    Morphologically different hydroxyapatite nanoparticles exert differential genotoxic effects in Drosophila

    Altres ajuts: acords transformatius de la UABHydroxyapatite (HAP) occurs naturally in sedimentary and metamorphic rocks and constitutes the hard structures in many organisms. Since synthetic nano-sized HAP (HAP-NPs) are used in orthopedic applications and for heavy metal remediation in aquatic and terrestrial media, both environment and humans are exposed to them. Due to the concerns about their potential hazards, the genotoxic effects that round/rod forms of HAP-NPs were investigated in Drosophila using the wing-spot and the comet assays. Furthermore, caspase activities were evaluated to examine the activation of cell death pathways. As a novelty, the expression of 36 genes involved in DNA repair was investigated, as a tool to indirectly determine DNA damage induction. Obtained sizes were 35-60 nm (roundHAP-NPs) and 45-90 nm (rodHAP-NPs) with a low Zeta-potential (-1.65 and 0.37 mV, respectively). Genotoxicity was detected in the wing-spot (round form), and in the comet assay (round and rod-like HA-NPs). In addition, increased expression of Caspases 3/7, 8, and 9 activities were observed. For both HAP forms, increased changes in the expression were observed for mismatch repair genes, while decreased expression was observed for genes involved in ATM, ATR, and cell cycle pathways. The observed changes in the repair pathways would reinforce the view that HAP-NPs have genotoxic potential, although more markedly in the round form. Thus, the environmental presence of engineered nanoparticles, including HAPs, raises concerns about potential effects on human health. It is essential that the effects of their use are carefully assessed and monitored to ensure safety and to mitigate any potential adverse effects

    Anatextrix monstrabilis sp. nov. (Araneae: Agelenidae) is described and illustrated, based on male and female specimens collected from Adana Province, Türkiye. The new species has an L-shaped male palpal femur bearing multiple apophyses, which is a rare trait in spiders. Anatextrix monstrabilis sp. nov. is the second species of the recently described genus Anatextrix Kaya, Zamani, Yağmur & Marusik, 2023, currently known only from southern Türkiye

    Behavioral toxicity evaluation of MgO nanoparticles on drosophila melanogaster

    Günümüzde nanopartiküllerin üstün fizikokimyasal özelliklerinin belirlenmesinden sonra nanopartiküller ve nanoteknolojiye olan ilgi de hızlı bir şekilde artmış ve kullanım alanları da yaygınlaşmıştır. Üç fiziksel boyutundan en az biri 1-100 nm aralığında olan ve belirli nano-ölçekli özellikler gösterebilen maddeler olarak tanımlanan nanopartiküller, tıp, elektronik, kozmetik, çevresel temizlik gibi birçok farklı alanda yaygın kullanım alanına sahiptir. Bu yaygın kullanımdan dolayı insanların da nanopartiküllere olan maruziyeti gün geçtikçe artmaktadır. Ayrıca nanopartiküllerin çevreye yayılması doğada ki birçok canlı için risk oluşturmaktadır. Bu sebeple de nanopartiküllerin yol açabileceği olumlu/olumsuz biyolojik etkilerin anlaşılabilmesi için yapılan çalışmalar da hız kazanmıştır. Bu bağlamda yapılan çalışmada MgO nanopartiküllerinin farklı konsantrasyonlarının (2, 5 ve 10 mM) Drosophila melanogaster üzerindeki davranışsal toksisiteye etkisinin araştırılması amaçlanmıştır. Davranışsal toksisitenin belirlenmesinde larval ağırlık ve hareketi, ergin birey ağırlığı, pupa oluşturma başarısı, pupa pozisyonu, pupadan çıkış başarısı, negatif jeotaksis ve ömür uzunluğu deneyleri gerçekleştirilerek değerlendirilmiştir. Çalışma sonucunda MgO NP’lerinin genel olarak Drosophila melanogaster’da negatif bir etkiye neden olduğu tespit edilmiştir. Larval hareketin incelendiği deney kapsamında çalışma kapsamında en yüksek doz olarak belirlenen 10 mM’lık derişimin istatistiksel olarak anlamlı bir şekilde azalmaya neden olduğu tespit edilmiştir.Nowadays, after the determination of the superior physicochemical properties of nanoparticles, interest in nanoparticles and nanotechnology has increased rapidly and their usage areas have become widespread. Nanoparticles, which are defined as substances that have at least one of their three physical dimensions in the range of 1-100 nm and can show certain nanoscale properties, have a widespread use in many different fields such as medicine, electronics, cosmetics, environmental cleaning. Due to this widespread use, the exposure of humans to nanoparticles is increasing day by day. In addition, the spread of nanoparticles into the environment poses a risk to many living things in nature. For this reason, studies to understand the positive / negative biological effects that nanoparticles can cause have also gained momentum. In this context, it was aimed to investigate the effect of different concentrations of MgO nanoparticles (2, 5 and 10 mM) on behavioral toxicity on Drosophila melanogaster. In determining behavioral toxicity, larval weight and movement, adult weight, pupa formation success, pupal position, exit success, negative geotaxis and longevity tests were performed and evaluated. As a result of the study, it was determined that MgO NPs generally cause a negative effect on Drosophila melanogaster. Within the scope of the experiment in which the larval movement was examined, it was determined that the concentration of 10 mM, which was determined as the highest dose within the scope of the study, caused a statistically significant decrease

