    Analyzing directionality of influence among ensemble musicians using Granger Causality

    In small musical groups, performers can seem to coordinate their movements almost effortlessly in remarkable exhibits of joint action and entrainment. To achieve a common musical goal, co-performers interact and communicate using non-verbal means such as upper-body movements, and particularly head motion. Studying these phenomena in naturalistic contexts can be challenging since most techniques make use of motion capture technologies that can be intrusive and costly. To investigate an alternative method, we analyze video recordings of a professional instrumental ensemble by extracting trajectory information using pose estimation algorithms. We examine Kansei perspectives such as the analysis of non-verbal expression conveyed by bodily movements and gestures, and test for causal relationships and directed influence between performers using the Granger Causality method. We compute weighted probabilities representing the likelihood that each performer Granger Causes co-performers’ movements. Effects of different aspects of musical textures were examined and results indicated stronger directionality for homophonic textures (clear melodic leader) than polyphonic (ambiguous leadership)

    NIHAO IV: Core creation and destruction in dark matter density profiles across cosmic time

    We use the NIHAO simulations to investigate the effects of baryonic physics on the time evolution of Dark Matter central density profiles. The sample is made of 70\approx 70 independent high resolution hydrodynamical simulations of galaxy formation and covers a wide mass range: 1e10< Mhalo <1e12, i.e., from dwarfs to L* . We confirm previous results on the dependence of the inner dark matter density slope, α\alpha, on the ratio between stellar-to-halo mass. We show that this relation holds approximately at all redshifts (with an intrinsic scatter of ~0.18 in α\alpha). This implies that in practically all haloes the shape of their inner density profile changes quite substantially over cosmic time, as they grow in stellar and total mass. Thus, depending on their final stellar-to-halo mass ratio, haloes can either form and keep a substantial density core (size~1 kpc), or form and then destroy the core and re-contract the halo, going back to a cuspy profile, which is even steeper than CDM predictions for massive galaxies (~1e12 Msun). We show that results from the NIHAO suite are in good agreement with recent observational measurements of α\alpha in dwarf galaxies. Overall our results suggest that the notion of a universal density profile for dark matter haloes is no longer valid in the presence of galaxy formation.Comment: 11 pages, 13 figures.     Risk factors for liposomal bupivacaine resistance after total hip or knee arthroplasties: A retrospective observational cohort in 237 patients

    Purpose: Liposomal bupivacaine demonstrated promise decreasing postoperative pain in total hip and total knee arthroplasty (THA/TKA). Some randomized trials have shown non-superior results; however, confounding variables were not accounted for in such analyses. This study attempts to determine risk factors associated with failure of pain management in patients receiving liposomal bupivacaine. Methods: Postoperative pain scores were collected following primary or revision arthroplasties between January 2016 and December 2017. Retrospective analysis of institutional total joint quality and outcomes registry was screened and patients undergoing primary or revision arthroplasties who completed a multi-modal pain management including liposomal bupivacaine were included in the study. Patients with a history of infection/deviated from the institutional pain management protocol were excluded. Results: A total of 237 patients were included for analysis. Younger patients less than 64 years old had significantly higher pain scores between 0 and 12 h and \u3e 24 h. Active smokers had significantly higher pain scores between 0 and 6 h and \u3e 24 h. Patients with a history of opioid use/pain management had significantly higher pain scores at 6-12 h and 24-48 h. Regression analysis indicated risk factors for resistance to liposomal bupivacaine are younger patients less than 64 years old, those undergoing primary THA, and patients with a history of smoking/pain management/opioid use. Conclusion: We identify risk factors for resistance to liposomal bupivacaine, which include younger age less than 64 years old, history of smoking/pain management/opioid use. Future studies should use these risk factors as exclusion criteria when using liposomal bupivacaine or initiating any randomized trials regarding efficacy

    Digitalisierungsfortschritt föderaler Einheiten : ein Vergleich der Ausgestaltungen kantonaler ePlattformen in der Schweiz

