    Comparative proteomic profiling of myofibrillar proteins in dry-cured ham with different proteolysis indices and adhesiveness

    Excessive proteolysis during dry-cured ham processing may lead to high adhesiveness and consumer dissatisfaction. The aim of this research is to identify biomarkers for proteolysis and adhesiveness. Two hundred biceps femoris porcine muscle samples from Spanish dry-cured ham were firstly evaluated for various physicochemical parameters, including their proteolysis indices and instrumental adhesiveness. Proteins of samples with extreme proteolysis indices were separated by two-dimensional electrophoresis and identified by tandem mass spectrometry (MALDI-TOF/TOF). We found that hams of higher proteolysis index had statistically significant increased adhesiveness. Proteomic analysis revealed statistically significant qualitative and quantitative differences between sample groups. Thus, protein fragments increased remarkably in samples with higher proteolysis index scores. In addition, higher proteolysis index hams showed increased degradation for a total of five non-redundant myofibrillar and sarcoplasmic proteins. However, myosin-1, α-actin and myosin-4 proteins were the biomarkers that underwent the most intense response to proteolysis and adhesiveness.info:eu-repo/semantics/acceptedVersio

    Evaluation of the performance of Euro-CORDEX Regional Climate Models for assessing hydrological climate change impacts in Great Britain: A comparison of different spatial resolutions and quantile mapping bias correction methods

    Regional Climate Models (RCMs) are an essential tool for analysing regional climate change impacts, such as hydrological change, as they provide simulations with more small-scale details and expected smaller errors than global climate models. There has been much effort to increase the spatial resolution and simulation skill of RCMs (i.e. through bias correction), yet the extent to which this improves the projection of hydrological change is unclear. Here, we evaluate the skill of five reanalysis-driven Euro-CORDEX RCMs in simulating precipitation and temperature, and as drivers of a hydrological model to simulate river flow on four UK catchments covering different physical, climatic and hydrological characteristics. We use a comprehensive range of evaluation indices for aspects of the distribution such as means and extremes, as well as for the structure of time series. We test whether high-resolution RCMs provide added value, through analysis of two RCM resolutions, 0.44° (50 km) and 0.11° (12.5 km), which are also bias-corrected employing the parametric quantile-mapping (QM) method, using the normal distribution for temperature, and the Gamma (GQM) and Double Gamma (DGQM) distributions for precipitation. The performance of these is considered for a range of meteorological variables and for the skill in simulating hydrological impacts at the catchment scale. In a small catchment with complex topography, the 0.11° RCMs clearly outperform their 0.44° version for precipitation and temperature, but when used in combination with the hydrological model, fail to capture the observed river flow distribution. In the other (larger) catchments, only one high-resolution RCM consistently outperforms its low-resolution version, implying that in general there is no added value from using the high-resolution RCMs in those catchments. Both resolutions produce river flow simulations that cover the observed flow duration curve, but the ensemble spread is large and therefore the simulations are difficult to use in practice. GQM decreases most of the simulation biases, except for extreme precipitation and high flows, which are further decreased by DGQM, which also reduces the multi-model simulation spread. Bias correction does not improve the representation of daily temporal variability measured by the Nash-Sutcliffe Efficiency Index, but it does for monthly variability, in particular when applying DGQM, which reduces most of the simulation biases. Overall, an increase in RCM resolution does not imply a better simulation of hydrology and bias-correction represents an alternative to ease decision-making

    Cognitive changes in patients with epilepsy identified through the MoCA test during neurology outpatient consultation

    Introduction Epilepsy is a chronic neurological disorder that may occur alongside cognitive changes, with effects on multiple cognitive domains. Objective To compare the cognitive performance of patients with epilepsy and healthy controls through Montreal Cognitive Assessment (MoCA) during outpatient consultation at a reference diagnostic center in Colombia and analyze and the influencing factors. Materials and methodology One-hundred and four patients during neurology outpatient consultation in the city of Cartagena, Colombia, were assessed with the (MoCA) test, i.e., 54 people who consulted for headache and have not been diagnosed with epilepsy (NEP) and 50 with a diagnosis of epilepsy (EPs) according to the diagnostic criteria of the International League Against Epilepsy (ILAE). Results Significant differences were found in the total mean scores of the (MoCA) between (EPs) and (NPE) groups (t = 4.72; p < 0.01), particularly in attention (t = 3.22; p < 0.02) and memory (t = 5.04; p < 0.01) dimensions. Additionally, a significant association was observed between years of schooling and (MoCA) scores (p = 0,019) but not between socioeconomic level (p = 0,510), age (p = 0,452) and the frequency of seizures (p = 0,471). Discussion Patients with epilepsy show lower scores in several cognitive domains in respect of the control group. The (MoCA) has proven its appropriateness for cognitive screening in the contexts of clinical neurology outpatient consultation

