Células Mato na retina : caracterização morfológica e funcional de uma nova população de células perivasculares autofluorescentes em situação fisiológica e de retinopatia

perivasculares e células da micróglia. Neste trabalho, descreve-se a presença de um novo
subtipo de macrófagos residentes, distintos das células da micróglia, localizados no espaço
perivascular, em retinas humanas e de murganho. Estas células emitem uma fluorescência
específica e expressam constitutivamente receptores scavenger da classe A. Exibem um
movimento alternado constante, ao longo dos vasos sanguíneos, providenciando um
revestimento adicional a áreas mais delgadas da parede vascular. Para além disso,
acumulam peroxidase de rábano e lipoproteínas de baixa densidade acetiladas em
circulação, sem rotura da barreira hemato-retiniana. No conjunto, estes resultados sugerem
que estas células, com função scavenger, contribuem para o funcionamento da barreira
hemato-retiniana. Acumulam, também, ferro, em lisossomas, provavelmente por um
mecanismo de endocitose de ferritina, mediada por receptores TIM-2, sugerindo o seu
envolvimento na homeostasia do ferro na retina e em situações de alteração desta. Em
fases iniciais de degenerescência dos fotorreceptores, estas células migram para o local das
lesões, sugerindo a sua participação nos processos de retinopatia. Todos estes aspectos
são semelhantes aos descritos para as células Mato cerebrais. Por conseguinte, este estudo
estabelece, pela primeira vez, a presença de células Mato na retina.ABSTRACT -

retinopathy --
The retina contains distinct populations of monocyte-derived cells: perivascular macrophages
and microglia. In this work we describe the presence in mouse and human healthy retinas of
a new subtype of resident macrophages, present in the perivascular space, different from
microglia. These cells emitted specific autofluorescence and constitutively expressed the
scavenger receptor class A. They were seen to move in an oscillatory manner along blood
vessels, providing an additional coating to thinner areas of the vascular wall. Furthermore,
these macrophages also accumulated blood-borne horseradish peroxidase and acetylated
low-density lipoprotein without blood-retinal barrier pathological disruption. Taken together
these findings suggest that these cells fulfil a major role in the scavenging of blood-borne
molecules contributing to blood-retinal barrier function. In addition, they stored iron in
lysosomes, probably by the TIM-2 mediated binding of ferritin, indicating they probably are
involved in retinal iron homeostasis and related disorders. These cells were also early
detected at lesions during photoreceptor degeneration, suggesting a possible role in the
onset and development of retinopathy. All these features are consistent with those described
for brain Mato cells. Thus, this study shows, for the first time, the presence of autofluorescent

A distributed order viscoelastic model for small deformations
In this work we discuss the connection between classical, fractional and dis-
tributed order viscoelastic Maxwell models, presenting the basic theory supporting these
constitutive equations, and establishing some background on the admissibility of the dis-
tributed order Maxwell model. We derive the storage and loss modulus functions for the
distributed order viscoelastic model and perform a fitting to experimental data. The fitting

O ciclo da escrita
O presente trabalho foca a aprendizagem da expressão escrita pelos alunos do 1º Ciclo do Ensino Básico. Após a construção de um quadro sobre modelos e práticas de escrita, efectuamos um estudo com alunos do Primeiro Ciclo, 3.º ano, turmas B e C. Na senda, do processo de escrita e alicerçado no modelo de Flower e Hayes validamos o ciclo da escrita com dois grupos seleccionados para o efeito, servindo um grupo de estudo e o outro o controle do mesmo.This work focus on the learning of writing by students of the first cycle of the