    Die Digitalisierung hält nicht nur in Wirtschaft und Gesellschaft Einzug, sondern auch im öffentlichen Sektor finden Digitalisierungsprozesse statt. Electronic Government (eGovernment) bezeichnet die Gesamtheit der elektronischen Dienstleistungen öffentlicher Verwaltungen. Mittels eGovernment sollen Prozesse sowohl optimiert als auch durchgeführt werden. In föderalen Staaten wie der Schweiz unterliegt die Implementierung technischer Systeme dem Subsidiaritätsprinzip. Es stellt sich deshalb die Frage, ob und worin sich der Digitalisierungsfortschritt von Kantonen unterscheidet. Ziel der Studie ist es, eine systematische Übersicht über Charakteristika verschiedener kantonaler ePlattform-Lösungen in der Schweiz zu erstellen. Gleichzeitig wird die Ausgestaltung von eGovernment-Lösungen mit Hilfe verschiedener Faktoren der technischen Umsetzung beurteilt. Mittels Dokumentenanalysen sowie einer Online-Befragung unter eGovernment-Verantwortlichen wurden Umfang und Ausgestaltung sämtlicher 26 kantonalen ePlattformen untersucht. Aus dem Modell zu Übernahmekategorien wurden Faktoren zur Ausgestaltung der technischen Umsetzung von eGovernment-Lösungen abgeleitet und auf den öffentlichen Sektor angewendet, um die eGovernment-Reife anhand der Ausprägungen der Kantone abzubilden. Das Modell der Übernahmekategorien unterteilt Systemmitglieder auf Basis ihrer Innovationsbereitschaft in fünf unterschiedliche Kategorien: Innovatoren, Frühe Übernehmer, Frühe Mehrheit, Späte Mehrheit und Nachzügler. Die Ergebnisse offenbaren einen heterogenen Innovationsgrad der Ausgestaltungen kantonaler ePlattformen. So entsprechen nur 4% der Kantone der Übernahmekategorie Innovatoren, wohingegen sich 23% der Kantone der Kategorie Nachzügler zuordnen lassen. Die Ergebnisse zur Beschaffenheit der Übernahmekategorien erlauben Schlussfolgerungen betreffend des Innovationsgrads verschiedener ePlattformen im schweizerischen Vergleich. Dadurch können die Ergebnisse dazu beitragen, dass sich die als Nachzügler identifizierten Kantone an den ePlattform Lösungen der Innovatoren als Vorbilder orientieren

    Applying scale-free mass estimators to the Local Group in Constrained Local Universe Simulations

    We use the recently proposed scale-free mass estimators to determine the masses of the Milky Way (MW) and Andromeda (M31) galaxy in a dark matter only Constrained Local UniversE Simulation (CLUES). While these mass estimators work rather well for isolated spherical host systems, we examine here their applicability to a simulated binary system with a unique satellite population similar to the observed satellites of MW and M31. We confirm that the scale-free estimators work also very well in our simulated Local Group galaxies with the right number of satellites which follow the observed radial distribution. In the isotropic case and under the assumption that the satellites are tracking the total gravitating mass, the power-law index of the radial satellite distribution N(<r)r3γN(<r)\propto r^{3-\gamma} is directly related to the host's mass profile M(<r)r1αM(<r)\propto r^{1-\alpha} as α=γ2\alpha=\gamma-2. The use of this relation for any given γ\gamma leads to highly accurate mass estimations which is a crucial point for observer, since they do not know a priori the mass profile of the MW and M31 haloes. We discuss possible bias in the mass estimators and conclude that the scale-free mass estimators can be satisfactorily applied to the real MW and M31 system.Comment: 14 pages, 6 figures, 6 tables.     Radially Extended Kinematics in the S0 Galaxy NGC 2768 from Planetary Nebulae, Globular Clusters and Starlight

    There are only a few tracers available to probe the kinematics of individual early-type galaxies beyond one effective radius. Here we directly compare a sample of planetary nebulae (PNe), globular clusters (GCs) and galaxy starlight velocities out to ~4 effective radii, in the S0 galaxy NGC 2768. Using a bulge-to-disk decomposition of a K-band image we assign PNe and starlight to either the disk or the bulge. We show that the bulge PNe and bulge starlight follow the same radial density distribution as the red subpopulation of GCs, whereas the disk PNe and disk starlight are distinct components. We find good kinematic agreement between the three tracers to several effective radii (and with stellar data in the inner regions). Further support for the distinct nature of the two galaxy components come from our kinematic analysis. After separating the tracers into bulge and disk components we find the bulge to be a slowly rotating pressure-supported system, whereas the disk reveals a rapidly rising rotation curve with a declining velocity dispersion profile. The resulting V/sigma ratio for the disk resembles that of a spiral galaxy and hints at an origin for NGC 2768 as a transformed late-type galaxy. A two-component kinematic analysis for a sample of S0s will help to elucidate the nature of this class of galaxy.Comment: 10 pages, 5 figures, accepted for publication in MRA

