    Sheep as an experimental model: individual housing allowing visual, auditory, olfactory and tactile contact is not an obstacle for studies involving hormonal interrelationships

    Thirty-two ewes were used to determine whether individual housing, allowing contact with neighbours, induces a stress response. Ewes were housed in individual pens designed to allow the ewes to see, hear, smell and touch adjacent animals, and were distributed into four groups (n=8/group): ewes with subcutaneous implants containing melatonin and oestradiol (M+E), melatonin (M), oestradiol (E) and non-implanted control ewes (C). Heart rate, stress indicators (plasma cortisol, glucose, lactate and creatine kinase (CK) concentrations) and luteinizing hormone (LH) concentrations were measured hourly and compared with the resting values (before and after pen housing). Heart rate increased significantly during the introduction into the pen (P<0.001) in all groups, in comparison with the resting values. No significant differences between groups were observed for cortisol concentrations, with the exception of the M group, which showed the highest response (P<0.001) when ewes were introduced into the pens. Lactate, CK and glucose changes in comparison with the resting values were similar between groups. LH concentrations during pen housing decreased significantly in all groups in comparison with resting values. In conclusion, individual confinement of sheep allowing visual, auditory, olfactory and tactile contact with their neighbouring animals was not an obstacle for investigating particular hormonal interrelationships with multiple sampling procedures. However further investigations are required to determine if this conclusion applies to other hormone systems in sheep

    Comparative uptake of fluoride ion into enamel from various topical fluorides in vitro

An external link to the publisher’s copy is included.Background: There are many forms of topical fluoride available today, making the decision as to which is most effective to manage the immediate caries risk problem at hand, very difficult. The objective of this project was to determine the concentration and pattern of fluoride ion uptake into enamel from a variety of categories of topical fluoride recently available in Australia. Methods: Extracted, intact molar teeth were sectioned to provide six plates of smooth surface enamel. Windows of enamel 2 x 6mm were exposed to a variety of topical fluorides for periods simulating those used in vivo. Following drying, the slates of enamel were exposed to 2ml of 0.1M HCl as a chemical biopsy agent for incremental periods of time. The concentrations of fluoride ion in the biopsy solutions for both test and background (control) slates of enamel were determined directly using a fluoride combination selective electrode in conjunction with a high impedance pH meter. Cumulative amounts of fluoride were determined for each topical fluoride agent. Results: The concentrations of fluoride ion taken up into enamel were generally proportional to those present in each agent. However, those from APF gel greatly exceeded the amounts taken up from NaF gel. Also, the concentrations taken up from some of the highly concentrated metal fluorides were surprisingly low. Prior etching of enamel increased uptake and prolonged application of APF gel provided no extra benefit. Conclusions: Some topical fluorides, e.g., APF gel, provided a greatly increased uptake and to a greater depth than other self-application products.     The Mayari-Baracoa Ophiolitic Belt (MBOB, eastern Cuba) is composed of two large, chromite-rich massifs: Mayari-Cristal and Moa-Baracoa. The chromitites and hosting dunites were firstly affected by a regional serpentinization event, a subsequent episode of hydrothermal alteration (chloritization mainly) and, finally, these already altered bodies were crosscut by thin calcite-dominated veins. Analysed serpentines from serpentinized chromitites and dunites present very similar isotopic compositions (delta(18)O= + 4.7 parts per thousand to + 6.3 parts per thousand and deltaD = - 67 parts per thousand to - 60 parts per thousand), suggesting that the serpentinization process took place at moderate temperatures, in an oceanic environment. Serpentine formation by interaction with ocean water is also supported by the isotopic composition of chlorite and calcite. These results suggest that the serpentinization, chloritization and fracture filling processes of the Mayari-Baracoa Ophiolite Belt took place in a subocean floor scenario and, thus, that the Mayari-Baracoa serpentines represent a good example of serpentine formed during interaction with seawater. The oceanic origin of the serpentines from serpentinized chromitites and dunites from the MBOB indicate that the serpentinization of the mantle sequence occurred pre-thrusting (pre-emplacement in age).     Severe accordion effect: Myocardial ischemia due to wire complication during percutaneous coronary intervention: A case report

    A mechanical alteration during manoeuvring of stiff guidewires in tortuous coronary arteries frequently induces vessel wall shortening and coronary psedostenosis, referred as accordion phenomenon. Subtraction of the guidewires normally leads to the entire resolution of the lesions. A case of this transient angiographic finding, during percutaneous coronary intervention in a tortuous right coronary artery, which resulted in a flow limiting effect and myocardial ischemia, is described in the present report. Differential diagnosis from potential procedure complications and interventional methodology issues are discussed, while similar reports are reviewed

    Mycobacterium tuberculosis is a major human pathogen and the causative agent for the pulmonary disease, tuberculosis (TB). Current treatment programs to combat TB are under threat due to the emergence of multi-drug and extensively-drug resistant TB. As part of our efforts towards the discovery of new anti-tubercular leads, a number of potent tetrahydropyrazolo[1,5-a]pyrimidine-3-ca​rboxamide(THPP) and N-benzyl-6′,7′-dihydrospiro[piperidine-4,​4′-thieno[3,2-c]pyran](Spiro) analogues were recently identified against Mycobacterium tuberculosis and Mycobacterium bovis BCG through a high-throughput whole-cell screening campaign. Herein, we describe the attractive in vitro and in vivo anti-tubercular profiles of both lead series. The generation of M. tuberculosis spontaneous mutants and subsequent whole genome sequencing of several resistant mutants identified single mutations in the essential mmpL3 gene. This ‘genetic phenotype’ was further confirmed by a ‘chemical phenotype’, whereby M. bovis BCG treated with both the THPP and Spiro series resulted in the accumulation of trehalose monomycolate. In vivo efficacy evaluation of two optimized THPP and Spiro leads showed how the compounds were able to reduce >2 logs bacterial cfu counts in the lungs of infected mice

