    Ozone autohaemotherapy protects against ketamine hydrochloride® induced liver and muscle damage in baboons

    Ozone is currently under scrutiny because of various claims of beneficial effect in disease. In order to shed some light on this we assessed the acute and chronic effect of O3 autohaemotherapy (AHT) on liver and muscle damage in baboons. Five percent of the total blood volume of a baboon was treated with O2 and O3. Eleven baboons were acutely treated with an O2/O3 gas mixture containing 20, 40 and 80ìg/ml ozone. Five were treated with pure O2 and three received no treatment to assess the effect of the ketamine hydrochloride anaesthesia. Blood samples were collected before treatment and after 4, 24 and 48 h. Anaesthesia increased aspartate aminotransferase (AST), alanine aminotransferase (ALT) and creatine kinase (CK) levels markedly. O3-AHT had a protective effect, since enzyme levels were lower. O2-AHT had no protective effect on liver and muscle damage. An O2/O3 gas mixture containing 40 ìg/ml O3 was used for chronic O3-AHT (n=6) treatment. The animals were treated at 0, 24 and 48 h. Blood was collected before treatment and again after 4, 24, 28, 48, 52, 72 and 96 h. ALT levels increased and remained elevated. AST levels increased during the four hours following each treatment and remainedelevated. CK levels increased markedly during the four hours following treatment, but decreased after treatment was stopped. The magnitude of changes was small and does not support the view that infusion of ozonated of blood is toxic

    Agtergrond Kindermishandeling is \'n sosiale probleem met verreikende gevolge. Ernstige onderrapportering van gevalle vind plaas, en geen studies kon opgespoor word wat die Suid-Afrikaanse konteks van die probleem ondersoek nie. In hierdie studie is gepoog om die profiel van die slagoffers van noodlottige kindermishandeling in die Suid-Vrystaat te ondersoek. Metodes Alle kindersterftes van kinders van tien jaar en jonger is uit die register van sterftes by die SAPD- (Suid-Afrikaanse Polisiediens-) lykshuis, Bloemfontein, geïdentifiseer. Hierdie sterftes se verslae is vir gevalle van noodlottige kindermishandeling geëvalueer. Irrelevante verslae is geëlimineer totdat \'n groep moontlike gevalle van noodlottige kindermishandeling verkry is. In gevalle waar daar twyfel oor die oorsaak van dood was, is \'n kliniese assistent in Geregtelike Geneeskunde geraadpleeg. In sommige gevalle het die assistent aanbeveel dat \'n geskiedenis uit polisieverslae verkry moes word. Indien gevalle nie op so \'n wyse bevestig kon word nie, is dit in die sogenaamde ‘grys area\' ingesluit. Negentien gevalle is geïdentifiseer en vier is in die grys area geplaas. Resultate Uit die 19 gevalle was die meerderheid vroulik (14). Die mediaanouderdom van slagoffers was twee jaar. Die slag-offers toon \'n eweredige verspreiding t.o.v. ras. Geen patroon kon geïdentifiseer word in die jaarlikse en maandelikse verspreiding van sterftes nie. Die meeste van die slagoffers, 73.7% (14 van 19 gevalle), was van normale massa, en geen slagoffers het aan \'n wanvoedingsiekte gely nie. Hoofbese-rings was die algemeenste oorsaak van sterftes (63.2%), gevolg deur veelvuldige beserings (21.2%). Gevolgtrekking Indien dokters bedag is op die tekens van kindermishandeling, kan gevalle vroeg geïdentifiseer word. Sodoende kan \'n beduidende verlaging in mortaliteit en morbiditeit bewerkstellig Introduction Child abuse is a social problem with far-reaching consequences. Serious underreporting of cases occurs, and no studies could be found which investigates the South African context of the problem. In this study we aimed to determine the profile of victims of fatal child abuse in the Southern Free State. Methods All child deaths of children 10 years and younger in the period 1 January 1995 to 31 December 2003 were identified in the register of deaths at the SA Police Mortuary, Bloemfontein. The reports of these deaths were evaluated to identify possible cases of fatal child abuse. Irrelevant reports were eliminated until a group of possible cases of fatal child abuse were identified. In cases where there was uncertainty about the cause of death, a registrar in Forensic Medicine was consulted. In some cases she advised that a history be obtained from police reports. Cases which could not be confirmed in this way were included in a so-called grey area. Nineteen cases were identified, and a further 4 placed in the grey area. Results Of the 19 cases, the majority were female (14). The median age of cases was 2 years. Cases showed an even distribution regarding race. No patterns regarding annual or monthly distribution were found. Most victims, 73.7% (14 of 19 cases), were of normal weight and no victims suffered from malnutrition. Head injuries were the most common cause of death (63.2%) followed by multiple injuries (21.2%). Conclusion If doctors are aware of the signs of child abuse cases can be identified early.     Identifying sequence variation in cation channel sperm associated genes in Cape mountain zebra (Equus zebra zebra)

