    Aide au diagnostic des pathologies neurologiques par traitement d'images couleur de fibres de myéline

    L’analyse par diffraction des rayons X sur des échantillons de biopsie du nerf sural fournit une aide au diagnostic des neuropathologies. Ce procédé coûteux en personnels et en moyens matériels ne donne cependant que des mesures globales des composantes de la fibre nerveuse. Afin d’obtenir des mesures plus précises de la distribution des surfaces et des constituants des fibres, nous proposons une méthode automatisée par analyse d’images numériques couleur en microscopie optique. Notre méthode repose sur une classification couleur non supervisée qui nous permet de segmenter les images des fascicules nerveux en différentes régions correspondant aux structures histologiques à mesurer. Nos résultats montrent la faisabilité de la méthode proposée et fournissent une mesure rapide et précise pour l’aide au diagnostic des neuropathologies

    Study of Plants Growth by Image Analysis

    Analysis of plants growth is an important step for the evaluation of plants performance and productivity. This paper presents study of plants growth by image analysis from seeds and up to photosynthesis. To extract parameters two different segmentation methods are used, one for the seeds, and one for the plants, combined with morphological operations. The first image segmentation is achieved with Fuzzy C-Means clustering method and the second is achieved with entropy based method, using histogram evaluation. In tests, the segmented image is transformed in binary image and the relevant regions are extracted after applying morphological operation. The following parameters are calculated: the projected surface, the contour’s perimeter, the seed’s mass center, the projected surface, and the cotyledon’s and hyper cotyledon’slength for the plants

    Inhibition of granulomatous inflammation and prophylactic treatment of schistosomiasis with a combination of edelfosine and Praziquantel

    This is an open access article distributed under the terms of the Creative Commons Attribution License.[Background]: Schistosomiasis is the third most devastating tropical disease worldwide caused by blood flukes of the genus Schistosoma. This parasitic disease is due to immunologic reactions to Schistosoma eggs trapped in tissues. Egg-released antigens stimulate tissue-destructive inflammatory and granulomatous reactions, involving different immune cell populations, including T cells and granulocytes. Granulomas lead to collagen fibers deposition and fibrosis, resulting in organ damage. Praziquantel (PZQ) is the drug of choice for treating all species of schistosomes. However, PZQ kills only adult Schistosoma worms, not immature stages. The inability of PZQ to abort early infection or prevent re-infection, and the lack of prophylactic effect prompt the need for novel drugs and strategies for the prevention of schistosomiasis. [Methodology/Principal Findings]: Using in vitro and in vivo approaches, we have found that the alkylphospholipid analog edelfosine kills schistosomula, and displays anti-inflammatory activity. The combined treatment of PZQ and edelfosine during a few days before and after cercariae infection in a schistosomiasis mouse model, simulating a prophylactic treatment, led to seven major effects: a) killing of Schistosoma parasites at early and late development stages; b) reduction of hepatomegaly; c) granuloma size reduction; d) down-regulation of Th1, Th2 and Th17 responses at late post-infection times, thus inhibiting granuloma formation; e) upregulation of IL-10 at early post-infection times, thus potentiating anti-inflammatory actions; f) down-regulation of IL-10 at late post-infection times, thus favoring resistance to re-infection; g) reduction in the number of blood granulocytes in late post-infection times as compared to infected untreated animals.     Exercise Capacity in Children and Adolescents With Congenital Heart Disease: A Systematic Review and Meta-Analysis

