    Because of his public leadership of the philosophe party in eighteenth-century France, Voltaire stands today as the iconic example of the French Enlightenment philosopher. Denis Diderot (1713–1784) is often seen as Voltaire’s second in that role since it was around both men that the Enlightenment philosophes rallied as a movement after 1750. The epochal project, which Diderot jointly pursued with Jean le Rond D’Alembert, to “change the common way of thinking” through a comprehensive Encyclopedia, or Reasoned Dictionary of the Arts, Sciences, and Trades provided the emergent philosophe movement with the cause around which they would coalesce. Diderot also fought vigorously with Voltaire on behalf of the Encyclopédie project and its principles, becoming as a result a public leader of the Enlightenment philosophical party in France alongside Voltaire. He also worked, like Voltaire, as a writer and critical intellectual who willingly positioned himself against the grain of established authority, and one who used philosophy as a vehicle for political and social activism. Yet Diderot’s philosophy pursued many more agendas and dimensions than Voltaire’s. He also left behind a corpus of philosophical writings that marks him out as arguably the most sophisticated of all the Enlightenment philosophes, and as one of the great philosophical thinkers of the eighteenth-century. Despite the obvious sophistication of Diderot’s philosophy, his legacy has suffered because of the historical differences separating his writings from the discipline of philosophy as it is practiced today. Enlightenment philosophie was something very different from what professional academic philosophers mean by that term today, and Diderot’s writings are often ignored by modern philosophers because they do not appear to be philosophy as they know it. Like many Enlightenment philosophes, Diderot also worked as an homme de lettres first and foremost, and only as a philosopher narrowly construed in certain instances. He also never authored any recognizable work of “systematic philosophy” if by that term we mean writing in the vein of his contemporaries such as David Hume in his Treatise or Immanuel Kant in his Critiques. Yet Diderot made important contributions to modern philosophy, and if they are to be grasped, the historical differences separating his writing from philosophy today must be transcended, and his eclectic manner of working accepted and embraced. Diderot wrote works that we recognize today as philosophy, but he also wrote a great deal more than that, and the challenge presented by his eighteenth-century philosophie is to see the modern philosophy contained in all of it. For Diderot did not simply write plays, art criticism, prose fictions, and highly imaginative works of literature alongside his work in philosophy; he pursued philosophie through these ostensibly literary works as well. He experimented with genres, including philosophical genres, when crafting his thought, and his writing overall is redolent with a self-consciousness that makes any easy separation of his explicitly philosophical writings from his literary work well-nigh impossible. His publishing habits were similarly complex, for as a writer who suffered personally under censorship that made the traffic in illicit ideas a prosecutable offense in Old Regime France, Diderot often had very good reason to leave his work unpublished—and very often did. At the same time, censorship alone does not explain the peculiar mix of published and unpublished writings found in Diderot’s oeuvre. This historical complexity has given rise to some difficulty in assessing Diderot’s writings according to the disciplinary canon of modern philosophy. Condillac, Helvétius, and d’Holbach are the Enlightenment philosophes most commonly studied within philosophy departments because their writings appear to conform better with conventional understandings of what philosophy should look like as a genre and a linguistic idiom. By contrast, the works of Diderot tend to be studied only in literature or history departments. This is unfortunate, for the treatment of Diderot’s philosophie as something different from modern philosophy has cut contemporary philosophers off from the work of one of the most sophisticated, subtle, and complex philosophical thinkers of the eighteenth century. To some extent, the way in which Diderot’s philosophical work employs different genres but also, challenges the idea of genre itself, has made it seem (perhaps too easily) congenial to a more “Continental” philosophical tradition, and foreign to a more formally oriented “analytic” tradition. But that would ignore Diderot’s naturalistic commitments and the role the Encyclopédie played, e.g., in the self-image of philosophers of science in the Vienna Circle. Our entry seeks to go beyond such oppositions in dealing with Diderot as a philosopher. Neither perspective alone fully grasps the richness of Diderot’s contributions to modern philosophy, so in order to fully situate his philosophie within philosophy writ large, a flexible and reflexive attitude regarding his writings must be adopted. Every text in Diderot’s oeuvre needs to be treated as a participant in both his philosophie and his philosophical work, and our conventional understanding of the boundaries isolating art and literature from science and philosophy also needs to be suspended because very often these modern distinctions do not apply in Diderot’s case. He also manifests an awareness of the new and emergent disciplinary taxonomy arising at the time, targeting his philosophie on many occasions at an interrogation of these developing epistemological divisions. This reflexivity often makes his thought even more relevant today than it was when it was written. To capture the complexity of Diderot’s philosophie as philosophy, this article adopts this reflexive approach. It will proceed in two parts. An overview of Diderot’s life and major texts is offered in Part I so as to present his work and writings as particular episodes in a coherent eighteenth-century life and career. To simplify the reading of this biography, the text is offered in a two-level presentation. A short overview of the highlights of Diderot’s life and work is offered in Section 1 to give readers a schematic overview, but a more extensive presentation of his biography is available in the Biographical Supplement. A comprehensive analysis of Diderot’s major philosophical preoccupations as revealed in his writings is then offered in Section 2 so as to outline the contours of Diderot’s place within Enlightenment philosophie and modern philosophy overall. This is followed by brief concluding remarks in Section 3

