    Da confiança organizacional ao comprometimento com o supervisor e com a organização:um modelo integrativo para a Administração Pública

    Dissertação de Mestrado em Administração PúblicaEste estudo pretende avaliar em que medida a relação de influência da confiança organizacional sobre o comprometimento organizacional é mediada pelo comprometimento com o supervisor, verificar as relações entre as componentes dos três constructos e procurar diferenças nos níveis de confiança e de comprometimentos no local de trabalho na Administração Pública Central e Autónoma Local. Foi aplicado um questionário a 904 sujeitos em diversas entidades do Sector Público e com base na investigação quantitativa é proposto um modelo, com recurso a modelagem por equações estruturais. Os resultados obtidos sugerem a possibilidade por um lado de estabelecer relações de influência da confiança organizacional sobre algumas componentes dos comprometimentos investigados, por outro de identificar algumas relações em que o comprometimento com o supervisor é um mediador da confiança organizacional sobre o comprometimento organizacional e por último que existem algumas diferenças estatisticamente significativas nas relações dos constructos entre os dois sectores públicos estudados. Existe a possibilidade de inferir que as recentes alterações gestionárias na Administração Pública portuguesa, na generalidade têm plasmado o Modelo da Nova Gestão Pública, estando ainda num estado embrionário o Modelo da Governança Pública, no que diz respeito a processos colaborativos no seio organizacional.This study aims to assess the extent to which the relationship of influence of organizational trust on the organizational commitment is mediated by the commitment to the supervisor, check the relationships between the components of the three constructs and look for differences in confidence levels and commitments in the workplace in Central and Local Public administration. It was applied a questionnaire to 904 subjects in several public sector entities and based on quantitative research, proposed a model using modeling for structural equation. The results suggest the possibility on one hand to establish relations of influence of organizational trust on some components of the investigated commitments, on the other to identify some relationships in which commitment to the supervisor mediates organizational trust on the organizational commitment and finally that there are some significant differences in the relations of the constructs between the two studied public sectors. It is possible to infer that recent management changes in the Portuguese Administration in general have shaped the model of New Public Management, still being in an embryonic state the Model of Public Governance, regarding the collaborative processes within the organization.N/

    Análise de desempenho de faixa preferencial para bicicletas

    The bicycle is a non-motorized type of transportation ideal for the concept of sustainable urban mobility. However, its insertion in the road system still lacks of public policies and greater actions to encourage the use in the form of implementation of acceptable infrastructures in roads. The concept of preferred bicycle traffic lane is an alternative for use in shared traffic. The present research seeks to analyze the performance of the use of a shared road with the presence of this type of lane in the city of João Pessoa-PB. The presence of the preferred use for cyclists at the right traffic lane inhibited the use of cars in this traffic range. After five years of implantation of this traffic lane, cyclists still feel insecure about riding bicycle at shared traffic road, after all, most of them still circulate in the lane at a distance of about 75 centimeters from the edge of the track, and instead of circulate at the center of the lane. Although 63% of car drivers respect the minimum recommended distance to overcome cyclists, only 30% have complied with one of the principles of using the preferred lane. To improve performance, enforcement actions and the promotion of educational campaigns on respect in traffic and on the principles of using a shared traffic system are necessary.A bicicleta é um meio de transporte não-motorizado ideal para o conceito de mobilidade urbana sustentável. Entretanto, a sua inserção no sistema viário ainda carece de políticas públicas e de maiores ações de incentivo ao uso na forma de implantação de infraestruturas adequadas nas vias. O conceito de faixa preferencial para bicicletas surge como alternativa de utilização em um tráfego compartilhado. A pesquisa buscou analisar o desempenho de utilização deste tipo de faixa em uma via compartilhada na cidade de João Pessoa-PB. A presença do uso preferencial para ciclistas na faixa da direita da via inibiu a utilização de carros nesta faixa de tráfego. Porém, após cinco anos da implantação do uso preferencial para bicicletas na faixa, os ciclistas ainda se sentem inseguros no trajeto de bicicleta na via compartilhada, afinal, a maioria deles ainda circulam a uma distância em torno de 75 centímetros do bordo da pista, ao em vez do centro da faixa. Apesar de 63% dos condutores de carros respeitarem a distância mínima recomendada ao ultrapassar os ciclistas, apenas 30% respeitaram um dos princípios de utilização da faixa preferencial. Para melhorar o desempenho, ações de fiscalização na via e da promoção de campanhas educativas sobre o respeito no trânsito e sobre os princípios de utilização de um sistema de tráfego compartilhado são necessárias

