    Meropenem heteroresistance in clinical isolates of OXA-48–producing Klebsiella pneumoniae

    OXA-48–producing Klebsiella pneumoniae isolates often show growth of colonies within inhibition zones in carbapenem diffusion assays. The nature of these colonies was investigated in a series of clinical isolates of OXA-48–producing K. pneumoniae obtained in the context of a hospital outbreak, and they were found to be persistent colonies that reproduced again the same phenotype when they were collected and tested in diffusion assays again. The frequency of mutations conferring resistance to meropenem (8 µg/mL) was determined for the same isolates. The average mutation frequency was 5.47·10-6 (range: 2.59·10-8–5.87·10-5), and the analysis of several resistant mutants showed that all of them had mutations in the ompK36 porin gene. Heteroresistance was investigated using population analysis profiling. The profiles were compatible with mutation frequency assays, and all the colonies analyzed were resistant mutants. In OXA-48–producing K. pneumoniae, the growth of persisters seems to be specific of diffusion assays

    Sex-Specific Effects of Early Life Stress on Brain Mitochondrial Function, Monoamine Levels and Neuroinflammation

    Sex differences have been reported in the susceptibility to early life stress and its neurobiological correlates in humans and experimental animals. However, most of the current research with animal models of early stress has been performed mainly in males. In the present study, prolonged maternal separation (MS) paradigm was applied as an animal model to resemble the effects of adverse early experiences in male and female rats. Regional brain mitochondrial function, monoaminergic activity, and neuroinflammation were evaluated as adults. Mitochondrial energy metabolism was greatly decreased in MS females as compared with MS males in the prefrontal cortex, dorsal hippocampus, and the nucleus accumbens shell. In addition, MS males had lower serotonin levels and increased serotonin turnover in the prefrontal cortex and the hippocampus. However, MS females showed increased dopamine turnover in the prefrontal cortex and increased norepinephrine turnover in the striatum, but decreased dopamine turnover in the hippocampus. Sex differences were also found for pro-inflammatory cytokine levels, with increased levels of TNF-α and IL-6 in the prefrontal cortex and hippocampus of MS males, and increased IL-6 levels in the striatum of MS females.     Candida albicans Hyphal Extracellular Vesicles Are Different from Yeast Ones, Carrying an Active Proteasome Complex and Showing a Different Role in Host Immune Response.

    Candida albicans is the principal causative agent of lethal fungal infections, predominantly in immunocompromised hosts. Extracellular vesicles (EVs) have been described as crucial in the interaction of microorganisms with their host. Since the yeast-to-hypha transition is an important virulence trait with great impact in invasive candidiasis (IC), we have addressed the characterization of EVs secreted by hyphal cells (HEVs) from C. albicans, comparing them to yeast EVs (YEVs). YEVs comprised a larger population of bigger EVs with mainly cell wall proteins, while HEVs were smaller, in general, and had a much higher protein diversity. YEVs were able to rescue the sensitivity of a cell wall mutant against calcofluor white, presumably due to the larger amount of cell wall proteins they contained. On the other hand, HEVs also contained many cytoplasmic proteins related to protein metabolism and intracellular protein transport and the endosomal sorting complexes required for transport (ESCRT) pathway related to exosome biogenesis, pointing to an intracellular origin of HEVs. Interestingly, an active 20S proteasome complex was secreted exclusively in HEVs. Moreover, HEVs contained a greater number of virulence-related proteins. As for their immunogenic role, both types of EV presented immune reactivity with human sera from patients suffering invasive candidiasis; however, under our conditions, only HEVs showed a cytotoxic effect on human macrophages and could elicit the release of tumor necrosis factor alpha (TNF-α) by these macrophages. IMPORTANCE This first analysis of HEVs of C. albicans has shown clear differences between them and the YEVs of C. albicans, showing their relevance and possible use in the discovery of new diagnostic markers and treatment targets against C. albicans infections. The data obtained point to different mechanisms of biogenesis of YEVs and HEVs, as well as different involvements in cell biology and host interaction. YEVs played a more relevant role in cell wall maintenance, while HEVs were more closely related to virulence, as they had greater effects on human immune cells. Importantly, an active 20S proteosome complex was described as a fungal-EV cargo. A deeper study of its role and those of many other proteins exclusively detected in HEVs and involved in different relevant biological processes of this fungus could open up interesting new areas of research in the battle against C. albicans

    Effectiveness of a program of physical activity in people with serious mental disorder

