Enteric Delivery of Regenerating Family Member 3 alpha Alters the Intestinal Microbiota and Controls Inflammation in Mice With Colitis
Background & Aims Paneth cell dysfunction causes deficiencies in intestinal C-type lectins and antimicrobial peptides, which leads to dysbiosis of the intestinal microbiota, alters the mucosal barrier, and promotes development of inflammatory bowel diseases. We investigated whether transgenic (TG) expression of the human regenerating family member 3 alpha gene ( REG3A ) alters the fecal microbiota and affects development of colitis in mice. Methods We performed studies with C57BL/6 mice that express human regenerating family member 3 alpha (hREG3A) in hepatocytes, via the albumin gene promoter. In these mice, hREG3A travels via the bile to the intestinal lumen. Some mice were given dextran sodium sulfate (DSS) to induce colitis. Feces were collected from mice and the composition of the microbiota was analyzed by 16S ribosomal RNA sequencing. The fecal microbiome was also analyzed from mice that express only 1 copy of human REG3A transgene but were fed feces from control mice (not expressing hREG3A) as newborns. Mice expressing hREG3A were monitored for DSS-induced colitis after cohousing or feeding feces from control mice. Colitis was induced in another set of control and hREG3A-TG mice by administration of trinitrobenzene sulfonic acid; some mice were given intrarectal injections of the hREG3A protein. Colon tissues were collected from mice and analyzed by histology and immunohistochemistry to detect mucin 2, as well as by 16S ribosomal RNA fluorescence in situ hybridization, transcriptional analyses, and quantitative polymerase chain reaction. We measured levels of reactive oxygen species (ROS) in bacterial cultures and fecal microbiota using 2âČ,7âČ-dichlorofluorescein diacetate and flow cytometry. Results The fecal microbiota of mice that express hREG3A had a significant shift in composition, compared with control mice, with enrichment of Clostridiales (Ruminococcaceae, Lachnospiraceae) and depletion of Bacteroidetes (Prevotellaceae); the TG mice developed less-severe colitis following administration of DSS than control mice, associated with preserved gut barrier integrity and reduced bacterial translocation, epithelial inflammation, and oxidative damage. A similar shift in the composition of the fecal microbiota occurred after a few months in TG mice heterozygous for REG3A that harbored a wild-type maternal microbiota at birth; these mice developed less-severe forms of colitis following DSS administration. Cohoused and germ-free mice fed feces from REG3A- TG mice and given DSS developed less-severe forms of colitis and had reduced lipopolysaccharide activation of the toll-like receptor 4 and increased survival times compared with mice not fed feces from REG3A -TG mice. REG3A TG mice developed only mild colonic inflammation after exposure to 2,4,6-trinitrobenzene sulfonic acid, compared with control mice. Control mice given intrarectal hREG3A and exposed to 2,4,6-trinitrobenzene sulfonic acid showed less colon damage and inflammation than mice not given intrarectal hREG3A. Fecal samples from REG3A- TG mice had lower levels of ROS than feces from control mice during DSS administration. Addition of hREG3A to bacterial cultures reduced levels of ROS and increased survival of oxygen-sensitive commensal bacteria ( Faecalibacterium prausnitzii and Roseburia intestinalis ). Conclusions Mice with hepatocytes that express hREG3A, which travels to the intestinal lumen, are less sensitive to colitis than control mice. We found hREG3A to alter the colonic microbiota by decreasing levels of ROS. Fecal microbiota from REG3A -TG mice protect non-TG mice from induction of colitis. These findings indicate a role for reduction of oxidative stress in preserving the gut microbiota and its ability to prevent inflammation
COVID-19 symptoms at hospital admission vary with age and sex: results from the ISARIC prospective multinational observational study
Background:
The ISARIC prospective multinational observational study is the largest cohort of hospitalized patients with COVID-19. We present relationships of age, sex, and nationality to presenting symptoms.