    Collective Animal Behavior from Bayesian Estimation and Probability Matching

    Animals living in groups make movement decisions that depend, among other factors, on social interactions with other group members. Our present understanding of social rules in animal collectives is based on empirical fits to observations and we lack first-principles approaches that allow their derivation. Here we show that patterns of collective decisions can be derived from the basic ability of animals to make probabilistic estimations in the presence of uncertainty. We build a decision-making model with two stages: Bayesian estimation and probabilistic matching.&#xd;&#xa;In the first stage, each animal makes a Bayesian estimation of which behavior is best to perform taking into account personal information about the environment and social information collected by observing the behaviors of other animals. In the probability matching stage, each animal chooses a behavior with a probability given by the Bayesian estimation that this behavior is the most appropriate one. This model derives very simple rules of interaction in animal collectives that depend only on two types of reliability parameters, one that each animal assigns to the other animals and another given by the quality of the non-social information. We test our model by obtaining theoretically a rich set of observed collective patterns of decisions in three-spined sticklebacks, Gasterosteus aculeatus, a shoaling fish species. The quantitative link shown between probabilistic estimation and collective rules of behavior allows a better contact with other fields such as foraging, mate selection, neurobiology and psychology, and gives predictions for experiments directly testing the relationship between estimation and collective behavior

    The effect of post-discharge educational intervention on patients in achieving objectives in modifiable risk factors six months after discharge following an episode of acute coronary syndrome, (CAM-2 Project): a randomized controlled trial

    <p>Abstract</p> <p>Objectives</p> <p>We investigated whether an intervention mainly consisting of a signed agreement between patient and physician on the objectives to be reached, improves reaching these secondary prevention objectives in modifiable cardiovascular risk factors six-months after discharge following an acute coronary syndrome.</p> <p>Background</p> <p>There is room to improve mid-term adherence to clinical guidelines' recommendations in coronary heart disease secondary prevention, specially non-pharmacological ones, often neglected.</p> <p>Methods</p> <p>In CAM-2, patients discharged after an acute coronary syndrome were randomly assigned to the intervention or the usual care group. The primary outcome was reaching therapeutic objectives in various secondary prevention variables: smoking, obesity, blood lipids, blood pressure control, exercise and taking of medication.</p> <p>Results</p> <p>1757 patients were recruited in 64 hospitals and 1510 (762 in the intervention and 748 in the control group) attended the six-months follow-up visit. After adjustment for potentially important variables, there were, between the intervention and control group, differences in the mean reduction of body mass index (0.5 vs. 0.2; p < 0.001) and waist circumference (1.6 cm vs. 0.6 cm; p = 0.05), proportion of patients who exercise regularly and those with total cholesterol below 175 mg/dl (64.7% vs. 56.5%; p = 0.001). The reported intake of medications was high in both groups for all the drugs considered with no differences except for statins (98.1% vs. 95.9%; p = 0.029).</p> <p>Conclusions</p> <p>At least in the short term, lifestyle changes among coronary heart disease patients are achievable by intensifying the responsibility of the patient himself by means of a simple and feasible intervention.</p

    Mitochondrial Diabetes in Children: Seek and You Will Find It

    Maternally Inherited Diabetes and Deafness (MIDD) is a rare form of diabetes due to defects in mitochondrial DNA (mtDNA). 3243 A>G is the mutation most frequently associated with this condition, but other mtDNA variants have been linked with a diabetic phenotype suggestive of MIDD. From 1989 to 2009, we clinically diagnosed mitochondrial diabetes in 11 diabetic children. Diagnosis was based on the presence of one or more of the following criteria: 1) maculopathy; 2) hearing impairment; 3) maternal heritability of diabetes/impaired fasting glucose and/or hearing impairment and/or maculopathy in three consecutive generations (or in two generations if 2 or 3 members of a family were affected). We sequenced the mtDNA in the 11 probands, in their mothers and in 80 controls. We identified 33 diabetes-suspected mutations, 1/33 was 3243A>G. Most patients (91%) and their mothers had mutations in complex I and/or IV of the respiratory chain. We measured the activity of these two enzymes and found that they were less active in mutated patients and their mothers than in the healthy control pool. The prevalence of hearing loss (36% vs 75–98%) and macular dystrophy (54% vs 86%) was lower in our mitochondrial diabetic adolescents than reported in adults. Moreover, we found a hitherto unknown association between mitochondrial diabetes and celiac disease. In conclusion, mitochondrial diabetes should be considered a complex syndrome with several phenotypic variants. Moreover, deafness is not an essential component of the disease in children. The whole mtDNA should be screened because the 3243A>G variant is not as frequent in children as in adults. In fact, 91% of our patients were mutated in the complex I and/or IV genes. The enzymatic assay may be a useful tool with which to confirm the pathogenic significance of detected variants