    En el presente estudio se analiza el impacto que ha tenido un Programa de actividad física y deporte en un grupo de 28 personas con Trastorno Mental Grave (TMG). Se realizó una evaluación antes del comienzo del Programa y otra a los cuatro meses, tomando medidas antropométricas, de capacidad física funcional, equilibrio y coordinación, así como del funcionamiento social y las actividades de la vida cotidiana. Los resultados mostraron mejorías estadísticamente significativas en aquellas personas que asistieron a más del 50% de las sesiones en todas las variables antropométricas, y de condición física funcional y cualidades motrices, así como en la Escala de Funcionamiento Personal y Social (PSP).Por otro lado, la asistencia al programa fue alta, manteniéndose en el mismo el 80% de las personas que lo iniciaron. Se discute la relevancia de los resultados y la necesidad de implantar este tipo de intervenciones en el TMGIn this paper the effectiveness of a physical activity and sport program is analyzed in a group of 28 people with Severe Mental Illness (SMI). An assessment was made before the start of the program and again after four months, measuring anthropometric variables, physical functional ability, balance and coordination, and social functioning and activities of daily life. The results showed statistically significant improvements in those who attended more than 50% of the sessions in all anthropometric variables, and functional fitness and motor skills, as well as the Personal and Social Performance Scale (PSP).     Using radio astronomical receivers for molecular spectroscopic characterization in astrochemical laboratory simulations: A proof of concept

    We present a proof of concept on the coupling of radio astronomical receivers and spectrometers with chemical reactorsand the performances of the resulting setup for spectroscopy and chemical simulations in laboratory astrophysics. Several experiments including cold plasma generation and UV photochemistry were performed in a 40\,cm long gas cell placed in the beam path of the Aries 40\,m radio telescope receivers operating in the 41-49 GHz frequency range interfaced with fast Fourier transform spectrometers providing 2 GHz bandwidth and 38 kHz resolution. The impedance matching of the cell windows has been studied using different materials. The choice of the material and its thickness was critical to obtain a sensitivity identical to that of standard radio astronomical observations. Spectroscopic signals arising from very low partial pressures of CH3OH, CH3CH2OH, HCOOH, OCS,CS, SO2 (<1E-03 mbar) were detected in a few seconds. Fast data acquisition was achieved allowing for kinetic measurements in fragmentation experiments using electron impact or UV irradiation. Time evolution of chemical reactions involving OCS, O2 and CS2 was also observed demonstrating that reactive species, such as CS, can be maintained with high abundance in the gas phase during these experiments.Comment: Accepted for publication in Astronomy and Astrophysics in September 21, 2017. 16 pages, 18 figure

    A hybrid intelligent system for PID controller using in a steel rolling process

    With the aim to improve the steel rolling process performance, this research presents a novel hybrid system for selecting the best parameters for tuning in open loop a PID controller. The novel hybrid system combines rule based system and Artificial Neural Networks. With the rule based system, it is modeled the existing knowledge of the PID controller tuning in open loop and, with Artificial Neural Network, it is completed the rule based model that allow to choose the optimal parameters for the controller. This hybrid model is tested with a long dataset to obtain the best fitness. Finally, the novel research is validated on a real steeling roll process applying the hybrid model to tune a PID controller which set the input speed in each of the gearboxes of the process

    Costs and benefits of transgenerational induced resistance in arabidopsis

    Recent evidence suggests that stressed plants employ epigenetic mechanisms to transmit acquired resistance traits to their progeny. However, the evolutionary and ecological significance of transgenerational induced resistance (t-IR) is poorly understood because a clear understanding of how parents interpret environmental cues in relation to the effectiveness, stability, and anticipated ecological costs of t-IR is lacking. Here, we have used a full factorial design to study the specificity, costs, and transgenerational stability of t-IR following exposure of Arabidopsis thaliana to increasing stress intensities by a biotrophic pathogen, a necrotrophic pathogen, and salinity. We show that t-IR in response to infection by biotrophic or necrotrophic pathogens is effective against pathogens of the same lifestyle. This pathogen-mediated t-IR is associated with ecological costs, since progeny from biotroph-infected parents were more susceptible to both necrotrophic pathogens and salt stress, whereas progeny from necrotroph-infected parents were more susceptible to biotrophic pathogens. Hence, pathogen-mediated t-IR provides benefits when parents and progeny are in matched environments but is associated with costs that become apparent in mismatched environments. By contrast, soil salinity failed to mediate t-IR against salt stress in matched environments but caused non-specific t-IR against both biotrophic and necrotrophic pathogens in mismatched environments. However, the ecological relevance of this non-specific t-IR response remains questionable as its induction was offset by major reproductive costs arising from dramatically reduced seed production and viability. Finally, we show that the costs and transgenerational stability of pathogen-mediated t-IR are proportional to disease pressure experienced by the parents, suggesting that plants use disease severity as an environmental proxy to adjust investment in t-IR

    Dynamic Ordering and Transverse Depinning of a Driven Elastic String in a Disordered Media

    We examine the dynamics of an elastic string interacting with quenched disorder driven perpendicular and parallel to the string. We show that the string is the most disordered at the depinning transition but with increasing drive partial ordering is regained. For low drives the noise power is high and we observe a 1/f^2 noise signature crossing over to a white noise character with low power at higher drives. For the parallel driven moving string there is a finite transverse critical depinning force with the depinning transition occuring by the formation of running kinks.Comment: 4 pages, 4 postscript figure
