    Phenotypic Characterization of Crossed Goats F1- (Desert Goat and Damascus)

    This study was conducted in North Kordofan state, Western Sudan, during the period Feb 2016- March 2020 to study the Phenotypic Characterization of crossed goats (F1-Desert goat and Damascus).44 female Desert goats of average age of 3-4 years and average body weights33.18±0.84 kg, were purchased from the local markets. Two mature sexual Damascus bucks were used in the experiment in addition to two castrated local young ones (teasers) all animals were identified using ear tags, tested for brucellosis and treated against external and internal parasites. During gestation period goats were closely observed up to the time of kidding. Data concerning with crossed kids (phenotypic characers, body measurements, performance up to maturity) were recorded. Data were analyzed using Statistical Package for Social Sciences, software package (SPSS, V23, 2017)). The results indicated that Male kids were significantly (P<0.01) heavier than females (2.74±0.48 visas 2.04±0.14kg). 27% mortality rate was recorded in kids. 71% of crossed goats (first generation) had light blue eye color. The flock had different coat colors (light brown, reddish brown and dark brown). The udder was somewhat small and attached to the abdomen the average body weight at kidding was 26.28 ± 1.56 and 22.03± 1.27 kg for single and twins, respectively

    Do grain reserves necessarily contribute to prices stability and food security in Sudan? An assessment

    AbstractMost governments in Africa implement policies aiming to stabilize the prices of staple foods, which often include building up grain reserves, besides other trade measures insulating their domestic market from the world market. The mechanism should ideally work as follows, grains should be bought and stored from areas, during the surplus seasons (after harvest) so as to assure fair prices to producers and should be distributed during deficit seasons, in deficit areas besides in cases of emergencies. However, ideal approaches are not necessarily followed in many developing countries due to different constraints and situations. The Strategic Reserve Corporation (SRC) is an institution that is established ten years ago to play such a role in Sudan. This paper tries to assess the performance of the SRC against the overall goals and to study the related obstacles if any. We use a sample of 112 respondents from the SRC staff, related and grain farmers as our data source. Results of the research revealed numerous financial and administrative constraints that obstruct SRC from playing the intended role, which need to be considered so as to contribute to price stability and food security in Sudan

    Epidemiology of strongyle nematode infections and first report of benzimidazole resistance in Haemonchus contortus in goats in South Darfur State, Sudan

    Background Since pastoralists in South Darfur, Sudan, had complained about lack of albendazole (ABZ) efficacy to control nematodes in goats, the frequency of infection with gastrointestinal helminths was studied before in vivo faecal egg count reduction tests (FECRT) were conducted using ABZ orally either at the dose recommended for sheep, 5 mg/kg body weight (bw) or at 10 mg/kg bw. Experiments included goats naturally infected with gastrointestinal nematodes or experimentally infected with local Haemonchus contortus isolates. Three study areas (Nyala, Beleil and Kass) were visited in autumn or winter. Results Out of 478 screened goats, 82.4% were infected with gastrointestinal helminths and 82% were shedding eggs of strongyle nematodes with 90% of the strongyle larvae representing Haemonchus spp. A FECRT using naturally infected goats (n = 225: 71 untreated, 104 and 50 treated with 5 and 10 mg ABZ/kg bw, respectively) detected reduced ABZ efficacy in Nyala and Kass. Paired and unpaired FECRT calculations detected reductions of 72–92% with samples taken at 8 days post treatment with 5 mg ABZ/kg bw and of 85–94% with 10 mg ABZ/kg bw. The FECRT based on day 14 post treatment samples showed reductions of 69–77% with 5 mg/kg and of 75–87% with 10 mg ABZ/kg bw. In Beleil, ABZ efficacy was 95%. In the egg hatch test EC50 values for Nyala and Kass ranged from 0.12–0.24 μg thiabendazole/ml, corresponding to benzimidazole resistant phenotypes. Only Haemonchus spp. larvae were present after treatments in coprocultures. When the efficacy was evaluated experimentally using isolates of H. contortus from Nyala and Kass, the 5 mg ABZ/kg dose revealed reductions of 76–78% on day 8 and of 62–70% on day 14 with the unpaired method. Using 10 mg ABZ/kg, the FECR was still only 77–82%. Conclusions Both, in vivo and in vitro methods detected resistant H. contortus populations in goats from South Darfur State. The time point 14 days post treatment was more sensitive for detection of ABZ resistance than 8 days post treatment. This is the first report on the occurrence of anthelmintic resistance in Sudan confirming that anthelmintic resistance selection is occurring in African subsistence farming systems

    The impact of the soft errors in convolutional neural network on GPUS: Alexnet as case study

