    Treten bei vorwiegend sitzenden Tätigkeiten während der Arbeitszeit, z.B. an Bildschirmarbeitsplätzen, über kurz oder lang körperliche Beschwerden auf und kann man diese im Rahmen der betrieblichen Gesundheitsförderung beeinflussen? Diese Frage wurde im Rahmen einer Studienarbeit , die während der Physiotherapieausbildung an der staatlichen Schule für Physiotherapie des Universitätsklinikums Münster durchgeführt wurde, untersucht. Den Probanden wurden Übungen aus der Funktionellen Bewegungslehre gezeigt, die Auswertung ergab keine unmittelbare Schmerzlinderung, aber eine Steigerung des körperlichen Wohlbefindens

    Improved silencing properties using small internally segmented interfering RNAs

    RNA interference is mediated by small interfering RNAs (siRNAs) that upon incorporation into the RNA-induced silencing complex (RISC) can target complementary mRNA for degradation. Standard siRNA design usually feature a 19–27 base pair contiguous double-stranded region that is believed to be important for RISC incorporation. Here, we describe a novel siRNA design composed of an intact antisense strand complemented with two shorter 10–12 nt sense strands. This three-stranded construct, termed small internally segmented interfering RNA (sisiRNA), is highly functional demonstrating that an intact sense strand is not a prerequisite for RNA interference. Moreover, when using the sisiRNA design only the antisense strand is functional in activated RISC thereby completely eliminating unintended mRNA targeting by the sense strand. Interestingly, the sisiRNA design supports the function of chemically modified antisense strands, which are non-functional within the context of standard siRNA designs. This suggests that the sisiRNA design has a clear potential of improving the pharmacokinetic properties of siRNA in vivo

    A novel physiological role for ARF1 in the formation of bidirectional tubules from the Golgi.

    Capitalizing on CRISPR/Cas9 gene-editing techniques and super-resolution nanoscopy, we explore the role of the small GTPase ARF1 in mediating transport steps at the Golgi. Besides its well-established role in generating COPI vesicles, we find that ARF1 is also involved in the formation of long (∼3 µm), thin (∼110 nm diameter) tubular carriers. The anterograde and retrograde tubular carriers are both largely free of the classical Golgi coat proteins coatomer (COPI) and clathrin. Instead, they contain ARF1 along their entire length at a density estimated to be in the range of close packing. Experiments using a mutant form of ARF1 affecting GTP hydrolysis suggest that ARF1[GTP] is functionally required for the tubules to form. Dynamic confocal and stimulated emission depletion imaging shows that ARF1-rich tubular compartments fall into two distinct classes containing 1) anterograde cargoes and clathrin clusters or 2) retrograde cargoes and coatomer clusters

    Non-equilibrium dynamics and floral trait interactions shape extant angiosperm diversity.

    Why are some traits and trait combinations exceptionally common across the tree of life, whereas others are vanishingly rare? The distribution of trait diversity across a clade at any time depends on the ancestral state of the clade, the rate at which new phenotypes evolve, the differences in speciation and extinction rates across lineages, and whether an equilibrium has been reached. Here we examine the role of transition rates, differential diversification (speciation minus extinction) and non-equilibrium dynamics on the evolutionary history of angiosperms, a clade well known for the abundance of some trait combinations and the rarity of others. Our analysis reveals that three character states (corolla present, bilateral symmetry, reduced stamen number) act synergistically as a key innovation, doubling diversification rates for lineages in which this combination occurs. However, this combination is currently less common than predicted at equilibrium because the individual characters evolve infrequently. Simulations suggest that angiosperms will remain far from the equilibrium frequencies of character states well into the future. Such non-equilibrium dynamics may be common when major innovations evolve rarely, allowing lineages with ancestral forms to persist, and even outnumber those with diversification-enhancing states, for tens of millions of years

    An outflow in the Seyfert ESO 362-G18 revealed by Gemini-GMOS/IFU Observations

    We present two-dimensional stellar and gaseous kinematics of the inner 0.7 ×\times 1.2 kpc2^{2} of the Seyfert galaxy ESO 362-G18, derived from optical spectra obtained with the GMOS/IFU on the Gemini South telescope at a spatial resolution of ≈\approx170 pc and spectral resolution of 36 km s−1^{-1}. ESO 362-G18 is a strongly perturbed galaxy of morphological type Sa or S0/a, with a minor merger approaching along the NE direction. Previous studies have shown that the [OIII] emission shows a fan-shaped extension of ≈\approx 10\arcsec\ to the SE. We detect the [OIII] doublet, [NII] and Hα{\alpha} emission lines throughout our field of view. The stellar kinematics is dominated by circular motions in the galaxy plane, with a kinematic position angle of ≈\approx137∘^{\circ}. The gas kinematics is also dominated by rotation, with kinematic position angles ranging from 122∘^{\circ} to 139∘^{\circ}. A double-Gaussian fit to the [OIII]λ\lambda5007 and Hα{\alpha} lines, which have the highest signal to noise ratios of the emission lines, reveal two kinematic components: (1) a component at lower radial velocities which we interpret as gas rotating in the galactic disk; and (2) a component with line of sight velocities 100-250 km s−1^{-1} higher than the systemic velocity, interpreted as originating in the outflowing gas within the AGN ionization cone. We estimate a mass outflow rate of 7.4 ×\times 10−2^{-2} M⊙_{\odot} yr−1^{-1} in the SE ionization cone (this rate doubles if we assume a biconical configuration), and a mass accretion rate on the supermassive black hole (SMBH) of 2.2 ×\times 10−2^{-2} M⊙_{\odot} yr−1^{-1}. The total ionized gas mass within ∼\sim84 pc of the nucleus is 3.3 ×\times 105^{5} M⊙_{\odot}; infall velocities of ∼\sim34 km s−1^{-1} in this gas would be required to feed both the outflow and SMBH accretion.Comment: 18 pages, 14 figure

