    Experimental study of the electrolysis of silicate melts

    Melting and electrolyzing lunar silicates yields oxygen gas and potentially can be practiced in situ to produce oxygen. With the present experiments conducted with simulant oxides at 1425-1480 C, it was ascertained that oxygen can be obtained anodically at feasible rates and current efficiencies. An electrolysis cell was operated with platinum anodes in a sealed vessel, and the production of gas was monitored. In these electrolysis experiments, stability of anodes remained a problem, and iron and silicon did not reduce readily into the liquid silver cathode

    A Rule-driven Approach for Defining the Behavior of Negotiating Software Agents

    One problem with existing agent-mediated negotiation systems is that they rely on ad hoc, static, non-adaptive, and hardcoded schemes to represent the behaviour of agents. This limitation is probably due to the complexity of the negotiation task itself. Indeed, while negotiating, software (human) agents face tough decisions. These decisions are based not only on the information made available by the negotiation server, but on the behaviour of the other participants in the negotiation process as well. The information and the behaviour in question are constantly changing and highly uncertain. In the first part of the paper, we propose a rule-driven approach to represent, manage and explore negotiation strategies and coordination information. For that, we divide the behaviour of negotiating agents into protocols, strategies and coordination. Among the many advantages of the proposed solution, we can cite the high level of abstraction, the closeness to human understanding, the versatility, and the possibility to modify the agents' behaviour during the negotiation process. To validate our solution, we ran many agent tournaments, and used the rule-driven approach to implement bidding strategies that are common in the English and Dutch auctions. We also implemented simple coordination schemes across several auctions. The ongoing validation work is detailed and discussed in the second part of the paper. Un des inconvénients qu'on retrouve fréquemment dans les systèmes de négociation par agents est qu'ils reposent sur des schémas ad-hoc, non adaptatifs et figés dans le code pour représenter le comportement des agents. Cette limitation est probablement due à la complexité de l'activité de négociation elle-même. En effet, au cours de la négociation, les agents logiciels (humains) ont des décisions difficiles à prendre. Ces décisions ne sont pas seulement basées sur l'information disponible sur le serveur de négociation, mais aussi sur le comportement des autres participants durant le processus de négociation. L'information et le comportement en question changent constamment et sont très incertains. Dans la première partie de l'article, nous proposons une approche à base de règles pour représenter, gérer et explorer les stratégies de négociation ainsi que l'information de coordination. Parmi les nombreux avantages de la solution proposée, on peut citer le haut niveau d'abstraction, la proximité avec la compréhension humaine, la souplesse d'utilisation et la possibilité de modifier le comportement des agents durant le processus de négociation. Pour valider notre solution, nous avons effectué plusieurs tournois entre agents et utilisé l'approche à base de règles pour implémenter des stratégies simples applicables à l'enchère anglaise et à l'enchère hollandaise. Nous avons aussi implémenté des schémas simples de coordination impliquant plusieurs enchères. Design patterns capture the expertise for reusable design solutions, but there is no methodical approach to providing conceptual design building blocks in tangible and composable form. Design components have been suggested to address this problem. We suggest compositional design reuse, which is a combined approach utilizing the ideas of design components and role models. We claim that design expertise in composable form with explicit documentation provides many advantages.     Synthesis of large scale 3D microscopic images of 3D cell cultures for training and benchmarking

    The analysis of 3D microscopic cell culture images plays a vital role in the development of new therapeutics. While 3D cell cultures offer a greater similarity to the human organism than adherent cell cultures, they introduce new challenges for automatic evaluation, like increased heterogeneity. Deep learning algorithms are able to outperform conventional analysis methods in such conditions but require a large amount of training data. Due to data size and complexity, the manual annotation of 3D images to generate large datasets is a nearly impossible task. We therefore propose a pipeline that combines conventional simulation methods with deep-learning-based optimization to generate large 3D synthetic images of 3D cell cultures where the labels are known by design. The hybrid procedure helps to keep the generated image structures consistent with the underlying labels. A new approach and an additional measure are introduced to model and evaluate the reduced brightness and quality in deeper image regions. Our analyses show that the deep learning optimization step consistently improves the quality of the generated images. We could also demonstrate that a deep learning segmentation model trained with our synthetic data outperforms a classical segmentation method on real image data. The presented synthesis method allows selecting a segmentation model most suitable for the user’s data, providing an ideal basis for further data analysis

