    Docosahexaenoic acid (DHA) has been reported to have a positive impact on many diet-related disease risks, including metabolic syndrome. Although many DHA-enriched foods have been marketed, the impact of different food matrices on the effect of DHA is unknown. As well, the possibility to enhance DHA effectiveness through the co-administration of other bioactives has seldom been considered. We evaluated DHA effects on the serum metabolome administered to volunteers at risk of metabolic syndrome as an ingredient of three different foods. Foods were enriched with DHA alone or in combination with oat beta-glucan or anthocyanins and were administered to volunteers for 4 weeks. Serum samples collected at the beginning and end of the trial were analysed by NMR-based metabolomics. Multivariate and univariate statistical analyses were used to characterize modifications in the serum metabolome and to evaluate bioactive-bioactive and bioactive-food matrix interactions. DHA administration induces metabolome perturbation that is influenced by the food matrix and the co-presence of other bioactives. In particular, when co-administered with oat beta-glucan, DHA induces a strong rearrangement in the lipoprotein profile of the subjects. The observed modifications are consistent with clinical results and indicate that metabolomics represents a possible strategy to choose the most appropriate food matrices for bioactive enrichment

    Extension and application of an observation-based local climate index aimed to anticipate the impact of El Niño–Southern Oscillation events on Colombia

    The Tumaco multivariate index (TMI) is a multidecadal monthly index constructed with unique time series of sea surface temperature, surface air temperature and rain measured at Tumaco bay, in the southern Pacific coast of Colombia, and available since 1961. In this work, this index is re-evaluated after the addition of in situ sea level data, and its properties for different standardization periods are compared against oceanic El Niño–Southern Oscillation (ENSO) and other derived indices. In particular, we propose a modified TMI, hereinafter referred as TMI4, whose potential to identify the expected sign and the amount of future variations of rain induced by ENSO events in Colombia is analysed for selected extreme episodes. Results indicate that after the inclusion of sea level data, TMI4 can anticipate the development of El Niño events before the ENSO 3 and some other sea surface temperature-based regional indices, although its predictability depends on the ENSO type (canonical or Modoki). The explanation is that sea level includes new information into TMI4 on the onset of El Niño events. In particular, the signal of intraseasonal sea level anomalies carried by downwelling Kelvin waves is detected at Tumaco tide-gauge. Moreover, the analysis of the differences, both in magnitude and spatial distribution, of rainfall anomalies induced by positive (El Niño) and negative (La Niña) ENSO events characterized by TMI4 are regionally presented. As a result, we find that TMI4 is especially suited for extensive northern and western areas of mainland Colombia.     Guidelines for nutritional intervention studies on BEF effectiveness with a special focus on pilot studies

    The guidelines presented here have been developed through consultation with PATHWAY-27 project partners to provide practical guidance for designing, implementing and reporting pilot intervention studies using bioactive enriched foods (BEF). These guidelines update and complement previously published guidance documents, but are unique in providing practical information and troubleshooting based on the PATHWAY-27 pilot studies. The aim of PATHWAY-27 is to evaluate the effectiveness of BEF containing docosahexaenoic acid (DHA), anthocyanins (AC) and beta-glucan (BG) alone or in combination, on improving risk factors of the metabolic syndrome (MetS). These compounds were chosen for their known effects as single compounds, and considered as ingredients of BEF in three different widely consumed food matrices (dairy, bakery and egg products). This approach of using BEF, compared to single pure compounds, will allow a better understanding of possible bioactive synergisms and bioactive-matrix interactions. This knowledge will increase the potential for exploitation of bioactive and BEF commercialization by the food industry. A pilot trial is a small-scale study that is used to test the physiological effects and participant acceptance of prototype formulations in order to select food products to be tested in a larger scale trial. The pilot is also a great opportunity to test the data collection methods, procedures and logistics that can provide early identification of problems in order to find suitable solutions that can be implemented to improve the quality and efficiency of the main trial. With examples gathered from the PATHWAY-27 project, this guidance document describes the requirements for the formulation and production of BEF, the design and conductance of a pilot trial, and how the data and information gathered can be used to inform further larger trials and the best practice for public reporting

    Systematic Review and Meta-Analysis on the Effects of Astaxanthin on Human Skin Ageing

    Context: Astaxanthin (ASX), a xanthophyll carotenoid derived from microalgae Haematococcus pluvialis, mitigating skin photoaging and age-related skin diseases by its antioxidant and anti-inflammatory effects in animal studies. Objective: The aim was to systematically evaluate if ASX applications have anti-ageing effects in humans. Methods: A comprehensive search of PubMed, Scopus and Web of Science found a total of eleven studies. Nine randomised, controlled human studies assessed oral ASX effects and two open-label, prospective studies evaluated topical, oral-topical ASX effects on skin ageing. GetData Graph Digitizer was used to extract mean values and standard deviations of baseline and endpoint, and Cochrane Collaboration’s tool assessed RoB for all included studies. Review Manager 5.4 was used to conduct meta-analysis of RCTs; the results were reported as effect size ± 95% confidence interval. Results: Oral ASX supplementation significantly restored moisture content (SMD = 0.53; 95% CI = 0.05, 1.01; I2 = 52%; p = 0.03) and improved elasticity (SMD = 0.77; 95% CI = 0.19, 1.35; I2 = 75%; p = 0.009) but did not significantly decrease wrinkle depth (SMD = −0.26; 95% CI = −0.58, 0.06; I2 = 0%; p = 0.11) compared to placebo. Open-label, prospective studies suggested slightly protective effects of topical and oral-topical ASX applications on skin ageing. Conclusions: Ingestion and/or topical usages of ASX may be effective in reducing skin ageing and have promising cosmetical potential, as it improves moisture content and elasticity and reduces wrinkles