Diagnostic assessment of the comfort of palliative patients: scoping review protocol
Introduction: The competence to evaluate is essential in any profession in the health area. The concept of comfort is intrinsically linked to palliative care, being a concern shared by any health professional. These, when analyzing, must know what they are looking for and what they should expect to find. Objectives: To map how health professionals assess people's comfort in palliative and end-of-life situations. Methods: will follow the methodology proposed by the Johanna Briggs Institute (JBI) for scoping reviews. Considered studies in Spanish, English and Portuguese, without temporal definition. Databases to search: MEDLINE Complete, Academic Search Complete, CINAHL Complete and Mediclatina (EBSCOhost), Scopus and Web of Science Core Collection. For unpublished studies, the RCAAP (open access repository in Portugal) will be used. Two independent reviewers will evaluate the articles and extract the data using a specific tool created. In case of discrepancies, a third reviewer will be requested. Results: It is expected to map the data used by health professionals in assessing the comfort of people in palliative care. Conclusions: This review will make it possible to identify instruments used to collect data and what is the professional competence of the group that has in its practice the assessment of comfort. This can form the basis for a systematic review and/or help identify gaps where it may be important to invest in the future. Virulencia, enzimas ligninolíticas y perfil metabólico de cryphonectria parasitica en cepas virulentas e hypovirulentas convertidas por CHV1 hipovirus
Cryphonectria parasitica, fungo responsável pelo cancro do castanheiro, causa lesões necróticas (cancros corticais) no tronco e ramos das árvores hospedeiras. O hipovírus, Cryphonectria hypovirus 1 (CHV1) reduz a virulência (hipovirulência) e altera a morfologia do fungo em cultura (redução da pigmentação e esporulação). As estirpes hipovirulentas CHV1 são utilizadas com sucesso na Europa como agentes de controlo biológico do Cancro do castanheiro.
Objetivos: O objetivo deste trabalho foi compreender o efeito do hipovírus na virulência e metabolismo do fungo, comparando a produção de algumas enzimas lenhinolíticas e os perfis metabólicos de estirpes virulentas e estirpes isogénicas de C. parasitica convertidas (com CHV1).
Métodos: A virulência de cada isolado foi avaliada por inoculação de micélio do fungo em maçãs (cv. Golden Delicious) e em ramos destacados de castanheiro com um ano de crescimento. Para a deteção da atividade de enzimas lenhinolíticas (lacases, peroxidases e celulases) foram usados vários substratos e compostos indicadores. O perfil metabólico de C. parasitica foi avaliado pelo sistema Biolog FF microplates pela utilização de 95 fontes diferentes de carbono.
Resultados: A utilização de MicroPlacas FF (Biolog, Inc.) indicaram que a utilização de 95 fontes de carbono pelos cinco isolados de C. parasitica, foram significativamente diferentes (p <0,001), quando os substratos foram agrupados em seis tipos de compostos químicos. Os maiores valores de AWCD foram obtidos para os hidratos de carbono, ácidos carboxílicos e polímeros, e os menores valores para os grupos aminas / amidas, aminoácidos e compostos diversos.
Conclusões: A avaliação da virulência de isolados de C. parasitica é importante para o estudo dos processos de hipovirulência mediados pelo hipovírus CHV1. Os ramos destacados de castanheiro foram, em nosso estudo, mais adequados que o teste em maçã para diferenciar as estirpes hipovirulentas das virulentas de C. parasitica. Os isolados virulentos evidenciaram sempre uma maior atividade de lacase induzida por ácido tânico (Lac3) e de outras enzimas lenhinolíticas (LiP, MnP e celulase) quando comparadas com os hipovirulentos. Os resultados da análise dos perfis metabólicos mostram que alguns grupos de substratos foram mais consumidos por isolados hipovirulentos. Estes estudos abrem novas perspetivas para entender o processo biológico usado pelo hipovírus, e sugerem que este é um método para discriminar estirpes hipovirulentas, e estudar a ecologia e a aptidão em campo destes isolados do fungo.Cryphonectria parasitica, the causal agent of chestnut blight, causes necrotic lesions (so-called cankers) on the bark of stems and branches of susceptible host trees. Cryphonectria hypovirus 1 (CHV1) infects C. parasitica and reduces the fungus virulence (hypovirulence) and alters the fungus morphology in culture (pigmentation and sporulation capacity). By these characteristics, the strains with hypovirus CHV1 are used in Europe as a biological control agent of chestnut blight.
Objectives: The aim of this work is to understand the effect of hypovirus on fungi virulence by comparing the production of some lignin-degrading enzymes and the metabolic profiles of some isogenic virulent and hypovirulent (converted and original) strains.
Methods: The virulence of each strain was evaluated by mycelial inoculation on apple fruits (cv. Golden Delicious) and on detached, one year old, chestnut branches. To detect the activity of ligninolytic enzymes (laccases, peroxidases and cellulases), various substrates and indicator compounds were used. The metabolic profile of C. parasitica was evaluated by the Biolog FF system using 95 different carbon sources.
