    Threshold Models for Genome-Enabled Prediction of Ordinal Categorical Traits in Plant Breeding

    Categorical scores for disease susceptibility or resistance often are recorded in plant breeding. The aim of this study was to introduce genomic models for analyzing ordinal characters and to assess the predictive ability of genomic predictions for ordered categorical phenotypes using a threshold model counterpart of the Genomic Best Linear Unbiased Predictor (i.e., TGBLUP). The threshold model was used to relate a hypothetical underlying scale to the outward categorical response. We present an empirical application where a total of nine models, five without interaction and four with genomic x environment interaction (G·E) and genomic additive x additive x environment interaction (GxGxE), were used. We assessed the proposed models using data consisting of 278 maize lines genotyped with 46,347 single-nucleotide polymorphisms and evaluated for disease resistance [with ordinal scores from 1 (no disease) to 5 (complete infection)] in three environments (Colombia, Zimbabwe, and Mexico). Models with GxE captured a sizeable proportion of the total variability, which indicates the importance of introducing interaction to improve prediction accuracy. Relative to models based on main effects only, the models that included GxE achieved 9–14% gains in prediction accuracy; adding additive x additive interactions did not increase prediction accuracy consistently across locations

    Acute stress assessment using infrared thermography in fattening rabbits reacting to handling under winter and summer conditions

    Aim of study: This study assesses acute stress by measuring, through infrared thermography in summer and winter, the temperature of the eye, outer ear, inner ear and nose in 40 fattening rabbits before and after handling.Area of study: Seville (Spain).Material and methods: Body thermographic temperatures were recorded during a 38-day fattening period twice weekly and twice a day, before and after the handler held the rabbits in their arms for one minute. Ambient temperature and relative humidity were also recorded, and their influence on body temperatures was assessed. For each anatomical part, the variation of the temperature between the handled and undisturbed rabbit, and the differential temperature between the anatomical part in the undisturbed rabbit and the ambient temperature were calculated.Main results: The variation in temperatures between handled and undisturbed rabbits ranged from 0.25±0.041 ºC for eye to 3.09±0.221 ºC for outer ear in summer and -0.41±0.182 ºC for nose to 2.09±0.178 ºC for outer ear in winter. The day of the fattening period influenced all the temperature traits during summer and winter, except for the inner ear in winter. In summer, unlike winter, the temperature variation at the end of fattening period between handled and undisturbed rabbits was lower than at weaning (-0.04 to 1.94 ºC vs. 0.54 to 5.52 ºC, respectively). The temperatures in undisturbed rabbits were correlated with ambient temperature.Research highlights: Measuring body temperature with infrared thermography is a useful tool to evaluate acute stress in handled rabbits, with the inner ear and eye the most reliable body parts for measuring it

