    Experiences with a Commercial ECDIS

    The integration of accurate positioning and electronic charts provides an extremely valuable tool to the mariner when navigating under hazardous conditions. This paper describes the experiences of two organizations in Canada who work under such conditions. On the St. Lawrence River, the Canadian Coast Guard is using the PINS 9000, a commercial ECDIS, for ice breaking chores with few, if any, other aids and has demonstrated operational savings. Marine Atlantic, Inc., uses the same system for ferry navigation into and out of Port aux Basques, Newfoundland, where it not only assists in meeting a tight schedule, but also enhances safety when operating in difficult conditions

    Novel analysis of global DNA methylation in the limbic system of the bovine brain

Global DNA methylation has been measured in brains of several species, but has yet to be examined in bovine. The objective of this study was to characterize global DNA methylation in 9 regions of the limbic system in the bovine brain: amygdala, bed nucleus of the stria terminalis, cingulate gyrus, dorsal raphe, hippocampus, hypothalamus, nucleus accumbens, periaqueductal gray, and prefrontal cortex. DNA was extracted from brain and blood samples of 6 Red Angus Ă— Simmental steers (less than 20 mo of age), using the DNA Extraction Kit from Agilent Technologies (Santa Clara, CA) and a phenol chloroform extraction. Percent of global DNA methylation was determined using the MethylFlash Methylated DNA Quantification Kit (Colormetric) from Epigentek (Farmingdale, NY). Varying amounts of global DNA methylation were observed among the 9 functionally distinct regions of the bovine limbic system. Amygdala, bed nucleus of the stria terminalis, cingulate gyrus, dorsal raphe, periaqueductal gray, prefrontal cortex, and nucleus accumbens are all significantly different (P < 0.05) from 1 or more brain tissue type, using a paired t test in SPSS (IBM, Armonk, NY). Conversely, global DNA methylation of blood was not significantly different (P < 0.05) from any brain tissue type. This study shows significant differences in global DNA methylation among different tissue types in the limbic system of the bovine brain. Understanding the differences in global DNA methylation within different tissues in the brain will facilitate future research involving the effects of differential methylation with regard to economically important traits

    Cavity magnonics deals with the interaction of magnons - elementary excitations in magnetic materials - and confined electromagnetic fields. We introduce the basic physics and review the experimental and theoretical progress of this young field that is gearing up for integration in future quantum technologies. Much of its appeal is derived from the strong magnon-photon coupling and the easily-reached nonlinear regime in microwave cavities. The interaction of magnons with light as detected by Brillouin light scattering is enhanced in magnetic optical resonators, which can be employed to manipulate magnon distributions. The cavity photon-mediated coupling of a magnon mode to a superconducting qubit enables measurements in the single magnon limit

    Epidemiologic investigation of immune-mediated polyradiculoneuropathy among abattoir workers exposed to porcine brain

    Background In October 2007, a cluster of patients experiencing a novel polyradiculoneuropathy was identified at a pork abattoir (Plant A). Patients worked in the primary carcass processing area (warm room); the majority processed severed heads (head-table). An investigation was initiated to determine risk factors for illness. Methods and Results Symptoms of the reported patients were unlike previously described occupational associated illnesses. A case-control study was conducted at Plant A. A case was defined as evidence of symptoms of peripheral neuropathy and compatible electrodiagnostic testing in a pork abattoir worker. Two control groups were used - randomly selected non-ill warm-room workers (n = 49), and all non-ill head-table workers (n = 56). Consenting cases and controls were interviewed and blood and throat swabs were collected. The 26 largest U.S. pork abattoirs were surveyed to identify additional cases. Fifteen cases were identified at Plant A; illness onsets occurred during May 2004–November 2007. Median age was 32 years (range, 21–55 years). Cases were more likely than warm-room controls to have ever worked at the head-table (adjusted odds ratio [AOR], 6.6; 95% confidence interval [CI], 1.6–26.7), removed brains or removed muscle from the backs of heads (AOR, 10.3; 95% CI, 1.5–68.5), and worked within 0–10 feet of the brain removal operation (AOR, 9.9; 95% CI, 1.2–80.0). Associations remained when comparing head-table cases and head-table controls. Workers removed brains by using compressed air that liquefied brain and generated aerosolized droplets, exposing themselves and nearby workers. Eight additional cases were identified in the only two other abattoirs using this technique. The three abattoirs that used this technique have stopped brain removal, and no new cases have been reported after 24 months of follow up. Cases compared to controls had higher median interferon-gamma (IFNγ) levels (21.7 pg/ml; vs 14.8 pg/ml, P<0.001). Discussion This novel polyradiculoneuropathy was associated with removing porcine brains with compressed air. An autoimmune mechanism is supported by higher levels of IFNγ in cases than in controls consistent with other immune mediated illnesses occurring in association with neural tissue exposure. Abattoirs should not use compressed air to remove brains and should avoid procedures that aerosolize CNS tissue. This outbreak highlights the potential for respiratory or mucosal exposure to cause an immune-mediated illness in an occupational setting

