Analysis of BMP4 and BMP7 signaling in breast cancer cells unveils time-dependent transcription patterns and highlights a common synexpression group of genes

<p>Abstract</p> <p>Background</p> <p>Bone morphogenetic proteins (BMPs) are members of the TGF-beta superfamily of growth factors. They are known for their roles in regulation of osteogenesis and developmental processes and, in recent years, evidence has accumulated of their crucial functions in tumor biology. BMP4 and BMP7, in particular, have been implicated in breast cancer. However, little is known about BMP target genes in the context of tumor. We explored the effects of BMP4 and BMP7 treatment on global gene transcription in seven breast cancer cell lines during a 6-point time series, using a whole-genome oligo microarray. Data analysis included hierarchical clustering of differentially expressed genes, gene ontology enrichment analyses and model based clustering of temporal data.</p> <p>Results</p> <p>Both ligands had a strong effect on gene expression, although the response to BMP4 treatment was more pronounced. The cellular functions most strongly affected by BMP signaling were regulation of transcription and development. The observed transcriptional response, as well as its functional outcome, followed a temporal sequence, with regulation of gene expression and signal transduction leading to changes in metabolism and cell proliferation. Hierarchical clustering revealed distinct differences in the response of individual cell lines to BMPs, but also highlighted a synexpression group of genes for both ligands. Interestingly, the majority of the genes within these synexpression groups were shared by the two ligands, probably representing the core molecular responses common to BMP4 and BMP7 signaling pathways.</p> <p>Conclusions</p> <p>All in all, we show that BMP signaling has a remarkable effect on gene transcription in breast cancer cells and that the functions affected follow a logical temporal pattern. Our results also uncover components of the common cellular transcriptional response to BMP4 and BMP7. Most importantly, this study provides a list of potential novel BMP target genes relevant in breast cancer.</p

Keittiöt osana kerrostalon rakennussuunnittelun lähtötietoja

Kerrostaloasuntokohteen sijainnin, hinnan ja asumiskustannusten lisäksi asiakkaan osto-päätökseen vaikuttavat myös asunnon tilaratkaisut ja materiaalit. Erottuakseen joukosta asuntorakentajan tulee miettiä asunnon pohja- ja kalusteratkaisuja. Asunnon tilaratkaisuista merkittävimmäksi nousee keittiö, sen toimivuus ja ulkonäkö. Arkkitehti laatii asuntorakentamisessa keittiökalustekuvat, joiden mukaan kalustetoimittaja tekee tuotantokuvat. Suunnittelun lähtötiedoiksi ei kuitenkaan tavanomaisesti anneta pintamateriaalien laatutasoa tarkempia tietoja, vaan kalustesuunnittelu toteutetaan perinteisin ratkaisuin mahdollisimman tehokkaalla tilankäytöllä, keittiön minimivaatimuksia silmällä pitäen. Suunnittelijalla harvoin on tiedossa tietyn kalustetoimittajan käyttämiä tarkkoja tilavaatimuksia eri kalusteille. Tästä johtuen on hyvin yleistä, että kalusteiden mittoja joudutaan muuttamaan siinä vaiheessa, kun kalustetoimittaja tekee tuotantokuvia ja todetaan, ettei asunnon tilavaraukset riitä. Suunnittelun ohjauksessa keittiökalusteiden mitoituksen ja sijoittelun tarkastelulle ei aina jää tarpeeksi aikaa, jolloin keittiön kalusteratkaisut eivät välttämättä toteudu parhaalla mahdollisella tavalla. Myös asunnon ostajan teettämät lisä- ja muutostyöt keskittyvät suurelta osin keittiön kalustemuutoksiin, mikä kuormittaa asiakaspalveluinsinöörien työmäärää. Työn taustaksi haastateltiin Arkta Rakennuttajat Oy:n arkkitehteja, joilta tiedusteltiin heidän kokemuksiaan ja toiveitaan suunnittelunohjauksesta, suunnittelua varten annettavista lähtötiedoista sekä mielipiteitä ajatuksesta käyttää valmiita keittiömalleja lähtötietoina. Lisäksi työssä tarkasteltiin nettiselainpohjaista materiaalivalintaohjelmaa ja sen tuomaa hyötyä asiakaspalveluinsinöörin näkökulmasta materiaalivalintojen ja asiakas-muutostöiden hallinnassa. Jotta keittiöistä saataisiin mahdollisimman toimivat ja näyttävät, suunniteltiin tässä opinnäytetyössä erikokoisiin kerrostaloasuntoihin soveltuvia keittiömalleja, joita voitaisiin käyttää suuntaviivoina suunnittelulle. Osasta malleista suunniteltiin myös korkeamman laatutason versio, jota voi hyödyntää joko vakiokeittiömallina tai vaihtoehtoisesti valmiiksi hinnoiteltuna asukasmuutoskeittiönä. Materiaalivalintaohjelmaa ja sen tuomaa hyötyä punnittiin niin asiakaspalveluinsinöörin kuin asiakkaidenkin näkökulmasta. Keittiöiden kalustekuvat pidettiin luottamuksellisena aineistona eivätkä ne sisälly julkiseen raporttiin.In addition to location, price and cost of living, the layout and materials also influence the customers’ decisions to buy a flat. The construction company needs to think about the floor design and the fixtures of the flat to stand out from the crowd. The most significant room in the flat is the kitchen, its functionality and look. In housing development, the architect makes the fixture drawings for the kitchen and the supplier makes the production line drawings on the basis of these. Usually only the quality standards of the surface materials are provided as the basis of the design instructions. Thus, the fixtures of the flats are made in traditional ways, keeping an eye on the minimum demands of the kitchen fixtures and making the most out of the layout. The architect rarely knows the exact fixture requirements of a particular supplier so it is common that the measurements of the kitchen fixtures need to be changed a little when the production drawings are made and a replicate measurement in the flat is made by the supplier. Sometimes there is not enough time in design management to consider the kitchen fixture sizing and placing. Thus, the result is not always the best possible. More-over, the additional and alteration works that customers order are mostly related to the kitchen’s fixtures. These increase the workload of customer service engineers. Architects of Arkta Rakennuttajat Oy were interviewed to collect background data for this thesis. Their experiences and wishes for the design management, given design instructions and their opinions on the idea of the ready-made kitchen modules as a part of design instructions were asked. In addition, a web-based program was in use to test whether it could be a useful tool for customer service engineers in handling customers' material selection processes, additional works and alterations. To ensure the functionality and appearance of the kitchens, different kitchen models were designed for different-sized flats. These models could be used as guidelines for construction planning. There were some models that were designed for higher quality targets but they could also be used as pre-priced alternative kitchens that can be ordered by the customer. The material selection software was considered both from a customer service engineer’s and customer’s point of view. The kitchen fixture drawings are confidential material and thus were not included in the public report

