    Birth elicits a conserved neuroendocrine response with implications for perinatal osmoregulation and neuronal cell death

    Long-standing clinical findings report a dramatic surge of vasopressin in umbilical cord blood of the human neonate, but the neural underpinnings and function(s) of this phenomenon remain obscure. We studied neural activation in perinatal mice and rats, and found that birth triggers activation of the suprachiasmatic, supraoptic, and paraventricular nuclei of the hypothalamus. This was seen whether mice were born vaginally or via Cesarean section (C-section), and when birth timing was experimentally manipulated. Neuronal phenotyping showed that the activated neurons were predominantly vasopressinergic, and vasopressin mRNA increased fivefold in the hypothalamus during the 2–3 days before birth. Copeptin, a surrogate marker of vasopressin, was elevated 30-to 50-fold in plasma of perinatal mice, with higher levels after a vaginal than a C-section birth. We also found an acute decrease in plasma osmolality after a vaginal, but not C-section birth, suggesting that the difference in vasopressin release between birth modes is functionally meaningful. When vasopressin was administered centrally to newborns, we found an ~ 50% reduction in neuronal cell death in specific brain areas. Collectively, our results identify a conserved neuroendocrine response to birth that is sensitive to birth mode, and influences peripheral physiology and neurodevelopment.Fil: Hoffiz, Yarely C..     Do Dimensions of Ethnic Identity Mediate the Association Between Perceived Ethnic Group Discrimination and Depressive Symptoms?

    Ethnic group discrimination represents a notable risk factor that may contribute to mental health problems among ethnic minority college students. However, cultural resources (e.g., ethnic identity) may promote psychological adjustment in the context of group-based discriminatory experiences. In the current study, we examined the associations between perceptions of ethnic group discrimination and depressive symptoms, and explored dimensions of ethnic identity (i.e., exploration, resolution, and affirmation) as mediators of this process among 2,315 ethnic minority college students (age 18 to 30 years; 37% Black, 63% Latino). Results indicated that perceived ethnic group discrimination was associated positively with depressive symptoms among students from both ethnic groups. The relationship between perceived ethnic group discrimination and depressive symptoms was mediated by ethnic identity affirmation for Latino students, but not for Black students. Ethnic identity resolution was negatively and indirectly associated with depressive symptoms through ethnic identity affirmation for both Black and Latino students. Implications for promoting ethnic minority college students’ mental health and directions for future research are discussed

    Mapping gravitational-wave backgrounds using methods from CMB analysis: Application to pulsar timing arrays

    We describe an alternative approach to the analysis of gravitational-wave backgrounds, based on the formalism used to characterise the polarisation of the cosmic microwave background. In contrast to standard analyses, this approach makes no assumptions about the nature of the background and so has the potential to reveal much more about the physical processes that generated it. An arbitrary background can be decomposed into modes whose angular dependence on the sky is given by gradients and curls of spherical harmonics. We derive the pulsar timing overlap reduction functions for the individual modes, which are given by simple combinations of spherical harmonics evaluated at the pulsar locations. We show how these can be used to recover the components of an arbitrary background, giving explicit results for both isotropic and anisotropic uncorrelated backgrounds. We also find that the response of a pulsar timing array to curl modes is identically zero, so half of the gravitational-wave sky will never be observed using pulsar timing, no matter how many pulsars are included in the array. An isotropic, unpolarised and uncorrelated background can be accurately represented using only three modes, and so a search of this type will be only slightly more complicated than the standard cross-correlation search using the Hellings and Downs overlap reduction function. However, by measuring the components of individual modes of the background and checking for consistency with isotropy, this approach has the potential to reveal much more information. Each individual mode on its own describes a background that is correlated between different points on the sky.     Implementation and evaluation of various demons deformable image registration algorithms on GPU

