    Optimization of molecular detection of GD2 synthase mRNA in retinoblastoma

    Extraocular dissemination is the main cause of death in patients with retinoblastoma in developing countries and there are few molecular markers that could be used for evaluation of minimal disseminated disease. The expression of the ganglioside GD2 is present in retinoblastoma cells metastatic to the bone marrow and the enzyme GD2 synthase activity is detected in neuroblastoma, which shares many phenotypic features with retinoblastoma. Our purpose was to optimize the detection of GD2 synthase expression by reverse transcription-polymerase chain reaction (RT-PCR) followed by nested-PCR in human retinoblastoma cell lines and patient samples. The optimization strategy was carried out by using the retinoblastoma cell lines Y79 and WERI-Rb1 and specific primers designed for the human sequence of the GD2 synthase mRNA. We detected GD2 synthase expression with at least 200 pg and 40 pg of total RNA extracted from cultured retinoblastoma cells, using a first round of RT-PCR amplification and a second round of nested-PCR, respectively. We have also confirmed the detection of GD2 synthase by RT-PCR and immunohistochemical expression of the ganglioside in human retinoblastoma tumors xenotransplanted in nude mice. In a study from tumor bank specimens from 8 retinoblastoma patients, we were able to demonstrate the presence of GD2 synthase mRNA in blood and cerebrospinal fluid samples in cases of extraocular dissemination of the tumor. The sequence was not detected in samples from children with low-risk disease or healthy adult volunteers. The detection of GD2 synthase mRNA through an optimized nested RT-PCR assay may be a promising tool for the assessment of minimal disseminated disease in enucleated patients.Fil: Laurent, Viviana Eunice.     Caracterización de los pacientes con neoplasias mieloproliferativas crónicas en San Cristóbal

    Introducción: las neoplasias mieloproliferativas crónicas son un conjunto de hemopatías con origen en una célula madre pluripotencial de la hematopoyesis. Objetivo: describir el comportamiento clínico y de los parámetros de laboratorio en pacientes diagnosticados con neoplasias mieloproliferativas crónicas durante diez años en el Hospital General Docente Comandante Pinares del municipio San Cristóbal, Artemisa. Material y métodos: se realizó un estudio observacional, descriptivo y transversal que comprendió el período entre enero de 2008 y diciembre de 2018, en el que fueron incluidos 52 casos de pacientes vivos mayores de dieciocho con confirmación histológica de neoplasias mieloproliferativas crónicas en cualquiera de sus formas. Se analizaron variables clínicas, sociodemográficas y de laboratorio. Se calcularon frecuencias absolutas y relativas, medidas de tendencia central y de distribución para variables cuantitativas Resultados: la policitemia vera fue la neoplasia más incidente con 29 casos (55,7 %). El grupo de 55 a 64 años fue el más afectado (32,7 %) para una media de 57 ± 15 años, con predominio del sexo masculino con 32 pacientes (61,6 %). Los síntomas generales y la esplenomegalia fueron comunes en el 89,5 % de las leucemias mieloides crónicas. En todos los casos hubo panmielosis asociada a la proliferación excesiva de las tres líneas hematopoyéticas, fibrosis reactiva de la médula ósea, hiperuricemia frecuente y aumento en los valores séricos de lactato deshidrogenasa. Conclusiones: el diagnóstico de estas entidades descansa en la indisoluble unión entre los hallazgos clínicos y los exámenes de laboratorio

    Evaluation of Etest for Susceptibility Testing of Multidrug-Resistant Isolates of Mycobacterium tuberculosis

    To prescribe effective treatment schemes for patients with tuberculosis, more-efficient susceptibility testing techniques for Mycobacterium tuberculosis are needed, especially in regions with multidrug resistance. Etest (AB BIODISK, Solna, Sweden) is a simple technique that provides quantitative drug susceptibility results for M. tuberculosis in 5 to 10 days from a culture grown at low cost. The performance of Etest was compared to that of the reference proportion method, using 95 M. tuberculosis clinical isolates of which 42.1% (40 of 95) were resistant to at least one antibiotic by the reference method. Overall agreement between Etest and the reference method was 98.9% (94 of 95) for detection of multidrug resistance; for resistance to individual drugs, agreement was 97.9% (93 of 95) for rifampin, 96.0% (92 of 95) for ethambutol, 94.7% (90 of 95) for isoniazid, and 85.3% (81 of 95) for streptomycin. This study supports the utility of Etest for timely detection of drug resistance in M. tuberculosis and for use in tuberculosis control programs

    Se realizó un estudio descriptivo prospectivo de los niños de 0-14 años, accidentados durante el período de enero a julio del 2006, en 16 consultorios del Consejo Popular Jesús Menéndez de Bayamo, con el objetivo de determinar el comportamiento de los accidentes durante el tiempo antes señalado. La muestra quedó constituida por 58 pacientes. Para la recolección de la información fue elaborado un formulario donde se incluyeron las variables edad, sexo, lugar del accidente, tipo de lesión y estado del accidentado. En el procesamiento de los resultados se utilizó el método de conteo simple y se le realizó el cálculo porcentual, se llegó a la conclusión que la mayor cantidad de accidentes ocurrió en niños de 1-4 años (55.1%), en le hogar (51.5%), las lesiones predominantes fueron las heridas (31.4%), seguidas de las contusiones (24.0%). El carácter leve de las lesiones fue más frecuente constituyendo el 82.8%.It was performed a descriptive-prospective study of accidents in children from o to 14 years old, from 16 Health Areas of  “Jesus Menendez” in Bayamo, during the period from January to July 2006, with the objective to determine the behaviour of the accidents during that period. The sample was constituted by 58 patients. To compile the information, it was elaborated a formulary where recessed variables were included: age, sex, place of accident, type of lesion and accident stage. It was used the simple count method and it was made the percentage calculation in the prosecution of the result, it is reached the conclusion that the greater quantity of accidents occurred in children from 1-4 years (55.1%) at home (51.5%). The prevailing lesions were wounded (31.4%), followed by contusions (24.0%). The characters of the lesions were more frequent (82.8%)

