    Elektroniskā versija nesatur pielikumusTrimetilamÄ«ns ir ticis identificēts kā viens no baktēriju metabolÄ«tiem ar potenciāli negatÄ«vu ietekmi uz cilvēka veselÄ«bu, kas padara to par aktuālu pētniecÄ«bas objektu. Å ajā darbā tika veikti bakteriālo trimetilamÄ«nu producējoÅ”o enzÄ«mu pētÄ«jumi. Tika veikta Klebsiella pneumoniae CutC holÄ«na liāzes 3D kristālstruktÅ«ras noteikÅ”ana divās dažādās konformācijās. Tika pētÄ«ti arÄ« ar CutC holÄ«na liāzi saistÄ«to GRM2 tipa bakteriālo mikrokompartmentu paÅ”salikÅ”anās un iepakoÅ”anas mehānismi. Mēs atklājām, ka serdes enzÄ«mi tiek iepakota hierarhiskā manierē, prezentējam krio-EM 3D struktÅ«ru 3.3 ƅ izŔķirtspējā un pētÄ«jām priekÅ”noteikumus rekobinantu čaulu izveidoÅ”anai. Mēs arÄ« analizējām CntA oksigenāzes substrātu paneļus no četriem dažādiem saimniekorganismiem, tādējādi atrisinot dažas iepriekŔējas neskaidrÄ«bas par Å”o enzÄ«mu.Trimethylamine has been identified as a bacterial metabolite with a potential negative impact on health, making it a target of interest. In this study the investigation of bacterial trimethylamine producing enzymes was performed. We solved the 3D crystal structures of two different conformations of CutC choline lyase from Klebsiella pneumoniae. The assembly and encapsulation mechanisms of the CutC associated GRM2 type bacterial microcompartments were also investigated. We discovered that the core enzymes are encapsulated in a hierachial manner, present a cryo-EM 3D structure of a pT=4 quasi symmetric shell particle at 3.3 ƅ resolution and studied the requirements for recombinant shell formation. We also analyzed the substrate panels of CntA oxygenase from four different hosts, thus clearing some previous controversies about this enzyme

    System Dynamic Modelling of the Strategic Development of Agriculture

    In the paper two main development models are shown: agriculture industry development model based on the estimations of experts (model of experts) and agriculture science development model. The experts of the Republic of Latvia Ministry of Agriculture take part in the preparation of the model data, and the results of the models are used in Latvia agriculture industry and science development strategy. The model results show that it is possible to reach profit per employee as the EU average. For this the number of employees have to diminish not less than two times. The level of profit for living in the countries lifts to average EU level. It is possible to develop the alternative agricultural production also. The above mentioned can be achieved only along with comprehensive developments of the countries: education, infrastructure, technologies, etc. Agriculture industry development is constrained to the potential of scientific research in the industry. Furthermore, the number of scientists is declining.     Large-Scale Recombinant Production of the SARS-CoV-2 Proteome for High-Throughput and Structural Biology Applications

    The highly infectious disease COVID-19 caused by the Betacoronavirus SARS-CoV-2 poses a severe threat to humanity and demands the redirection of scientific efforts and criteria to organized research projects. The international COVID19-NMR consortium seeks to provide such new approaches by gathering scientific expertise worldwide. In particular, making available viral proteins and RNAs will pave the way to understanding the SARS-CoV-2 molecular components in detail. The research in COVID19-NMR and the resources provided through the consortium are fully disclosed to accelerate access and exploitation. NMR investigations of the viral molecular components are designated to provide the essential basis for further work, including macromolecular interaction studies and high-throughput drug screening. Here, we present the extensive catalog of a holistic SARS-CoV-2 protein preparation approach based on the consortiumā€™s collective efforts. We provide protocols for the large-scale production of more than 80% of all SARS-CoV-2 proteins or essential parts of them. Several of the proteins were produced in more than one laboratory, demonstrating the high interoperability between NMR groups worldwide. For the majority of proteins, we can produce isotope-labeled samples of HSQC-grade. Together with several NMR chemical shift assignments made publicly available on covid19-nmr.com, we here provide highly valuable resources for the production of SARS-CoV-2 proteins in isotope-labeled form

