Genomic blueprints of sponge-prokaryote symbiosis are shared by low abundant and cultivatable Alphaproteobacteria
Marine sponges are early-branching, filter-feeding metazoans that usually host complex microbiomes comprised of several, currently uncultivatable symbiotic lineages. Here, we use a low-carbon based strategy to cultivate low-abundance bacteria from Spongia officinalis. This approach favoured the growth of Alphaproteobacteria strains in the genera Anderseniella, Erythrobacter, Labrenzia, Loktanella, Ruegeria, Sphingorhabdus, Tateyamaria and Pseudovibrio, besides two likely new genera in the Rhodobacteraceae family. Mapping of complete genomes against the metagenomes of S. officinalis, seawater, and sediments confirmed the rare status of all the above-mentioned lineages in the marine realm. Remarkably, this community of low-abundance Alphaproteobacteria possesses several genomic attributes common to dominant, presently uncultivatable sponge symbionts, potentially contributing to host fitness through detoxification mechanisms (e.g. heavy metal and metabolic waste removal, degradation of aromatic compounds), provision of essential vitamins (e.g. B6 and B12 biosynthesis), nutritional exchange (especially regarding the processing of organic sulphur and nitrogen) and chemical defence (e.g. polyketide and terpenoid biosynthesis). None of the studied taxa displayed signs of genome reduction, indicative of obligate mutualism. Metagenome-assembled genome sequences of three uncultured planktomarina sp. strains from the Northeast Atlantic Ocean
We report three metagenome-assembled genomes (MAGs) of Planktomarina strains from coastal seawater (Portugal) to help illuminate the functions of understudied Rhodobacteraceae bacteria in marine environments. Comparative genomics reveals complex natural product biosynthesis capacities and carbon metabolism across host-associated and free-living Aquimarina (Bacteroidetes, Flavobacteriaceae) species
This study determines the natural product biosynthesis and full coding potential within the bacterial genus Aquimarina. Using comprehensive phylogenomics and functional genomics, we reveal that phylogeny instead of isolation source [host-associated (HA) vs. free-living (FL) habitats] primarily shape the inferred metabolism of Aquimarina species. These can be coherently organized into three major functional clusters, each presenting distinct natural product biosynthesis profiles suggesting that evolutionary trajectories strongly underpin their secondary metabolite repertoire and presumed bioactivities. Aquimarina spp. are highly versatile bacteria equipped to colonize HA and FL microniches, eventually displaying opportunistic behaviour, owing to their shared ability to produce multiple glycoside hydrolases from diverse families. We furthermore uncover previously underestimated, and highly complex secondary metabolism for the genus by detecting 928 biosynthetic gene clusters (BGCs) across all genomes, grouped in 439 BGC families, with polyketide synthases (PKSs), terpene synthases and non-ribosomal peptide synthetases (NRPSs) ranking as the most frequent BGCs encoding drug-like candidates. We demonstrate that the recently described cuniculene (trans-AT PKS) BGC is conserved among, and specific to, the here delineated A. megaterium-macrocephali-atlantica phylogenomic clade. The use of BASILICA Technique to prevent coronary obstruction in a TAVI-TAVI procedure
Transcatheter aortic valve implantation (TAVI) to manage structural bioprosthetic valve deterioration has been successful in mitigating the risk of a redo cardiac surgery. However, TAVI-in-TAVI is a complex intervention, potentially associated with feared complications such as coronary artery obstruction. Coronary obstruction risk is especially high when the previously implanted prosthesis had supra-annular leaflets and/or the distance between the prosthesis and the coronary ostia is short. The BASILICA technique (bioprosthetic or native aortic scallop intentional laceration to prevent iatrogenic coronary artery obstruction) was developed to prevent coronary obstruction during native or valve-in-valve interventions but has now also been considered for TAVI-in-TAVI interventions. Despite its utility, the technique requires a not so widely available toolbox. Herein, we discuss the TAVI-in-TAVI BASILICA technique and how to perform it using more widely available tools, which could spread its use
Glacier Monitoring Based on Multi-Spectral and Multi-Temporal Satellite Data: A Case Study for Classification with Respect to Different Snow and Ice Types
Remote sensing techniques are frequently applied for the surveying of remote areas, where the use of conventional surveying techniques remains difficult and impracticable. In this paper, we focus on one of the remote glacier areas, namely the Tyndall Glacier area in the Southern Patagonian Icefield in Chile. Based on optical remote sensing data in the form of multi-spectral Sentinel-2 imagery, we analyze the extent of different snow and ice classes on the surface of the glacier by means of pixel-wise classification. Our study comprises three main steps: (1) Labeled Sentinel-2 compliant data are obtained from theoretical spectral reflectance curves, as there are no training data available for the investigated area; (2) Four different classification approaches are used and compared in their ability to identify the defined five snow and ice types, thereof two unsupervised approaches (k-means clustering and rule-based classification via snow and ice indices) and two supervised approaches (Linear Discriminant Analysis and Random Forest classifier); (3) We first focus on the pixel-wise classification of Sentinel-2 imagery, and we then use the best-performing approach for a multi-temporal analysis of the Tyndall Glacier area. While the achieved classification results reveal that all of the used classification approaches are suitable for detecting different snow and ice classes on the glacier surface, the multi-temporal analysis clearly reveals the seasonal development of the glacier. The change of snow and ice types on the glacier surface is evident, especially between the end of ablation season (April) and the end of accumulation season (September) in Southern Chile
Automatic Serialization of Dynamic Structures in Ada
Serialization is the process that transforms the state of a software object into a sequence of bytes. Serialization is useful, for example, to store the value of an object in persistent memory or to send it through a network channel. Ada provides object serialization by means of the Streams package. However, default Ada serialization does not address the problem of serializing dynamic structures automatically. The streaming of an access variable in Ada flattens the value of the access variable itself and not the value of the object referenced by it. It is the responsibility of the programmer to write custom serialization routines for the lists, trees, stacks or any other dynamic structure requiring serialization. In this paper, we present an algorithm for the automatic generation of serialization subprograms for dynamic structures in Ada. We will use ASIS for analyzing the structure of the types to be serialized