Results: Virulent strains were found to cause more significant necrotic lesions in chestnut branches (p<0.05) and to produce larger lignin-degrading enzymes. The use of Biolog FF MicroPlates indicated that the use of 95 carbon sources five isolates of C. parasitica were significantly different (p <0.001), when the substrates were grouped into six types of chemical compounds. The highest AWCD values were obtained for carbohydrates, carboxylic acids and polymers, and the lowest values for amines/amides, amino acids and miscellaneous.
Conclusions: Virulence evaluation of C. parasitica strains is important to study the hypovirulence processes mediated by the hypovirus CHV1. The detached branches of chestnut were, in our study, more suitable than apple fruits test in discriminating hypovirulent from virulent strains of C. parasitica. Virulent strains showed higher activity of acid-tannic inducible laccase (Lac3) and other lignin-degrading enzymes (LiP, MnP, and cellulase) when compared with hypovirulent ones. The results of the metabolic profiles studies may lead to new perspectives for understanding the biological process used by the hypovirus therefore, this may suggest a method for discriminating hypovirulent strains and study ecology and field fitness in this fungal strains.Cryphonectria parasitica, el agente causal del Chancro del castaño, causa lesiones con necrosis (chancros corticales) en la corteza de los troncos y ramas de los árboles de hospedadores susceptibles. Exploring the role of Portuguese brands' actions on social media on consumer engagement
With the progressive evolution of technology, digital communications and social media quickly
turned into an indispensable practice of people's daily lives. At the same time, from a consumer
point of view, individuals crave more than just transactional exchanges, they want to bond,
connect with brands and engage in two-way communications. As a result, marketers saw a real
opportunity in social media, as an instrument to nurture relationships, interact with consumers
and increase engagement.
However, due to the lack of literature concerning the impact of brand-owned communities on
users' behaviour, this study aims to comprehend the role of firm-generated content on consumer
engagement, investigating for that purpose, three Portuguese brand fan pages on Facebook and
Instagram.
In order to do so, two complementary topics connected to the subject were also investigated online relationship cultivation strategies, and the combination of traditional and digital
communications- to achieve a more realistic perspective regarding marketing communications
in the digital world.
Results showed that brands implement interactivity and involvement strategies on Facebook
and Instagram as a way to connect and boost consumer's participation. Additionally, fans
respond better to photo-based posts and most commonly react to those posts through "likes".
Finally, regarding engagement rate, Instagram scored higher than Facebook.Com a progressiva evolução da tecnologia, as comunicações digitais e os "social media"
rapidamente se tornaram numa parte indispensável da rotina diária da maioria dos cidadãos. Ao
mesmo tempo, enquanto consumidores, os indivíduos procuram estabelecer relações com as
marcas que se estendam para além das transações. Ambicionam conseguir identificar-se e
relacionar-se, envolvendo-se num diálogo que valorize a comunicação entre as duas partes.
Consequentemente, os "marketers" vislumbraram uma excelente oportunidade nos social media
como uma ferramenta útil para cultivar relações, interagir com os consumidores e aumentar o
compromisso ("engagement").
No entanto, devido à falta de literatura acerca do impacto das comunidades "online" geridas por
marcas no comportamento dos usuários, este estudo pretende compreender o papel do conteúdo
gerado pelas empresas ("firm-generated content") no compromisso dos consumidores (consumer
engagement). Para tal, 3 páginas de marcas portuguesas vão ser estudadas no Facebook e no
Instagram.
Adicionalmente, dois tópicos complementares a este tema foram investigados - estratégias de
cultivação de relações e a combinação entre comunicações tradicionais e digitais - de modo a
obter uma perspectiva mais realista acerca das comunicações em marketing no mundo digital.
Os resultados demonstram que as marcas implementam estratégias de interatividade no
Facebook e Instagram de modo a criar ligações e aumentar a participação dos consumidores.
Foi também possível verificar que os seguidores preferem interagir com "posts" que contenham
fotografias e que a reação mais comum a esses posts se dá através de gostos/"likes". Por fim,
analisando a taxa de compromisso, foi possível verificar que o Instagram permite atingir níveis
mais elevados desta variável do que o Facebook
Financial constraints and financial crises: the case of Portuguese listed companies
The purpose of this study is to analyze the degree of financial constraints faced by the companies included on the Portuguese Stock General Index when accessing to external financing, especially after the beginning and during the most recent financial crisis that affected the world financial markets from 2007. According to this aim, a longitudinal database is collected from the SABI database and was analyzed under panel data methodology. The final sample is panel data of 430 firm-year observations, related to 43 companies, during the period 2006-2015. In line with previous literature, our results provide evidence that the payout ratio is an efficient measure of the degree of financial constraints; companies that pay out less (or no) dividends display higher sensitivity of the investment to the cash flow. Moreover, we also found that the investment sensitivity to cash flow intensifies immediately after and during the most recent financial crisis.(undefined