    Changes in toxin content, biomass and pigments of the dinoflagellate Alexandrium minutum during nitrogen refeeding and growth into nitrogen or phosphorus stress

    Two strains oi the paralytic shellfish toxin (PST) producing dinoflagellate Alexandrium minutum Halim (highly toxic ALl V and weakly toxic AL2V) were grown in batch culture with either nitrate or phosphate as the limiting nutrient. In comparison with cells of the strain AL1V, cells of AL2V grew at a similar C-specific Tale, had a higher C/N ratio, and lower ratios of chl a/chl C2and chl a/peridinin. Neither chlorophylls flor carotenoids could be used to estimate C-biomass, N-biomass or toxin content for this organismo The toxin profile for both strains was dominated (up to 95 %) by the gonyautoxin GTX4, with smaller proportions of GTX1, GTX2 and GTX3. The Tale of toxin synthesis for both strains was greatest 1 to 2 d after the N-refeeding of N-deprived cells, with the net Tale of toxin syn- .thesis exceeding that of C-biomass and cell division by a factor of up to 4. Toxin synthesis was not enhanced by short-term P-stress. N-stress alone led to a decrease in toxin cell-I, but P-stress followed by N-stress did not result in such a decline, implicating phosphorus in the regulation of toxin metabolism. Although arginine is a majar precursor for PST synthesis, taurine, glycine, glutamine, and cell N showed similar relations to that observed for arginine with respect to toxin contento Furthermore, the mole ratio of arginine/toxin could vary by a factor of up to 5 between AL1V and AL2V at peak values of toxin cell-1, and by more than 5 within a strain when growing under different conditions. These observations suggest that the relationship between free arginine content and toxin content is complex. No explanation for the higher toxin content of AL1V is apparent, except that AL1V has a higher N-content per cell and this may be conducive to a higher Tale of synthesis of the N-rich toxins.Publicado

    Functional traits driving species role in the structure of terrestrial vertebrate scavenger networks

    Species assemblages often have a non-random nested organization, which in vertebrate scavenger (carrion-consuming) assemblages is thought to be driven by facilitation in competitive environments. However, not all scavenger species play the same role in maintaining assemblage structure, as some species are obligate scavengers (i.e., vultures) and others are facultative, scavenging opportunistically. We used a database with 177 vertebrate scavenger species from 53 assemblages in 22 countries across five continents to identify which functional traits of scavenger species are key to maintaining the scavenging network structure. We used network analyses to relate ten traits hypothesized to affect assemblage structure with the “role” of each species in the scavenging assemblage in which it appeared. We characterized the role of a species in terms of both the proportion of monitored carcasses on which that species scavenged, or scavenging breadth (i.e., the species “normalized degree”), and the role of that species in the nested structure of the assemblage (i.e., the species “paired nested degree”), therefore identifying possible facilitative interactions among species. We found that species with high olfactory acuity, social foragers, and obligate scavengers had the widest scavenging breadth. We also found that social foragers had a large paired nested degree in scavenger assemblages, probably because their presence is easier to detect by other species to signal carcass occurrence.     Uniqueness of diffeomorphism invariant states on holonomy-flux algebras

    Loop quantum gravity is an approach to quantum gravity that starts from the Hamiltonian formulation in terms of a connection and its canonical conjugate. Quantization proceeds in the spirit of Dirac: First one defines an algebra of basic kinematical observables and represents it through operators on a suitable Hilbert space. In a second step, one implements the constraints. The main result of the paper concerns the representation theory of the kinematical algebra: We show that there is only one cyclic representation invariant under spatial diffeomorphisms. While this result is particularly important for loop quantum gravity, we are rather general: The precise definition of the abstract *-algebra of the basic kinematical observables we give could be used for any theory in which the configuration variable is a connection with a compact structure group. The variables are constructed from the holonomy map and from the fluxes of the momentum conjugate to the connection. The uniqueness result is relevant for any such theory invariant under spatial diffeomorphisms or being a part of a diffeomorphism invariant theory.Comment: 38 pages, one figure. v2: Minor changes, final version, as published in CM

    Energy band structure and intrinsic coherent properties in two weakly linked Bose Einstein Condensates