    Cepten yapılan sağlık harcaması, hanelerin veya bireylerin sağlık hizmetine ulaşabilmek için direkt olarak hizmet sunucusuna yaptıkları ve kendilerine geri ödenmeyecek olan harcamalardır. Sağlık hizmetleri finansman yöntemlerinden biri olan cepten yapılan sağlık harcamaları, hem bireyleri hem de sağlık sektörü finansmanını etkilediği için bu harcamaların boyutu tespit edilmelidir. Bu çalışmada sağlık, sağlık hizmetleri, sağlık ekonomisi, Türkiye'de sağlık sistemi ve finansmanı üzerinde durulmuştur. Türkiye'de hanelerin yaptığı sağlık harcamalarını etkileyen faktörlerin belirlenmesi amacıyla TÜİK tarafından yayınlanan 2010 yılı Hanehalkı Bütçe Anketi mikro veri seti kullanılarak, lojistik regresyon ile model tahmin edilmeye çalışılmıştır. Yapılan çalışmalar sonucunda; Türkiye'de hane reisinin isteğe bağlı sigorta kapsamında olduğu, kadın olduğu ve evli olduğu haneler ile hanede 5 yaş ve altı, 65 yaş ve üstü birey bulunan ve hanede günlük faaliyete engel fiziksel ya da zihinsel problemi olan bireyin olduğu hanelerin, diğer hanelere göre cepten sağlık harcaması yapma olasılıklarının daha yüksek olduğu tespit edilmiştir. Ayrıca gelir düzeyi arttıkça hanelerin cepten sağlık harcaması yapma olasılığının arttığı sonucuna ulaşılmıştır.Out-of-pocket health expenditure refers to non-refundable direct payments to health-care service providers, paid by households or individuals to access to health-care services. Because out-of-pocket health expenditures, one of the methods of financing health-care services, affect both individuals and health-care financing, the size of out-of-pocket health expenditures must be determined. In this study, health, health-care services, health economics, Turkey's health system and its financing is focused on. In order to determine the factors that affect health expenditures of households in Turkey, a logistic regression model is estimated by using micro data set of 2010 Household Budget Survey, published by TurkStat. The results indicate that, compared to other households, households whose head has optional insurance, households whose head is female or married, households that have family members 5 years old and under or 65 years old and over and households that have family members having physical or mental problem that prevents daily activity are more likely to come across with out-of-pocket health expenditure. In addition, it is concluded that as the level of income of households increases, the probability of those households to disburse out-of-pocket health expenditure also rises

    Sudden Death Due To Rupture Of Dissection Of The Aorta: Three Cases

    Acute aorta dissection is an urgent condition that presented with chest pain, back or abdominalpain and usually resulted with death. We presented three autopsy performed cases thatdetermined cardiac tamponade developed secondary to aortic dissection. Three of them weremale and their ages were 36, 49 and 62 years. Death was occurred after back and epigastric painin two patients while other patient was found dead in home. All of them did not have any diseasein history. Death rates could be reduced under to %50 with intervention on time so the possibilityof aorta dissection must not be forgotten in cases with abdomen, thorax or back pain onadmission

