    Proposed herein is a spring-tab mechanism that can be provided for Universal Serial Bus (USB) connectors that can provide an improved electrical bond between USB connectors/housings for previously designed products that are already in-use/deployed in commercial environments. The electrical bond that can be created via the spring-tab connectors proposed herein may create a stronger hold than standard USB connectors that are typically used in products, which can help to prevent accidental and/or undesired removal of a USB device when it is in-use

    Genetic Changes to a Transcriptional Silencer Element Confers Phenotypic Diversity within and between Drosophila Species

    The modification of transcriptional regulation has become increasingly appreciated as a major contributor to morphological evolution. However, the role of negative-acting control elements (e.g. silencers) in generating morphological diversity has been generally overlooked relative to positive-acting “enhancer” elements. The highly variable body coloration patterns among Drosophilid insects represents a powerful model system in which the molecular alterations that underlie phenotypic diversity can be defined. In a survey of pigment phenotypes among geographically disparate Japanese populations of Drosophila auraria, we discovered a remarkable degree of variation in male-specific abdominal coloration. In testing the expression patterns of the major pigment-producing enzymes, we found that phenotypes uniquely correlated with differences in the expression of ebony, a gene required for yellow-colored cuticle. Assays of ebony’s transcriptional control region indicated that a lightly pigmented strain harbored cis-regulatory mutations that caused correlated changes in its expression. Through a series of chimeric reporter constructs between light and dark strain alleles, we localized function-altering mutations to a conserved silencer that mediates a male-specific pattern of ebony repression. This suggests that the light allele was derived through the loss of this silencer’s activity. Furthermore, examination of the ebony gene of D. serrata, a close relative of D. auraria which secondarily lost male-specific pigmentation revealed the parallel loss of this silencer element. These results demonstrate how loss-of-function mutations in a silencer element resulted in increased gene expression. We propose that the mutational inactivation of silencer elements may represent a favored path to evolve gene expression, impacting morphological traits

    Concentrating Membrane Proteins Using Asymmetric Traps and AC Electric Fields

    Membrane proteins are key components of the plasma membrane and are responsible for control of chemical ionic gradients, metabolite and nutrient transfer, and signal transduction between the interior of cells and the external environment. Of the genes in the human genome, 30% code for membrane proteins (Krogh et al. J. Mol. Biol.2001, 305, 567). Furthermore, many FDA-approved drugs target such proteins (Overington et al. Nat. Rev. Drug Discovery2006, 5, 993). However, the structure-function relationships of these are notably sparse because of difficulties in their purification and handling outside of their membranous environment. Methods that permit the manipulation of membrane components while they are still in the membrane would find widespread application in separation, purification, and eventual structure-function determination of these species (Poo et al. Nature1977, 265, 602). Here we show that asymmetrically patterned supported lipid bilayers in combination with AC electric fields can lead to efficient manipulation of charged components. We demonstrate the concentration and trapping of such components through the use of a “nested trap” and show that this method is capable of yielding an approximately 30-fold increase in the average protein concentration. Upon removal of the field, the material remains trapped for several hours as a result of topographically restricted diffusion. Our results indicate that this method can be used for concentrating and trapping charged membrane components while they are still within their membranous environment. We anticipate that our approach could find widespread application in the manipulation and study of membrane proteins

    When should a mechanical tricuspid valve replacement be considered?

    BackgroundIsolated mechanical tricuspid valve replacement (mTVR) is uncommon, early mortality is reported to be high, and little is known regarding the long-term outcome. We sought to evaluate the long-term outcome of mTVR.MethodsFrom 1980 to 2007, isolated mTVR was performed in 64 patients (33 men) at our institution; the median age was 45.5 years (6-71 years). There were 2192 tricuspid valve (TV) repairs and 137 isolated bioprosthetic TV replacements during the same time interval. Valve dysfunction was caused by congenital TV abnormality in 45 patients (70%), carcinoid heart disease in 13 (20%), traumatic TV regurgitation in 3 (5%), and other reason in 3 (5%). Twenty-three patients (36%) had at least 1 previous cardiac procedure (TV repair in 8 and bioprosthetic TV replacement in 7).ResultsMechanical prostheses used included Starr-Edwards (before 1993) in 36 patients (56%) and bileaflet prostheses in 28 (44%). Concomitant procedures included atrial septal defect closure in 28 (44%), arrhythmia surgery in 11 (17%), and pulmonary valvectomy for carcinoid disease in 10 patients (16%). Early mortality occurred in 5 patients (7.8%). Early morbidity included a permanent pacemaker in 9 (14%) and reexploration for bleeding in 2 patients (3%). Mean follow-up was 6 years (maximum 22.4 years). Five- and 10-year survival was 65% and 58%, respectively. There was no valve-related mortality. Late morbidity included valve thrombosis in 5 patients (8%); 3 were managed nonoperatively and 2 underwent TV rereplacement.ConclusionsIsolated mTVR still leads to increased early mortality. A mechanical valve can be considered in select situations when anticoagulation is necessary and in the presence of good right ventricular function

