    Intra- and interspecies interactions between prion proteins and effects of mutations and polymorphisms

    Recently, crystallization of the prion protein in a dimeric form was reported. Here we show that native soluble homogenous FLAG-tagged prion proteins from hamster, man and cattle expressed in the baculovirus system are predominantly dimeric. The PrP/PrP interaction was confirmed in Semliki Forest virus-RNA transfected BHK cells co-expressing FLAG- and oligohistidine-tagged human PrP. The yeast two-hybrid system identified the octarepeat region and the C-terminal structured domain (aa90-aa230) of PrP as PrP/PrP interaction domains. Additional octarepeats identified in patients suffering from fCJD reduced (wtPrP versus PrP+90R) and completely abolished (PrP+90R versus PrP+90R) the PrP/PrP interaction in the yeast two-hybrid system. In contrast, the Met/Val polymorphism (aa129), the GSS mutation Pro102Leu and the FFI mutation Asp178Asn did not affect PrP/PrP interactions. Proof of interactions between human or sheep and bovine PrP, and sheep and human PrP, as well as lack of interactions between human or bovine PrP and hamster PrP suggest that interspecies PrP interaction studies in the yeast two-hybrid system may serve as a rapid pre-assay to investigate species barriers in prion diseases

    Transport of skyrmions by surface acoustic waves

    Magnetic skyrmions in thin films with perpendicular magnetic anisotropy are promising candidates for magnetic memory and logic devices, making the development of ways to transport skyrmions efficiently in a desired trajectory of significant interest. Here, we investigate the transport of skyrmions by surface acoustic waves (SAWs) via several modalities using micromagnetic simulations. We show skyrmion pinning sites created by standing SAWs at anti-nodes and skyrmion Hall-like motion without pinning driven by traveling SAWs. We also show how orthogonal SAWs formed by combining a longitudinal traveling SAW and a transverse standing SAW can be used for the 2D positioning of skyrmions. Our results also suggest SAWs offer a viable approach to the transport of multiple skyrmions along a multichannel racetrack

    Do on-farm natural, restored, managed and constructed wetlands mitigate agricultural pollution in Great Britain and Ireland?: a systematic review

    Wetlands in agricultural landscapes offer a number of benefits to the landscape function in which they are set, reducing nutrient runoff, providing additional habitat mosaics and offering various ecosystem services. They require careful planning and maintenance in order to perform their optimum design function over a prolonged period of time. They should be treated as functional units of farm infrastructure rather than fit-and-forget systems. A high priority topic within the Department for Environment, Food and Rural Affairs (DEFRA) water quality programme is the mitigation of pollution from agriculture. This programme was set up to meet the requirements of the European Water Framework Directive (WFD) EU (2000). Nutrient loss from agricultural land has been suggested as a major cause of elevated nutrient concentrations in surface waters in the UK. Nitrogen (N) and phosphorus (P) are of particular concern as an excess of either nutrient can lead to eutrophication of freshwater systems and coastal waters. Agriculture has also been identified as a significant source of suspended sediment (SS) concentrations in UK rivers and agriculturally derived sediment has been identified as a source of increased bed-sediment P concentrations in rivers. High bed sediments loads have other negative impacts, such as clogging river gravels reducing fish spawning. There is considerable evidence in the published and grey literature that wetlands have the ability to remove nutrients and sediment and thus reduce the load on receiving waters. Wetlands have also been reported to perform other ecosystem services, such as reducing floods, supporting biodiversity and sequestering carbon. A policy to promote the conservation, management, restoration or construction of wetlands could help to mitigate the impacts of N, P and SS from agriculture delivering requirements of WFD through Catchment Sensitive Farming following an Ecosystem Approach and Catchment Based Approach promoted by Defra. It could also meet other commitments such as implementing the Ramsar and Biodiversity Conventions to which the UK is a signatory. However, the term wetlands covers a wide range of habitat types and it is important that policy makers are provided with accurate, robust and independently reviewed information on the degree to which different types of wetland perform these services under different circumstances, so that policy can most best targeted. This systematic review assesses the available evidence on the performance of various wetland types on farms to reduce nutrient input and suspended sediments to receiving waters. It provides a defensible evidence base on which to base policy. The studies reviewed cover different input loads and the analysis compares performance of these wetland systems in respect of % reduction efficiency. In England and Wales, Defra, working closely with the Environment Agency and Natural England, has commissioned this systematic review on how effective, and what influences the effectiveness of wetlands at mitigating N, P and SS inputs from agriculture to receiving freshwater in the United Kingdom and Ireland

