    APPLICATION OF MICROSATELLITE MARKERS IN IDENTIFICATION AND PARENTAGE ASSIGNMENT OF SHEEP

    The accuracy of the parentage reference is the early step of information needed for genotyping study.Inaccurate data of recording sheep numbers is often occurred. Sometimes, we found sheep or lambs with uncertain original parents. This study was designed to identify the parentages of sheep and to find the putative parents of lambs. Using an advance DNA technology in molecular genetics particularly in genotyping study, those purposes can be clarified. DNA of sheep were isolated from fresh blood samples in EDTA using a high salt method. A concentration of 50ng/(il DNA was used for amplification. Several microsatellite markers were applied in the amplification. Using a DNA analyser and sequencer, PCR products were analysed to look at the figures of alleles (bands). Results showed that using microsatellite markers could inform that parents of several experimental lambs were incorrect. It was found that putative parents of some lambs were generated from incorrect parents. This study suggests that microsatellite marker can be used to clarify unknown original parents

    Activity and social interactions in a wideranging specialist scavenger, the Tasmanian devil (Sarcophilus harrisii), revealed by animalborne video collars

    Observing animals directly in the field provides the most accurate understanding of animal behaviour and resource selection. However, making prolonged observation of undisturbed animals is difficult or impossible for many species. To overcome this problem for the Tasmanian devil (Sarcophilus harrisii), a cryptic and nocturnal carnivore, we developed animal-borne video collars to investigate activity patterns, foraging behaviour and social interactions. We collected 173 hours of footage from 13 individual devils between 2013 and 2017. Devils were active mostly at night, and resting was the most common behaviour in all diel periods. Devils spent more time scavenging than hunting and exhibited opportunistic and flexible foraging behaviours. Scavenging occurred mostly in natural vegetation but also in anthropogenic vegetation and linear features (roads and fence lines). Scavenging frequency was inversely incremental with size e.g. small carcasses were scavenged most frequently. Agonistic interactions with conspecifics occurred most often when devils were traveling but also occurred over carcasses or dens. Interactions generally involved vocalisations and brief chases without physical contact. Our results highlight the importance of devils as a scavenger in the Tasmanian ecosystem, not just of large carcasses for which devils are well known but in cleaning up small items of carrion in the bush. Our results also show the complex nature of intraspecific interactions, revealing greater detail on the context in which interactions occur. In addition, this study demonstrates the benefits of using animal-borne imaging in quantifying behaviour of elusive, nocturnal carnivores not previously seen using conventional field methods

    Vaginal use of micronized progesterone for luteal support. A randomized study comparing utrogestan® and crinone® 8%

    Background and Objective. Luteal phase physiology is distorted by in vitro fertilization (IVF) cycles using gonadotropin-releasing hormone (GnRH) agonists and antagonists, Controlled ovarian hyperstimulation leads to luteal phase defect and for this reason, luteal phase support is now an integral part of IVF/ICSI-ET programs. The support is provided by hCG, progesterone or GnRH-a. This study compared the efficiency, safety and tolerance of two vaginal micronized progesterones, Utrogestan and Crinone 8%. Methods. 111 women, 18-40 years old, FSH < 10 IU/L and normal uterus findings were included. The efficiency of the two preparations to provide luteal support was evaluated by the fertilization, implantation, pregnancy and take-home baby rates. The safety was compared through the results of vaginal findings and vaginal inflammation markers before and after treatment. Comparison of tolerance was made by evaluating 21 subjective patient questionnaire parameters. Results. There were no significant differences between the preparations in terms of efficiency or safety though Crinone 8% was better tolerated. Conclusion. The outcomes of this study suggest that a vaginal gel with micronized progesterone (Crinone 8%) is the optimal choice at this time for luteal support. © 2017, PALACKY UNIV. All rights reserved

    Morphological and molecular changes in the murine placenta exposed to normobaric hypoxia throughout pregnancy.