    Current challenges and future directions in data assimilation and reanalysis

    The first Joint WCRP1-WWRP2 Symposium on Data Assimilation and Reanalysis took place on13-17 September 2021, and it was organized in conjunction with the ECMWF Annual Seminaron observations. The last WCRP/WWRP-organized meetings were held separately for data assimilation and reanalysis in 2017 (Buizza et al. 2018; Cardinali et al. 2019). Since then, commonchallenges and new emerging topics have increased the need to bring these communities together toexchange new ideas and findings. Thus, a symposium involving the aforementioned communitieswas jointly organized by DWD3, HErZ4, WCRP, WWRP, and the ECMWF annual seminar. Majorgoals were to increase diversity, provide early career scientists with opportunities to present theirwork and extend their professional network, and bridge gaps between the various communities.The online format allowed more than 500 participants from over 50 countries to meet in avirtual setting, using the gathertown 5 platform as the central tool to access the meeting. A virtualconference center was created where people could freely move around and talk to other close-byparticipants. A lobby served as the main hub and it connected the poster halls and the conferencerooms for the oral presentations and the ECMWF seminar talks. The feedback from the participantswas overwhelmingly positive.Scientifically, the meeting offered opportunities to bring together the communities of Earth systemdata assimilation, reanalysis and observations to identify current challenges, seek opportunitiesfor collaboration, and strategic planning on more integrated systems for the longer term. Thecontributions totalled 140 oral and over 150 poster presentations covering a large variety oftopics with increased interest in Earth system approaches, machine learning and increased spatial resolutions.     Patient-derived xenografts and organoids model therapy response in prostate cancer.

    Therapy resistance and metastatic processes in prostate cancer (PCa) remain undefined, due to lack of experimental models that mimic different disease stages. We describe an androgen-dependent PCa patient-derived xenograft (PDX) model from treatment-naïve, soft tissue metastasis (PNPCa). RNA and whole-exome sequencing of the PDX tissue and organoids confirmed transcriptomic and genomic similarity to primary tumor. PNPCa harbors BRCA2 and CHD1 somatic mutations, shows an SPOP/FOXA1-like transcriptomic signature and microsatellite instability, which occurs in 3% of advanced PCa and has never been modeled in vivo. Comparison of the treatment-naïve PNPCa with additional metastatic PDXs (BM18, LAPC9), in a medium-throughput organoid screen of FDA-approved compounds, revealed differential drug sensitivities. Multikinase inhibitors (ponatinib, sunitinib, sorafenib) were broadly effective on all PDX- and patient-derived organoids from advanced cases with acquired resistance to standard-of-care compounds. This proof-of-principle study may provide a preclinical tool to screen drug responses to standard-of-care and newly identified, repurposed compounds

    SARS-CoV-2 susceptibility and COVID-19 disease severity are associated with genetic variants affecting gene expression in a variety of tissues

    Variability in SARS-CoV-2 susceptibility and COVID-19 disease severity between individuals is partly due to genetic factors. Here, we identify 4 genomic loci with suggestive associations for SARS-CoV-2 susceptibility and 19 for COVID-19 disease severity. Four of these 23 loci likely have an ethnicity-specific component. Genome-wide association study (GWAS) signals in 11 loci colocalize with expression quantitative trait loci (eQTLs) associated with the expression of 20 genes in 62 tissues/cell types (range: 1:43 tissues/gene), including lung, brain, heart, muscle, and skin as well as the digestive system and immune system. We perform genetic fine mapping to compute 99% credible SNP sets, which identify 10 GWAS loci that have eight or fewer SNPs in the credible set, including three loci with one single likely causal SNP. Our study suggests that the diverse symptoms and disease severity of COVID-19 observed between individuals is associated with variants across the genome, affecting gene expression levels in a wide variety of tissue types