    Energy averages and fluctuations in the decay out of superdeformed bands

    We derive analytic formulae for the energy average (including the energy average of the fluctuation contribution) and variance of the intraband decay intensity of a superdeformed band. Our results may be expressed in terms of three dimensionless variables: Γ/ΓS\Gamma^{\downarrow}/\Gamma_S, ΓN/d\Gamma_N/d, and ΓN/(ΓS+Γ)\Gamma_N/(\Gamma_S+\Gamma^{\downarrow}). Here Γ\Gamma^{\downarrow} is the spreading width for the mixing of a superdeformed (SD) state 0>|0> with the normally deformed (ND) states Q>|Q> whose spin is the same as 0>|0>'s. The Q>|Q> have mean level spacing dd and mean electromagnetic decay width ΓN\Gamma_N whilst 0>|0> has electromagnetic decay width ΓS\Gamma_S. The average decay intensity may be expressed solely in terms of the variables Γ/ΓS\Gamma^{\downarrow}/\Gamma_S and ΓN/d\Gamma_N/d or, analogously to statistical nuclear reaction theory, in terms of the transmission coefficients T0(E)T_0(E) and TNT_N describing transmission from the Q>|Q> to the SD band via 0|0\angle and to lower ND states. The variance of the decay intensity, in analogy with Ericson's theory of cross section fluctuations depends on an additional variable, the correlation length \Gamma_N/(\Gamma_S+\Gamma^{\downarrow})=\frac{d}{2\pi}T_N/(\Gamma_S+\Gamma^{\d ownarrow}). This suggests that analysis of an experimentally obtained variance could yield the mean level spacing dd as does analysis of the cross section autocorrelation function in compound nuclear reactions.     Tracking azimuthons in nonlocal nonlinear media

    We study the formation of azimuthons, i.e., rotating spatial solitons, in media with nonlocal focusing nonlinearity. We show that whole families of these solutions can be found by considering internal modes of classical non-rotating stationary solutions, namely vortex solitons. This offers an exhaustive method to identify azimuthons in a given nonlocal medium. We demonstrate formation of azimuthons of different vorticities and explain their properties by considering the strongly nonlocal limit of accessible solitons.Comment: 11 pages, 7 figure

    Polycyclic aromatic hydrocarbons in the dwarf galaxy IC 10

    Infrared observations from the Spitzer Space Telescope archive are used to study the dust component of the interstellar medium in the IC~10 irregular galaxy. Dust distribution in the galaxy is compared to the distributions of Hα\alpha and [SII] emission, neutral hydrogen and CO clouds, and ionizing radiation sources. The distribution of polycyclic aromatic hydrocarbons (PAH) in the galaxy is shown to be highly non-uniform with the mass fraction of these particles in the total dust mass reaching 4%. PAHs tend to avoid bright HII regions and correlate well with atomic and molecular gas. This pattern suggests that PAHs form in the dense interstellar gas. We propose that the significant decrease of the PAH abundance at low metallicity is observed not only globally (at the level of entire galaxies), but also locally (at least, at the level of individual HII regions). We compare the distribution of the PAH mass fraction to the distribution of high-velocity features, that we have detected earlier in wings of Hα\alpha and SII lines, over the entire available galaxy area. No conclusive evidence for shock destruction of PAHs in the IC~10 galaxy could be found.Comment: Accepted for publication in Astronomy Report