    Convolutional Neural Networks (CNNs) have been increasingly deployed in many applications, including safety critical system such as healthcare and autonomous vehicles. Meanwhile, the vulnerability of CNN model to soft errors (e.g., caused by radiation mduced) rapidly increases, thus reliability is crucial especially in real-tmie system. There are many traditional techniques for miprove the reliability of the system, e.g.. Triple Modular Redundancy, but these techniques incur high overheads, which makes them hard to deploy. In tins paper, we experimentally evaluate the vulnerable parts of Alexnet mode (e.g., fault mjector). Results show that FADD and LD are the top vulnerable mstructions against soft errors for Alexnet model, both mstruetions generate at least 84% of injected faults as SDC errors. Thus, these the only parts of the Alexnet model that need to be hardened mstead of usmg fully duplication solutions

    Antiapoptotic and Antioxidant Properties of Orthosiphon stamineus Benth (Cat's Whiskers): Intervention in the Bcl-2-Mediated Apoptotic Pathway

    Antiapoptotic and antioxidant activities of aqueous-methanolic extract (CAME) of Orthosiphonstamineus Benth(OS), and its hexane (HF), chloroform (CF), n-butanol (NBF), ethyl acetate (EAF) and water (WF) fractions were investigated. Antioxidant properties were evaluated using the assays of Folin-Ciocalteu, aluminiumtrichloride, β-carotene bleaching and DPPH. The role of OS against hydrogen peroxide induced apoptosis on MDA-M231 epithelial cells was examined using MTT assay, phase contrast microscope, colorimetric assay of caspase-3, western blot and quantitative real-time PCR. Results showed that EAF showed the highest total phenolic content followed by CAME, NBF, WF, CF and HF, respectively. Flavonoid content was in the order of the CF > EAF > HF > CAME > NBF > WF. The IC50 values on DPPH assay for different extract/fractions were 126.2 ± 23, 31.25 ± 1.2, 15.25 ± 2.3, 13.56 ± 1.9, 23.0 ± 3.2, and 16.66 ± 1.5 μg/ml for HF, CF, EAF, NBF, WF and CAME, respectively. OSreduced the oxidation of β-carotene by hydroperoxides. Cell death was dose-dependently inhibited by pretreatment with OS. Caspase-3 and distinct morphological features suggest the anti-apoptotic activities of OS. This plant not only increased the expression of Bcl-2, but also decreased Bax expression, and ultimately reduced H2O2-induced apoptosis. The current results showed that phenolics may provide health and nutritional benefits

    Acellular Bi-Layer Silk Fibroin Scaffolds Support Tissue Regeneration in a Rabbit Model of Onlay Urethroplasty

    Acellular scaffolds derived from Bombyx mori silk fibroin were investigated for their ability to support functional tissue regeneration in a rabbit model of urethra repair. A bi-layer silk fibroin matrix was fabricated by a solvent-casting/salt leaching process in combination with silk fibroin film casting to generate porous foams buttressed by homogeneous silk fibroin films. Ventral onlay urethroplasty was performed with silk fibroin grafts (Group 1, N = 4) (Width×Length, 1×2 cm2) in adult male rabbits for 3 m of implantation. Parallel control groups consisted of animals receiving small intestinal submucosa (SIS) implants (Group 2, N = 4) or urethrotomy alone (Group 3, N = 3). Animals in all groups exhibited 100% survival prior to scheduled euthanasia and achieved voluntary voiding following 7 d of initial catheterization. Retrograde urethrography of each implant group at 3 m post-op revealed wide urethral calibers and preservation of organ continuity similar to pre-operative and urethrotomy controls with no evidence of contrast extravasation, strictures, fistulas, or stone formation. Histological (hematoxylin and eosin and Masson's trichrome), immunohistochemical, and histomorphometric analyses demonstrated that both silk fibroin and SIS scaffolds promoted similar extents of smooth muscle and epithelial tissue regeneration throughout the original defect sites with prominent contractile protein (α-smooth muscle actin and SM22α) and cytokeratin expression, respectively. De novo innervation and vascularization were also evident in all regenerated tissues indicated by synaptophysin-positive neuronal cells and vessels lined with CD31 expressing endothelial cells. Following 3 m post-op, minimal acute inflammatory reactions were elicited by silk fibroin scaffolds characterized by the presence of eosinophil granulocytes while SIS matrices promoted chronic inflammatory responses indicated by mobilization of mononuclear cell infiltrates. The results of this study demonstrate that bi-layer silk fibroin scaffolds represent promising biomaterials for onlay urethroplasty, capable of promoting similar degrees of tissue regeneration in comparison to conventional SIS scaffolds, but with reduced immunogenicity

    Susceptible trichostrongyloid species mask presence of benzimidazole-resistant Haemonchus contortus in cattle

