The genome of Spraguea lophii and the basis of host-microsporidian interactions

Although they are increasingly recognized as economically and medically important parasites, the molecular basis of microsporidian pathogenicity is almost completely unknown and no genetic manipulation system is currently available. The fish-infecting microsporidian Spraguea lophii shows one of the most striking host cell manipulations known for these parasites, converting host nervous tissue into swollen spore factories known as xenomas. In order to investigate the basis of these interactions between microsporidian and host, we sequenced and analyzed the S. lophii genome. Although, like other microsporidia, S. lophii has lost many of the protein families typical of model eukaryotes, we identified a number of gene family expansions including a family of leucine-rich repeat proteins that may represent pathogenicity factors. Building on our comparative genomic analyses, we exploited the large numbers of spores that can be obtained from xenomas to identify potential effector proteins experimentally. We used complex-mix proteomics to identify proteins released by the parasite upon germination, resulting in the first experimental isolation of putative secreted effector proteins in a microsporidian. Many of these proteins are not related to characterized pathogenicity factors or indeed any other sequences from outside the Microsporidia. However, two of the secreted proteins are members of a family of RICIN B-lectin-like proteins broadly conserved across the phylum. These proteins form syntenic clusters arising from tandem duplications in several microsporidian genomes and may represent a novel family of conserved effector proteins. These computational and experimental analyses establish S. lophii as an attractive model system for understanding the evolution of host-parasite interactions in microsporidia and suggest an important role for lineage-specific innovations and fast evolving proteins in the evolution of the parasitic microsporidian lifecycle.This work was supported by a BBSRC studentship to SEC (http://www.bbsrc.ac.uk), a Marie Curie postdoctoral fellowship to TAW (http://cordis.europa.eu/fp7/home_en.html) and a Royal Society University Research Fellowship to BAPW (http://royalsociety.org)

Characterizing organic carbon dynamics during biostimulation of a uranium contaminated field site
Uranium contamination of groundwater remains a pressing problem at many former uranium mining and milling operations, such as the Rifle, Integrated Field Research Challenge (IFRC) site. Biostimulation of the subsurface with an organic carbon source such as acetate, followed by the microbially-induced reductive precipitation of uranium has been proposed as an effective remediation strategy. While uranium bioreduction has been studied in several field experiments, the transformation and fate of injected carbon remains poorly understood. This study evaluated the impact of added organic carbon on the long-term biogeochemical attenuation of uranium in the subsurface of a former mill tailings site. Fluorescence and ultravioletâvisible absorbance analyses were used together with dissolved organic carbon (DOC) measurements to track organic carbon dynamics during and post-biostimulation of the 2011 Rifle IFRC experiment. An electron mass balance was performed on well CD01 to account for any unidentified carbon sinks. Measured DOC values increased to 1.76 mM-C during biostimulation, and to 3.18 mM-C post-biostimulation over background DOC values of 0.3â0.4 mM-C. Elevated DOC levels persisted for 90 days after acetate injections ceased. The electron mass balance revealed that assumed electron acceptors would not account for the total amount of acetate consumed. Excitationâemission matrices showed an increase in signals associated with soluble microbial products, during biostimulation, which disappeared post-biostimulation despite an increase in total DOC. Specific ultraviolet absorbance analyses, indicated that DOC present post-biostimulation is less aromatic in nature, compared to background DOC. Our results suggest that microbes convert injected acetate into a form of solid phase organic matter that may be available to sustain iron reduction post-stimulation
Identifying geochemical hot moments and their controls on a contaminated river floodplain system using wavelet and entropy approaches
Geochemical hot moments are defined here as short periods of time that are associated with disproportionally high levels of concentrations (biogeochemically-driven or transport-related) relative to longer intervening time periods. We used entropy and wavelet techniques to identify temporal variability in geochemical constituents and their controls along three transects within a contaminated floodplain system near Rifle CO. Results indicated that transport-dominated hot moments drove overall geochemical processing in the contaminated groundwater and seep zones. These hot moments were associated with seasonal hydrologic variability (âŒ4 months) in the contaminated aquifer and with annual hydrologic cycle and residence times in the seep zone. Hot moments associated with a naturally reduced zone within the aquifer were found to be biogeochemically-driven, with a different dominant frequency (âŒ3 months) and no correlation to hydrologic or weather variations, in contrast to what is observed in other regions of the floodplain
Midday measurements of leaf water potential and stomatal conductance are highly correlated with daily water use of Thompson Seedless grapevines
A study was conducted to determine the relationship between midday measurements of vine water status and daily water use of grapevines measured with a weighing lysimeter. Water applications to the vines were terminated on August 24th for 9 days and again on September 14th for 22 days. Daily water use of the vines in the lysimeter (ETLYS) was approximately 40 L vineâ1 (5.3 mm) prior to turning the pump off, and it decreased to 22.3 L vineâ1 by September 2nd. Pre-dawn leaf water potential (ΚPD) and midday Κl on August 24th were â0.075 and â0.76 MPa, respectively, with midday Κl decreasing to â1.28 MPa on September 2nd. Leaf g s decreased from ~500 to ~200 mmol mâ2 sâ1 during the two dry-down periods. Midday measurements of g s and Κl were significantly correlated with one another (r = 0.96) and both with ETLYS/ETo (r = ~0.9). The decreases in Κl, g s, and ETLYS/ETo in this study were also a linear function of the decrease in volumetric soil water content. Improving the Representation of Cross-Boundary Transport of Anthropogenic Pollution in East Asia Using Radon-222.
We report on 10 years of hourly atmospheric radon, CO, and SO2 observations at Gosan Station, Korea. An improved radon detector was installed during this period and performance of the detectors is compared. A technique is developed whereby the distribution of radon concentrations from a fetch region can be used to select air masses that have consistently been in direct contact with land-based emissions, and have been least diluted en route to the measurement site. Hourly radon concentrations are used to demonstrate and characterise contamination of remote-fetch pollution observations by local emissions at this key WMO GAW site, and a seasonally-varying 5-hour diurnal sampling window is proposed for
days on which diurnal cycles are evident to minimise these effects. The seasonal variability in mixing depth and
âbackgroundâ pollutant concentrations are characterised. Based on a subset of observations most representative of the
important regional fetch areas for this site, and least affected by local emissions, seasonal estimates of CO and SO2 in air