Our objective was to propose and apply a novel diet quality indicator (DQIn) using an a priori approach for NHs. Methods: the QUality Index for Nutrition in Nursing homes (QUINN) was implemented in a public NH located in Valladolid, Spain, during a 5-week period (n = 137 subjects). The choice of the QUINN components was based on a rapid review. The QUINN was based on 15 dietary components — 12 were basic (vegetables, fruits, legumes, olive oil, cereals, dairy, white fish and seafood, white-meat, eggs/positive; other fats, red and processed meat, and sweets/negative), and 3 were supplementary (fruits and vegetables variety, oily-fish, and whole-grains/positive). Each component was classified into 4-categories (0, 1, 2 o 3 points; range: 0-45 points). Results: the QUINN was tested on a menu offered by a NH giving a result of 34 points (good diet). The components with the highest scores were related to the Mediterranean diet (high consumption of legumes, olive oil, white fish and shellfish; low intake of other fats; and a wide variety of fruits and vegetables), together with cereals, white meat, dairy, and eggs. The components that required a major change were red- and processed-meats, sweets, and whole grains. Conclusion: the menu of this Spanish NH showed a good DQ according to the QUINN. The assessment of the DQ in NHs using QUINN will allow the proposal of interventions aimed at improving their diet.Antecedentes: la valoración de la calidad de la dieta es fundamental para el estudio de las asociaciones enfermedad-dieta, y es necesario implantar una herramienta de fácil aplicación en las residencias de ancianos. Nuestro objetivo fue proponer y aplicar un nuevo indicador de calidad de la dieta (diet quality indicator, DQIn) utilizando un enfoque a priori para su utilización en residencias de ancianos. Métodos: el Índice de Calidad Nutricional en Residencias de Ancianos (QUality Index for Nutrition in Nursing homes, QUINN) se aplicó en una residencia pública de Valladolid durante un periodo de 5 semanas (n = 137 sujetos). La elección de los componentes del QUINN se basó en una revisión rápida. En el QUINN se consideraron 15 componentes dietéticos, 12 básicos (verduras, frutas, legumbres, aceite de oliva, cereales, lácteos, pescado blanco y marisco, carnes blancas, huevos/positivos; otras grasas, carnes rojas y procesadas, y dulces/negativos) y 3 adicionales (variedad de frutas y verduras, pescado azul, y cereales integrales/positivos). Cada componente se clasificó en 4 categorías (0, 1, 2 o 3 puntos; rango: 0-45 puntos). Resultados: el QUINN se aplicó en el menú ofertado por una residencia de ancianos dando un resultado de 34 puntos (dieta de buena calidad). Los componentes con mayor puntuación estaban relacionados con la dieta mediterránea (alto consumo de legumbres, aceite de oliva, pescado blanco y marisco, bajo consumo de otras grasas y variedad de frutas y verduras), junto con los cereales, las carnes blancas, los lácteos y los huevos. Los componentes que requerían un cambio importante fueron las carnes rojas y procesadas, los dulces y los cereales integrales. Conclusión: el menú de esta residencia de ancianos situada en España mostró una calidad de la dieta buena según el QUINN. La evaluación de la calidad de la dieta en las residencias de ancianos mediante el QUINN permitirá proponer intervenciones para mejorar la dieta

    Waveguide manufacturing technologies for next-generation millimeter-wave antennas

    Some recent waveguide-based antennas are presented in this paper, designed for the next generation of communication systems operating at the millimeter-wave band. The presented prototypes have been conceived to be manufactured using different state-of-the-art techniques, involving subtractive and additive approaches. All the designs have used the latest developments in the field of manufacturing to guarantee the required accuracy for operation at millimeter-wave frequencies, where tolerances are extremely tight. Different designs will be presented, including a monopulse antenna combining a comparator network, a mode converter, and a spline profile horn; a tunable phase shifter that is integrated into an array to implement reconfigurability of the main lobe direction; and a conformal array antenna.     Detailed energy analysis of a sheet-metal-forming press from electrical measurements

    This paper presents a methodology that allows for the detection of the state of a sheet-metal-forming press, the parts being produced, their cadence, and the energy demand for each unit produced. For this purpose, only electrical measurements are used. The proposed analysis is conducted at the level of the press subsystems: main motor, transfer module, cushion, and auxiliary systems, and is intended to count, classify, and monitor the production of pressed parts. The power data are collected every 20 ms and show cyclic behavior, which is the basis for the presented methodology. A neural network (NN) based on heuristic rules is developed to estimate the press states. Then, the production period is determined from the power data using a least squares method to obtain normalized harmonic coefficients. These are the basis for a second NN dedicated to identifying the parts in production. The global error in estimating the parts being produced is under 1%. The resulting information could be handy in determining relevant information regarding the press behavior, such as energy per part, which is necessary in order to evaluate the energy performance of the press under different production conditions.Xunta de Galicia | Ref.     USP49 deubiquitinase regulates the mitotic spindle checkpoint and prevents aneuploidy.

    The spindle assembly checkpoint (SAC) is an essential mechanism that ensures the accurate chromosome segregation during mitosis, thus preventing genomic instability. Deubiquitinases have emerged as key regulators of the SAC, mainly by determining the fate of proteins during cell cycle progression. Here, we identify USP49 deubiquitinase as a novel regulator of the spindle checkpoint. We show that loss of USP49 in different cancer cell lines impairs proliferation and increases aneuploidy. In addition, USP49-depleted cells overcome the arrest induced by the SAC in the presence of nocodazole.     Mechanically reconfigurable linear phased array antenna based on single-block waveguide reflective phase shifters with tuning screws