    Exploring differential item functioning in the SF-36 by demographic, clinical, psychological and social factors in an osteoarthritis population

    The SF-36 is a very commonly used generic measure of health outcome in osteoarthritis (OA). An important, but frequently overlooked, aspect of validating health outcome measures is to establish if items work in the same way across subgroup of a population. That is, if respondents have the same 'true' level of outcome, does the item give the same score in different subgroups or is it biased towards one subgroup or another. Differential item functioning (DIF) can identify items that may be biased for one group or another and has been applied to measuring patient reported outcomes. Items may show DIF for different conditions and between cultures, however the SF-36 has not been specifically examined in an osteoarthritis population nor in a UK population. Hence, the aim of the study was to apply the DIF method to the SF-36 for a UK OA population. The sample comprised a community sample of 763 people with OA who participated in the Somerset and Avon Survey of Health. The SF-36 was explored for DIF with respect to demographic, social, clinical and psychological factors. Well developed ordinal regression models were used to identify DIF items. Results: DIF items were found by age (6 items), employment status (6 items), social class (2 items), mood (2 items), hip v knee (2 items), social deprivation (1 item) and body mass index (1 item). Although the impact of the DIF items rarely had a significant effect on the conclusions of group comparisons, in most cases there was a significant change in effect size. Overall, the SF-36 performed well with only a small number of DIF items identified, a reassuring finding in view of the frequent use of the SF-36 in OA. Nevertheless, where DIF items were identified it would be advisable to analyse data taking account of DIF items, especially when age effects are the focus of interest

    Individual Differences and Metacognitive Knowledge of Visual Search Strategy

    A crucial ability for an organism is to orient toward important objects and to ignore temporarily irrelevant objects. Attention provides the perceptual selectivity necessary to filter an overwhelming input of sensory information to allow for efficient object detection. Although much research has examined visual search and the ‘template’ of attentional set that allows for target detection, the behavior of individual subjects often reveals the limits of experimental control of attention. Few studies have examined important aspects such as individual differences and metacognitive strategies. The present study analyzes the data from two visual search experiments for a conjunctively defined target (Proulx, 2007). The data revealed attentional capture blindness, individual differences in search strategies, and a significant rate of metacognitive errors for the assessment of the strategies employed. These results highlight a challenge for visual attention studies to account for individual differences in search behavior and distractibility, and participants that do not (or are unable to) follow instructions