    Å Ä« darba mērÄ·is bija pētÄ«t vÄ«rusiem lÄ«dzÄ«go daļiņu (VLD) piegādāŔanas iespējas uz Ŕūnām, izmantojot preS1 sekvenci un tās fragmentus. Otrs uzdevums bija Latvijas Organiskā Sintēzes InstitÅ«tā (LOSI) sintezētu transfekcijas lÄ«dzekļu izpēte proteÄ«nu un to kompleksu virzÄ«Å”anai uz Ŕūnām. Varēja secināt, ka preS1 sekvences adresējoÅ”ais efekts uz HepG2 Ŕūnām nav liels. Pilna garuma preS1 sekvencei piemita spēja nodroÅ”ināt VLD uzņemÅ”anu atseviŔķās HepG2 un Hek 293 Ŕūnās, kamēr preS1 sekvences imunodominantais 10-36 aminoskābju fragments to nenodroÅ”ināja pie tiem paÅ”iem apstākļiem. ProteÄ«nu ransfekcijas lÄ«dzekļu efektivitāte tika raksturota kvantitatÄ«vi un kvalitatÄ«vi salÄ«dzinājumā ar komerciāliem lÄ«dzekļiem. Tālākai pētÄ«Å”anai varēja tikt izvirzÄ«ti vairāki transfekcijas lÄ«dzekļi, kas bija efektÄ«vi salÄ«dzinājumā ar komerciālajiem lÄ«dzekļiem. Atslēgas vārdi: bakteriofāgs AP 205, hepatÄ«ta B vÄ«russ, bakteriofāgs GA, katjoniskie lipÄ«di, vÄ«rusiem lÄ«dzÄ«gās daļiņas, preS1 sekvence, piegāde uz Ŕūnām.One of the aims of this work was to study delivery of virus-like particles (VLP) to cells using HBV preS1 sequence. Second aim of this work was study in LOSI produced transfection reagent effectiveness in delivering proteins and protein complexes to eukaryotic cells. It was concluded that preS1 sequence is not effective in addresing virus-like particles to HepG2 cells. Full length preS1 sequence mediates virus-like particle uptake in few HepG2 and Hek293 cells, while immunodomnant 10-36 amino acid fragment could not in the same experimental conditions. Protein transfection was studied using both quantitative and qualitative methods, and compared to commercial transfection reagents. For further study could be distinguished several transfection reagents with activity equal or above in comparision to commercial transfection reagents.     Å Ä« darba mērÄ·is bija pētÄ«t ar preS1 sekvences fragmentiem dekorēto AP 205 bakteriofāga apvalka proteÄ«na un hepatÄ«ta B vÄ«rusa core proteÄ«na veidoto vÄ«rusiem lÄ«dzÄ«go daļiņu (VLD) mijiedarbÄ«bu ar dažādu eikariotu Ŕūnu lÄ«nijām ā€“HepG2, Hek293, Jurkat, Namalva un BHK. Tika izstrādātas metodes VLD iezÄ«mÄ“Å”anai ar fluorescento iezÄ«mi un to saistÄ«bas pētÄ«Å”anai ar eikariotu Ŕūnām, izmantojot fluorimetrijas, Ŕūnu citometrijas, fluorescences mikroskopijas un lāzerskenējoŔās konfokālās mikroskopijas metodes. Varēja secināt, ka no HBV core proteÄ«na atvasināto VLD mijiedarbÄ«bas rezultātu ietekmē DNS saistoÅ”ais domēns. Savukārt uz VLD virsmas eksponēti preS1 sekvences fragmenti pētÄ«tajās konstrukcijās nenodroÅ”ina pārliecinoÅ”i augstāku mijiedarbÄ«bu ar Ŕūnām, taču visgarākā preS1 fragmenta klātbÅ«tne VLD sastāvā nodroÅ”ināja daļiņu internalizāciju Ŕūnās. Å is darbs tika izstrādāts Latvijas BiomedicÄ«nas petÄ«jumu un studiju centrā laikā no 2010. gada septembra lÄ«dz 2011. gada maijam. Atslēgas vārdi: bakteriofāgs AP 205, hepatÄ«ta B vÄ«russ, vÄ«rusiem lÄ«dzÄ«gās daļiņas, preS1 sekvence, piegādāŔana uz Ŕūnām.The aim of this work was to study AP205 bacteriophage coat protein and hepatitis B virus core protein virus-like particles, decorated with preS1 sequence fragments, interaction with different eukaryotic cell lines: HepG2, Hek293, Jurkat, Namalva and BHK. There were developed methods for labeling virus-like particles with FITC label and studying their interaction with eukaryotic cell lines, using fluorimetry, cell cytometry, fluorescent microscopy, confocal laser-scanning microscopy and immunofluorescence methods. It was concluded that HBV core protein DNA-binding domain is essential for VLP interaction with cells. It was also concluded that decorating AP 205 phage coat protein VLP with preS1 sequence fragments does not statistically significantly improve the ability of VLP to interact with cells, but the presence of the longest preS1 sequence fragment mediate VLP internalisation into cells.     Metamodes in Design of GFRP Composite Stiffened Deck Structure

    This paper deals with the comparison of parametric and non-parametric metamodels used in design procedure of pultruded glass fibre reinforced plastic (GFRP) deck panels. Stiffened deck structure loaded under the three-point bending and uniform distributed load has been examined. A set of five hundred sample points has been evaluated by means of finite element analysis and stiffness and strength responses extracted for building of metamodels. Moreover, set of complimentary physical experiments has been carried out in order to verify the numerical model. Both advantages and disadvantages of parametric and non-parametric metamodels have been drawn for a specific engineering application