    Adult infiltrating gliomas with WHO 2016 integrated diagnosis: additional prognostic roles of ATRX and TERT

    The “integrated diagnosis” for infiltrating gliomas in the 2016 revised World Health Organization (WHO) classification of tumors of the central nervous system requires assessment of the tumor for IDH mutations and 1p/19q codeletion. Since TERT promoter mutations and ATRX alterations have been shown to be associated with prognosis, we analyzed whether these tumor markers provide additional prognostic information within each of the five WHO 2016 categories. We used data for 1206 patients from the UCSF Adult Glioma Study, the Mayo Clinic and The Cancer Genome Atlas (TCGA) with infiltrative glioma, grades II-IV for whom tumor status for IDH, 1p/19q codeletion, ATRX, and TERT had been determined. All cases were assigned to one of 5 groups following the WHO 2016 diagnostic criteria based on their morphologic features, and IDH and 1p/19q codeletion status. These groups are: (1) Oligodendroglioma, IDH-mutant and 1p/19q-codeleted; (2) Astrocytoma, IDH-mutant; (3) Glioblastoma, IDH-mutant; (4) Glioblastoma, IDH-wildtype; and (5) Astrocytoma, IDH-wildtype. Within each group, we used univariate and multivariate Cox proportional hazards models to assess associations of overall survival with patient age at diagnosis, grade, and ATRX alteration status and/or TERT promoter mutation status. Among Group 1 IDH-mutant 1p/19q-codeleted oligodendrogliomas, the TERT-WT group had significantly worse overall survival than the TERT-MUT group (HR: 2.72, 95% CI 1.05–7.04, p&nbsp;=&nbsp;0.04). In both Group 2, IDH-mutant astrocytomas and Group 3, IDH-mutant glioblastomas, neither TERT mutations nor ATRX alterations were significantly associated with survival. Among Group 4, IDH-wildtype glioblastomas, ATRX alterations were associated with favorable outcomes (HR: 0.36, 95% CI 0.17–0.81, p&nbsp;=&nbsp;0.01). Among Group 5, IDH-wildtype astrocytomas, the TERT-WT group had significantly better overall survival than the TERT-MUT group (HR: 0.48, 95% CI 0.27–0.87), p&nbsp;=&nbsp;0.02). Thus, we present evidence that in certain WHO 2016 diagnostic groups, testing for TERT promoter mutations or ATRX alterations may provide additional useful prognostic information

    A tiered-layered-staged model for informed consent in personal genome testing

    In recent years, developments in genomics technologies have led to the rise of commercial personal genome testing (PGT): broad genome-wide testing for multiple diseases simultaneously. While some commercial providers require physicians to order a personal genome test, others can be accessed directly. All providers advertise directly to consumers and offer genetic risk information about dozens of diseases in one single purchase. The quantity and the complexity of risk information pose challenges to adequate pre-test and post-test information provision and informed consent. There are currently no guidelines for what should constitute informed consent in PGT or how adequate informed consent can be achieved. In this paper, we propose a tiered-layered-staged model for informed consent. First, the proposed model is tiered as it offers choices between categories of diseases that are associated with distinct ethical, personal or societal issues. Second, the model distinguishes layers of information with a first layer offering minimal, indispensable information that is material to all consumers, and additional layers offering more detailed information made available upon request. Finally, the model stages informed consent as a process by feeding information to consumers in each subsequent stage of the process of undergoing a test, and by accommodating renewed consent for test result updates, resulting from the ongoing development of the science underlying PGT. A tiered-layered-staged model for informed consent with a focus on the consumer perspective can help overcome the ethical problems of information provision and informed consent in direct-to-consumer PGT.European Journal of Human Genetics advance online publication, 21 November 2012; doi:10.1038/ejhg.2012.237