    Bone Sialoprotein Shows Enhanced Expression in Early, High-Proliferation Stages of Three-Dimensional Spheroid Cell Cultures of Breast Cancer Cell Line MDA-MB-231

    Normally, bone sialoprotein (BSP) is an important contributor to bone micro-calcification. However, it is also highly expressed in bone-metastatic malignancies, including prostate, lung, and breast cancer. In these disorders, BSP correlates with poor prognosis. Its expression in triple-negative breast cancer cells is enhanced by the transcription factor RUNX2, and both, BSP and RUNX2 are under control of IGF-1 and TGFβ1. Knockdown of BSP or its inactivation by specific antibodies were found to reduce the metastatic potential of MDA-MB-231 triple-negative breast cancer cells in xenografts. While the role of BSP in bone metastasis was studied using such in vivo models, valid in vitro test systems to investigate BSP biology have been lacking since this protein is expressed at very low levels in classical 2D cell cultures and the frequently used breast cancer cell line MDA-MB-231 is difficult to grow in 3D. Here, we have developed a long-term 3D spheroid culture model using MDA-MB-231 cells in a sandwich approach using cell embedding between a non-adherent surface and basement membrane extracts. This allowed consistent growth of spheroids for more than 21 days. Also, co-culturing of MDA-MB-231 with CCD-1137Sk fibroblasts yielded stably growing spheroids, suggesting the importance of extracellular matrix (ECM) in this process. In addition, we have set up a novel and simple open source analysis tool to characterize protein expression in 2D cultures and spheroids by immunofluorescence. Using this approach in combination with Western blot analysis, the expression profile of BSP was analyzed. BSP was enriched at the rims of spheroids, both in mono- and co-cultures and its abundance in general correlated with that of TGFβ1 under different conditions, including spheroid maturation, cytostatic treatment, and fibroblast co-culture. Conversely, correlation of IGF-1 and BSP was limited to mono-culture time course profiles. In conclusion, we present novel tools to study the regulation of gene expression in combination with cell proliferation and apoptosis in a long-term 3D model of breast cancer and find dynamic abundance profiles of the metastasis-relevant protein BSP and its regulators

    Anthropogenic disturbance as a driver of microspatial and microhabitat segregation of cytotypes of Centaurea stoebe and cytotype interactions in secondary contact zones

    Background and Aims In a mixed-ploidy population, strong frequency-dependent mating will lead to the elimination of the less common cytotype, unless prezygotic barriers enhance assortative mating. However, such barriers favouring cytotype coexistence have only rarely been explored. Here, an assessment is made of the mechanisms involved in formation of mixed-ploidy populations and coexistence of diploid plants and their closely related allotetraploid derivates from the Centaurea stoebe complex (Asteraceae). Methods An investigation was made of microspatial and microhabitat distribution, life-history and fitness traits, flowering phenology, genetic relatedness of cytotypes and intercytotype gene flow (cpDNA and microsatellites) in six mixed-ploidy populations in Central Europe. Key Results Diploids and tetraploids were genetically differentiated, thus corroborating the secondary origin of contact zones. The cytotypes were spatially segregated at all sites studied, with tetraploids colonizing preferentially drier and open microhabitats created by human-induced disturbances. Conversely, they were rare in more natural microsites and microsites with denser vegetation despite their superior persistence ability (polycarpic life cycle). The seed set of tetraploid plants was strongly influenced by their frequency in mixed-ploidy populations. Triploid hybrids originated from bidirectional hybridizations were extremely rare and almost completely sterile, indicating a strong postzygotic barrier between cytotypes. Conclusions The findings suggest that tetraploids are later immigrants into already established diploid populations and that anthropogenic activities creating open niches favouring propagule introductions were the major factor shaping the non-random distribution and habitat segregation of cytotypes at fine spatial scale. Establishment and spread of tetraploids was further facilitated by their superior persistence through the perennial life cycle. The results highlight the importance of non-adaptive spatio-temporal processes in explaining microhabitat and microspatial segregation of cytotype