Methods:
International, prospective observational study of 60â109 hospitalized symptomatic patients with laboratory-confirmed COVID-19 recruited from 43 countries between 30 January and 3 August 2020. Logistic regression was performed to evaluate relationships of age and sex to published COVID-19 case definitions and the most commonly reported symptoms.
Results:
âTypicalâ symptoms of fever (69%), cough (68%) and shortness of breath (66%) were the most commonly reported. 92% of patients experienced at least one of these. Prevalence of typical symptoms was greatest in 30- to 60-year-olds (respectively 80, 79, 69%; at least one 95%). They were reported less frequently in children (â€â18 years: 69, 48, 23; 85%), older adults (â„â70 years: 61, 62, 65; 90%), and women (66, 66, 64; 90%; vs. men 71, 70, 67; 93%, each Pâ<â0.001). The most common atypical presentations under 60 years of age were nausea and vomiting and abdominal pain, and over 60 years was confusion. Regression models showed significant differences in symptoms with sex, age and country.
Interpretation:
This international collaboration has allowed us to report reliable symptom data from the largest cohort of patients admitted to hospital with COVID-19. Adults over 60 and children admitted to hospital with COVID-19 are less likely to present with typical symptoms. Nausea and vomiting are common atypical presentations under 30 years. Confusion is a frequent atypical presentation of COVID-19 in adults over 60 years. Women are less likely to experience typical symptoms than men
L'influence de la présentation des données dans un énoncé de problÚme d'arithmétique
Ce mémoire porte sur le domaine des mathématiques à l'école. Je m'intéresse à l'influence que peuvent avoir des modifications de l'énoncé d'un problÚme d'arithmétique sur sa résolution. Mes lectures montrent que plusieurs variables influencent la résolution de problÚmes : le type de problÚme, la place de la question, le contexte de l'énoncé, la forme de l'énoncé, la présence de mots inducteurs... Dans ce travail, je me focalise sur la variable des données. L'objectif principal est d'étudier dans quelle mesure la maniÚre de présenter les données influence leur prise en compte par les élÚves. Pour cela, je me suis plus précisément intéressée à trois facteurs : le choix d'une écriture en lettres ou en chiffres des données, leur ordre d'apparition dans l'énoncé et la présence de données inutiles. Mon étude a porté sur un échantillon de 22 élÚves. Je leur ai soumis un certain nombre de problÚmes de nature variée en jouant sur les facteurs précédemment identifiés. Leur tùche était de les résoudre. Je me suis appuyée sur leurs productions écrites pour relever parmi les données celles qui étaient utilisées par les élÚves dans leur procédure de résolution. Les données recueillies ont montré un impact plus ou moins important des variables étudiées sur le travail des élÚves
The NIS iodide transporter in non thyroid carcinogenesis : new functions, new issues
Le symporteur sodium iodure (NIS), une glycoprotĂ©ine membranaire, connue pour son rĂŽle dans la biosynthĂšse des hormones thyroĂŻdiennes, catalyse le transport actif de lâiodure dans la thyroĂŻde. Cette activitĂ© de transport lui confĂšre des propriĂ©tĂ©s Ă la base du diagnostic, du suivi par imagerie radio-isotopique et du traitement par radiothĂ©rapie Ă lâiode 131 des cancers thyroĂŻdiens. Plusieurs Ă©tudes ont montrĂ© une expression de NIS dans un grand nombre de cancers non-thyroĂŻdiens, oĂč il prĂ©sente en gĂ©nĂ©ral un dĂ©faut dâadressage membranaire. Nous avons identifiĂ© une nouvelle fonction biologique de NIS, indĂ©pendante de sa fonction de transport, dans la migration et lâinvasion cellulaires via une activation de la GTPase RhoA connue pour son implication dans lâoncogenĂšse. Nous montrons Ă©galement que NIS est une protĂ©ine associĂ©e aux jonctions serrĂ©es qui transite rapidement dans le cytoplasme ou au front de migration des cellules motiles. Cette Ă©tude rĂ©vĂšle un nouveau rĂŽle de NIS dans la carcinogenĂšse et dĂ©voile certains facteurs qui gouvernent le trafic intracellulaire de NIS.The Natrium iodide symporter (NIS) is a transmembrane glycoprotein known for its role in thyroid hormones biosynthesis, catalyzes active iodide transport in the thyroid. The iodide transport activity of NIS allows its clinical applications for diagnosis, follow-up using radio-isotopic imaging and treatment by 131I radiotherapy of thyroid cancers. Several studies described NIS expression in non thyroidal tumour tissues frequently displaying a cytoplasmic localization. In the current study, we reveal that, independently to its transport activity, NIS habors new biological functions in migration and invasion through RhoGTPase signalling pathway, a pathway involved in carcinogenesis. In addition, we establish that NIS is a tight junction-associated protein, which dynamically shuttles between tight junctions and cytoplasm during cell migration, and localizes at the leading edge of the metastatic cancer cells. Overall, these findings offer a novel appraisal of the potential role of NIS in carcinogenesis, and of the factors governing NIS intracellular trafficking

When the pupils write, what are they concerned with, in the first place ? What space is made for spelling corrections in the period dedicated to writing ? Which status can you give them from an operating point of view ? Which incidences have they on the semantic evolution of the text ?