    Background Benzimidazole (BZ) anthelmintics are widely used to control infections with parasitic nematodes, but BZ resistance is an emerging threat among several nematode species infecting humans and animals. In Sudan, BZ-resistant Haemonchus contortus populations were recently reported in goats in South Darfur State. The objective of this study was to collect data regarding the situation of BZ resistance in cattle parasitic nematodes in South Darfur using phenotypic and molecular approaches, besides providing some epidemiological data on nematodes in cattle. Methods The faecal egg count reduction test and the egg hatch test (EHT) were used to evaluate benzimidazole efficacy in cattle nematodes in five South Darfur study areas: Beleil, Kass, Nyala, Rehed Al-Birdi and Tulus. Genomic DNA was extracted from pools of third-stage larvae (L3) (n = 40) during trials, before and after treatment, and pools of adult male Haemonchus spp. (n = 18) from abattoirs. The polymorphisms F167Y, E198A and F200Y in isotype 1 β-tubulin genes of H. contortus and H. placei were analysed using Sanger and pyrosequencing. Results Prevalence of gastro-intestinal helminths in cattle was 71% (313/443). Reduced albendazole faecal egg count reduction efficacy was detected in three study areas: Nyala (93.7%), Rehed Al-Birdi (89.7%) and Tulus (88.2%). In the EHT, EC50 values of these study areas ranged between 0.032 and 0.037 µg/ml thiabendazole. Genus-specific PCRs detected the genera Haemonchus, Trichostrongylus and Cooperia in L3 samples collected after albendazole treatment. Sanger sequencing followed by pyrosequencing assays did not detect elevated frequencies of known BZ resistance-associated alleles in codon F167Y, E198A and F200Y in isotype 1 β-tubulin gene of H. placei (≤ 11.38%). However, polymorphisms were detected in H. contortus and in samples with mixed infections with H. contortus and H. placei at codon 198, including E198L (16/58), E198V (2/58) and potentially E198Stop (1/58). All pooled L3 samples post-albendazole treatment (n = 13) were identified as H. contortus with an E198L substitution at codon 198. Conclusions To the knowledge of the authors, this is the first report of reduced albendazole efficacy in cattle in Sudan and is the first study describing an E198L substitution in phenotypically BZ-resistant nematodes collected from cattle

    Impact-based forecasting in South East Asia – what underlies impact perceptions?

    The move towards impact-based forecasting presents a challenge for forecasters, who must combine information not just on what the weather might be, but also on what the weather might do. Yet different hazards and impacts are qualitatively distinct, meaning such information cannot be easily or straightforwardly integrated. The present study aimed to provide a way of characterising seemingly disparate impacts. In a collaboration between UK psychologists and partners from three meteorological organisations in Indonesia, Malaysia and the Philippines, the psychometric paradigm was employed to investigate how forecasters and stakeholders perceive weather-related impacts. Participants provided ratings of nine categories of impacts on a total of 10 characteristics, as well as providing an overall impact severity rating. Principal components analysis revealed differing component solutions across countries, which explained around 75% of the variance in perceptions. There were some similarities across all countries, with the characteristics ‘worry’ and ‘destructiveness’ loading positively together, as well as ‘likelihood of harm’ and ‘seriousness of harm’. We did not find strong evidence to indicate that forecasters and stakeholders perceive impacts in different ways. Our results highlight the complex nature of impact perceptions, which are characterised not just by objective factors such as impact scope and duration, but also subjective factors, such as worry and perceived severity

    CXCL1: A new diagnostic biomarker for human tuberculosis discovered using Diversity Outbred mice.

    More humans have died of tuberculosis (TB) than any other infectious disease and millions still die each year. Experts advocate for blood-based, serum protein biomarkers to help diagnose TB, which afflicts millions of people in high-burden countries. However, the protein biomarker pipeline is small. Here, we used the Diversity Outbred (DO) mouse population to address this gap, identifying five protein biomarker candidates. One protein biomarker, serum CXCL1, met the World Health Organization\u27s Targeted Product Profile for a triage test to diagnose active TB from latent M.tb infection (LTBI), non-TB lung disease, and normal sera in HIV-negative, adults from South Africa and Vietnam. To find the biomarker candidates, we quantified seven immune cytokines and four inflammatory proteins corresponding to highly expressed genes unique to progressor DO mice. Next, we applied statistical and machine learning methods to the data, i.e., 11 proteins in lungs from 453 infected and 29 non-infected mice. After searching all combinations of five algorithms and 239 protein subsets, validating, and testing the findings on independent data, two combinations accurately diagnosed progressor DO mice: Logistic Regression using MMP8; and Gradient Tree Boosting using a panel of 4: CXCL1, CXCL2, TNF, IL-10. Of those five protein biomarker candidates, two (MMP8 and CXCL1) were crucial for classifying DO mice; were above the limit of detection in most human serum samples; and had not been widely assessed for diagnostic performance in humans before. In patient sera, CXCL1 exceeded the triage diagnostic test criteria (\u3e90% sensitivity; \u3e70% specificity), while MMP8 did not. Using Area Under the Curve analyses, CXCL1 averaged 94.5% sensitivity and 88.8% specificity for active pulmonary TB (ATB) vs LTBI; 90.9% sensitivity and 71.4% specificity for ATB vs non-TB; and 100.0% sensitivity and 98.4% specificity for ATB vs normal sera.
