Bridge over the Chvalšinský Brook

Účelem bakalářské práce je návrh mostní konstrukce přes Chvalšinský potok v obci Chvalšiny. Konstrukce je navržena ve třech variantách, z nichž byla vybrána varianta předpjaté deskové konstrukce s délkou přemostění 13,5 m. Výpočet zatížení je proveden počítačovým programem Scia Engineer a porovnán s ručním výpočtem. Posouzení konstrukce je řešeno dle platných norem ČSN EN 1992-1-1.The project of the bridge construction aross the Chvalsinsky stream in the village Chvalsiny is the target of the bachelor’s thesis. The construction is designed in the three variations, all of which were chosen the variant prestressed slab structure with the length of bridging 13.5 m. The load calculation is performed by the computer program Scia Engineer and compared with manual calculations. The assessment of the structure is solved according to the standards EN 1992-1-1.

Bridge over a local road and brook

Předmětem diplomové práce je návrh mostní konstrukce, která převádí silnici I/11 přes Bystrý potok a místní komunikaci mezi Nebory a Oldřichovicemi. Konstrukce je navržena ve čtyřech variantách, z nichž byla vybrána varianta předpjaté jednotrámové konstrukce o čtyřech polích s délkou přemostění 106,6 m. Výpočet účinků zatížení je proveden počítačovým programem Scia Engineer. Posouzení konstrukce je řešeno dle platných evropských norem ČSN EN 1992-1-1. Dále je zpracována přehledná výkresová dokumentace, stavební postup a vizualizace mostu.The Diploma thesis deals with the design of the bridge construction that conveys the road I/11 across the Bystry brook and the local road between Nebory and Oldřichovice. The construction is designed in four options, one of which was selected is the variation of the prestressed onebeam construction of four fields and the lenght of bridging 106,6 m. Calculation of the effects of the load is carried by a computer program Scia Engineer. Assessing the design is done in accordance with applicable European standards EN 1992-1-1. It is also elaborated well arranged drawing documentation, construction process and visualization of the bridge.

Optimization of the Degassing Process in a Foundry

Diplomová práce byla vypracována ve společnosti Brembo Czech s.r.o. a zaměřuje se na dosažení optimálního nastavení technologického procesu odplynění slitiny hliníku ve stavu taveniny, která je následně v rámci společnosti využívána k výrobě podvozkových komponentů osobních automobilů. V první části diplomové práce jsou shrnuty teoretické základy k řešené problematice, na které v části druhé navazuje analýza současného stavu procesu, návrh a provedení experimentů, jejich vyhodnocení, aplikace optimálního nastavení a zhodnocení efektu pro výrobní proces.The Diploma thesis was done in corporation with company Brembo Czech s.r.o. and is focused on optimal setting of technological process for degassing aluminum alloy in melted state, which is then used for personal vehicles chassis components production. Cílem práce je za pomocí vybraných metod, kterými byli SWOT analýza, dotazník a rozhovor odhalit aktuální nedostatky. Následná komparace výsledků by měla přispět k jejich odstranění. Výsledky, ke kterým jsem dospěl na základě tohoto šetření, jsem prezentoval oběma manažerům.In my thesis I focused on a marketing analysis of chosen fitness centers in the city of Ostrava. The goal of this thesis was to use the methods of SWOT analysis, a survey and an interview to discover potential weaknesses in their marketing strategies. The following comparison of the results should assist in the elimination of said weaknesses. Možnosti využití trojrozměrné (3D) echokardiografie při měření dynamických změn funkční mitrální insuficience při zátěžové echokardiografii