    Life expectancy for people diagnosed with HIV has improved dramatically however the number of new infections in the UK remains high. Understanding patterns of sexual behaviour among people living with diagnosed HIV, and the factors associated with having condom-less sex, is important for informing HIV prevention strategies and clinical care. In addition, in view of the current interest in a policy of early antiretroviral treatment (ART) for all people diagnosed with HIV in the UK, it is of particular importance to assess whether ART use is associated with increased levels of condom-less sex. In this context the ASTRA study was designed to investigate current sexual activity, and attitudes to HIV transmission risk, in a large unselected sample of HIV-infected patients under care in the UK. The study also gathered background information on demographic, socio-economic, lifestyle and disease-related characteristics, and physical and psychological symptoms, in order to identify other key factors impacting on HIV patients and the behaviours which underpin transmission. In this paper we describe the study rationale, design, methods, response rate and the demographic characteristics of the participants. People diagnosed with HIV infection attending 8 UK HIV out-patient clinics in 2011-2012 were invited to participate in the study. Those who agreed to participate completed a confidential, self-administered pen-and-paper questionnaire, and their latest CD4 count and viral load test results were recorded. During the study period, 5112 eligible patients were invited to take part in the study and 3258 completed questionnaires were obtained, representing a response rate of 64% of eligible patients. The study includes 2248 men who have sex with men (MSM), 373 heterosexual men and 637 women. Future results from ASTRA will be a key resource for understanding HIV transmission within the UK, targeting prevention efforts, and informing clinical care of individuals living with HIV

    The purpose of this report is to summarize and synthesize activities and achievements of the CPWF through the end of 2006. The CPWF is a CGIAR Challenge Program designed to take on the global challenge of water scarcity and food security. It takes the form of an international, multi-institutional research-for-development initiative that brings together scientists, development specialists, and river basin communities in Africa, Asia and Latin America. It seeks to create and disseminate international public goods (IPGs) helpful in achieving food security, reducing poverty, improving livelihoods, reducing agriculture–related pollution, and enhancing environmental security. This Challenge Program is a three-phase, 15-year endeavor. Several years have passed since the start of Phase 1 (2003-2008) which began with an inception phase in 2003 and was followed by full CPWF launch in January 2004. Research projects began field operations in mid-2004. This synthesis report, then, only describes work carried out in the first two and a half years of the Program. During this time, CPWF has conducted its research on water and food in nine benchmark basins, organized around five different themes. This work is being implemented through “first call projects”, “basin focal projects”, “small grant projects” and “synthesis research”. This present report is one example of the latter. CPWF projects have made considerable progress in developing innovative technologies, policies and institutions to address water and food issues. Some projects focused on improving agricultural water productivity. Others focused on developing mechanisms to inform multi-stakeholder dialogue and negotiation, or explored ways to value water used to produce ecosystem services. Advances were also made in understanding water-foodpoverty links, and their regional and global policy context

    Universal Holographic Chiral Dynamics in an External Magnetic Field

    In this work we further extend the investigation of holographic gauge theories in external magnetic fields, continuing earlier work. We study the phenomenon of magnetic catalysis of mass generation in 1+3 and 1+2 dimensions, using D3/D7- and D3/D5-brane systems, respectively. We obtain the low energy effective actions of the corresponding pseudo Goldstone bosons and study their dispersion relations. The D3/D7 system exhibits the usual Gell-Mann--Oakes--Renner (GMOR) relation and a relativistic dispersion relation, while the D3/D5 system exhibits a quadratic non-relativistic dispersion relation and a modified linear GMOR relation. The low energy effective action of the D3/D5 system is related to that describing magnon excitations in a ferromagnet. We also study properties of general Dp/Dq systems in an external magnetic field and verify the universality of the magnetic catalysis of dynamical symmetry breaking.Comment: 41 pages, 11 figures, references adde

    Flight Testing of Nap of-the-Earth Unmanned Helicopter Systems

    This paper describes recent results from a partnership between the Sikorsky Aircraft Corporation and the Georgia Institute of Technology to develop, improve, and flight test a sensor, guidance, navigation, control, and real-time flight path optimization system to support high performance nap-of-the-Earth helicopter flight. The emphasis here is on optimization for a combination of low height above terrain/obstacles and high speeds. Multiple methods for generating the desired flight path were evaluated, including (1) a simple processing of each laser scan; and (2) a potential field based method. Simulation and flight test results have been obtained utilizing an onboard laser scanner to detect terrain and obstacles while flying at low altitude, and have successfully demonstrated obstacle avoidance in a realistic semi-urban environment at speeds up to 40 ft/s while maintaining a miss distance of 50 ft horizontally and vertically. These results indicate that the technical approach is sound, paving the way for testing of even lower altitudes, higher speeds, and more aggressive maneuvering in future work

    Sum rules, plasma frequencies and Hall phenomenology in holographic plasmas

    We study the AC optical and hall conductivities of Dp/Dq-branes intersections in the probe approximation and use sum-rules to study various associated transport coefficients. We determine that the presence of massive fundamental matter, as compared to massless fundamental matter described holographically by a theory with no dimensional defects, reduces the plasma frequency. We further show that this is not the case when the brane intersections include defects. We discuss in detail how to implement correctly the regularization of retarded Green's functions so that the dispersion relations are satisfied and the low energy behaviour of the system is physically realistic.Comment: 25 pages, 5 figures. v2.minor changes, published versio
