    Multi-scale Habitat Use of Male Ruffed Grouse in the Black Hills National Forest

    Ruffed grouse (Bonasa umbellus) are native upland game birds and a management indicator species (MIS) for aspen (Populus tremuloides) in the Black Hills National Forest (Black Hills). Our objective was to assess resource selection of male ruffed grouse to identify the most appropriate scale to manage for aspen and ruffed grouse in the Black Hills. During spring 2007 and 2008, we conducted drumming surveys throughout the central and northern Black Hills to locate used and unused sites from which we compared habitat characteristics at increasing spatial scales. Aspen with \u3e70% overstory canopy cover (OCC) was important to the occurrence of ruffed grouse across all spatial scales, but was most influential within 1600 m of drumming sites. Probability of a site being used was maximized when 20% of the 1600-m scale (~804 ha) had aspen with \u3e70% OCC. Ruffed grouse also selected for areas with many small, regular shaped patches of aspen over those with few large patches. At the smallest scale evaluated of 200 m (~12.5 ha), ruffed grouse selected drumming logs in close proximity to high stem densities of aspen with a minimal presence of roads. Ponderosa pine (Pinus ponderosa) had a negative influence on site selection at the 400-m (~50 ha), 1600-m (~804 ha), and 4800-m (~7200 ha) scales. Management for ruffed grouse in the Black Hills as the MIS for aspen should focus on increasing the extent of aspen with a goal of at least 20% occurrence on the landscape. Management efforts also should incorporate multiple age and size classes of aspen with an emphasis on enhancing early successional habitat to provide valuable cover through increased stem densities

    Crusticorallina gen. nov., a nongeniculate genus in the subfamily Corallinoideae (Corallinales, Rhodophyta)

    Molecular phylogenetic analyses of 18S rDNA (SSU) gene sequences confirm the placement of Crusticorallina gen. nov. in Corallinoideae, the first non-geniculate genus in an otherwise geniculate subfamily. Crusticorallina is distinguished from all other coralline genera by the following suite of morpho-anatomical characters: 1) sunken, uniporate gametangial and bi/tetrasporangial conceptacles, 2) cells linked by cell fusions, not secondary pit connections, 3) an epithallus of 1 or 2 cell layers, 4) a hypothallus that occupies 50% or more of the total thallus thickness, 5) elongate meristematic cells, 6) trichocytes absent. Four species are recognized based on rbcL, psbA and COI-5P sequences, C. painei sp. nov., the generitype, C. adhaerens sp. nov., C. nootkana sp. nov. and C. muricata comb. nov., previously known as Pseudolithophyllum muricatum. Type material of Lithophyllum muricatum, basionym of C. muricata, in TRH comprises at least two taxa, and therefore we accept the previously designated lectotype specimen in UC that we sequenced to confirm its identity. Crusticorallina species are very difficult to distinguish using morpho-anatomical and/or habitat characters, although at specific sites, some species may be distinguished by a combination of morpho-anatomy, habitat and biogeography.     Libraries often develop and implement new services for the public. Sometimes they succeed; other times they fail. Expecting community members to understand the benefits of a great new idea on their own may not always work. But, taking a concierge approach to the implementation and maintenance of an innovative service may make all the difference to its success. Using a case study from the Embry-Riddle Aeronautical University (ERAU) library as a backdrop, this presentation will highlight the effectiveness of taking a concierge approach to implementing a new service in any type of library. In 2013, ERAU launched Scholarly Commons to gather and provide open access to faculty and student research. Library administration was concerned that initial response from the university community might be slow based on the experience of other institutional repositories. Faculty who might be willing to submit their work to an open access repository often did not engage due to concerns about copyright violation, plagiarism, perceived quality of “free” online resources, or simply lack of time and inclination to learn something new. After researching different approaches other academic libraries took to support similar repositories, ERAU decided to offer a concierge approach to developing Scholarly Commons. Librarians would be available to help members of the community through every step of the process from locating copyright permission to submission. In the seven years of its existence, Scholarly Commons has grown to include journals and magazines, conferences, open educational resources, and datasets, all of which are supported by library staff. Members of the library concierge team will highlight their part in implementing and maintaining this very successful innovative service

    Invited review: Lying time and the welfare of dairy cows

    Adequate time lying down is often considered an important aspect of dairy cow welfare. We examine what is known about cows’ motivation to lie down and the consequences for health and other indicators of biological function when this behavior is thwarted. We review the environmental and animal-based factors that affect lying time in the context of animal welfare. Cows can be highly motivated to lie down. They show rebound lying behavior after periods of forced standing and will sacrifice other activities, such as feeding, to lie down for an adequate amount of time. They will work, by pushing levers or weighted gates, to lie down and show possible indicators of frustration when lying behavior is thwarted. Some evidence suggests that risk of lameness is increased in environments that provide unfavorable conditions for cows to lie down and cows are forced to stand. Lameness itself can result in longer lying times, whereas mastitis reduces it. Cow-based factors such as reproductive status, age, and milk production influence lying time, but the welfare implications of these differences are unknown. Lower lying times are reported in pasture-based systems, dry lots, and bedded packs (9 h/d) compared with tiestalls and freestalls (10 to 12 h/d) in cross-farm research. Unfavorable conditions, including too few lying stalls for the number of cows, hard or wet lying surfaces, inadequate bedding, stalls that are too small or poorly designed, heat, and rain all reduce lying time. Time constraints, such as feeding or milking, can influence lying time. However, more information is needed about the implications of mediating factors such as the effect of the standing surface (concrete, pasture, or other surfaces) and cow behavior while standing (e.g., being restrained, walking, grazing) to understand the effect of low lying times on animal welfare. Many factors contribute to the difficulty of finding a valid threshold for daily lying time to use in the assessment of animal welfare. Although higher lying times often correspond with cow comfort, and lower lying times are seen in unfavorable conditions, exceptions occur, namely when cows lie down for longer because of disease or when they spend more time standing because of estrus or parturition, or to engage in other behaviors. In conclusion, lying behavior is important to dairy cattle, but caution and a full understanding of the context and the character of the animals in question is needed before drawing firm conclusions about animal welfare from measures of lying time.Peer reviewe