Ohitusleikkauspotilaan preoperatiivinen ohjaus : Ohjausrunko Savonlinnan keskussairaalan osasto 5A:n sairaanhoitajille

Opinnäytetyön tarkoituksena oli tuottaa ohjausrunko ohitusleikkauspotilaan preoperatiiviseen ohjaukseen Savonlinnan keskussairaalan sisätautien osasto 5A:lle. Opinnäytetyön tehtävänä oli selvittää, mitä eri asioita osastolla työskentelevien sairaanhoitajien tulisi ottaa esille ohitusleikkaukseen menevän potilaan ohjauksessa ja millainen ohjauksen ajoituksen tulisi olla. Opinnäytetyön aihetta on käsitelty systemaattisen kirjallisuuskatsauksen muodossa, jonka avulla on ollut mahdollista löytää kattava otos tutkittua tietoa aiheesta. Opinnäytetyö rakentuu tuotekehitysprosessin ympärille, ja ohjausrunko on kehitetty tuotekehitysprosessin eri vaiheiden kautta. Ohitusleikkauspotilaan ohjauksen kannalta tärkeiksi asioiksi koettiin tieto sepelvaltimotaudista ja siitä, kuinka potilaan tulisi valmistautua leikkaukseen fyysisen-, psyykkisen- ja sosiaalisen hyvinvoinnin kannalta. Potilaan pitäisi myös saada tietoa leikkauksen jälkeisestä toipumisajasta, koska se parantaa potilaan motivoitumista tulevaan. Keskeisiksi asioiksi nousi myös potilaan yksilöllinen ohjaus ja sen ajoitus. Nämä asiat koettiin tärkeiksi, koska ne mahdollistavat potilaan nopeamman toipumisen ja näin vähentävät terveydenhuollon kustannuksia. Kehittämästämme ohjausrungosta hyötyvät hoitohenkilökunta ja potilaat. Ohjausrunkoa käytettäessä ohjaus on yhtenevä ja potilas saa laadukkaan ohjauksen.The purpose of this bachelor’s thesis was to provide a framework for a bypass surgery patient’s preoperative guidance on the medical ward 5A at Savonlinna Central Hospital. Our aim was to survey the issues nurses should include in the guidance of a bypass surgery patient as well as the appropriate timing. The topic was discussed through a systematic literature review, whereupon it was possible to find a comprehensive sample of research information on the subject. Our thesis was constructed around a product development process, and the guidance framework was developed through the different phases of the development cycle. The review revealed several important issues for a bypass surgery patient’s guidance, such as information on coronary artery disease as well as physical, mental and social preparation for surgery. The patient should also receive information on the postoperative recovery time because it improves the patient’s motivation to prepare for the upcoming operation. Essential are also individual guidance and timing. The above factors were important because they allow faster recovery and thereby reduce health care costs. The guidance framework will benefit medical staff and patients by enabling congruent guidance and high-quality counseling