    Online adaptive radiation therapy (ART) promises the ability to deliver an optimal treatment in response to daily patient anatomic variation. A major technical barrier for the clinical implementation of online ART is the requirement of rapid image segmentation. Deformable image registration (DIR) has been used as an automated segmentation method to transfer tumor/organ contours from the planning image to daily images. However, the current computational time of DIR is insufficient for online ART. In this work, this issue is addressed by using computer graphics processing units (GPUs). A grey-scale based DIR algorithm called demons and five of its variants were implemented on GPUs using the Compute Unified Device Architecture (CUDA) programming environment. The spatial accuracy of these algorithms was evaluated over five sets of pulmonary 4DCT images with an average size of 256x256x100 and more than 1,100 expert-determined landmark point pairs each. For all the testing scenarios presented in this paper, the GPU-based DIR computation required around 7 to 11 seconds to yield an average 3D error ranging from 1.5 to 1.8 mm. It is interesting to find out that the original passive force demons algorithms outperform subsequently proposed variants based on the combination of accuracy, efficiency, and ease of implementation.Comment: Submitted to Physics in Medicine and Biolog

    Measuring acute effects of subanesthetic ketamine on cerebrovascular hemodynamics in humans using TD-fNIRS

    Quantifying neural activity in natural conditions (i.e. conditions comparable to the standard clinical patient experience) during the administration of psychedelics may further our scientific understanding of the effects and mechanisms of action. This data may facilitate the discovery of novel biomarkers enabling more personalized treatments and improved patient outcomes. In this single-blind, placebo-controlled study with a non-randomized design, we use time-domain functional near-infrared spectroscopy (TD-fNIRS) to measure acute brain dynamics after intramuscular subanesthetic ketamine (0.75 mg/kg) and placebo (saline) administration in healthy participants (n = 15, 8 females, 7 males, age 32.4 ± 7.5 years) in a clinical setting. We found that the ketamine administration caused an altered state of consciousness and changes in systemic physiology (e.g. increase in pulse rate and electrodermal activity). Furthermore, ketamine led to a brain-wide reduction in the fractional amplitude of low frequency fluctuations, and a decrease in the global brain connectivity of the prefrontal region. Lastly, we provide preliminary evidence that a combination of neural and physiological metrics may serve as predictors of subjective mystical experiences and reductions in depressive symptomatology. Overall, our study demonstrated the successful application of fNIRS neuroimaging to study the physiological effects of the psychoactive substance ketamine in humans, and can be regarded as an important step toward larger scale clinical fNIRS studies that can quantify the impact of psychedelics on the brain in standard clinical settings

    A member of the TERMINAL FLOWER1/CENTRORADIALIS gene family controls sprout growth in potato tubers

    Potato tuber bud dormancy break followed by premature sprouting is a major commercial problem which results in quality losses and decreased tuber marketability. An approach to controlling premature tuber sprouting is to develop potato cultivars with a longer dormancy period and/or reduced rate of sprout growth. Our recent studies using a potato diploid population have identified several quantitative trait loci (QTLs) that are associated with tuber sprout growth. In the current study, we aim to characterize a candidate gene associated with one of the largest effect QTLs for rapid tuber sprout growth on potato chromosome 3. Underlying this QTL is a gene encoding a TERMINAL FLOWER 1/CENTRORADIALIS homologue (PGSC0003DMG400014322). Here, we use a transgenic approach to manipulate the expression level of the CEN family member in a potato tetraploid genotype (cv. Désirée). We demonstrate a clear effect of manipulation of StCEN expression, with decreased expression levels associated with an increased rate of sprout growth, and overexpressing lines showing a lower rate of sprout growth than controls. Associated with different levels of StCEN expression were different levels of abscisic acid and cytokinins, implying a role in controlling the levels of plant growth regulators in the apical meristem

    Sediment source apportionment following wildfire in an upland commercial forest catchment