    The Cape mountain zebra (Equus zebra zebra) has recovered from near extinction over more than eight decades. While their numbers have increased, populations remain isolated with low genetic diversity. With more than 75 new populations being founded and more than 4800 extant animals, conservation management strategies are being implemented to mitigate risk of losses in genetic diversity and reproductive fitness. One objective is to identify reproductive characteristics that may improve population growth. Cation channel sperm (CatSper) genes play an important role in hyperactivation of sperm during fertilization. Mutations in these genes lead to reduced fertility and even infertility. Ten male zebras were sampled from a group that were translocated in 2016 in order to found a new population. Single nucleotide polymorphisms (SNPs) were identified in three of the CatSper genes (1 - 3). Lack of variation was observed in all exons, with only four SNPs being identified in the intronic regions in close proximity to exons 1, 2, 7, 8, and 9 of CatSper 1. These results may contribute to the pre-identification of males for new founder populations to ensure population growth and viability, and may be a useful tool for selection against low-producing individuals

    Uptake and transport of novel amphiphilic polyelectrolyte-insulin nanocomplexes by caco-2 cells - towards oral insulin

Copyright SpringerPurpose: The influence of polymer architecture on cellular uptake and transport across Caco-2 cells of novel amphiphilic polyelectrolyte-insulin nanocomplexes was investigated. Method: Polyallylamine (PAA) (15 kDa) was grafted with palmitoyl chains (Pa) and subsequently modified with quaternary ammonium moieties (QPa). These two amphiphilic polyelectrolytes (APs) were tagged with rhodamine and their uptake by Caco-2 cells or their polyelectrolyte complexes (PECs) with fluorescein isothiocyanate-insulin (FITC-insulin) uptake were investigated using fluorescence microscopy. The integrity of the monolayer was determined by measurement of transepithelial electrical resistance (TEER). Insulin transport through Caco-2 monolayers was determined during TEER experiments. Result: Pa and insulin were co-localised in the cell membranes while QPa complexes were found within the cytoplasm. QPa complex uptake was not affected by calcium, cytochalasin D or nocodazole. Uptake was reduced by co-incubation with sodium azide, an active transport inhibitor. Both polymers opened tight junctions reversibly where the TEER values fell by up to 35 % within 30 minutes incubation with Caco-2 cells. Insulin transport through monolayers increased when QPa was used (0.27 ngmL-1 of insulin in basal compartment) compared to Pa (0.14 ngmL-1 of insulin in basal compartment) after 2 hours. Conclusion: These APs have been shown to be taken up by Caco-2 cells and reversibly open tight cell junctions. Further work is required to optimise these formulations with a view to maximising their potential to facilitate oral delivery of insulin.Peer reviewe

    Monitoring compliance of CITES lion bone exports from South Africa

    From 2008 to 2018, South Africa permitted the export of captive-bred African lion (Panthera leo) skeletons to Southeast Asia under CITES Appendix II. Legal exports rose from approximately 50 individuals in 2008 to a maximum of 1,771 skeletons in 2016, and has led to ongoing concerns over possible laundering of non-lion, multiple-source and wild-sourced bones. South Africa is required under its obligations to CITES to employ mechanisms for monitoring and reporting trade, and to limit the potential for illegal trade and laundering of lion and other large felid bones. Monitoring tools for legal trade are critical to compliance with CITES. Here we evaluate the CITES-compliance procedure implemented by South Africa for export of lion bones and identify six essential general points for consideration in the implementation of animal export quota compliance protocols. We provide specific insight into the South African lion bone export monitoring system through: i) outlining the protocols followed; ii) assessing the utility of cranial morphology to identify species; iii) evaluating skeleton consignment weight as a monitoring tool; and iv) presenting molecular (DNA) species assignment and pairwise-comparative sample matching of individuals. We describe irregularities and illicit behaviour detected in the 2017 and 2018 lion bone quotas. Notably, we report that the compliance procedure successfully identified and prevented the attempted laundering of a tiger (P. tigris) skeleton in 2018. We emphasise the utility of mixed-method protocols for the monitoring of compliance in CITES Appendix II export quota systems

    Differentiation of two South African otter species (Aonyx capensis and Lutra maculicollis) from spraint based on partial CytB primer sets

    AbstractAccurate species identification based on visual cues can be challenging due to morphological similarities and the cryptic nature of certain species. Thus a more conclusive method of identification is required, namely DNA barcoding. This is the case regarding two South African otter species, Cape Clawless otter (Aonyx capensis) and the spotted necked otter (Lutra maculicollis). Due to the cryptic nature of these animals faecal samples, known as spraints, are the easiest way of confirming the presence of the animal in an area. In this study, we compared results obtained for universal and partial CytB primer sets on collected spraint and tissue control samples. Universal CytB primers revealed a low percentage of amplified otter species from faecal samples (species specific amplification success of 10.9%) whereas, the partial CytB primer set resulted in successful amplification of 45 out of 55 (82%) samples. We were thus able to positively differentiate between the two otter species using the partial CytB primer set developed in this study. The ability to accurately identify species using partial DNA will be beneficial in understanding numerous aspects of the behaviour and ecological importance of animals in their environment

    A critique of the Global Pact for the Environment: A stillborn initiative or the foundation for Lex Anthropocenae?

