    5'-terminal structure of poliovirus polyribosomal RNA is pUp

    Poliovirus RNA purified from virus-specific polyribosomes does not contain m7G in a 5'-5'-pyrophosphate linkage at its 5'-end. The only potential 5'-end found in ribonuclease digests of this RNA is pUp, which is present in a yield of 1 mole/mole of poliovirus RNA. We conclude that a 5'-terminal m7G is not required for translation of at least one RNA species in animal cells

    Reliability and sensitivity to change of the bristol rheumatoid arthritis fatigue scales

    Objective. To examine the reliability (stability) and sensitivity of the Bristol Rheumatoid Arthritis Fatigue scales (BRAFs) and patient-reported outcome measures (PROMs) developed to capture the fatigue experience. The Multi-Dimensional Questionnaire (BRAF-MDQ) has a global score and four subscales (Physical Fatigue, Living with Fatigue, Cognitive Fatigue and Emotional Fatigue), while three numerical rating scales (BRAF-NRS) measure fatigue Severity, Effect and Coping. Methods. RA patients completed the BRAFs plus comparator PROMs. Reliability (study 1): 50 patients completed questionnaires twice. A same-day test-retest interval (minimum 60 min) ensured both time points related to the same 7 days, minimizing the capture of fatigue fluctuations. Reliability (study 2): 50 patients completed the same procedure with a re-worded BRAF-NRS Coping. Sensitivity to change (study 3): 42 patients being given clinically a single high dose of i.m. glucocorticoids completed questionnaires at weeks 0 and 2.Results. The BRAF-MDQ, its subscales and the BRAF-NRS showed very strong reliability (r = 0.82-0.95). BRAF-NRS Coping had lower moderate reliability in both wording formats (r = 0.62, 0.60). The BRAF-MDQ, its subscales and the BRAF-NRS Severity and Effect were sensitive to change, with effect sizes (ESs) of 0.33-0.56. As hypothesized, the BRF-NRS Coping was not responsive to the pharmaceutical intervention (ES 0.05). Preliminary exploration suggests a minimum clinically important difference of 17.5% for improvement and 6.1% for fatigue worsening. Conclusion. The BRAF scales show good reliability and sensitivity to change. The lack of BRAF-NRS Coping responsiveness to medication supports the theory that coping with fatigue is a concept distinct from severity and effect that is worth measuring separately. © The Author 2013.     Men, rheumatoid arthritis, psychosocial impact and self-management: A narrative review.

    Rheumatoid arthritis (RA) is a chronic disease affecting fewer men than women. We systematically reviewed the literature on impact and self-management of RA men. Twenty eight papers were included, and grouped into two categories: Psychosocial impact of RA; and Coping and self-management. This review finds gender differences relating to quality of life; work; distress; self-management; coping; and support. We conclude there is a dearth of literature focussing on RA men only, and mixed gender studies include insufficient men to draw strong conclusions about men. Thus, further research is needed to understand the support needs of men with RA in depth

    Tradeoffs May Counteract Effects of Telecommunication Towers on Osprey Nesting

    The number of telecommunication towers (TCTs) throughout the United States has and continues to increase exponentially. Increased TCT placement has been implicated in local population increases in platform nesting birds such as ospreys (Pandion haliaetus). However, osprey use of artificial structures can cause conflicts with humans because of economic costs and risks to human safety. Managers require better information regarding the effect of artificial nesting surfaces, such as TCTS, on osprey reproduction to make informed decisions regarding species conservation and to mitigate potential conflicts. The objective of our study was to evaluate the influence of TCTs on osprey productivity and adult nesting behaviors. From March to July 2019, we monitored 30 osprey nests located in western Kentucky, USA, on TCTs (n = 11), navigation channel markers (NCMs; n = 10), and natural surfaces (NATs; n = 9). We found no differences in nest success, productivity, or the proportion of male ospreys perched at the nest between surface types. Nests on NCMs received 1.9–2.1 times more fish than nests on NATs and TCTs, respectively. Moreover, NCMs experienced 5.4–8.0 times more disturbances per hour compared to NATs and TCTs, respectively. Our data suggest ospreys nesting on TCTs are productive contributors to local populations. We recommend deterrence of osprey nesting should be focused on locations that are unsafe for ospreys and/or pose insurmountable problems for communication infrastructure

    Identification of XMRV Infection-Associated microRNAs in Four Cell Types in Culture

    INTRODUCTION: XMRV is a gammaretrovirus that was thought to be associated with prostate cancer (PC) and chronic fatigue syndrome (CFS) in humans until recently. The virus is culturable in various cells of human origin like the lymphocytes, NK cells, neuronal cells, and prostate cell lines. MicroRNAs (miRNA), which regulate gene expression, were so far not identified in cells infected with XMRV in culture. METHODS: Two prostate cell lines (LNCaP and DU145) and two primary cells, Peripheral Blood Lymphocytes [PBL] and Monocyte-derived Macrophages [MDM] were infected with XMRV. Total mRNA was extracted from mock- and virus-infected cells at 6, 24 and 48 hours post infection and evaluated for microRNA profile in a microarray. RESULTS: MicroRNA expression profiles of XMRV-infected continuous prostate cancer cell lines differ from that of virus-infected primary cells (PBL and MDMs). miR-193a-3p and miRPlus-E1245 observed to be specific to XMRV infection in all 4 cell types. While miR-193a-3p levels were down regulated miRPlus-E1245 on the other hand exhibited varied expression profile between the 4 cell types. DISCUSSION: The present study clearly demonstrates that cellular microRNAs are expressed during XMRV infection of human cells and this is the first report demonstrating the regulation of miR193a-3p and miRPlus-E1245 during XMRV infection in four different human cell types

    Utilizing qualitative data from nominal groups: Exploring the influences on treatment outcome prioritization with rheumatoid arthritis patients

    The nominal group technique generates quantitative data through a process of experts ranking items of interest. This article focuses on the additional collection of qualitative data from nominal groups with rheumatoid arthritis (RA) patients, used to explore the influences on prioritizing treatment outcomes. Across all groups, the top five outcomes with the highest importance scores were identified as: pain; joint damage; fatigue; activities of daily living; and mobility. Qualitative findings showed that the personal impact of RA influenced decisions on how to rank specific outcomes through four domains: disease impact; adaptation to illness; external resources and stressors; and social expectations. © The Author(s) 2011

    Constructing a definition: Adolescent wellbeing from the perspective of the child and expert

    Background: Adolescent’s wellbeing is gaining increasing attention, with it being part of the United Nations sustainable goals (United Nations, 2019 [online]). The question regarding what adolescent’s wellbeing is, however, has received no rigorous answer (Alexandrova, 2017), even though it is regarded as a fundamental measure of learning and growing as a human being (Stevens & Jarden, 2019). Understanding what wellbeing means to adolescents is vital and an under-researched area (Dunlop-Bennett et al., 2019) which has implications for the development and validation of tools to measure their wellbeing. Construct definition development literature highlights that experts, in addition to the relevant population, need to be consulted about the subject area to form a representative and valid definition (Podsakoff et al., 2016). Method: Two groups were invited to interview with the aim to understand their conceptualisations of wellbeing. Group 1consisted of both expert practitioners and researchers from different disciplines in the field of wellbeing (n = 8) and Group 2were young people aged 11-16 (n = 41). Conclusion: Any definition needs to be reflective of the multi-faceted nature of wellbeing. Findings revealed three themes: holism, positive feelings, and external influences. Based on these findings a definition of wellbeing was formed. These conclusions should be used to underpin research with adolescents and the development of a measure of young people’s wellbeing