    Application of surface response analysis to the optimization of penicillin acylase purification in aqueous two-phase systems

    Penicillin acylase purification from an Escherichia coli crude extract using PEG 3350 – sodium citrate aqueous two phase systems was optimized. An experimental design was used to evaluate the influence of PEG, sodium citrate and sodium chloride on the purification parameters. A central composite design was defined centred on the previously found conditions for highest purification from an osmotic shock extract. Mathematical models for the partition coefficient of protein and enzyme, balance of protein and enzyme, yield and purification were calculated and statistically validated. Analysis of the contours of constant response as a function of PEG and sodium citrate concentrations for three different concentrations of NaCl revealed different effects of the three factors on the studied parameters. A maximum purification factor of 6.5 was predicted for PEG 3350, Sodium Citrate and NaCl concentrations of 15.1%, 11.0% and 8.52% respectively. However under these conditions the predicted yield was 61%. A better compromise between these two parameters can be found by superimposing the contour plots of the purification factor and yield for 10.3% NaCl. A region in the experimental space can be defined where the purification factor is always higher than 5.5 with yields exceeding 80%

    Partial purification of penicillin acylase from Escherichia Coli in poly(ethylene glycol)-sodium citrate aqueous two-phase systems

    Studies on the partition and purification of penicillin acylase from osmotic shock extract Escherichia coli were performed in poly (ethylene glycol)- citrate systems. Both partition behavior of the enzyme and total protein are similar to those described in other reports increasing ,with pH and tie-line length and decreasing with PEG molecular weight . However, some selectivity could be attained with PEG 1000 systems and long tie-line at pH 6.9. In these conditions 2.6 fold purification with 83% yield were achieved. Influence of pH on partition shows that is the composition of the system and not the net charge of the enzyme that determine the behaviour in these conditions. Addition of NaCl to PEG 3350 systems significantly increases the partition of the enzyme. Althought protein partition also increased, purification conditions were possible with 1.5 M NaCl where 5.7 fold purification and 85% yield was obtained. This was possible due to the higher hydrophobicity of the enzyme compared to that of most of contaminants proteins

    Carbon storage patterns and landscape sustainability in northeast portugal: a digital mapping approach

    This study investigated the impact of regional land abandonment in northeast Portugal. It specifically focused on carbon sequestration opportunities in the Upper Sabor RiverWatershed, situated in the northeast of Portugal, amidst agricultural land abandonment. The study involved mapping the distribution of soil organic carbon (SOC) across four soil layers (0–5 cm, 5–10 cm, 10–20 cm, and 20–30 cm) at 120 sampling points. The quantification of SOC storage (measured in Mg C ha−1) allowed for an analysis of its relationship with various landscape characteristics, including elevation, land use and land cover (LULC), normalized difference vegetation index (NDVI), modified soil-adjusted vegetation index (MSAVI), topographic wetness index (TWI), and erosion risk (ER). Six statistical tests were employed, including multivariate approaches like Cubist and Random Forest, within different scenarios to assess carbon distribution within the watershed’s soils. These modeling results were then utilized to propose strategies aimed at enhancing soil carbon storage. Notably, a significant discrepancy was observed in the carbon content between areas at higher elevations (>1000 m) and those at lower elevations (<800 m). Additionally, the study found that the amount of carbon stored in agricultural soils was often significantly lower than in other land use categories, including forests, mountain herbaceous vegetation, pasture, and shrub communities. Analyzing bi- and multivariate scenarios, it was determined that the scenario with the greatest number of independent variables (set 6) yielded the lowest RMSE (root mean squared error), serving as a key indicator for evaluating predicted values against observed values. However, it is important to note that the independent variables used in set 4 (elevation, LULC, and NDVI) had reasonably similar values.     Production of 6-aminopenicillanic acid in aqueous two-phase systems by recombinant Escherichia coli with intracellular penicillin acylase