Intraoperative radiotherapy in the treatment of breast cancer: a review of the evidence
The surgical treatment of early breast cancer has evolved from the removal of the entire breast and surrounding tissues (mastectomy) to the removal of the tumour together with a margin of healthy tissue (lumpectomy). Adjuvant radiotherapy, however, is still mainly given to the whole breast. Furthermore, external beam radiotherapy is often given several months after initial surgery and requires the patient to attend the radiotherapy centre daily for several weeks. A single fraction of radiotherapy given during surgery directly to the tumour bed (intraoperative radiotherapy) avoids these problems. The rationale and level-1 evidence for the safety and efficacy of the technique are reviewed
The effect of a professional cricket match schedule on selected immune parameters
Objective. The impact of a professional cricket match schedule on white blood cell (WBC) distribution and lymphocyte activation (CD69 expression) was investigated.
Methods. After a 3-month pre-season training period, physical and immune parameters were determined in 14 male cricketers before (B) and after (A) an intensive 5- week match schedule.
Results. Exercise test results were unchanged from B to A. Total WBC counts were similar, but total lymphocyte and lymphocyte subpopulation counts decreased significantly. The CD4:CD8 ratio did not change. After in vitro stimulation, percentage CD4+CD69+ cells increased (B: 54.4 ± 9.7%, A: 64.0 ± 8.5%, p < 0.01), but absolute CD4+CD69+ cell counts did not change from B to A. In contrast, both the %CD8+CD69+ cells and absolute CD8+CD69+ cell count remained similar.
Conclusion. A strenuous, interregional, professional cricket match schedule resulted in a decreased number of lymphocytes, but relatively increased in vitro reactivity of CD4+ cells, thus maintaining the absolute capacity of the CD4+ cells to become activated on stimulation. In cricketers who suffered upper respiratory tract symptoms during the match schedule (N = 7), none of the immune parameters investigated differed significantly from the others at B or A.