    In May 2018 the process which may ultimately lead to the negotiation of a legally binding Global Pact for the Environment formally commenced under the auspices of the United Nations General Assembly. Expectations for the Pact are high, evidenced in particular by its multiple and overlapping objectives: to serve as a generic binding instrument of international environmental law (IEL) principles ; to integrate, consolidate, unify and ultimately entrench many of the fragmented principles of IEL; and to constitute the first global environmental human rights instrument. In the wake of the impending intergovernmental process, the paper offers a thorough critique of the draft Pact in its present iteration. We do so with the aim of evaluating the strengths and weaknesses of the present draft Pact by interrogating: a) its diplomatic and symbolic relevance and possible unique contribution at the policy level to global environmental law and governance; and b) its potential at the operational level of IEL and global environmental governance, focusing on the extent to which the draft Pact accommodates both existing and more recent rules and principles for environmental protection. As the Pact’s primary ambition is to become a universally binding global treaty, it would be churlish not to recognise its potential for innovation, as well as the considerable opportunity that the negotiation of the Pact will have to generate broad-sweeping and positive impacts. However, our central thesis is that only if the Global Pact were to incorporate ambitious normative provisions to strengthen those public and private global governance efforts that aim to halt the deterioration of Earth system integrity, as well as to maintain and improve integrity, will it be able to offer a firm foundation of the type of Anthropocene Law, termed here as the Lex Anthropocenae, required to confront head-on the deep socio-ecological crisis of the Anthropocene

    Ethnomedicinal use of African pangolins by traditional medical practitioners in Sierra Leone

    Abstract Background: Pangolins (Manidae) have long been used for traditional medicinal purposes in most parts of sub-Saharan Africa. However, very little is known about the extent of this use, the body parts that are used and the ailments these practices are attempting to cure or alleviate. Pangolin body parts are used extensively and frequently by traditional medical practitioners in Sierra Leone. Methods: A total of 63 traditional medical practitioners consented and were interviewed using semi-structured questionnaires on the traditional medicinal use of pangolin body parts. The use value, informant agreement ratio and use agreement value for each pangolin part was calculated to ascertain the most sought after body part, the level of knowledge dissemination among traditional medical practitioners about body parts and the most culturally significant body part. Results: It was found that 22 pangolin parts are used to treat various ailments and conditions under 17 international categories of diseases. The highest use value was recorded for scales while eyes had the highest level of consensus among the traditional medical practitioners. The highest use value and informant agreement ratio for scales were recorded for spiritual ailments. Scales were the most culturally significant body part according to the use agreement value

    The comparative gastrointestinal morphology of Jaculus jaculus (Rodentia) and Paraechinus aethiopicus (Erinaceomorpha)

    Jaculus jaculus (Lesser Egyptian jerboa) and Paraechinus aethiopicus (Desert hedgehog) are small mammals which thrive in desert conditions and are found, amongst others, in the Arabian Peninsula. J. jaculus is omnivorous while P. aethiopicus is described as being insectivorous. The study aims to describe the gastrointestinal tract (GIT) morphology of these animals which differ in diet and phylogeny. The GITs of J. jaculus (n=8) and P. aethiopicus (n=7) were weighed, photographed, and the length, basal surface areas and luminal surface areas of each of the anatomically distinct gastrointestinal segments were determined. The internal aspects of each area were examined and photographed while representative histological sections of each area were processed to wax and stained using haematoxylin and eosin. Both species had a simple unilocular stomach which was confirmed as wholly glandular on histology sections. P. aethiopicus had a relatively simple GIT which lacked a caecum. The caecum of J. jaculus was elongated, terminating in a narrow cecal appendix which contained lymphoid tissue on histological examination. The internal aspect of the proximal colon revealed distinct V-shaped folds.     Extinctions, genetic erosion and conservation options for the black rhinoceros (Diceros bicornis)

    The black rhinoceros is again on the verge of extinction due to unsustainable poaching in its native range. Despite a wide historic distribution, the black rhinoceros was traditionally thought of as depauperate in genetic variation, and with very little known about its evolutionary history. This knowledge gap has hampered conservation efforts because hunting has dramatically reduced the species’ once continuous distribution, leaving five surviving gene pools of unknown genetic affinity. Here we examined the range-wide genetic structure of historic and modern populations using the largest and most geographically representative sample of black rhinoceroses ever assembled. Using both mitochondrial and nuclear datasets, we described a staggering loss of 69% of the species’ mitochondrial genetic variation, including the most ancestral lineages that are now absent from modern populations. Genetically unique populations in countries such as Nigeria, Cameroon, Chad, Eritrea, Ethiopia, Somalia, Mozambique, Malawi and Angola no longer exist. We found that the historic range of the West African subspecies (D. b. longipes), declared extinct in 2011, extends into southern Kenya, where a handful of individuals survive in the Masai Mara. We also identify conservation units that will help maintain evolutionary potential. Our results suggest a complete re-evaluation of current conservation management paradigms for the black rhinoceros