    Background model systematics for the Fermi GeV excess

    The possible gamma-ray excess in the inner Galaxy and the Galactic center (GC) suggested by Fermi-LAT observations has triggered a large number of studies. It has been interpreted as a variety of different phenomena such as a signal from WIMP dark matter annihilation, gamma-ray emission from a population of millisecond pulsars, or emission from cosmic rays injected in a sequence of burst-like events or continuously at the GC. We present the first comprehensive study of model systematics coming from the Galactic diffuse emission in the inner part of our Galaxy and their impact on the inferred properties of the excess emission at Galactic latitudes 2<b<202^\circ<|b|<20^\circ and 300 MeV to 500 GeV. We study both theoretical and empirical model systematics, which we deduce from a large range of Galactic diffuse emission models and a principal component analysis of residuals in numerous test regions along the Galactic plane. We show that the hypothesis of an extended spherical excess emission with a uniform energy spectrum is compatible with the Fermi-LAT data in our region of interest at 95%95\% CL. Assuming that this excess is the extended counterpart of the one seen in the inner few degrees of the Galaxy, we derive a lower limit of 10.010.0^\circ (95%95\% CL) on its extension away from the GC. We show that, in light of the large correlated uncertainties that affect the subtraction of the Galactic diffuse emission in the relevant regions, the energy spectrum of the excess is equally compatible with both a simple broken power-law of break energy 2.1±0.22.1\pm0.2 GeV, and with spectra predicted by the self-annihilation of dark matter, implying in the case of bˉb\bar{b}b final states a dark matter mass of 495.4+6.449^{+6.4}_{-5.4} GeV.Comment: 65 pages, 28 figures, 7 table

    Evolutionary aspects of population structure for molecular and quantitative traits in the freshwater snail Radix balthica.

    Detecting the action of selection in natural populations can be achieved using the QST-FST comparison that relies on the estimation of FST with neutral markers, and QST using quantitative traits potentially under selection. QST higher than FST suggests the action of directional selection and thus potential local adaptation. In this article, we apply the QST-FST comparison to four populations of the hermaphroditic freshwater snail Radix balthica located in a floodplain habitat. In contrast to most studies published so far, we did not detect evidence of directional selection for local optima for any of the traits we measured: QST calculated using three different methods was never higher than FST. A strong inbreeding depression was also detected, indicating that outcrossing is probably predominant over selfing in the studied populations. Our results suggest that in this floodplain habitat, local adaptation of R. balthica populations may be hindered by genetic drift, and possibly altered by uneven gene flow linked to flood frequency

    We report a measurement of cross section σ(νμ+nucleusμ+X)\sigma(\nu_{\mu}+{\rm nucleus}\rightarrow\mu^{-}+X) and the first measurements of the cross section σ(νˉμ+nucleusμ++X)\sigma(\bar{\nu}_{\mu}+{\rm nucleus}\rightarrow\mu^{+}+X) and their ratio R(σ(νˉ)σ(ν))R(\frac{\sigma(\bar \nu)}{\sigma(\nu)}) at (anti-)neutrino energies below 1.5 GeV. We determine the single momentum bin cross section measurements, averaged over the T2K νˉ/ν\bar{\nu}/\nu-flux, for the detector target material (mainly Carbon, Oxygen, Hydrogen and Copper) with phase space restricted laboratory frame kinematics of θμ\theta_{\mu}500 MeV/c. The results are σ(νˉ)=(0.900±0.029(stat.)±0.088(syst.))×1039\sigma(\bar{\nu})=\left( 0.900\pm0.029{\rm (stat.)}\pm0.088{\rm (syst.)}\right)\times10^{-39} and $\sigma(\nu)=\left( 2.41\ \pm0.022{\rm{(stat.)}}\pm0.231{\rm (syst.)}\ \right)\times10^{-39}inunitsofcm in units of cm^{2}/nucleonand/nucleon and R\left(\frac{\sigma(\bar{\nu})}{\sigma(\nu)}\right)= 0.373\pm0.012{\rm (stat.)}\pm0.015{\rm (syst.)}$.Comment: 18 pages, 8 figure