"Sie et tiiä miltä miusta tuntuu!" : - opas nuorten seksuaalisesta kehityksestä

Opinnäytetyön tarkoituksena oli tuottaa nuorille ja heidän vanhemmilleen tutkit-tuun tietoon perustuva opas, joka käsittelee nuoren psyykkistä kasvua ja kehitystä seksuaalisuuteen. Tavoitteena oli oppaan avulla lisätä nuorten tietoutta it-seensä ja muihin kohdistuvista ajatuksista, tunteista, asenteista ja kokemuksista. Opinnäytetyöraportissa käsitellään nuorten seksuaalioikeuksia, heidän psyykkistä kehittymistä seksuaalisuuteen sekä seksuaalikasvatusta. Tässä opinnäytetyössä keskityttiin tekemään nuorille suunnattu, helposti ymmärrettävä ja helposti luettava opas. Opas sisältää johdannon, nuorten seksu-aalioikeudet sekä seksuaalisuuden portaat. Seksuaalisuuden portaat on esitetty Etelä-Karjalassa puhuttavalla murteella. Opas pohjautuu sen takakannessa oleviin linkkeihin, jotka toimiva lukijalle lisätiedon lähteinä. Oppaassa ja opinnäytetyössä lähteinä on käytetty lähteitä, jossa käsitellään nuorten seksuaalista kehittymistä. Aineistoa etsittiin kirjallisuudesta ja Internetistä, ja käyttöön valittiin tuoretta ja tieteellistä aineistoa luotettavista lähteistä. Opinnäytetyönä tuotettua opasta voidaan hyödyntää kouluterveydenhuollossa nuorten seksuaalikasvatuksen lisänä ja tukena sekä jakaa nuorille ja heidän vanhemmilleen.The purpose of this study was to create a guide to young people and their parents that was based on research. The guide deals with young people’s psycho-logical growth and development into sexuality. The aim of the study was to increase young people’s knowledge of thoughts, feelings, attitudes and experi-ences they have towards themselves and each other with the help of the guide. This functional thesis consists of two parts; thesis report and a guide. This thesis focused on creating a guide for young people that would be easily understood and easy to read. The guide has three parts: introduction, young people’s sexual rights and steps of sexuality. The steps of sexuality was written in South Carelian dialect. The guide was based on links that can be found from its back cover. The links also provide additional information to the reader. The reference of the thesis and the guide was a material that deals with young people’s sexual development. The material was searched from literature and the Internet. Only new and scientific material from reliable sources was chosen as a reference. The guide can be utilized in school health care as an addition to young people’s sexual education. The guide can also be distributed to young people and their parents

CD marker expression profiles of human embryonic stem cells and their neural derivatives, determined using flow-cytometric analysis, reveal a novel CD marker for exclusion of pluripotent stem cells

AbstractHuman embryonic stem cells (hESCs) are pluripotent cells that can differentiate into neural cell lineages. These neural populations are usually heterogeneous and can contain undifferentiated pluripotent cells that are capable of producing teratomas in cell grafts. The characterization of surface protein profiles of hESCs and their neural derivatives is important to determine the specific markers that can be used to exclude undifferentiated cells from neural populations. In this study, we analyzed the cluster of differentiation (CD) marker expression profiles of seven undifferentiated hESC lines using flow-cytometric analysis and compared their profiles to those of neural derivatives. Stem cell and progenitor marker CD133 and epithelial adhesion molecule marker CD326 were more highly expressed in undifferentiated hESCs, whereas neural marker CD56 (NCAM) and neural precursor marker (chemokine receptor) CD184 were more highly expressed in hESC-derived neural cells. CD326 expression levels were consistently higher in all nondifferentiated hESC lines than in neural cell derivatives. In addition, CD326-positive hESCs produced teratomas in SCID mouse testes, whereas CD362-negative neural populations did not. Thus, CD326 may be useful as a novel marker of undifferentiated hESCs to exclude undifferentiated hESCs from differentiated neural cell populations prior to transplantation