The disease spreads locally and systemically in a very short time. It is resistant to almost all available cytostatics. There is an urgent need for chemicals with an effective, low toxicity profile. Since Docetaxel (DX) is partially effective in the current routine, it is used to prolong survival in patients with ATK. In this study, the antimetastatic property of tetrahydrocurcumin (THC), the active metabolite of Curcuma Longa, was compared with docetaxel and the presence of synergism in the combination of the two substances wasinvestigated in vitro. Materials-Methods: IC25 and IC50 concentrations of THC and (DX) in CAL62 and 8505C ATC cells were determined by the modified MTT method we developed in our laboratory. These 2 concentrations were compared with the control in the treatment groups. Cell motility as an indicator of metastatic potential was evaluated by in vitro wound healing method. Free oxygen radicals were evaluated by TAS and TOS analyses.Results: It was observed that cell adhesions decreased in 8505 cell lines starting from the 2nd hour in the treatment groups, more so in the groups combined with THC and curcumin. It was found that the combined dose prevented migration and invasion in CAL62 cells in the first hours (24) statistically, and the 72-hour IC25 dose did not have a statistically significant difference with the DX50 dose. There was no difference in Total Oxidant level between the combination and control groups. It was observed that the most effective dose that significantly reduced the oxidative level was DX50. Conclusions: THC in ATK cells has an antimetastatic potential close to Docetaxel; It was observed that docetaxel decreased oxidative stress.Keywords: Anaplastic thyroid cancer (ATC), Docetaxel (DX), tetrahydrocurcumin (THC), Total Antioksidan Status (TAS), Total Antioksidan Status (TOS) Amaç: Anaplastik tiroid kanseri (ATK) ortalama sağkalımbirkaç ayla sınırlı olduğu, bilinen en agresiv kanser türlerinden birisidir. Hastalık oldukça kısa sürede lokal ve sistemik yayılım yapmaktadır. Mevcut sitostatiklerin neredeyse tamamına dirençlidir. Etkin, düşük toksisite profiline sahip kimyasallara acil ihtiyaç bildirlmektedir. Güncel rutinde Docetaksel (DX) kısmen etkinlik gösterdiğinden, ATK lı olgularda sağkalımı uzatmak amacıyla kullanılmaktadır. Bu çalışmada Curcuma Longanın aktif metaboliti olan tetrahidrokurkuminin(THC) antimetastatik özelliği docetaksel ile karşılaştırmak ve iki madde kombinasyonunda sinerjizmin varlığı in vitro incelenmiştir.Gereç-Yöntem: CAL62 ve 8505C ATC hücrelerinde THC ve (DX) ait IC25 ve IC50 konsantrasyonları, laboratuvarımızda geliştirdiğimiz modifiye MTT yöntemi ile belirlendi. Bu 2 konsantrasyon, tedavi gruplarında kontrol ile karşılaştırıldı. Metastatik potansiyelin bir göstergesi olarak hücre motilitesi invitro yara iyileşme yöntemi ile değerlendirildi. Hücresel sitotoksiste TAS ve TOS analizleri ile değerlendirildi.Bulgular:8505 hücre hattında tedavi gruplarında 2. Saatten itibaren THC ve kurkuminle kombin gruplarda daha fazla olmak üzere hücre adezyonlarının azaldığı gözlendi.CAL62 hücrelerinde kombine dozun ilk saatlerde (24) migrasyonu ve invazyonu istatistiksel anlamlı engellediği, 72 sa lik IC25 dozunun DX50ndozu ile anlamlı istatistiksel farka sahip olmadığı bulgularına ulaşıldı.Total Oksidan seviyede kombinasyon ve kontrol grupları arasında farki izlenmedi.     Anti-Cancer Potential of Curcumin in Anaplastic Thyroid Cancer

    BACKGROUND AND AIM: Anaplastic Thyroid Cancer (ATK);It is one of the aggressive malignancies whose potential molecularmechanisms have not been elucidated and is the 9th among common cancers. It is resistant to existing cytostatic agents.To supportthe current therapy, the antimetastatic property of Tetrahydrocurcumin (THC) was compared with DX and in-vitro studies wereconducted to demonstrate synergism in combination use.MATERIALS and METHODS: IC25 and IC50 concentrations ofTHC and DX in CAL62 and 8505C cell lines were determined bythe modified MTT method we developed. Cell motility, indicative of metastatic potential; In-vitro wound healing method and 3Dcolony formation potential were evaluated with Agarose Gel Spheroid Model. The effects of treatments on oxidation on cells wereevaluated by TAS and TOS analyzes. SPSS program was used inthe evaluation. Compliance with normal distribution (Kolmogorov-Smirnov/Shapiro-Wilk tests), comparison of groups was evaluated with ANOVA, Significance, Holm-Sidak test.RESULTS: In migration analysis; It was observed that the THC50dose decreased the migration significantly compared to the DX50and DX+THC (Combined) groups (P&lt;0.05). DX+THC; It was observed that it reduced spheroid formation the most, and there was aclose effect between this dose effect and the THC25 dose in termsof spheroid formation and numbers (P&lt;0.05). There was no statistically significant difference in Total Oxidant level between thecombination and control.CONCLUSION: We predict that THC may be effective in metastasis and tumor growth at low doses in ATK cells.
