Sin embargo, la tradición mecanicista, anti vitalista, se halla enfrentada históricamente a otra, vitalista, que hunde sus raíces en Grecia y que tiene como hitos importantes el «primer Renacimiento» y la Naturphilosop-hie. Las dificultades con que tropiezan los biólogos a la hora de intentar definir la vida muestran que el problema planteado conserva plena vigencia.     The sinking of the supertanker Prestige off the coast of Spain in November 2002 was one of the most devastating oil spills ever worldwide. During any crisis most of the decisions have to be taken with limited information and they can benefit greatly from a rational methodology based on the available information. The main mechanisms involved in oil spill impacts are reviewed and we propose a decision tree based only on scientific criteria and data that are expected to be available in the first moments of an oil spill crisis. In the Prestige crisis, one of the decisions taken was to sail the Prestige out to sea. This paper makes an independent analysis of that decision to illustrate the methodology proposed, which may help decision making in future emergency situations.El hundimiento del petrolero Prestige frente a las costas españolas en noviembre de 2002 ha constituido uno de los vertidos más devastadores producidos en el mundo. Durante una crisis, la mayor parte de las decisiones han de ser tomadas con información limitada y pueden beneficiarse enormemente de una metodología racional que se base en la información disponible. Este artículo revisa los principales mecanismos que afectan al impacto de un vertido de petróleo y propone un árbol de decisión basado sólo en criterios científicos y datos que se puede esperar estarán disponibles en los primeros instantes de una crisis por vertido. En la crisis del Prestige, una de las decisiones tomadas fue la de conducir al barco mar adentro. Este artículo hace un análisis independiente de esa decisión con el fin de ilustrar la metodología propuesta, que puede ayudar a tomar la mejor decisión en futuras situaciones de emergencia

    Stress Increases Vulnerability to Inflammation in the Rat Prefrontal Cortex

    Inflammation could be involved in some neurodegenerative disorders that accompany signs of inflammation. However, because sensitivity to inflammation is not equal in all brain structures, a direct relationship is not clear. Our aim was to test whether some physiological circumstances, such as stress, could enhance susceptibility to inflammation in the prefrontal cortex (PFC), which shows a relative resistance to inflammation. PFC is important in many brain functions and is a target for some neurodegenerative diseases. We induced an inflammatory process by a single intracortical injection of 2 μg of lipopolysaccharide (LPS), a potent proinflammogen, in nonstressed and stressed rats. We evaluated the effect of our treatment on inflammatory markers, neuronal populations, BDNF expression, and behavior of several mitogen-activated protein (MAP) kinases and the transcription factor cAMP response element-binding protein. Stress strengthens the changes induced by LPS injection: microglial activation and proliferation with an increase in the levels of the proinflammatory cytokine tumor necrosis factor-α; loss of cells such as astroglia, seen as loss of glial fibrillary acidic protein immunoreactivity, and neurons, studied by neuronal-specific nuclear protein immunohistochemistry and GAD67 and NMDA receptor 1A mRNAs expression by in situ hybridization. A significant increase in the BDNF mRNA expression and modifications in the levels of MAP kinase phosphorylation were also found. In addition, we observed a protective effect from RU486 [mifepristone (11β-[p-(dimethylamino)phenyl]-17β-hydroxy-17-(1-propynyl)estra-4,9-dien-3-one)], a potent inhibitor of the glucocorticoid receptor activation. All of these data show a synergistic effect between inflammation and stress, which could explain the relationship described between stress and some neurodegenerative pathologies.España,Ministerio de Educación y Ciencia Grants SAF2002-01952 and SAF2004-0660

    The complement system (CS) includes more than 50 proteins and its main function is to recognize and protect against foreign or damaged molecular components. Other homeostatic functions of CS are the elimination of apoptotic debris, neurological development, and the control of adaptive immune responses. Pathological activation plays prominent roles in the pathogenesis of most autoimmune diseases such as systemic lupus erythematosus, antiphospholipid syndrome, rheumatoid arthritis, dermatomyositis, and ANCA-associated vasculitis. In this review, we will review the main rheumatologic autoimmune processes in which complement plays a pathogenic role and its potential relevance as a therapeutic target

    Impact of Dynamic Capabilities on Customer Satisfaction through Digital Transformation in the Automotive Sector

    Technology has impacted businesses in different areas, and, consequently, many companies have found it necessary to make changes in their structures and business models to improve customer satisfaction. The objective was to quantify the effect of dynamic capabilities on customer satisfaction, through digital transformation within the automotive sector. A random sample of 42 questionnaires on 127 surveyed industries was collected during the period 2019–2020 in a pre-COVID-19 context. A structural equation model (SEM) in two stages was applied. In the first stage, two reflective models were built. In a second stage, a structural equation model was evaluated. The results obtained in this study showed that the capabilities of sensing, seizing and innovation were suitably grouped in a construct called “Dynamic Capabilities”. A positive influence of Dynamic Capabilities on customer satisfaction was found. Therefore, the companies in this industry should focus on developing dynamic capabilities to improve customer satisfaction. Once the opportunities have been identified, managers take advantage of their potential (seizing) to transform and exploit knowledge in the creation, innovation, process improvement, and definition of strategies to combine new knowledge with that already existing. The digital transformation has contributed to identify the real needs for customers, to contact them and solve their problems, as well as offering products and services by anticipating their needs

    Peripheral Inflammation Enhances Microglia Response and Nigral Dopaminergic Cell Death in an in vivo MPTP Model of Parkinson’s Disease

    The impact of systemic inflammation in nigral dopaminergic cell loss remains unclear. Here, we have investigated the role of peripheral inflammation induced by systemic lipopolysaccharide (LPS) administration in the MPTP-based model of Parkinson’s disease. Brain inflammation, microglia and astroglia activation, disruption of the blood–brain barrier (BBB) and integrity of the nigrostriatal dopaminergic system were evaluated in response to i.p. injection of LPS, MPTP or the combination of both. Our results showed that combinative treatment exacerbates microglia activation and enhances (i) the appearance of galectin-3-positive microglia, recently identified as microglial disease-associated phenotypic marker, (ii) the up-regulation of pro-inflammatory cytokines, (iii) the occurrence of A1 neurotoxic astrocytes, (iv) the breakdown of the BBB, and (v) the loss of dopaminergic neurons in the substantia nigra. Microglia activation was triggered earlier than other degenerative events, suggesting that over-activation of microglia (including different polarization states) may induce dopaminergic neuron loss by itself, initiating the endless cycle of inflammation/degeneration. Our study revitalizes the importance of peripheral inflammation as a potential risk factor for Parkinson’s disease and raises the possibility of using new anti-inflammatory therapies to improve the course of neurodegenerative diseases, including those directly aimed at modulating the deleterious activity of disease-associated microglia.España MINECO y FEDER SAF2015-64171-
