    Iridium Cyclooctene Complex Forms a Hyperpolarization Transfer Catalyst Before Converting to a Binuclear C-H Bond Activation Product Responsible for Hydrogen Isotope Exchange

    [IrCl(COE)2]2 ( 1 ) reacts with pyridine and H2 to form crystallo-graphically characterized IrCl(H)2(COE)(py)2 ( 2 ). 2 undergoes pyridine loss to form 16-electron IrCl(H)2(COE)(py) (3) with equivalent hydride ligands. When this reaction is studied with parahydrogen, 1 efficiently achieves the hyperpolarization of free pyridine (and nicotinamide, nicotine, 5-aminopyrimidine and 3,5-lutudine) via signal amplification by reversible exchange (SABRE) and hence reflects a simple and readily available precatayst for this process. 2 reacts further over 48 hrs at 298 K to form crystallographically characterized (Cl)(H)(py)(μ-Cl)(μ-H)(κ-μ-NC5H4)Ir(H)(py)2 (4). This dimer is shown to be active in the hydrogen isotope exchange process that is used in radiophar-maceutical preparations. Furthermore, while [Ir(H)2(COE)(py)3]PF6 ( 6 ) forms on addition of AgPF6 to 2 , its stability precludes its efficient involvement in SABRE

    Modelling the seismic performance of rooted slopes from individual root–soil interaction to global slope behaviour

    Many natural and man-made slopes are planted with vegetation, and it is known that this can increase the stability of slopes under static conditions. There is anecdotal evidence that vegetated slopes also perform better than fallow slopes during earthquakes. However, the study of the dynamic behaviour of slopes planted with species having dichotomous (‘woody’) roots is relatively rare owing to the extreme expense and difficulty involved in conducting full-scale dynamic testing on shrubs and trees. In this paper, dynamic centrifuge testing and supporting numerical modelling have been conducted to study this problem. In the centrifuge modelling, ABS plastic rods are used to simulate repeatably the mechanical properties of real roots. The numerical modelling work consisted of two parts. First, a computationally-efficient beam-on-non-linear-Winkler-foundation (BNWF) model using existing p–y formulations from piling engineering was employed to produce a macro-element describing the individual root and soil interaction both pre- and post-failure. By adding contributions from the different root analogues of different diameters, smeared continuum properties were derived that could be included in a fully dynamic, plane-strain continuum, finite-element model in a straightforward way. The BNWF approach was validated against large direct shear tests having stress conditions simulating those in the centrifuge at different potential slip plane depths. The conversion to smeared properties for global time-history analysis of the slope was validated by comparing the continuum finite-element results with the centrifuge test data in terms of both the dynamic response and permanent deformations at the crest, and these demonstrated good agreement. Owing to the simplicity of the BNWF approach and its ability to consider variable root geometries and properties, along with variation of soil properties with depth, it is suggested that the validated approach described will be useful in linking individual root–soil interaction characteristics (root strength and stiffness, diameter variation, root spacing and so on) to global slope behaviour

    Regional phylogeographic studies have long been conducted in the southeastern United States for a variety of species. With some exceptions, many of these studies focus on single species or single clades of organisms, and those considering multiple species tend to focus on deep historical breaks causing differentiation. However, in many species more recent factors may be influencing genetic diversity. To understand the roles of historic and contemporary processes in structuring genetic diversity, we reanalyzed existing genetic data from Southeast of North America using approaches gleaned from phylogeographic and landscape genetic literature that were implemented across species including AMOVAs, PCoAs, Species Distribution Modelling, and tests of isolation by distance, environment, and habitat instability. Genetic variance was significantly partitioned by ecoregions, watersheds, and across phylogeographic breaks in the majority of species. Similarly, genetic variation was significantly associated with some combination of geographic or environmental distance or habitat instability in most species. Patterns of genetic variation were largely idiosyncratic across species. While habitat instability over time is significantly correlated with genetic diversity in some species, it appears generally less important than isolation by geographic or environmental distance. Our results suggest that many factors, both historical and contemporary, impact genetic diversity within a species, and more so, that these patterns aren’t always similar in closely related species. This supports the importance of species- specific factors and cautions against assumptions that closely related species will respond to historical and contemporary forces in similar ways

    Altered urothelial ATP signaling in a major subset of human overactive bladder patients with pyuria

    Overactive Bladder (OAB) is an idiopathic condition, characterized by urgency, urinary frequency, and urgency incontinence, in the absence of routinely traceable urinary infection. We have described microscopic pyuria (≥10 wbc/μl) in patients suffering from the worst symptoms. It is established that inflammation is associated with increased ATP release from epithelial cells, and extracellular ATP originating from the urothelium following increased hydrostatic pressure is a mediator of bladder sensation. Here, using bladder biopsy samples, we have investigated urothelial ATP signaling in OAB patients with microscopic pyuria. Basal, but not stretch-evoked, release of ATP was significantly greater from the urothelium of OAB patients with pyuria than from non-OAB patients or OAB patients without pyuria (<10 wbc/μl). Basal ATP release from the urothelium of OAB patients with pyuria was inhibited by the P2 receptor antagonist suramin and abolished by the hemichannel blocker carbenoxolone, which differed from stretch-activated ATP release. Altered P2 receptor expression was evident in the urothelium from pyuric OAB patients. Furthermore, intracellular bacteria were visualized in shed urothelial cells from ∼80% of OAB patients with pyuria. These data suggest that increased ATP release from the urothelium, involving bacterial colonization, may play a role in the heightened symptoms associated with pyuric OAB patients