    Bioconversion of Penicillin G in PEG 20000-Dextran T 70 aqueous two-phase systems was achieved using the recombinant Escherichia coli A56 (ppA22) with intracellular penicillin acylase as catalyst. The best conversion conditions were attained for: 7%(w/v) substrate (penicillin G), enzyme activity in bottom phase 52 U/ml, pH 7.8, temperature 37°C, reaction time 40 min. Five repeated batches could be performed in these conditions. Conversions ratios between 0.902-0.985mol of 6-aminopenicillanic acid (6-APA) per mol of penicillin G, were obtained and specific productivity was 3.6-4.6 μmol/min•ml.     O presente artigo tem como objetivo apresentar as reflexões de um professor de Educação Física em (constante) formação albino, negro e com baixa visão acerca de suas experiências e trajetórias formativas a partir de uma narrativa (auto)biográfica. O referencial teórico parte de um conceito de inclusão amplo, processual, infindável e dialético (SAWAIA, 2017; BOOTH E AINSCOW, 2012; SANTOS FONSECA E MELO, 2009),  permeado pelas elaborações sobre formação docente na e para perspectiva inclusiva (FONSECA, 2021). Optamos por uma parceria na escrita entre orientadora-orientando, mas enfatizando o protagonismo da narrativa (auto)biográfica em primeira pessoa, entremeado com referenciais teóricos que apoiam as reflexões sobre os desafios e as potências durante o percurso na educação básica, no curso de Licenciatura em Educação Física e na prática profissional, ao reconhecer-se negro, albino e com deficiência visua

    Population analysis of xenobiotic metabolizing genes in South Brazilian Euro and Afro-descendants

    Individual variability in xenobiotic metabolism has been associated with susceptibility to developing complex diseases. Genes involved in xenobiotic metabolism have been evaluated in association studies; the difficulty of obtaining accurate gene frequencies in mixed populations makes interpretation of the results difficult. We sought to estimate population parameters for the cytochrome P450 and glutathione S-transferase gene families, thus contributing to studies using these genes as markers. We describe the frequencies of six genes (CYP1A1, CYP2D6, CYP2E1, GSTM1, GSTT1, and GSTP1) and estimate population parameters in 115 Euro-descendants and 196 Afro-descendants from Curitiba, South of Brazil. PCR-based methods were used for genotyping, and statistical analysis were performed by AMOVA with ARLEQUIN software. The mutant allele frequencies in the Afro-descendants and Euro-descendants, respectively, were: CYP1A1*2A = 30.1% and 15.2%; CYP2D6*4 = 14.5% and 21.5%; CYP2E1*5B = 7.9% and 5%; GSTP1*B = 37.8% and 28.3%. The null genotype frequencies were: GSTM1*0 = 36.8% and 46.1%; GSTT1*0 = 24.2% and 17.4%

    Individual Acclimatization of Apis mellifera L. to the Thermal Homeostasis of the Colony

    Bees play an important role in maintaining biodiversity by promoting the pollination of numerous plant species. Recent global climate changes are affecting the average air temperature, thereby altering the biological processes of many species. The objective of this study was to evaluate the adaptation of Apis mellifera L. bees to temperature increases and their responses to thermal homeostasis in the colony. Research was performed at the Federal University of Paraíba Laboratory of Bees using three treatments: Control, 33 °C and 40 °C. For the latter two treatments, colonies were kept in a 24 m² climate chamber with an opening at the hive entrance, giving the bees access to the outside environment. The following parameters were evaluated: difference between internal and external hive temperature, thorax surface temperature and total protein concentration in the hemolymph. Internal colony temperature varied according to the external hive temperature. Nurse bees that care for larvae exhibited higher heat production, expressed as thorax surface temperature. Total protein content in the hemolymph was highest in the 40 °C treatment and decreased with ambient temperature. External hive temperature influences internal hive temperature, and nurse bees have higher capacities for thermogenesis