The analysis proposed is based on reconstruction of the writing of texts by children in CM2. The tool to collect is Genesis of the text, a word processor that saves the whole of writing procedures and allows observing in detail the tracks of the writing of a text : once the text is done, the scriptural procedures are chronologically reconstructed with the pauses, the crossing out, the returns backward, the advances.
Certain characteristics of this corpus are similar to those in manuscripts. Others are different : at the macro-procedural level, one can observe more moves than on manuscripts and the modifications of a great extent seem more frequent. At the micro-procedural level, the use of a word processor provokes the appearance of specific mistakes, typing errors, of which the correction takes a larger space when the typists are young.
Starting from examples, we will sketch analysis axis of pupils' writings on word processors while questioning the rÎle of procédures of which the semantic range is very small, nay nil, in the constitution of a text. We shall see at what times the formal modifications take place, what cornes after, and which semantic modifications their making is likely to have generated them. The examining of back and forth movements between modifications related to spelling and related to meaning, will allow us to work out some aspects of interactions between the conceptualisation activity and the constraints of scripting.Quand les élÚves écrivent, de quoi se préoccupent-ils d'abord ? Quelle place est faite aux corrections orthographiques dans la durée de l'écriture ? Quel statut leur accorder d'un point de vue opératoire ? Quelles incidences ont-elles sur l'évolution sémantique du texte ?
L'analyse proposée se base sur des reconstitutions d'écriture de textes par des enfants de CM2. L'outil de recueil est GenÚse du texte, un traitement de texte qui enregistre l'ensemble des opérations d'écriture et permet d'observer en détail les traces de l'écriture d'un texte : une fois le texte fini, les opérations scripturales se reconstituent chronologiquement avec les pauses, les ratures, les retours en arriÚre, les avancées.
Certaines caractéristiques de ce corpus sont similaires à celles des manuscrits. D'autres en diffÚrent : au niveau macro-procédural, on observe davantage de déplacements que sur les manuscrits et les modifications de grande ampleur paraissent plus fréquentes ; au niveau micro-procédural, l'usage du traitement de texte provoque l'apparition d'erreurs spécifiques, les erreurs de frappe, dont la correction prend une place d'autant plus grande que les scripteurs sont jeunes. à partir d'exemples, nous esquisserons des axes d'analyse de l'écriture des élÚves sur traitement de texte en nous interrogeant sur le rÎle d'opérations dont la portée sémantique est trÚs faible, voire nulle, dans la constitution du texte. Nous verrons à quels moments interviennent les modifications formelles, ce qui les suit, et quelles modifications sémantiques leur effectuation est susceptible d'avoir engendrées. L'examen d'allers-retours entre modifications liées à l'orthographe et modifications liées au sens, nous permettra de cerner certains aspects de l'interaction entre l'activité de conceptualisation et les contraintes de la scription.Doquet-Lacoste Claire.