    This work presents the design and prototyping of a reconfigurable phased array in Ku band (16 to 18 GHz) implemented in waveguide technology. The design is based on the use of a novel seamless waveguide module integrating four reconfigurable phase shifters to adjust the relative phase shift between the unitary elements of a linear array, which are illuminated uniformly by a corporate waveguide feeding network. The phase shifters are implemented by a 90º hybrid coupler in waveguide technology where two of its ports are loaded with a tunable reactive load, implemented in this proof of concept with a tuning screw. The four phase shifters have been manufactured in a single part using direct metal laser sintering, avoiding the losses related to bad electric contacts and misalignments associated to multipart devices. This also simplifies the assembly of the full phased array, leading to a modular approach with three parts whose design can be addressed separately. The experimental results for the complete array antenna show great performance and demonstrate that the main-lobe of the radiation pattern can be effectively scanned continuously between the angles - 25º and 25º, with a high efficiency in the whole design band thanks to the proposed waveguide implementationThis work was supported by the Spanish Government, Agencia Estatal de Investigación, Fondo Europeo de Desarrollo Regional: AEI/FEDER, UE, under Grant TEC2016-76070-C3-1-

    Crambescin C1 Acts as A Possible Substrate of iNOS and eNOS Increasing Nitric Oxide Production and Inducing In Vivo Hypotensive Effect

    Crambescins are guanidine alkaloids from the sponge Crambe crambe. Crambescin C1 (CC) induces metallothionein genes and nitric oxide (NO) is one of the triggers. We studied and compared the in vitro, in vivo, and in silico effects of some crambescine A and C analogs. HepG2 gene expression was analyzed using microarrays. Vasodilation was studied in rat aortic rings. In vivo hypotensive effect was directly measured in anesthetized rats. The targets of crambescines were studied in silico. CC and homo-crambescine C1 (HCC), but not crambescine A1 (CA), induced metallothioneins transcripts. CC increased NO production in HepG2 cells. In isolated rat aortic rings, CC and HCC induced an endothelium-dependent relaxation related to eNOS activation and an endothelium-independent relaxation related to iNOS activation, hence both compounds increase NO and reduce vascular tone. In silico analysis also points to eNOS and iNOS as targets of Crambescin C1 and source of NO increment. CC effect is mediated through crambescin binding to the active site of eNOS and iNOS. CC docking studies in iNOS and eNOS active site revealed hydrogen bonding of the hydroxylated chain with residues Glu377 and Glu361, involved in the substrate recognition, and explains its higher binding affinity than CA. The later interaction and the extra polar contacts with its pyrimidine moiety, absent in the endogenous substrate, explain its role as exogenous substrate of NOSs and NO production.     Effects of lng Mutations on LngA Expression, Processing, and CS21 Assembly in Enterotoxigenic Escherichia coli E9034A

    Enterotoxigenic Escherichia coli (ETEC) is a major cause of morbidity in children under 5 years of age in low- and middle-income countries and a leading cause of traveler's diarrhea worldwide. The ability of ETEC to colonize the intestinal epithelium is mediated by fimbrial adhesins, such as CS21 (Longus). This adhesin is a type IVb pilus involved in adherence to intestinal cells in vitro and bacterial self-aggregation. Fourteen open reading frames have been proposed to be involved in CS21 assembly, hitherto only the lngA and lngB genes, coding for the major (LngA) and minor (LngB) structural subunit, have been characterized. In this study, we investigated the role of the LngA, LngB, LngC, LngD, LngH, and LngP proteins in the assembly of CS21 in ETEC strain E9034A. The deletion of the lngA, lngB, lngC, lngD, lngH, or lngP genes, abolished CS21 assembly in ETEC strain E9034A and adherence to HT-29 cells was reduced 90%, compared to wild-type strain. Subcellular localization prediction of CS21 proteins was similar to other well-known type IV pili homologues. We showed that LngP is the prepilin peptidase of LngA, and that ETEC strain E9034A has another peptidase capable of processing LngA, although with less efficiency. Additionally, we present immuno-electron microscopy imagens to show that the LngB protein could be localized at the tip of CS21, and probably helps to control CS21 length. In conclusion, our results demonstrate that the LngA, LngB, LngC, LngD, LngH, and LngP proteins are essential for CS21 assembly, as well as for bacterial aggregation and adherence to HT-29 cells
