    Impact of different light sources on broiler rearing environment.

    Broiler production is highly dependent on the use of artificial light. The light source may affect the effectiveness of housing conditions due to increasing ambient temperature and concentration of noxious gases. This research aimed to evaluate the effects of different bulb types on the thermal, aerial, and acoustic environment of broiler aviaries. The experiment was carried out at a commercial broiler farm in Dourados, Mato Grosso do Sul State, Brazil. Three aviaries were used, and two flocks of male broilers from Cobb® genetic strain were reared from the first day to slaughter. Each aviary was equipped with a different light source, comprising the adopted treatments (A1 - incandescent light bulb, control; A2 - sodium vapor light bulb; A3 - fluorescent light bulb). The aviaries were divided into nine quadrants, and the environmental data (ambient dry bulb temperature and relative humidity), litter surface temperature, CO2 and NH3 concentrations, and bird sound pressure behavior were recorded in each quadrant. The aviary with incandescent light presented higher air and litter temperatures, and concentration of gases than the other tested alternatives. It also presented higher level of sound pressure in the second week of the growing period; however, from this period up to slaughter, there was no effect of the light source on the results of broiler sound pressure level

    Aim: describe the methodological approach adopted to build a Brazilian database of LCTFs in the country. Methods: This exploratory research was conducted between August 2020 and 2021 based on primarily publicly accessible data. First, the database of the Sistema Único de Assistência Social for 2019 was adopted as the primary source of information. In addition, public agencies and managers were consulted and invited to share their databases. Likewise, researchers and private entities also collaborated by making their spreadsheets available. The information collected was placed in individual spreadsheets for each Brazilian state. LTCFs not catering to older adults (aged 60 and over) were excluded. Duplicate data were excluded when overlaps were identified for each new aggregated source. Results & Discussion: This brief communication describes the methodology adopted for mapping the current status of Brazilian LTCFs. Despite its caveats, this study represents an important advance in the identification, characterization, and monitoring of these services nationwide. A total of 5769 facilities were found in the 2019 SUAS census. After excluding facilities not caring for residents aged 60 or over, this total decreased to 2381 LTCFs. Consolidating and filtering the information from multiple data sources led to the identification of 7029 LTCFs for the country as a whole

    Endocytosis of DNA-Hsp65 Alters the pH of the Late Endosome/Lysosome and Interferes with Antigen Presentation

    BACKGROUND: Experimental models using DNA vaccine has shown that this vaccine is efficient in generating humoral and cellular immune responses to a wide variety of DNA-derived antigens. Despite the progress in DNA vaccine development, the intracellular transport and fate of naked plasmid DNA in eukaryotic cells is poorly understood, and need to be clarified in order to facilitate the development of novel vectors and vaccine strategies. METHODOLOGY AND PRINCIPAL FINDINGS: Using confocal microscopy, we have demonstrated for the first time that after plasmid DNA uptake an inhibition of the acidification of the lysosomal compartment occurs. This lack of acidification impaired antigen presentation to CD4 T cells, but did not alter the recruitment of MyD88. The recruitment of Rab 5 and Lamp I were also altered since we were not able to co-localize plasmid DNA with Rab 5 and Lamp I in early endosomes and late endosomes/lysosomes, respectively. Furthermore, we observed that the DNA capture process in macrophages was by clathrin-mediated endocytosis. In addition, we observed that plasmid DNA remains in vesicles until it is in a juxtanuclear location, suggesting that the plasmid does not escape into the cytoplasmic compartment. CONCLUSIONS AND SIGNIFICANCE: Taken together our data suggests a novel mechanism involved in the intracellular trafficking of plasmid DNA, and opens new possibilities for the use of lower doses of plasmid DNA to regulate the immune response

    Reducing the environmental impact of surgery on a global scale: systematic review and co-prioritization with healthcare workers in 132 countries

    Abstract Background Healthcare cannot achieve net-zero carbon without addressing operating theatres. The aim of this study was to prioritize feasible interventions to reduce the environmental impact of operating theatres. Methods This study adopted a four-phase Delphi consensus co-prioritization methodology. In phase 1, a systematic review of published interventions and global consultation of perioperative healthcare professionals were used to longlist interventions. In phase 2, iterative thematic analysis consolidated comparable interventions into a shortlist. In phase 3, the shortlist was co-prioritized based on patient and clinician views on acceptability, feasibility, and safety. In phase 4, ranked lists of interventions were presented by their relevance to high-income countries and low–middle-income countries. Results In phase 1, 43 interventions were identified, which had low uptake in practice according to 3042 professionals globally. In phase 2, a shortlist of 15 intervention domains was generated. In phase 3, interventions were deemed acceptable for more than 90 per cent of patients except for reducing general anaesthesia (84 per cent) and re-sterilization of ‘single-use’ consumables (86 per cent). In phase 4, the top three shortlisted interventions for high-income countries were: introducing recycling; reducing use of anaesthetic gases; and appropriate clinical waste processing. In phase 4, the top three shortlisted interventions for low–middle-income countries were: introducing reusable surgical devices; reducing use of consumables; and reducing the use of general anaesthesia. Conclusion This is a step toward environmentally sustainable operating environments with actionable interventions applicable to both high– and low–middle–income countries

    ESTUDO DA INFLUÊNCIA DE ONDAS DE CALOR SOBRE A PRODUÇÃO DE LEITE NO ESTADO DE SÃO PAULO

    O presente trabalho objetivou estimar as perdas da produção leiteira, em função da ocorrência de ondas de calor, baseado no Índice de Temperatura e Umidade (ITU). Inicialmente, foi determinada a freqüência de ocorrências de ITU acima do nível crítico (81), e elaborado um histórico de ocorrência de onda de calor no Estado de São Paulo, baseado em dados climatológicos, de intervalo variando de três a dez anos. Foi também realizada a validação do uso do ITU médio através do cálculo do declínio na produção de leite, (DPL) utilizando dados do controle leiteiro de uma fazenda comercial. Os resultados da ocorrência de ondas de calor mostraram que os municípios do Vale do Paraíba e a cidade de Presidente Prudente foram os que apresentaram o maior número de ondas de calor por ano e que nem sempre as ondas de calor de maior duração apresentaram maior intensidade de ocorrência. Concluiu-se na validação do ITU médio que este índice por si só, não é uma medida acurada para a redução da produção de leite em decorrência de estresse térmico e que o estudo da ocorrência de ondas de calor pode mostrar de forma mais efetiva o efeito nocivo do ambiente térmico.</p