    Chronic hypoxia is a common complication of pregnancy, arising through malperfusion of the placenta or pregnancy at high altitude. The present study investigated the effects of hypoxia on the growth of the placenta, which is the organ that interfaces between the mother and her fetus. Mice were housed in an hypoxic environment for the whole of gestation. An atmosphere of 13% oxygen induced fetal growth restriction (1182 ± 9 mg, n = 90 vs. 1044 ± 11 mg, n = 62, P < 0.05) but enhanced placental weight (907 ± 11 mg, n = 90 vs. 998 ± 15 mg, n = 62,P < 0.05). Stereological analyses revealed an increase in the volume of maternal blood spaces in the placenta, consistent with increased flow. At the molecular level, we observed activation of the protein kinase B (Akt)-mechanistic target of rapamycin growth and proliferation pathway. Chronic hypoxia also triggered mild endoplasmic reticulum stress, a conserved homeostatic response that mediates translational arrest through phosphorylation of eukaryotic initiation factor 2 subunit α. Surprisingly, although subunits of the mitochondrial electron transport chain complexes were reduced at the protein level, there was no evidence of intracellular energy depletion. Finally, we demonstrated sex-specific placental responses to chronic hypoxia. Placentas from male fetuses were heavier (1082 ± 2 mg, n = 30 vs. 928 ± 2 mg, n = 34, P < 0.05) and less susceptible to hypoxia-induced oxidative stress than those from females. Their capacity to adapt may explain why male fetuses were significantly less growth restricted at embryonic day 18.5 than their female counterparts. These findings are consistent with the concept that male fetuses are more aggressive with respect to their nutrient demands, which may place them at greater risk of adverse outcomes under limiting conditions.This study was supported by a grant from the Wellcome Trust (084804/2/08/Z).This is the final version of the article.     Comparative Morphology of the Penis and Clitoris in Four Species of Moles (Talpidae).

    The penile and clitoral anatomy of four species of Talpid moles (broad-footed, star-nosed, hairy-tailed, and Japanese shrew moles) were investigated to define penile and clitoral anatomy and to examine the relationship of the clitoral anatomy with the presence or absence of ovotestes. The ovotestis contains ovarian tissue and glandular tissue resembling fetal testicular tissue and can produce androgens. The ovotestis is present in star-nosed and hairy-tailed moles, but not in broad-footed and Japanese shrew moles. Using histology, three-dimensional reconstruction, and morphometric analysis, sexual dimorphism was examined with regard to a nine feature masculine trait score that included perineal appendage length (prepuce), anogenital distance, and presence/absence of bone. The presence/absence of ovotestes was discordant in all four mole species for sex differentiation features. For many sex differentiation features, discordance with ovotestes was observed in at least one mole species. The degree of concordance with ovotestes was highest for hairy-tailed moles and lowest for broad-footed moles. In relationship to phylogenetic clade, sex differentiation features also did not correlate with the similarity/divergence of the features and presence/absence of ovotestes. Hairy-tailed and Japanese shrew moles reside in separated clades, but they exhibit a high degree of congruence. Broad-footed and hairy-tailed moles reside within the same clade but had one of the lowest correlations in features and presence/absence of ovotestes. Thus, phylogenetic affinity and the presence/absence of ovotestes are poor predictors for most sex differentiation features within mole external genitalia

    Influence of speed of sample processing on placental energetics and signalling pathways: implications for tissue collection.

    INTRODUCTION: The placenta is metabolically highly active due to extensive endocrine and active transport functions. Hence, placental tissues soon become ischaemic after separation from the maternal blood supply. Ischaemia rapidly depletes intracellular ATP, and leads to activation of stress-response pathways aimed at reducing metabolic demands and conserving energy resources for vital functions. Therefore, this study aimed to elucidate the effects of ischaemia ex vivo as may occur during tissue collection on phosphorylation of placental proteins and kinases involved in growth and cell survival, and on mitochondrial complexes. METHODS: Eight term placentas obtained from normotensive non-laboured elective caesarean sections were kept at room-temperature and sampled at 10, 20, 30 and 45 min after delivery. Samples were analyzed by Western blotting. RESULTS: Between 10 and 45 min the survival signalling pathway intermediates, P-AKT, P-GSK3α and β, P-4E-BP1 and P-p70S6K were reduced by 30-65%. Stress signalling intermediates, P-eIF2α increased almost 3 fold after 45 min. However, other endoplasmic reticulum stress markers and the Heat Shock Proteins, HSP27, HSP70 and HSP90, did not change. Phosphorylation of AMPK, an energy sensor, was elevated 2 fold after 45 min. Contemporaneously, there was an ∼25% reduction in mitochondrial complex IV subunit I. DISCUSSION AND CONCLUSIONS: These results suggest that for placental signalling studies, samples should be taken and processed within 10 min of caesarean delivery to minimize the impact of ischaemia on protein phosphorylation