    File replaced (incorrect version) on 09/05/2022 by KT (LDS).Purpose: Wildfires can have major impacts on water scarcity and water quality linked to off-site transfer of polluting ash and nutrients. Understanding sediment sources in burnt landscapes can help to develop mitigation strategies, especially in catchments planted with introduced species that are prone to fire. We investigated sediment sources activated by post-fire rainfall in a small-forested catchment that was impacted by a severe wildfire. The aim was to use environmental radionuclides and elemental geochemistry as tracers to apportion sediment sources within burnt plantation systems. Methods: Surficial (0–2 cm) topsoil (n = 9), sub-surficial (2–4 cm) topsoil (i.e. below the burnt layer; n = 8) samples from burnt hillslopes and forest roads (n = 5) and stream banks (n = 5) soil samples were taken in the Quivolgo catchment, El Maule region, Chile. Sediment samples (n = 9) were collected from behind a v-notched weir on three dates after the fire: May 2017, July 2017 and October 2017. Soil and sediment samples were analysed by gamma spectrometry and wavelength-dispersive X-ray fluorescence (WD-XRF) used to obtain tracer properties. These were evaluated visually and statistically to identify potential non-conservative tracers. Sediment apportionment was undertaken using the MixSIAR mixing model. Results: The tracer selection procedure resulted in ten tracers being used for sediment apportionment. Tracer suitability was based on (i) weak and non-significant linear relationship between tracer concentrations and specific surface area (SSA) and soil organic matter (SOM), and (ii) conservative behaviour supported by the inclusion of sediment samples within source convex hull. Sediments from sub-surface layer (2–4 cm) were the dominant source during the first two periods contributing 55 ± 11 and 78 ± 10% respectively, whereas road contribution was only important in the last period (71 ± 14%). Apportionment showed a shift in sediment source (i.e. from forest roads to hillslopes) compared to a previous study in the same catchment before wildfire. The main driver of erosion was attributed to overland flow convergence and consequent rill erosion across burnt hillslopes. Conclusion: The study demonstrated combined use of environmental radionuclides with elemental geochemistry for sediment apportionment within burnt forest plantations and highlighted a switch in predominant source (e.g. sub-surface burnt soil) activated by post-fire rainfall events. The findings in this research will help forest companies to develop strategies to reduce off-site impacts of sediment release after wildfire in forest plantations

    Eutrophication and acidification: Do they induce changes in the dissolvedorganic matter dynamics in the coastal Mediterranean Sea?

    Original research paperTwo mesocosms experiments were conducted in winter 2010 and summer 2011 to examine how increased pCO2and/or nutrient concentrations potentially perturbate dissolved organic matter dynamics in natural microbialassemblages. Thefluorescence signals of protein- and humic-like compounds were used as a proxy for labileand non-labile material, respectively, while the evolution of bacterial populations, chlorophylla(Chla) anddissolved organic carbon (DOC) concentrations were used as a proxy for biological activity. For both seasons,the presence of elevated pCO2did not cause any significant change in the DOC dynamics (p-valueb0.05). Theconditions that showed the greatest changes in prokaryote abundances and Chlacontent were those amendedwith nutrients, regardless of the change in pH. The temporal evolution offluorophores and optical indices re-vealed that the degree of humification of the organic molecules and their molecular weight changed significantlyin the nutrient-amended treatment. The generation of protein-like compounds was paired to increases in theprokaryote abundance, being higher in the nutrient-amended tanks than in the control. Different patterns inthe magnitude and direction of the generation of humic-like molecules suggested that these changes dependedon initial microbial populations and the availability of extra nutrient inputs. Based on our results, it is expected that in the future projected coastal scenarios the eutrophication processes will favor the transformations of labile and recalcitrant carbon regardless of changes in pCO2.MINECO, European Union, Generalitat de Catalunya, CSICVersión del editor3,25

    Mapping gravitational-wave backgrounds in modified theories of gravity using pulsar timing arrays

    We extend our previous work on applying CMB techniques to the mapping of gravitational-wave backgrounds to backgrounds which have non-GR polarisations. Our analysis and results are presented in the context of pulsar-timing array observations, but the overarching methods are general, and can be easily applied to LIGO or eLISA observations using appropriately modified response functions. Analytic expressions for the pulsar-timing response to gravitational waves with non-GR polarisation are given for each mode of a spin-weighted spherical-harmonic decomposition of the background, which permit the signal to be mapped across the sky to any desired resolution. We also derive the pulsar-timing overlap reduction functions for the various non-GR polarisations, finding analytic forms for anisotropic backgrounds with scalar-transverse ("breathing") and vector-longitudinal polarisations, and a semi-analytic form for scalar-longitudinal backgrounds. Our results indicate that pulsar-timing observations will be completely insensitive to scalar-transverse mode anisotropies in the polarisation amplitude beyond dipole, and anisotropies in the power beyond quadrupole. Analogously to our previous findings that pulsar-timing observations lack sensitivity to tensor-curl modes for a transverse-traceless tensor background, we also find insensitivity to vector-curl modes for a vector-longitudinal background.Comment: 35 pages, 8 figures, submitted to Phys.