    The energy band structure and energy splitting due to quantum tunneling in two weakly linked Bose-Einstein condensates were calculated by using the instanton method. The intrinsic coherent properties of Bose Josephson junction were investigated in terms of energy splitting. For EC/EJ1E_{C}/E_{J}\ll 1, the energy splitting is small and the system is globally phase coherent. In the opposite limit, EC/EJ1E_{C}/E_{J}\gg 1, the energy splitting is large and the system becomes a phase dissipation. Our reslults suggest that one should investigate the coherence phenomna of BJJ in proper condition such as EC/EJ1E_{C}/E_{J}\sim 1.Comment: to appear in Phys. Rev. A, 2 figure

    A recombinant Yellow Fever 17D vaccine expressing Lassa virus glycoproteins

    AbstractThe Yellow Fever Vaccine 17D (YFV17D) has been used as a vector for the Lassa virus glycoprotein precursor (LASV-GPC) resulting in construction of YFV17D/LASV-GPC recombinant virus. The virus was replication-competent and processed the LASV-GPC in cell cultures. The recombinant replicated poorly in guinea pigs but still elicited specific antibodies against LASV and YFV17D antigens. A single subcutaneous injection of the recombinant vaccine protected strain 13 guinea pigs against fatal Lassa Fever. This study demonstrates the potential to develop an YFV17D-based bivalent vaccine against two viruses that are endemic in the same area of Africa

    Late glacial and post-glacial deposits of the Navamuno peatbog (Iberian Central System): Chronology and paleoenvironmental implications

    The Navamuno peatbog (Sierra de Bejar, western Spain) is a ~14 ha pseudo-endorheic depression with boundaries defined by a lateral moraine of the Cuerpo de Hombre paleoglacier and fault-line scarps on granite bedrock. The stratigraphy of the Navamu~no peatbog system is characterized here using borehole data to a depth of 20 m. An integrated interpretation from direct-push coring, dynamic probing boreholes and handheld auger drillings advances our knowledge of the Navamu~no polygenetic infill. Correlating this data with those obtained in other studies of the chronology and evolutionary sequence of the Cuerpo de Hombre paleoglacier has enabled us to establish the sequence of the hydrological system in the Navamuno depression. During the Late Pleistocene (MIS2), the depression was dammed by the Cuerpo de Hombre glacier and fed by its lateral meltwaters, and was filled with glaciolacustrine deposits. The onset of the Holocene in Navamuno is linked to a flat, fluviotorrential plain with episodes of local shallow pond/peat bog sedimentation. This evolutionary sequence is congruent with the age model obtained from available radiocarbon dating, obtaining 19 ages from ~800 cal yr BP (at depth 1.11 m) to ~16800 cal yr BP (at depth 15.90e16.0 m). Finally, the sedimentary record enabled interpretation of the environmental changes occurring in this zone during the late glacial (from the Older Dryas to the Younger Dryas) and postglacial (Holocene) stages, placing them within the paleoclimatic context of the Iberian Peninsula and Mediterranean regions

    Effect of sugammadex on processed EEG parameters in patients undergoing robot-assisted radical prostatectomy

    Background: Sugammadex has been associated with increases in the bispectral index (BIS). We evaluated the effects of sugammadex administration on quantitative electroencephalographic (EEG) and electromyographic (EMG) measures. Methods: We performed a prospective observational study of adult male patients undergoing robot-assisted radical prostatectomy. All patients received a sevoflurane-based general anaesthetic and a continuous infusion of rocuronium, which was reversed with 2 mg kg1 of sugammadex i.v. BIS, EEG, and EMG measures were captured with the BIS Vista™ monitor. Results: Twenty-five patients were included in this study. Compared with baseline, BIS increased at 4e6 min (b coefficient: 3.63; 95% confidence interval [CI]: 2.22e5.04; P<0.001), spectral edge frequency 95 (SEF95) increased at 2e4 min (b coefficient: 0.29; 95% CI: 0.05e0.52; P¼0.016) and 4e6 min (b coefficient: 0.71; 95% CI: 0.47e0.94; P<0.001), and EMG increased at 4e6 min (b coefficient: 1.91; 95% CI: 1.00e2.81; P<0.001) after sugammadex administration. Compared with baseline, increased beta power was observed at 2e4 min (b coefficient: 93; 95% CI: 1e185; P¼0.046) and 4e6 min (b coefficient: 208; 95% CI: 116e300; P<0.001), and decreased delta power was observed at 4e6 min (b coefficient: 526.72; 95% CI: 778 to 276; P<0.001) after sugammadex administration. Neither SEF95 nor frequency band data analysis adjusted for EMG showed substantial differences. None of the patients showed clinical signs of awakening. Conclusions: After neuromuscular block reversal with 2 mg kg1 sugammadex, BIS, SEF95, EMG, and beta power showed small but statistically significant increases over time, while delta power decreased
