    The 38th Parallel: Penetrating the Line

    In July 1953, the armistice ended the Korean War that lasted for three years and established the Demilitarized Zone on either side of the demarcation line as a buffer between the two countries to prevent further military confrontation. However, the two sides remain at odds for half a century, and, despite the armistice, a state of war still exists between the two Koreas. As Koreans have dreamed of a united nation, the division has been described as a ‘temporary’ term to Koreans, yet the process of it has been much more obscure. Half a century has passed by, and South Korea has become a nation in which all facets of economic, political, and cultural identity are delineated in opposition to North Korea. What the future was supposed to present to Koreans has shifted relentlessly creating a disparity between the individual and national dreams. With repetitive see-saw events of national tension and reconciliation, individuals find themselves in an ambivalent position between series of oppositions: people and state, real and unreal, unification and national division. Multiple narratives crossover, creating confusion of whether the ultimate dream of Korea is even appropriate. The thesis examines the two opposing conditions: the idealized dream of homogeneity, and the factual reality of heterogeneity. Four series of investigations are presented in this thesis: the condition, the cause, the response, and the location of the individual. First, the disparity between the two Koreas illustrates the external conditions of the situation. Then an investigation of the Korean identity is presented to analyze the cause of the condition. The indigenous identity of Korea and the desire to preserve it are presented as the creative forces behind the dichotomy of Korea. The ambivalence of the individual is understood as a response such conditions. The concept of ‘Han’ is employed as a possible vehicle of understanding Korean cultural despondency. Finally the design exploration of a very significant archaeological site in the Demilitarized Zone is undertaken in order to mediate the disparity between the Korean dream and reality for the individual. The design is intended to locate the individual within the Korean pathology. Playing on the previously studied Korean conditions, the design is an amplified display of the opposing conditions which will enable the individual to face the ambivalence of today’s Korea. The thesis does not suggest the solution or envision the end but aims to meditate and negotiate the present moment. It is not my intention to force either fantasy or reality as an absolute answer, but to create an understanding of both conditions in hopes that Koreans can start to break their ambivalence regarding their national reunification

    Translating Hanja Historical Documents to Contemporary Korean and English

    The Annals of Joseon Dynasty (AJD) contain the daily records of the Kings of Joseon, the 500-year kingdom preceding the modern nation of Korea. The Annals were originally written in an archaic Korean writing system, `Hanja', and were translated into Korean from 1968 to 1993. The resulting translation was however too literal and contained many archaic Korean words; thus, a new expert translation effort began in 2012. Since then, the records of only one king have been completed in a decade. In parallel, expert translators are working on English translation, also at a slow pace and produced only one king's records in English so far. Thus, we propose H2KE, a neural machine translation model, that translates historical documents in Hanja to more easily understandable Korean and to English. Built on top of multilingual neural machine translation, H2KE learns to translate a historical document written in Hanja, from both a full dataset of outdated Korean translation and a small dataset of more recently translated contemporary Korean and English. We compare our method against two baselines: a recent model that simultaneously learns to restore and translate Hanja historical document and a Transformer based model trained only on newly translated corpora. The experiments reveal that our method significantly outperforms the baselines in terms of BLEU scores for both contemporary Korean and English translations. We further conduct extensive human evaluation which shows that our translation is preferred over the original expert translations by both experts and non-expert Korean speakers.Comment: 2022 EMNLP Finding

    A lab-on-a-disc platform enables serial monitoring of individual CTCs associated with tumor progression during EGFR-targeted therapy for patients with NSCLC

    Rationale: Unlike traditional biopsy, liquid biopsy, which is a largely non-invasive diagnostic and monitoring tool, can be performed more frequently to better track tumors and mutations over time and to validate the efficiency of a cancer treatment. Circulating tumor cells (CTCs) are considered promising liquid biopsy biomarkers; however, their use in clinical settings is limited by high costs and a low throughput of standard platforms for CTC enumeration and analysis. In this study, we used a label-free, high-throughput method for CTC isolation directly from whole blood of patients using a standalone, clinical setting-friendly platform. Methods: A CTC-based liquid biopsy approach was used to examine the efficacy of therapy and emergent drug resistance via longitudinal monitoring of CTC counts, DNA mutations, and single-cell-level gene expression in a prospective cohort of 40 patients with epidermal growth factor receptor (EGFR)-mutant non-small cell lung cancer. Results: The change ratio of the CTC counts was associated with tumor response, detected by CT scan, while the baseline CTC counts did not show association with progression-free survival or overall survival. We achieved a 100% concordance rate for the detection of EGFR mutation, including emergence of T790M, between tumor tissue and CTCs. More importantly, our data revealed the importance of the analysis of the epithelial/mesenchymal signature of individual pretreatment CTCs to predict drug responsiveness in patients.     A microfluidic chip for screening individual cancer cells via eavesdropping on autophagyinducing crosstalk in the stroma niche