    Mid-mantle deformation inferred from seismic anisotropy

    With time, convective processes in the Earth's mantle will tend to align crystals, grains and inclusions. This mantle fabric is detectable seismologically, as it produces an anisotropy in material properties—in particular, a directional dependence in seismic-wave velocity. This alignment is enhanced at the boundaries of the mantle where there are rapid changes in the direction and magnitude of mantle flow, and therefore most observations of anisotropy are confined to the uppermost mantle or lithosphere and the lowermost-mantle analogue of the lithosphere, the D" region. Here we present evidence from shear-wave splitting measurements for mid-mantle anisotropy in the vicinity of the 660-km discontinuity, the boundary between the upper and lower mantle. Deep-focus earthquakes in the Tonga–Kermadec and New Hebrides subduction zones recorded at Australian seismograph stations record some of the largest values of shear-wave splitting hitherto reported. The results suggest that, at least locally, there may exist a mid-mantle boundary layer, which could indicate the impediment of flow between the upper and lower mantle in this region

    Differences in the signaling pathways of α1A- and α1B-adrenoceptors are related to different endosomal targeting

    Aims: To compare the constitutive and agonist-dependent endosomal trafficking of α1A- and α1B-adrenoceptors (ARs) and to establish if the internalization pattern determines the signaling pathways of each subtype. Methods: Using CypHer5 technology and VSV-G epitope tagged α1A- and α1B-ARs stably and transiently expressed in HEK 293 cells, we analyzed by confocal microscopy the constitutive and agonist-induced internalization of each subtype, and the temporal relationship between agonist induced internalization and the increase in intracellular calcium (determined by FLUO-3 flouorescence), or the phosphorylation of ERK1/2 and p38 MAP kinases (determined by Western blot). Results and Conclusions: Constitutive as well as agonist-induced trafficking of α1A and α1B ARs maintain two different endosomal pools of receptors: one located close to the plasma membrane and the other deeper into the cytosol. Each subtype exhibited specific characteristics of internalization and distribution between these pools that determines their signaling pathways: α1A-ARs, when located in the plasma membrane, signal through calcium and ERK1/2 pathways but, when translocated to deeper endosomes, through a mechanism sensitive to β-arrestin and concanavalin A, continue signaling through ERK1/2 and also activate the p38 pathway. α1B-ARs signal through calcium and ERK1/2 only when located in the membrane and the signals disappear after endocytosis and by disruption of the membrane lipid rafts by methyl-β-cyclodextrin

    Migraine headaches in Chronic Fatigue Syndrome (CFS): Comparison of two prospective cross-sectional studies

    <p>Abstract</p> <p>Background</p> <p>Headaches are more frequent in Chronic Fatigue Syndrome (CFS) than healthy control (HC) subjects. The 2004 International Headache Society (IHS) criteria were used to define CFS headache phenotypes.</p> <p>Methods</p> <p>Subjects in Cohort 1 (HC = 368; CFS = 203) completed questionnaires about many diverse symptoms by giving nominal (yes/no) answers. Cohort 2 (HC = 21; CFS = 67) had more focused evaluations. They scored symptom severities on 0 to 4 anchored ordinal scales, and had structured headache evaluations. All subjects had history and physical examinations; assessments for exclusion criteria; questionnaires about CFS related symptoms (0 to 4 scale), Multidimensional Fatigue Inventory (MFI) and Medical Outcome Survey Short Form 36 (MOS SF-36).</p> <p>Results</p> <p>Demographics, trends for the number of diffuse "functional" symptoms present, and severity of CFS case designation criteria symptoms were equivalent between CFS subjects in Cohorts 1 and 2. HC had significantly fewer symptoms, lower MFI and higher SF-36 domain scores than CFS in both cohorts. Migraine headaches were found in 84%, and tension-type headaches in 81% of Cohort 2 CFS. This compared to 5% and 45%, respectively, in HC. The CFS group had migraine without aura (60%; MO; CFS+MO), with aura (24%; CFS+MA), tension headaches only (12%), or no headaches (4%). Co-morbid tension and migraine headaches were found in 67% of CFS. CFS+MA had higher severity scores than CFS+MO for the sum of scores for poor memory, dizziness, balance, and numbness ("Neuro-construct", p = 0.002) and perceived heart rhythm disturbances, palpitations and noncardiac chest pain ("Cardio-construct"; p = 0.045, t-tests after Bonferroni corrections). CFS+MO subjects had lower pressure-induced pain thresholds (2.36 kg [1.95-2.78; 95% C.I.] n = 40) and a higher prevalence of fibromyalgia (47%; 1990 criteria) compared to HC (5.23 kg [3.95-6.52] n = 20; and 0%, respectively). Sumatriptan was beneficial for 13 out of 14 newly diagnosed CFS migraine subjects.</p> <p>Conclusions</p> <p>CFS subjects had higher prevalences of MO and MA than HC, suggesting that mechanisms of migraine pathogenesis such as central sensitization may contribute to CFS pathophysiology.</p> <p>Clinical Trial Registration</p> <p>Georgetown University IRB # 2006-481</p> <p>ClinicalTrials.gov <a href="http://www.clinicaltrials.gov/ct2/show/NCT00810329">NCT00810329</a></p