    Ergot alkaloids in feeds and forages are a worldwide concern. Consumption of ergot alkaloids by ruminant livestock can range from extreme cases that threaten the life of the animal to more frequent and chronic outcomes where livestock productivity is decreased. Consumption of ergot alkaloids alters ruminant physiology such that it outwardly manifests in lower animal intake and gain, decreased reproductive efficiency, and a compromised circulatory system. This talk will cover current research that is improving our understanding of how ergot alkaloids alter cell and tissue physiology that results in the compromised growth and reproduction observed at the whole animal level

    Absence of differential predation on rats by Malaysian Barn Owls in oil palm plantations

    Barn Owls (Tyto alba javanica) have been widely introduced in Malaysian oil palm plantations to control rodent pests. However, their effectiveness in regulating rodent populations is unknown. We investigated whether Barn Owls selected prey with respect to size and sex classes based on data from 128 pellets of Barn Owls compared to 1292 live-trapped rats in an oil palm plantation in Malaysia. The birds mostly fed on Rattus rail as diardii, the most commonly trapped species. Body mass of prey consumed was predicted based on models derived from measurements from trapped rats. Sex of prey was determined by pelvic measurements with reference to those taken from specimens of known gender. There was no clear selection of prey by Barn Owls in relation to size or sex of prey, and no difference in the body mass of prey between the owls' breeding and nonbreeding seasons. The absence of differential predation in Barn Owls may partly explain the lack of dear evidence that they regulate rodent populations and thus act as successful biological control agents

    Improving the hyperpolarization of (31)p nuclei by synthetic design

    Traditional (31)P NMR or MRI measurements suffer from low sensitivity relative to (1)H detection and consequently require longer scan times. We show here that hyperpolarization of (31)P nuclei through reversible interactions with parahydrogen can deliver substantial signal enhancements in a range of regioisomeric phosphonate esters containing a heteroaromatic motif which were synthesized in order to identify the optimum molecular scaffold for polarization transfer. A 3588-fold (31)P signal enhancement (2.34% polarization) was returned for a partially deuterated pyridyl substituted phosphonate ester. This hyperpolarization level is sufficient to allow single scan (31)P MR images of a phantom to be recorded at a 9.4 T observation field in seconds that have signal-to-noise ratios of up to 94.4 when the analyte concentration is 10 mM. In contrast, a 12 h 2048 scan measurement under standard conditions yields a signal-to-noise ratio of just 11.4. (31)P-hyperpolarized images are also reported from a 7 T preclinical scanner

    Alkylating agents constitute a major class of frontline chemotherapeutic drugs that inflict cytotoxic DNA damage as their main mode of action, in addition to collateral mutagenic damage. Numerous cellular pathways, including direct DNA damage reversal, base excision repair (BER) and mismatch repair (MMR), respond to alkylation damage to defend against alkylation-induced cell death or mutation. However, maintaining a proper balance of activity both within and between these pathways is crucial for a favourable response of an organism to alkylating agents. Furthermore, the response of an individual to alkylating agents can vary considerably from tissue to tissue and from person to person, pointing to genetic and epigenetic mechanisms that modulate alkylating agent toxicity

    Experiencing visual impairment in a lifetime home: an interpretative phenomenological inquiry

    Lifetime home standards (LTHS) are a set of standards aimed at making homes more accessible. Previous research, however, indicates that LTHS do not adequately meet the needs of those with sensory impairments. Now, with visual impairment set to increase globally and acknowledging the recognised link between quality of dwelling and wellbeing, this article aims to examine the experiences of visually impaired people living in lifetime homes. The objectives are to investigate existing lifetime homes and to identify whether LTHS meet occupants’ needs. Qualitative semi-structured interviews were carried out with six visually impaired people living in homes designed to LTHS in Northern Ireland. Collected data was analysed using interpretative phenomenological analysis identifying three super-ordinate themes: (1) living with visual impairment; (2) design considerations and (3) coping strategies. A core theme of balance between psychological and physical needs emerged through interconnection of super-ordinate themes. Although there are benefits to living in lifetime homes, negative aspects are also apparent with occupants employing several coping strategies to overcome difficulties. Whilst residents experience negative emotions following visual impairment diagnoses, results suggest that occupants still regard their homes as key places of security and comfort in addition to then highlighting the need for greater consideration of specific individual needs within general guidelines