    El seguimiento farmacoterapéutico es una actividad asistencial de los farmacéuticos cuya función es el manejo de la farmacoterapia aplicada al paciente, contribuyendo a la identificación, prevención y resolución de Problemas Relacionados con Medicamentos. El objetivo de la investigación fue caracterizar los problemas relacionados con medicamentos en gestantes con anemia atendidas en la consulta de atención prenatal del Policlínico “30 de Noviembre”, de Santiago de Cuba. Se realizó un estudio descriptivo, longitudinal y prospectivo. Las pacientes se caracterizaron según variables biosociales, clínicas y farmacoterapéuticas a partir de la revisión del historial clínico y de las entrevistas. La clasificación de los problemas relacionados con medicamentos se realizó según Cipolle, et al., 2012. Se analizaron los resultados clínicos alcanzados después de las intervenciones. Se empleó la estadística descriptiva. Predominaron las pacientes del grupo de 20 a 24 años (27,1%), en el primer trimestre de gestación (72,9%), con comorbilidades (48,2%) y consumo de más de cuatro medicamentos (71,8%), siendo los más prescritos el ácido fólico (97,6%) y la aspirina (77,6%). Se identificaron 150 Problemas Relacionados con los Medicamentos para una proporción de 1,7 PRM/paciente. El 56% fueron problemas de Adherencia, seguido de un 23,3% de Indicación, el 18% de Seguridad y el 2,7% de Efectividad. Los antianémicos, el ácido fólico y el carbonato de calcio resultaron los medicamentos más implicados en la aparición de estos problemas. Las intervenciones farmacéuticas oportunas contribuyeron a la solución de los problemas relacionados con medicamentos detectados y a la obtención de mejores resultados clínicos en las pacientes

    O trabalho expõe uma análise jornalística da realidade midiática (Agenda Setting) em torno às condições nas quais se expressa a democracia a partir de três categorias: qualidade de vida, cidadania e gênero. Se usaram os jornais El Zócalo de Piedras Negras em Coahuila e La Crónica de Mexicali em Baixa California.  Os resultados são os seguintes: somente em tempos de processo eleitorais é notória a preferência por dar voz às instancias governamentais; a condição de gênero não é objeto de atenção, agenda e incidência das problemáticas referidas a isso; a mulher não é sujeito de voz pública para temas políticos, o espaço outorgado por setores ou agenda de meios. Existe escassa possibilidade de atuação e inserção de políticas públicas como resposta institucionalizada para a resolução de problemáticas fronteiriças

    Endotoxin exposure during sensitization to Blomia tropicalis allergens shifts TH2 immunity towards a TH17-mediated airway neutrophilic inflammation: role of TLR4 and TLR2

    Experimental evidence and epidemiological studies indicate that exposure to endotoxin lipopolysaccharide (eLPS) or other TLR agonists prevent asthma. We have previously shown in the OVA-model of asthma that eLPS administration during alum-based allergen sensitization blocked the development of lung TH2 immune responses via MyD88 pathway and IL-12/IFN-γ axis. In the present work we determined the effect of eLPS exposure during sensitization to a natural airborne allergen extract derived from the house dust mite Blomia tropicalis (Bt). Mice were subcutaneously sensitized with Bt allergens co-adsorbed onto alum with or without eLPS and challenged twice intranasally with Bt. Cellular and molecular parameters of allergic lung inflammation were evaluated 24 h after the last Bt challenge. Exposure to eLPS but not to ultrapure LPS (upLPS) preparation during sensitization to Bt allergens decreased the influx of eosinophils and increased the influx of neutrophils to the airways. Inhibition of airway eosinophilia was not observed in IFN-γdeficient mice while airway neutrophilia was not observed in IL-17RA-deficient mice as well in mice lacking MyD88, CD14, TLR4 and, surprisingly, TLR2 molecules. Notably, exposure to a synthetic TLR2 agonist (PamCSK4) also induced airway neutrophilia that was dependent on TLR2 and TLR4 molecules. In the OVA model, exposure to eLPS or PamCSK4 suppressed OVA-induced airway inflammation. Our results suggest that B. tropicalis allergens engage TLR4 that potentiates TLR2 signaling. This dual TLR activation during sensitization results in airway neutrophilic inflammation associated with increased frequency of lung TH17 cells.     Allergen Micro-Bead Array for IgE Detection: A Feasibility Study Using Allergenic Molecules Tested on a Flexible Multiplex Flow Cytometric Immunoassay

    Background: Allergies represent the most prevalent non infective diseases worldwide. Approaching IgE-mediated sensitizations improved much by adopting allergenic molecules instead of extracts, and by using the micro-technology for multiplex testing. Objective and Methods: To provide a proof-of-concept that a flow cytometric bead array is a feasible mean for the detection of specific IgE reactivity to allergenic molecules in a multiplex-like way. A flow cytometry Allergenic Moleculebased micro-bead Array system (ABA) was set by coupling allergenic molecules with commercially available micro-beads. Allergen specific polyclonal and monoclonal antibodies, as well as samples from 167 allergic patients, characterized by means of the ISAC microarray system, were used as means to show the feasibility of the ABA. Three hundred and thirty-six sera were tested for 1 or more of the 16 selected allergens, for a total number of 1,519 tests on each of the two systems.