    Effects of an amylopectin and chromium complex on the anabolic response to a suboptimal dose of whey protein

    Background Previous research has demonstrated the permissive effect of insulin on muscle protein kinetics, and the enhanced insulin sensitizing effect of chromium. In the presence of adequate whole protein and/or essential amino acids (EAA), insulin has a stimulatory effect on muscle protein synthesis, whereas in conditions of lower blood EAA concentrations, insulin has an inhibitory effect on protein breakdown. In this study, we determined the effect of an amylopectin/chromium (ACr) complex on changes in plasma concentrations of EAA, insulin, glucose, and the fractional rate of muscle protein synthesis (FSR). Methods Using a double-blind, cross-over design, ten subjects (six men, four women) consumed 6 g whey protein + 2 g of the amylopectin-chromium complex (WPACr) or 6 g whey protein (WP) after an overnight fast. FSR was measured using a primed, continuous infusion of ring-d5-phenylalanine with serial muscle biopsies performed at 2, 4, and 8 h. Plasma EAA and insulin were assayed by ion-exchange chromatography and ELISA, respectively. After the biopsy at 4 h, subjects ingested their respective supplement, completed eight sets of bilateral isotonic leg extensions at 80% of their estimated 1-RM, and a final biopsy was obtained 4 h later. Results Both trials increased EAA similarly, with peak levels noted 30 min after ingestion. Insulin tended (p = 0.09) to be higher in the WPACr trial. Paired samples t-tests using baseline and 4-h post-ingestion FSR data separately for each group revealed significant increases in the WPACr group (+0.0197%/h, p = 0.0004) and no difference in the WP group (+0.01215%/hr, p = 0.23). Independent t-tests confirmed significant (p = 0.045) differences in post-treatment FSR between trials. Conclusions These data indicate that the addition of ACr to a 6 g dose of whey protein (WPACr) increases the FSR response beyond what is seen with a suboptimal dose of whey protein alone

    The use of common bean (Phaseolus vulgaris ) traditional varieties and their mixtures with commercial varieties to manage bean fly (Ophiomyia spp .) infestations in Uganda

    The bean fly (Ophiomyia spp.) is considered the most economically damaging field insect pest of common beans in Uganda. Despite the use of existing pest management approaches, reported damage has remained high. Forty-eight traditional and improved common bean varieties currently grown in farmers’ fields were evaluated for resistance against bean fly. Data on bean fly incidence, severity and root damage from bean stem maggot were collected. Generalized linear mixed model (GLMM) revealed significant resistance to bean fly in the Ugandan traditional varieties. A popular resistant traditional variety and a popular susceptible commercial variety were selected from the 48 varieties and evaluated in pure and mixed stands. The incidence of bean fly infestation on both varieties in mixtures with different arrangements (systematic random versus rows), and different proportions within each of the two arrangements, was measured and analysed using GLMMs. The proportion of resistant varieties in a mixture and the arrangement type significantly decreased bean fly damage compared to pure stands, with the highest decrease in damage registered in the systematic random mixture with at least 50 % of resistant variety. The highest reduction in root damage, obvious 21 days after planting, was found in systematic random mixtures with at least 50 % of the resistant variety. Small holder farmers in East Africa and elsewhere in the world have local preferences for growing bean varieties in genetic mixtures. These mixtures can be enhanced by the use of resistant varieties in the mixtures to reduce bean fly damage on susceptible popular varieties
