors over which the tests achieve a prescribed power. In the functional regression model this general methodology is applied to test some qualitative hypotheses on the regression function. For example, we test that the regression function is positive, increasing, convex, or more generally, satisfies a differential inequality. Uniform separation rates over classes of smooth functions are established and a comparison with other results in the literature is provided.     Shortly after Caliph ‘Umar ibn al-Khattab’s (579-644, caliph 634-644) arrival in Jerusalem in 638, he is said to have constructed a rudimentary mosque or prayer space south of the historical Rock now contained within the Dome of the Rock (completed 691) on the former Temple Mount or Bayt al-Maqdis known popularly since Mamluk and Ottoman times as the Haram al-Sharif. Though later textual evidence indicates that ‘Umar prayed somewhere south of the “rock” and later scholars suggest that he constructed a rudimentary prayer space on the site, there is no surviving physical evidence of that initial structure. After his appointment as Governor of Syria (bilad al-sham) by ‘Umar in 639/40, Mu‘awiya ibn Abi Sufyan (602-680, caliph 660-680) either expanded upon the Mosque of ‘Umar or constructed an entirely new mosque in Jerusalem between 640 and 660. This mosque was completed in time for his investiture in that mosque in 660 as the first Umayyad caliph. This article proposes that the seventh-century mosque of Jerusalem constructed between 640-660 has survived to today and that the official entrance to that mosque and the Bayt al-Maqdis precinct was the centrally placed eastern gate. Further, it will be demonstrated that there were four entrances to this mosque, one ceremonial and public from the east, leading to a northern entrance to the prayer space, a second public entrance and a third private entrance, both from south of the city. The physical evidence from the site itself, the newly established presence of Islamic rule in Jerusalem, and the persistence of religious memory of the earlier significance of the site contributed to the location of this mosque in the southeastern quadrant of the sanctuary. The building is variously known historically as masjid qadim, aqsa, Solomon’s Stables and is today the Marwani Musalla (prayer space)

    The Imperial Museum of Antiquities in Jerusalem, 1890-1930: An Alternative Narrative

    The creation of the first Museum of Antiquities in Jerusalem during the late Ottoman period is a fascinating story of archaeological pursuits in the region by both Ottoman government officialdom in Istanbul and foreign archaeologists working in Palestine for the British Palestine Exploration Fund. The Ottoman Museum called the Muze-I Humayun in Turkish or Imperial Museum (1901-1917) and its collection is continuous with the British Palestine Museum of Antiquities (1921-1930) and the Palestine Archaeological Museum. The construction of the last began in 1930 and was completed in 1935, but the museum, now known as the Rockefeller Museum, did not open until 1938. Between 1922 and 1935 the British encouraged the creation of a museum for Islamic Art (1922) and one for Jewish Art. The history of the museum from the Ottoman Period through the British Mandate Period (1917-1948) is well documented in sources located in the Ottoman National Archives in Istanbul, the Mandate Period Archives of the Department of Antiquities housed in the Rockefeller Museum in Jerusalem, the Palestine Exploration Fund (PEF) archives in London, and the Sijillat al-Mahkama al-Shar’iya (Religious Court Registers) of Jerusale

    A 1-year case-control study in patients with rheumatoid arthritis indicates prevention of loss of bone mineral density in both responders and nonresponders to infliximab

    The goal of the present study was to record changes in bone mineral density (BMD) and markers of bone turnover in patients with rheumatoid arthritis (RA) who were treated with methotrexate combined (or not combined) with infliximab. Included were 90 patients with RA who required anti-TNF-α therapy with infliximab because of persistent active disease despite treatment with methotrexate. The historical control group included 99 patients with RA who were treated with methotrexate at a time when anti-TNF-α treatment was not yet available. Lumbar and femoral neck BMD was measured using dual energy X-ray absorptiometry at baseline and 1 year later. Osteocalcin, C-terminal cross-linked telopeptide of type I collagen, parathyroid hormone and 25-hydroxycholecalciferol were measured in plasma at baseline and 1 year later. At 1 year BMD had decreased in the control group at spine (P < 0.01) and femoral neck (P < 0.001). In contrast, BMD at spine and femoral neck did not change after 1 year of infliximab treatment. At the same time point, no change in bone remodelling markers was observed. No association was observed between clinical response and changes in BMD, indicating that even those who did not respond clinically did not lose bone over a 1-year period. These data confirm the BMD decrease observed in RA patients treated with methotrexate alone. This bone loss was prevented by infliximab therapy. Importantly, this beneficial effect was also observed in apparent nonresponders

    The mode of action of nuclear receptors in living cells is an actively investigated field but much remains hypothetical due to the lack, until recently, of methods allowing the assessment of molecular mechanisms in vivo. However, these last years, the development of fluorescence microscopy methods has allowed initiating the dissection of the molecular mechanisms underlying gene regulation by nuclear receptors directly in living cells or organisms. Following our analyses on peroxisome proliferator activated receptors (PPARs) in living cells, we discuss here the different models arising from the use of these tools, that attempt to link mobility, DNA binding or chromatin interaction, and transcriptional activity

    A murine herpesvirus closely related to ubiquitous human herpesviruses causes T-cell depletion

    ABSTRACT The human roseoloviruses human herpesvirus 6A (HHV-6A), HHV-6B, and HHV-7 comprise the Roseolovirus genus of the human Betaherpesvirinae subfamily. Infections with these viruses have been implicated in many diseases; however, it has been challenging to establish infections with roseoloviruses as direct drivers of pathology, because they are nearly ubiquitous and display species-specific tropism. Furthermore, controlled study of infection has been hampered by the lack of experimental models, and until now, a mouse roseolovirus has not been identified. Herein we describe a virus that causes severe thymic necrosis in neonatal mice, characterized by a loss of CD4 + T cells. These phenotypes resemble those caused by the previously described mouse thymic virus (MTV), a putative herpesvirus that has not been molecularly characterized. By next-generation sequencing of infected tissue homogenates, we assembled a contiguous 174-kb genome sequence containing 128 unique predicted open reading frames (ORFs), many of which were most closely related to herpesvirus genes. Moreover, the structure of the virus genome and phylogenetic analysis of multiple genes strongly suggested that this virus is a betaherpesvirus more closely related to the roseoloviruses, HHV-6A, HHV-6B, and HHV-7, than to another murine betaherpesvirus, mouse cytomegalovirus (MCMV). As such, we have named this virus murine roseolovirus (MRV) because these data strongly suggest that MRV is a mouse homolog of HHV-6A, HHV-6B, and HHV-7. IMPORTANCE Herein we describe the complete genome sequence of a novel murine herpesvirus. By sequence and phylogenetic analyses, we show that it is a betaherpesvirus most closely related to the roseoloviruses, human herpesviruses 6A, 6B, and 7. These data combined with physiological similarities with human roseoloviruses collectively suggest that this virus is a murine roseolovirus (MRV), the first definitively described rodent roseolovirus, to our knowledge. Many biological and clinical ramifications of roseolovirus infection in humans have been hypothesized, but studies showing definitive causative relationships between infection and disease susceptibility are lacking. Here we show that MRV infects the thymus and causes T-cell depletion, suggesting that other roseoloviruses may have similar properties. </jats:p