    Dark Ages Cold Period: A literature review and directions for future research

    Several late Quaternary studies have recorded cold and disturbed climates centred during the mid-first millennium AD and discussed these conditions under the term ‘Dark Ages Cold Period’ (DACP). A review of 114 palaeoclimate papers indicated that cold climates were common in the Northern Hemisphere between AD 400 and 765. There are also suggestions that some regions may have been relatively wet during the DACP, while those around the Mediterranean and the China/Tibetan Plateau indicate coinciding droughts. A set of environmental responses, however, indicate a delayed DACP interval (AD 509–865) postdating the actual climate signal. Previously, the DACP has been linked with the North Atlantic ice-rafting event at about 1400 years ago, while some evidence suggests an involvement of the North Atlantic Oscillation and/or El Niño–Southern Oscillation. More recently, another proposed phase of widespread cooling, the ‘Late Antique Little Ice Age’ (LALIA), overlaps with the DACP and has been tentatively linked with volcanic aerosol and solar irradiance variations reinforcing the climatic downturn since AD 536. Importantly, a higher number of proxy records extending over the first millennium AD is required for more rigorous assessments of climate variability and the forcing during these centuries and to disentangle the DACP and LALIA fingerprints in the proxy data, particularly to determine whether the DACP and the LALIA are distinct features. Also a richer network of both climate and environmental proxies is needed to evaluate the human–environment interactions, during the historical Migration Period, and thus through the DACP

    Examining the Relationships among the Coaching Climate, Life Skills Development and Well-Being in Sport

    Using Benson and Saito's framework for positive youth development, we investigated the relationships between the coaching climate, young people's perceived life skills development within sport, and their psychological well-being. British youth sport participants (N=326, Mage=13.81, range=11–18 years) completed a survey assessing the coaching climate, participants' perceived life skills development (teamwork, goal setting, time management, emotional skills, interpersonal communication, social skills, leadership, and problem solving and decision making) and psychological well-being (self-esteem, positive affect, and satisfaction with life). In all analyses, the coaching climate was positively related to young peoples' perceived development of life skills within sport and their psychological well-being. Total life skills development (a summative score of all eight life skills scores) was positively related to all three psychological well-being indicators – providing support for the "pile-up" effect – and partially mediated the relationships between the coaching climate and participants' psychological well-being. Interpretation of the results indicated that coaches should foster the development of multiple life skills in youth sport participants, as they are associated with participants' psychological well-being. One way this can be achieved is through autonomy-supportive coaching behaviours

    Geometric phase outside a Schwarzschild black hole and the Hawking effect

    We study the Hawking effect in terms of the geometric phase acquired by a two-level atom as a result of coupling to vacuum fluctuations outside a Schwarzschild black hole in a gedanken experiment. We treat the atom in interaction with a bath of fluctuating quantized massless scalar fields as an open quantum system, whose dynamics is governed by a master equation obtained by tracing over the field degrees of freedom. The nonunitary effects of this system are examined by analyzing the geometric phase for the Boulware, Unruh and Hartle-Hawking vacua respectively. We find, for all the three cases, that the geometric phase of the atom turns out to be affected by the space-time curvature which backscatters the vacuum field modes. In both the Unruh and Hartle-Hawking vacua, the geometric phase exhibits similar behaviors as if there were thermal radiation at the Hawking temperature from the black hole. So, a measurement of the change of the geometric phase as opposed to that in a flat space-time can in principle reveal the existence of the Hawking radiation.Comment: 14 pages, no figures, a typo in the References corrected, version to appear in JHEP. arXiv admin note: text overlap with arXiv:1109.033