    Observation of nonequilibrium motion and equilibration in polariton rings

    We present a study of the macroscopic dynamics of a polariton condensate formed by nonresonant optical excitation in a quasi-one-dimensional ring-shaped microcavity. The presence of a gradient in the cavity photon energy creates a macroscopic trap for the polaritons in which a coherent condensate is formed which evolves into a single-mode condensate at late times. With time- and energy-resolved imaging we show the role of interactions in the motion of the condensate as it undergoes equilibration in the ring. These experiments also give a direct measurement of the polariton-polariton interaction strength above the condensation threshold. Our observations are compared to the open-dissipative one-dimensional Gross-Pitaevskii equation which shows excellent qualitative agreement

    Anthropometric Characteristics of Young Women in Yakutia, Depending on the Type of Sexual Dimorphism and Ethnos

    The aim of this study was to assess the anthropometric data of girls and young women in Yakutia, depending on the type of sexual dimorphism and ethnos. The study included 2,081 girls and young women aged from 16 to 20 years. All those examined were born and permanently resided in the territory of the Republic of Sakha (Yakutia); among them there were 1284 Yakuts and 797 Russians. Anthropometric measurements were carried out according to the method of V.V. Bunak (1941), adopted in the Institute of Anthropology of MSU. Absolute values of the main components of the body (the total amount of fat, muscle tissue, bone tissue) were calculated by the formula Matiegka (1921). The body type was determined in accordance with the Tanner index. Statistical analysis was performed using statistical software package SPSS version 17.0 (SPSS Inc, Chicago, IL) The data obtained, depending on the type of sexual dimorphism, indicate the different maturation rates of the morphofunctional structures in Yakut and Russian women in the extreme natural climatic conditions of Yakutia

    <p>Abstract</p> <p>Background</p> <p>The aim of this study was to investigate the prevalence of cerebral palsy (CP) as well as to characterize the CP population, its participation in a secondary prevention programme (CPUP) and to validate the CPUP database.</p> <p>Methods</p> <p>The study population was born 1990–1997 and resident in Skåne/Blekinge on Jan 1<sup>st </sup>2002. Multiple sources were used. Irrespective of earlier diagnoses, neuropaediatrician and other professional medical records were evaluated for all children at the child habilitation units. The CPUP database and diagnosis registers at hospital departments were searched for children with CP or psychomotor retardation, whose records were then evaluated. To enhance early prevention, CP/probable CP was searched for also in children below four years of age born 1998–2001.</p> <p>Results</p> <p>The prevalence of CP was 2.4/1,000 (95% CI 2.1–2.6) in children 4–11 years of age born in Sweden, excluding post-neonatally acquired CP. Children born abroad had a higher prevalence of CP with more severe functional limitations. In the total population, the prevalence of CP was 2.7/1,000 (95% CI 2.4–3.0) and 48% were GMFCS-level I (the mildest limitation of gross motor function).</p> <p>One third of the children with CP, who were born or had moved into the area after a previous study in 1998, were not in the CPUP database. The subtype classification in the CPUP database was adjusted in the case of every fifth child aged 4–7 years not previously reviewed.</p> <p>Conclusion</p> <p>The prevalence of CP and the subtype distribution did not differ from that reported in other studies, although the proportion of mild CP tended to be higher.</p> <p>The availability of a second opinion about the classification of CP/CP subtypes is necessary in order to keep a CP register valid, as well as an active search for undiagnosed CP among children with other impairments.</p

    Gene set of nuclear-encoded mitochondrial regulators is enriched for common inherited variation in obesity

    There are hints of an altered mitochondrial function in obesity. Nuclear-encoded genes are relevant for mitochondrial function (3 gene sets of known relevant pathways: (1) 16 nuclear regulators of mitochondrial genes, (2) 91 genes for oxidative phosphorylation and (3) 966 nuclear-encoded mitochondrial genes). Gene set enrichment analysis (GSEA) showed no association with type 2 diabetes mellitus in these gene sets. Here we performed a GSEA for the same gene sets for obesity. Genome wide association study (GWAS) data from a case-control approach on 453 extremely obese children and adolescents and 435 lean adult controls were used for GSEA. For independent confirmation, we analyzed 705 obesity GWAS trios (extremely obese child and both biological parents) and a population-based GWAS sample (KORA F4, n = 1,743). A meta-analysis was performed on all three samples. In each sample, the distribution of significance levels between the respective gene set and those of all genes was compared using the leading-edge-fraction-comparison test (cut-offs between the 50(th) and 95(th) percentile of the set of all gene-wise corrected p-values) as implemented in the MAGENTA software. In the case-control sample, significant enrichment of associations with obesity was observed above the 50(th) percentile for the set of the 16 nuclear regulators of mitochondrial genes (p(GSEA,50) = 0.0103). This finding was not confirmed in the trios (p(GSEA,50) = 0.5991), but in KORA (p(GSEA,50) = 0.0398). The meta-analysis again indicated a trend for enrichment (p(MAGENTA,50) = 0.1052, p(MAGENTA,75) = 0.0251). The GSEA revealed that weak association signals for obesity might be enriched in the gene set of 16 nuclear regulators of mitochondrial genes
