    Relação entre o estado nutricional de ferro de parturientes e seus recém-nascidos

    OBJECTIVE: To determine the relationship between iron nutritional status of pregnant women and their newborns using a combination of hematological and biochemical parameters for the diagnosis of iron deficiency. METHODS: A cross-sectional study was conducted in Jundiaí, Southeastern Brazil, in 2000. Venous blood samples collected from 95 pregnant women and from their umbilical cord and used for the determination of complete blood count, serum iron, total iron-binding capacity, serum ferritin, zinc protoporphyrin, and transferrin saturation. Women were classified into three groups: anemic, iron deficient and non-iron deficient. Statistical analysis included the Tukey-HSD test, Pearson's correlation coefficient and multiple linear regression analysis. RESULTS: Among pregnant women, 19% were anemic (97.9% mildly anemic and 2.1% moderately anemic) and 30.5% were iron deficient. No significant difference was seen in mean values of any parameter studied between newborns in the three groups (p>;0.05). Multiple linear regression analysis showed weak association between neonatal and maternal parameters. CONCLUSIONS: The iron nutritional status of pregnant women with iron deficiency or mild anemia does not seem to have a significant impact on the iron levels of their children.OBJETIVO: Determinar a relação entre os níveis de ferro de gestantes e seus filhos recém-nascidos, utilizando uma combinação de parâmetros hematológicos e bioquímicos para o diagnóstico da deficiência de ferro. MÉTODOS: Estudo transversal realizado em Jundiaí, SP, em 2000. Amostras de sangue venoso foram coletadas de 95 gestantes e do cordão umbilical de cada uma, e utilizadas na determinação de hemograma completo, ferro sérico, capacidade total de ligação do ferro, ferritina sérica, zinco-protoporfirina, e saturação de transferrina. As mulheres foram classificadas em três grupos: anêmicas, deficientes de ferro e não-deficientes de ferro. As análises estatísticas utilizadas foram o teste de Tukey-HSD, o coeficiente de correlação de Pearson e regressão linear múltipla. RESULTADOS: Entre as gestantes, 19% estavam anêmicas (97,9% levemente anêmicas e 2,1% moderadamente anêmicas) e 30,5% apresentavam deficiência de ferro. Não foi observada diferença significativa nas médias dos valores dos parâmetros estudados nos recém-nascidos dos três grupos (p>;0,05). A análise de regressão linear múltipla mostrou fraca associação entre os parâmetros maternos e neonatais. CONCLUSÕES: Os níveis de ferro de gestantes com deficiência de ferro ou com anemia leve/moderada parecem não influenciar de forma significativa os níveis de ferro de seus filhos

    Effects of Field Pea Diet and Immunocastration in Heavy Pigs on Fresh Pork and Dry-Cured Ham

    Peas are an alternative to soybeans to supply protein in livestock feeds. Immunocastration avoids surgical castration of male pigs and increases fat deposition in female pigs. This work aimed to assess the effects of pea inclusion on the amount of fat and fatty acid profile of loins and on weight losses of dry-cured hams in different sexes of pigs; in addition, growth performance and feeding behavior in immunocastrated female pigs were evaluated. Two experiments were conducted with crossbred immunocastrated female (IF) and immunocastrated (IM) or surgically castrated (CM) male pigs from Duroc dams sired by Berkshire, which were assigned to one of two diets (soybean vs. pea-based). The effect of castration type on the afore-mentioned variables was evaluated in male pigs, and the growth performance, and feeding behavior in female pigs. The pea-based diet in IF had no effect on average daily gain nor on carcass traits; although, it increased feeding time (p < 0.001) and reduced n-3 polyunsaturated fatty acid (PUFA) content (p < 0.05). In male pigs, the pea-based diet did not change carcass fatness either but reduced the n-3 PUFA levels (p < 0.05). Likewise, IM had lower (p < 0.001) monounsaturated fatty acid (MUFA) and higher (p < 0.05) n-6 PUFA than CM. Diet did not affect dry-cured ham weight losses during the process, while IM showed greater (p < 0.001) losses than CM. Pigs fed a pea-based diet complied with the requirements of cured ham production, while immunocastration in male pigs increased weight losses, partly explained by lower fat content and higher fatty acid unsaturation

    Structural Determinants for Functional Coupling Between the β and α Subunits in the Ca2+-activated K+ (BK) Channel

    High conductance, calcium- and voltage-activated potassium (BK, MaxiK) channels are widely expressed in mammals. In some tissues, the biophysical properties of BK channels are highly affected by coexpression of regulatory (β) subunits. The most remarkable effects of β1 and β2 subunits are an increase of the calcium sensitivity and the slow down of channel kinetics. However, the detailed characteristics of channels formed by α and β1 or β2 are dissimilar, the most remarkable difference being a reduction of the voltage sensitivity in the presence of β1 but not β2. Here we reveal the molecular regions in these β subunits that determine their differential functional coupling with the pore-forming α-subunit. We made chimeric constructs between β1 and β2 subunits, and BK channels formed by α and chimeric β subunits were expressed in Xenopus laevis oocytes. The electrophysiological characteristics of the resulting channels were determined using the patch clamp technique. Chimeric exchange of the different regions of the β1 and β2 subunits demonstrates that the NH3 and COOH termini are the most relevant regions in defining the behavior of either subunit. This strongly suggests that the intracellular domains are crucial for the fine tuning of the effects of these β subunits. Moreover, the intracellular domains of β1 are responsible for the reduction of the BK channel voltage dependence. This agrees with previous studies that suggested the intracellular regions of the α-subunit to be the target of the modulation by the β1-subunit