    The role of bioactives in energy metabolism and metabolic syndrome

    Some food bioactives potentially exert anti-obesity effects. Anthocyanins (ACN), catechins, β-glucan (BG) and n-3 long chain PUFA (LCPUFA) are among the most promising candidates and have been considered as a strategy for the development of functional foods counteracting body weight gain. At present, clinical trials, reviews and meta-analyses addressing anti-obesity effects of various bioactives or bioactive-rich foods show contradictory results. Abdominal obesity is an important criterion for metabolic syndrome (MetS) diagnosis along with glucose intolerance, dyslipidaemia and hypertension. Food bioactives are supposed to exert beneficial effects on these parameters, therefore representing alternative therapy approaches for the treatment of MetS. This review summarises outcomes on MetS biomarkers in recent clinical trials supplementing ACN, catechins, BG and n-3 LCPUFA, focusing mainly on anti-obesity effects. Overall, it is clear that the level of evidence for the effectiveness varies not only among the different bioactives but also among the different putative health benefits suggested for the same bioactive. Limited evidence may be due to the low number of controlled intervention trials or to inconsistencies in trial design, i.e. duration, dose and/or the method of bioactive supplementation (extracts, supplements, rich or enriched food). At present, the question ‘Are bioactives effective in weight management and prevention of metabolic syndrome?’ remains inconclusive. Thus, a common effort to harmonise the study design of intervention trials focusing on the most promising bioactive molecules is urgently needed to strengthen the evidence of their potential in the treatment of obesity, MetS and related diseases

    Singularities of surface mixing activity in the Western Mediterranean influence bluefin tuna larval habitats

    Understanding how the surface dynamics of the ocean influence the spawning and larval ecology of many large pelagic species, in particular tuna species, is a major challenge. For temperate tunas, the selection of geographically restricted spawning grounds is influenced by environmental conditions, but the influence of surface mixing properties on the early life stages of these species remains poorly understood. Here, based on ichthyoplankton samples collected over 4 yr and satellite-derived finite size Lyapunov exponents (FSLEs), we examined how horizontal mixing activity drives the probability of presence of Atlantic bluefin tuna Thunnus thynnus larvae. We further analyzed the spatial and temporal scales of the FSLE variability at which the relationship between larval presence and mesoscale activity is maximized. We found that moderate mixing activity strongly favors the spatial-temporal distribution of larval habitats, evidencing an optimal environmental window of bluefin tuna spawning and early life development within the mesoscale dynamics. During the spawning season, the Balearic Sea presents a unique spatial and temporal hydrodynamic scenario within the Western Mediterranean. These results can be used for developing oceanographic indicators and improving larval abundance indices that are currently used in Atlantic bluefin tuna stock assessments.En prensa2,48

    A Community Program of Integrated Care for Frail Older Adults : +AGIL Barcelona

    Objectives: To assess the 3-month impact on physical function of a program for community-dwelling frail older adults, based on the integration of primary care, geriatric medicine, and community resources, implemented in \u201creal life\u201d. Design: Interventional cohort study. Setting: Primary care in Barcelona, Spain. Participants: Individuals aged 6580 years (n=134), presenting at least one sign of frailty (i.e., slow gait speed, weakness, memory complaints, involuntary weight loss, poor social support). Intervention: After frailty screening by the primary care team, candidates were referred to a geriatric team (geriatrician + physical therapist), who performed a comprehensive geriatric assessment and designed a tailored multidisciplinary intervention in the community, including a) multi-modal physical activity (PA) sessions, b) promotion of adherence to a Mediterranean diet c) health education and d) medication review. Measurements: Participants were assessed based on a comprehensive geriatric assessment including physical performance (Short Physical Performance Battery -SPPB- and gait speed), at baseline and at a three month follow-up. Results: A total of 112 (83.6%) participants (mean age=80.8 years, 67.9% women) were included in this research. Despite being independent in daily life, participants\u2019 physical performance was impaired (SPPB=7.5, SD=2.1, gait speed=0.71, SD=0.20 m/sec). After three months, 90.2% of participants completed 657.5 physical activity sessions. The mean improvements were +1.47 (SD 1.64) points (p<0.001) for SPPB, +0.08 (SD 0.13) m/sec (p<0.001) for gait speed, 125.5 (SD 12.10) sec (p<0.001) for chair stand test, and 53% (p<0.001) improved their balance. Results remained substantially unchanged after stratifying the analyses according to the severity of frailty. Conclusions: Our results suggested that a \u201creal-world\u201d multidisciplinary intervention, integrating primary care, geriatric care, and community services may improve physical function, a marker of frailty, within 3 months. Further studies are needed to address the long-term impact and scalability of this implementation program
