    Modeling the material resistance of wood—part 2:Validation and optimization of the meyer-veltrup model

    Service life planning with timber requires reliable models for quantifying the effects of exposure-related parameters and the material-inherent resistance of wood against biotic agents. The Meyer-Veltrup model was the first attempt to account for inherent protective properties and the wetting ability of wood to quantify resistance of wood in a quantitative manner. Based on test data on brown, white, and soft rot as well as moisture dynamics, the decay rates of different untreated wood species were predicted relative to the reference species of Norway spruce (Picea abies). The present study aimed to validate and optimize the resistance model for a wider range of wood species including very durable species, thermally and chemically modified wood, and preservative treated wood. The general model structure was shown to also be suitable for highly durable materials, but previously defined maximum thresholds had to be adjusted (i.e., maximum values of factors accounting for wetting ability and inherent protective properties) to 18 instead of 5 compared to Norway spruce. As expected, both the enlarged span in durability and the use of numerous and partly very divergent data sources (i.e., test methods, test locations, and types of data presentation) led to a decrease in the predictive power of the model compared to the original. In addition to the need to enlarge the database quantity and improve its quality, in particular for treated wood, it might be advantageous to use separate models for untreated and treated wood as long as the effect of additional impact variables (e.g., treatment quality) can be accounted for. Nevertheless, the adapted Meyer-Veltrup model will serve as an instrument to quantify material resistance for a wide range of wood-based materials as an input for comprehensive service life prediction software

    Modelling the Material Resistance of Wood—Part 3: Relative Resistance in above- and in-Ground Situations—Results of a Global Survey

    Durability-based designs with timber require reliable information about the wood properties and how they affect its performance under variable exposure conditions. This study aimed at utilizing a material resistance model (Part 2 of this publication) based on a dose–response approach for predicting the relative decay rates in above-ground situations. Laboratory and field test data were, for the first time, surveyed globally and used to determine material-specific resistance dose values, which were correlated to decay rates. In addition, laboratory indicators were used to adapt the material resistance model to in-ground exposure. The relationship between decay rates in- and above-ground, the predictive power of laboratory indicators to predict such decay rates, and a method for implementing both in a service life prediction tool, were established based on 195 hardwoods, 29 softwoods, 19 modified timbers, and 41 preservative-treated timbers

    The Anticancer Plant Triterpenoid, Avicin D, Regulates Glucocorticoid Receptor Signaling: Implications for Cellular Metabolism

    Avicins, a family of apoptotic triterpene electrophiles, are known to regulate cellular metabolism and energy homeostasis, by targeting the mitochondria. Having evolved from “ancient hopanoids,” avicins bear a structural resemblance with glucocorticoids (GCs), which are the endogenous regulators of metabolism and energy balance. These structural and functional similarities prompted us to compare the mode of action of avicin D with dexamethasone (Dex), a prototypical GC. Using cold competition assay, we show that Avicin D competes with Dex for binding to the GC receptor (GR), leading to its nuclear translocation. In contrast to Dex, avicin-induced nuclear translocation of GR does not result in transcriptional activation of GC-dependent genes. Instead we observe a decrease in the expression of GC-dependent metabolic proteins such as PEPCK and FASN. However, like Dex, avicin D treatment does induce a transrepressive effect on the pro-inflammatory transcription factor NF-κB. While avicin's ability to inhibit NF-κB and its downstream targets appear to be GR-dependent, its pro-apoptotic effects were independent of GR expression. Using various deletion mutants of GR, we demonstrate the requirement of both the DNA and ligand binding domains of GR in mediating avicin D's transrepressive effects. Modeling of avicin-GR interaction revealed that avicin molecule binds only to the antagonist confirmation of GR. These findings suggest that avicin D has properties of being a selective GR modulator that separates transactivation from transrepression. Since the gene-activating properties of GR are mainly linked to its metabolic effects, and the negative interference with the activity of transcription factors to its anti-inflammatory and immune suppressive effects, the identification of such a dissociated GR ligand could have great potential for therapeutic use

    Über die Wirtschaft der Antike sind wir gut informiert. Für den Waren- und Geldverkehr standen in den Städten eigene Gebäude zur Verfügung, die im römischen Reich von Speicherbauten in bedeutenden Wirtschaftszentren und Hafenorten bis zu Bank- und Börsengebäuden an den zentralen Plätzen der Städte reichten. Diese Bauten wurden von Archäologen und Bauhistorikern vielfach untersucht, wobei Bauweise und Gestaltung im Vordergrund standen. Wirtschaftshistoriker befassten sich intensiv mit Handels- und Geldgeschäften, ohne Zusammenhänge mit Baulichkeiten in den Blick zu nehmen. Interdisziplinäre Zusammenarbeit eröffnet neue Perspektiven. Mit dem Wissen über wirtschaftliche Vorgänge lassen sich Bauten besser verstehen, und Bauwerke können ökonomische Vorgänge sichtbar machen. Das wurde beim erstmaligen Gedankenaustausch zwischen den verschiedenen Forschungsdisziplinen rasch deutlich. Der vorliegende Tagungsband enthält Beiträge über Handelsformen und Handelsbeziehungen, er stellt Speichergebäude und Kaufmannshäuser vor und beleuchtet mit dem Magdalensberg bei Kärnten ein bedeutendes Zentrum der Erzgewinnung und der Metallverarbeitung. Einen besonderen Platz nimmt die römische Basilika als großartiges Bank- und Börsengebäude ein, dessen Entwicklung um 200 v. Chr. quasi schlagartig am Forum Romanum begann und die neue Vormacht Rom als Zentrum der antiken Wirtschaft und zentrale Instanz des Wirtschafts- und Steuerrechts mit neuen Maßstäben auch städtebaulich prägte. Die beiden letzten Beiträge befassen sich mit der weiteren Entwicklung des Begriffes Basilika und seinen Nachwirkungen im Kirchenbau und in der Architekturtheorie bis heute

    Primary biliary cirrhosis (PBC) is a classical autoimmune liver disease for which effective immunomodulatory therapy is lacking. Here we perform meta-analyses of discovery data sets from genome-wide association studies of European subjects (n=2,764 cases and 10,475 controls) followed by validation genotyping in an independent cohort (n=3,716 cases and 4,261 controls). We discover and validate six previously unknown risk loci for PBC (Pcombined<5 × 10(-8)) and used pathway analysis to identify JAK-STAT/IL12/IL27 signalling and cytokine-cytokine pathways, for which relevant therapies exist