    Collagen Fiber Regulation in Human Pediatric Aortic Valve Development and Disease

    Congenital aortic valve stenosis (CAVS) affects up to 10% of the world population without medical therapies to treat the disease. New molecular targets are continually being sought that can halt CAVS progression. Collagen deregulation is a hallmark of CAVS yet remains mostly undefined. Here, histological studies were paired with high resolution accurate mass (HRAM) collagen-targeting proteomics to investigate collagen fiber production with collagen regulation associated with human AV development and pediatric end-stage CAVS (pCAVS). Histological studies identified collagen fiber realignment and unique regions of high-density collagen in pCAVS. Proteomic analysis reported specific collagen peptides are modified by hydroxylated prolines (HYP), a post-translational modification critical to stabilizing the collagen triple helix. Quantitative data analysis reported significant regulation of collagen HYP sites across patient categories. Non-collagen type ECM proteins identified (26 of the 44 total proteins) have direct interactions in collagen synthesis, regulation, or modification. Network analysis identified BAMBI (BMP and Activin Membrane Bound Inhibitor) as a potential upstream regulator of the collagen interactome. This is the first study to detail the collagen types and HYP modifications associated with human AV development and pCAVS. We anticipate that this study will inform new therapeutic avenues that inhibit valvular degradation in pCAVS and engineered options for valve replacement

    N-terminal β-strand underpins biochemical specialization of an ATG8 isoform

    Autophagy-related protein 8 (ATG8) is a highly conserved ubiquitin-like protein that modulates autophagy pathways by binding autophagic membranes and a number of proteins, including cargo receptors and core autophagy components. Throughout plant evolution, ATG8 has expanded from a single protein in algae to multiple isoforms in higher plants. However, the degree to which ATG8 isoforms have functionally specialized to bind distinct proteins remains unclear. Here, we describe a comprehensive protein-protein interaction resource, obtained using in planta immunoprecipitation (IP) followed by mass spectrometry (MS), to define the potato ATG8 interactome. We discovered that ATG8 isoforms bind distinct sets of plant proteins with varying degrees of overlap. This prompted us to define the biochemical basis of ATG8 specialization by comparing two potato ATG8 isoforms using both in vivo protein interaction assays and in vitro quantitative binding affinity analyses. These experiments revealed that the N-terminal β-strand-and, in particular, a single amino acid polymorphism-underpins binding specificity to the substrate PexRD54 by shaping the hydrophobic pocket that accommodates this protein's ATG8-interacting motif (AIM). Additional proteomics experiments indicated that the N-terminal β-strand shapes the broader ATG8 interactor profiles, defining interaction specificity with about 80 plant proteins. Our findings are consistent with the view that ATG8 isoforms comprise a layer of specificity in the regulation of selective autophagy pathways in plants

    Genomic surveillance uncovers a pandemic clonal lineage of the wheat blast fungus

    Wheat, one of the most important food crops, is threatened by a blast disease pandemic. Here, we show that a clonal lineage of the wheat blast fungus recently spread to Asia and Africa following two independent introductions from South America. Through a combination of genome analyses and laboratory experiments, we show that the decade-old blast pandemic lineage can be controlled by the Rmg8 disease resistance gene and is sensitive to strobilurin fungicides. However, we also highlight the potential of the pandemic clone to evolve fungicide-insensitive variants and sexually recombine with African lineages. This underscores the urgent need for genomic surveillance to track and mitigate the spread of wheat blast outside of South America and to guide preemptive wheat breeding for blast resistance

    Exome-Derived Adiponectin-Associated Variants Implicate Obesity and Lipid Biology

    Circulating levels of adiponectin, an adipocyte-secreted protein associated with cardiovascular and metabolic risk, are highly heritable. To gain insights into the biology that regulates adiponectin levels, we performed an exome array meta-analysis of 265,780 genetic variants in 67,739 individuals of European, Hispanic, African American, and East Asian ancestry. We identified 20 loci associated with adiponectin, including 11 that had been reported previously (p .60) spanning as much as 900 kb. To identify potential genes and mechanisms through which the previously unreported association signals act to affect adiponectin levels, we assessed cross-trait associations, expression quantitative trait loci in subcutaneous adipose, and biological pathways of nearby genes. Eight of the nine loci were also associated (p <1 x 10(-4)) with at least one obesity or lipid trait. Candidate genes include PRKAR2A, PTH1R, and HDAC9, which have been suggested to play roles in adipocyte differentiation or bone marrow adipose tissue. Taken together, these findings provide further insights into the processes that influence circulating adiponectin levels.Peer reviewe