The utility of three-dimensional (3D) colour Doppler echocardiography for the evaluation of dynamic changes of secondary mitral regurgitation during dynamic stress echocardiography Previous studies proved, that severe functional mitral regurgitation (FMR) at rest and/or its significant increase during exercise have been shown to be associated with reduced functional capacity and impaired prognosis. In these studies, FMR has been assessed using 2- dimensional (D) quantitative techniques: proximal isovelocity surface (PISA) and pulsed Doppler volumetry. These methods, however, have several known limitations. Recently, color Doppler 3-D echocardiography-derived vena contracta cross-sectional area (VCA) has emerged as a new method to assess FMR severity at rest. VCA at rest has been shown to have higher correlation with the 2-D integrative method or magnetic resonance-derived regurgitant volume than any single 2-D method. The prognostic significance of VCA at rest and its increase during exercise, however, has not been investigated. Therefore, the goal of our study was 1) to find out if the the relationship between exercise-induced changes of FMR assessed by VCA, clinical and Doppler characteristics is different from previous PISA derived ERO based studies, 2) to find resting predictors of exercise-induced...Možnosti využití trojrozměrné (3D) echokardiografie při měření dynamických změn funkční mitrální insuficience při zátěžové echokardiografii Předchozí práce prokázaly, že těžká funkční mitrální regurgitace (FMR) v klidu a/nebo její významné prohloubení při dynamické fyzické zátěži se pojí se sníženou funkční kapacitou a špatnou prognózou pacientů. V těchto studiích byla významnost FMR hodnocena pomocí 2- rozměrné (2D) echokardiografie: proximal isovelocity surface area (PISA) a pulzní Dopplerovská volumetrie. Tyto metody však mají četné limitace jako je např. předpoklad pravidelného kruhového tvaru efektivního regurgitačního ústí (ERO), podcenění významnosti ERO FMR, nízká reproducibilita s možným exponenciálním růstem chyby měření a nepřímé měření ERO. V nedávné minulosti zavedená metodika barevné trojrozměrné (3D) Dopplerovské echokardiografie umožňující měření plochy průřezu krčku regurgitačního jetu FMR (vena contracta area - VCA), jako nové metody hodnocení významnosti FMR v klidu., která umožňuje přímé měření ERO bez limitací spojených s tvarem regurgitačního ústí a průtokem. Consideration of the construction development and form of the castle core of Aueršperk (district Žďár nad Sázavou) on the basis of the discovery of an architectural element of the castle tower

The paper publishes the conclusions from a survey of the castle. It also deals with the finding of a stone console on the tower and its interpretation. Published archaeological material was also used. The detailed survey was supplemented by published historical reports. Subsequently, the castle served as an important settlement and fortified point of the Lords of Pernštejn. Abandonment of the castle did not occur until the second half of the 15th century

Brain abscess and pyocephalus: our experience with treatment

Introduction. Pyocephalus always presents serious complications in the treatment of brain abscesses, and is associated with high rates of mortality and morbidity. This study aimed to comprehensively evaluate this understandably feared complication from a purely medical perspective by using an evidence-based approach and drawing comparisons from the available literature, which mostly comprises case reports. Methods. This was a prospective monocentric study of all patients treated for brain abscesses at the Neurosurgery Clinic of the University Hospital Ostrava between 2012 and 2017. The cohort was divided into two groups for statistical comparison; one group comprised those in which pyocephalus occurred before or during treatment, while the other group comprised patients without this complication. Particular consideration was given to the effect of pyocephalus on morbidity and mortality rates and C-reactive protein levels, as well as to the identification of risk factors, and to its possible therapeutic influence. Patients were followed up for six months. Results. A total of 43 patients were treated for a brain abscess. An unequivocal diagnosis of pyocephalus was established via CT and MRI brain scans in five cases (11.6%). In the cohort as a whole, mortality and morbidity rates were 23.3% and 48.8% respectively. Among patients with pyocephalus the incidence of mortality and morbidity was 40% and 66.6% respectively. The presence of pyocephalus is not a significant predictor of either morbidity (p 0.575) or mortality (p 0.664). In patients with pyocephalus, we determined elevated CRP levels on the day of surgery (p 0.038). The occurrence of epileptic seizures in the acute phase of the disease is associated with a poor outcome (p 0.039). Conclusions. Pyocephalus will continue to be a serious complication in the treatment of brain abscesses, although we were unable to determine its utility as a prognostic factor. Apocynin and Diphenyleneiodonium Induce Oxidative Stress and Modulate PI3K/Akt and MAPK/Erk Activity in Mouse Embryonic Stem Cells

Reactive oxygen species (ROS) are important regulators of cellular functions. In embryonic stem cells, ROS are suggested to influence differentiation status. Regulated ROS formation is catalyzed primarily by NADPH-dependent oxidases (NOXs). Apocynin and diphenyleneiodonium are frequently used inhibitors of NOXs; however, both exhibit uncharacterized effects not related to NOXs inhibition. Interestingly, in our model of mouse embryonic stem cells we demonstrate low expression of NOXs. Therefore we aimed to clarify potential side effects of these drugs. Both apocynin and diphenyleneiodonium impaired proliferation of cells. Surprisingly, we observed prooxidant activity of these drugs determined by hydroethidine. Further, we revealed that apocynin inhibits PI3K/Akt pathway with its downstream transcriptional factor Nanog. Opposite to this, apocynin augmented activity of canonical Wnt signaling. On the contrary, diphenyleneiodonium activated both PI3K/Akt and Erk signaling pathways without affecting Wnt. Our data indicates limits and possible unexpected interactions of NOXs inhibitors with intracellular signaling pathways