    Autophagy is a cellular homeostatic mechanism where proteins and organelles are digested and recycled to provide an alternative source of building blocks and energy to cells. The role of autophagy in cancer microenvironment is still poorly understood. Here, we present a microfluidic system allowing monitoring of the crosstalk between single cells. We used this system to study how tumor cells induced autophagy in the stromal niche. Firstly, we could confirm that transforming growth factor beta 1 (TGF beta 1) secreted from breast tumor cells is a paracrine mediator of tumor-stroma interaction leading to the activation of autophagy in the stroma component fibroblasts. Through proof of concept experiments using TGF beta 1 as a model factor, we could demonstrate real time monitoring of autophagy induction in fibroblasts by single tumor cells. Retrieval of individual tumor cells from the microfluidic system and their subsequent genomic analysis was possible, allowing us to determine the nature of the factor mediating tumor-stroma interactions. Therefore, our microfluidic platform might be used as a promising tool for quantitative investigation of tumor-stroma interactions, especially for and high-throughput screening of paracrine factors that are secreted from heterogeneous tumor cell populations

    Efficacy of an Educational Material on Second Primary Cancer Screening Practice for Cancer Survivors: A Randomized Controlled Trial

    <div><h3>Background</h3><p>Cancer surivors have limited knowledge about second primary cancer (SPC) screening and suboptimal rates of completion of screening practices for SPC. Our objective was to test the efficacy of an educational material on the knowledge, attitudes, and screening practices for SPC among cancer survivors.</p> <h3>Methods</h3><p>Randomized, controlled trial among 326 cancer survivors from 6 oncology care outpatient clinics in Korea. Patients were randomized to an intervention or an attention control group. The intervention was a photo-novel, culturally tailored to increase knowledge about SPC screening. Knowledge and attitudes regarding SPC screening were assessed two weeks after the intervention, and screening practices were assessed after one year.</p> <h3>Results</h3><p>At two weeks post-intervention, the average knowledge score was significantly higher in the intervention compared to the control group (0.81 vs. 0.75, P<0.01), with no significant difference in their attitude scores (2.64 vs. 2.57, P = 0.18). After 1 year of follow-up, the completion rate of all appropriate cancer screening was 47.2% in both intervention and control groups.</p> <h3>Conclusion</h3><p>While the educatinal material was effective for increasing knowledge of SPC screening, it did not promote cancer screening practice among cancer survivors.     The same neuron may play different functional roles in the neural circuits to which it belongs. For example, neurons in the Tritonia pedal ganglia may participate in variable phases of the swim motor rhythms [1]. While such neuronal functional variability is likely to play a major role the delivery of the functionality of neural systems, it is difficult to study it in most nervous systems. We work on the pyloric rhythm network of the crustacean stomatogastric ganglion (STG) [2]. Typically network models of the STG treat neurons of the same functional type as a single model neuron (e.g. PD neurons), assuming the same conductance parameters for these neurons and implying their synchronous firing [3, 4]. However, simultaneous recording of PD neurons shows differences between the timings of spikes of these neurons. This may indicate functional variability of these neurons. Here we modelled separately the two PD neurons of the STG in a multi-neuron model of the pyloric network. Our neuron models comply with known correlations between conductance parameters of ionic currents. Our results reproduce the experimental finding of increasing spike time distance between spikes originating from the two model PD neurons during their synchronised burst phase. The PD neuron with the larger calcium conductance generates its spikes before the other PD neuron. Larger potassium conductance values in the follower neuron imply longer delays between spikes, see Fig. 17.Neuromodulators change the conductance parameters of neurons and maintain the ratios of these parameters [5]. Our results show that such changes may shift the individual contribution of two PD neurons to the PD-phase of the pyloric rhythm altering their functionality within this rhythm. Our work paves the way towards an accessible experimental and computational framework for the analysis of the mechanisms and impact of functional variability of neurons within the neural circuits to which they belong

    Changes in the Brain in Temporal Lobe Epilepsy with Unilateral Hippocampal Sclerosis: An Initial Case Series

    Temporal lobe epilepsy (TLE) is a network disorder of the brain. Network disorders predominately involve dysregulation of hippocampal function caused by neuronal hyperexcitability. However, the relationship between the macro- and microscopic changes in specific brain regions is uncertain. In this study, the pattern of brain atrophy in patients with TLE and hippocampal sclerosis (HS) was investigated using volumetry, and microscopic changes in specific lesions were observed to examine the anatomical correspondence with specific target lesions using diffusion tensor imaging (DTI) with statistical parametric mapping (SPM). This retrospective cross-sectional study enrolled 17 patients with TLE and HS. We manually measured the volumes of the hippocampus (HC), amygdala (AMG), entorhinal cortex, fornix, and thalamus (TH) bilaterally. The mean diffusivity and fractional anisotropy of each patient were then quantified and analyzed by a voxel-based statistical correlation method using SPM8. In right TLE with HS, there was no evidence of any abnormal diffusion properties associated with the volume reduction in specific brain regions. In left TLE with HS, there were significant changes in the volumes of the AMG, HC, and TH. Despite the small sample size, these differences in conditions were considered meaningful. Chronic left TLE with HS might cause structural changes in the AMG, HC, and TH, unlike right TLE with HS