    The detection of minimal residual disease (MRD) is an important prognostic factor in childhood acute lymphoblastic leukemia (ALL) providing crucial information on the response to treatment and risk of relapse. However, the high cost of these techniques restricts their use in countries with limited resources. Thus, we prospectively studied the use of flow cytometry (FC) with a simplified 3-color assay and a limited antibody panel to detect MRD in the bone marrow (BM) and peripheral blood (PB) of children with ALL. BM and PB samples from 40 children with ALL were analyzed on days (d) 14 and 28 during induction and in weeks 24-30 of maintenance therapy. Detectable MRD was defined as > 0.01% cells expressing the aberrant immunophenotype as characterized at diagnosis among total events in the sample. A total of 87% of the patients had an aberrant immunophenotype at diagnosis. On d14, 56% of the BM and 43% of the PB samples had detectable MRD. On d28, this decreased to 45% and 31%, respectively. The percentage of cells with the aberrant phenotype was similar in both BM and PB in T-ALL but about 10 times higher in the BM of patients with B-cell-precursor ALL. Moreover, MRD was detected in the BM of patients in complete morphological remission (44% on d14 and 39% on d28). MRD was not significantly associated to gender, age, initial white blood cell count or cell lineage. This FC assay is feasible, affordable and readily applicable to detect MRD in centers with limited resources.A detecção de doença residual mínima (DRM) é um importante fator prognóstico na leucemia linfóide aguda (LLA) infantil e fornece informações sobre a resposta ao tratamento e o risco de recaída. Entretanto, os altos custos das técnicas utilizadas limitam seu uso nos países em desenvolvimento. Desta forma, realizamos um estudo prospectivo para avaliar a citometria de fluxo (CF), utilizando três fluorescências e um painel limitado de anticorpos monoclonais, como método de detecção de DRM em medula óssea (MO) e sangue periférico (SP) de crianças com LLA. Amostras de MO e SP de 40 crianças portadoras de LLA foram analisadas nos dias (d)14 e d28 da indução e nas semanas 24-30 da terapia de manutenção. Foram consideradas como DRM+ as amostras que apresentaram > 0,01% das células com o fenótipo aberrante (FA). Oitenta e sete por cento dos pacientes apresentaram FA ao diagnóstico. No d14, 56% das amostras de MO e 43% do SP apresentaram DRM. No d28, foi detectada DRM em 45% e 31% das amostras de MO e SP, respectivamente. A porcentagem de DRM na MO foi similar à do SP nos casos de LLA-T, mas aproximadamente dez vezes maior na LLA de precursor-B. Foi detectada DRM na MO de 44% e 39% dos pacientes que estavam remissão morfológica nos d14 e d28, respectivamente. Não foi demonstrada associação significante entre a presença de DRM e sexo, idade, leucometria inicial e linhagem celular. Esta técnica de detecção de DRM por CF é relativamente barata e pode ser aplicada em centros com recursos limitados

    Comparação entre o HemoCue® e um contador automático para determinação de hemoglobina

    Com o objetivo de avaliar a precisão e concordância na determinação de hemoglobina (Hb) em sangue capilar e venoso, utilizando-se o HemoCue® e um contador automático, foram estudadas 29 gestantes. O HemoCue® apresentou baixa repetibilidade para as medidas em duplicata em sangue capilar (CR=0,53 g/dL; CV=13,6%) e venoso (CR=0,53 g/dL; CV=13,6%). As medidas de Hb obtidas em sangue capilar foram mais altas do que as obtidas em sangue venoso (12,4 e 11,7 g/dL, respectivamente; pTo determine the precision and agreement of the hemoglobin (Hb) measurements in capillary and venous blood samples by the HemoCue® and an automated counter. Hb was determined by both equipaments in blood samples of 29 pregnant women. The HemoCue® showed low repeatability of Hb measurements in duplicate in capillary (CR=0.53 g/dL, CV=13.6%) and venous blood (CR=0.53 g/dL, CV=13.6%).     Knowledge and perceptions of food sustainability in a Spanish university population

    In 2015, the United Nations adopted the 2030 Agenda for Sustainable Development, with 17 Sustainable Development Goals (SDGs) at its core. Besides tackling climate change and the fight to reduce inequality, the SDG number 12 is specifically focused to develop strategies toward food sustainability. The aim of this study, aligned with SDG number 12, was to analyze the level of knowledge and perceptions of food sustainability in a university community from Spain. A descriptive cross-sectional study, based on an online questionnaire, was carried out between July and November 2021 with convenience sampling. The survey included 28 items and was distributed among students, teachers, researchers and administrative staff from a Spanish university. A total of 1,220 participants completed the survey. 70.4% of the respondents heard about the environmental impact of food and more than 50% were aware of the existence of the SDGs. The different aspects related to diet that concerned them the most were food waste, plastic usage, and environmental impact. They reported that a sustainable diet should be mainly based on local and seasonal products and with a low environmental impact as well as no or the minimum food waste. When asked if they were following a sustainable diet, 77% answered affirmatively. Moreover, the food groups more involved in a sustainable diet should be vegetables and fruits, olive oil, legumes, and whole grains. Regarding food waste, 60% of the surveyed population claimed to generate it at home, with the use of leftovers and planning shopping and meals being some of the most important domestic actions to avoid it. Further initiatives must be implemented to increase the level of knowledge as well as to raise the awareness on the importance to translate it into individual and collective actions that allow a shift toward more sustainable practices