    Congenital heart disease (CHD) entails structural defects in the morphogenesis of the heart or its main vessels. Analyzing exercise capacity of children and adolescents with CHD is important to improve their functional condition and quality of life, since it can allow timely intervention on poor prognostic factors associated with higher risk of morbidity and mortality.To describe exercise capacity in children and adolescents with CHD compared with healthy controls.A systematic review was carried out. Randomized clinical trials and observational studies were included assessing exercise capacity through direct and indirect methods in children and adolescents between 5 and 17 years-old. A sensitive analysis was performed including studies with CHD repaired participants. Additionally, it was sub-analyzed by age range (< and ? 12 years old). Two independent reviewers analyzed the studies, extracted the data, and assessed the quality of the evidence.5619 articles were found and 21 were considered for the review. Eighteen articles used the direct exercise capacity measurement method by cardiopulmonary exercise test (CPET). The CHD group showed significant differences in peak oxygen consumption (VO2peak) with a value of -7.9 ml/Kg/min (95% CI: -9.9, -5.9, p = 0.00001), maximum workload (Wmax) -41.5 (95% CI: -57.9, -25.1 watts, p = 0.00001), ventilatory equivalent (VE/VCO2 ) slope 2.6 (95% CI: 0.3, 4.8), oxygen pulse (O2 pulse)-2.4 ml/beat (95% CI: -3.7, -1.1, p = 0.0003), and maximum heart rate (HRmax) -15 bpm (95% CI: -18, -12 bpm, p = 0.00001), compared with healthy controls. Adolescents (? 12 yrs) with CHD had a greater reduction in VO2peak (-10.0 ml/Kg/min (95% CI: -12.0, -5.3), p < 0.00001), Wmax (-45.5 watts (95% CI: -54.4, -36.7), p < 0.00001) and HRmax (-21 bpm (95% CI: -28, -14), p<0.00001).Suffering CHD in childhood and adolescence is associated with lower exercise capacity as shown by worse VO2peak, Wmax, VE/VCO2 slope, O2 pulse, and HRmax compared with matched healthy controls.     Thermography as non invasive functional imaging for monitoring seedling growth

    We demonstrate the value of thermal imaging for monitoring growth of seedling organs. An automated procedure is introduced to perform the segmentation of the organs of the seedlings from the thermal contrasts in the images. Results of the automated procedure were assessed visually by expert operators in a separate procedure. This constitutes a new application of thermal imaging as a non invasive imaging providing functional information on the physiology of the seedlings

    Evaluation of the in vitro and in vivo efficacy of ruthenium polypyridyl compounds against breast cancer

    [Abstract] The clinical success of cisplatin, carboplatin, and oxaliplatin has sparked the interest of medicinal inorganic chemistry to synthesize and study compounds with non-platinum metal centers. Despite Ru(II)–polypyridyl complexes being widely studied and well established for their antitumor properties, there are not enough in vivo studies to establish the potentiality of this type of compound. Therefore, we report to the best of our knowledge the first in vivo study of Ru(II)–polypyridyl complexes against breast cancer with promising results. In order to conduct our study, we used MCF7 zebrafish xenografts and ruthenium complexes [Ru(bipy)2(C12H8N6-N,N)][CF3SO3]2Ru1 and [{Ru(bipy)2}2(μ-C12H8N6-N,N)][CF3SO3]4Ru2, which were recently developed by our group. Ru1 and Ru2 reduced the tumor size by an average of 30% without causing significant signs of lethality when administered at low doses of 1.25 mg·L−1. Moreover, the in vitro selectivity results were confirmed in vivo against MCF7 breast cancer cells. Surprisingly, this work suggests that both the mono- and the dinuclear Ru(II)–polypyridyl compounds have in vivo potential against breast cancer, since there were no significant differences between both treatments, highlighting Ru1 and Ru2 as promising chemotherapy agents in breast cancer therapy.Xunta de Galicia; ED431C 2018/39Portugal.     Propensity score matched comparison of transcatheter aortic valve implantation versus conventional surgery in intermediate and low risk aortic stenosis patients: A hint of real-world

    Background: Recently, the use of transcatheter aortic valve implantation (TAVI) in inter­mediate-low risk patients has been evaluated in the PARTNER II randomized trial. However, in the last years, this therapy has been employed in this scenario with underreported results, as compared to surgical aortic valve replacement (SAVR). Methods: We enrolled 362 consecutive patients with severe symptomatic aortic stenosis and intermediate-low surgical risk (logEuroSCORE &lt; 20%), treated in our center with TAVI (103 patients) or single SAVR (259 patients) between 2009 and 2014. Patients were matched according to age, gender, logEuroSCORE, and use of bioprosthesis. Results: Mean age of the patients was 73 ± 10.4 years, and 40.3% were women. LogEuroSCORE and Society Thoracic Surgeons score were 7.0 ± 4.4% and 4.2 ± 2.5%, respectively, with mean left ventricular ejection fraction of 52 ± 9%. There were no differences regarding other comorbidities. The length-of-hospitalization was 11 ± 5 days after TAVI vs. 17 ± 9 days after SAVR (p = 0.003). After matched comparison, no differences in terms of in-hospital mortality (5.7% after TAVI vs. 2.9% after SAVR, p = 0.687) and 1-year mortality (11.4% vs. 7.1%, p = 0.381) were found. The combined endpoint of stroke and mortality at 1-year was also similar between both groups (15.7% in TAVI patients vs. 14.4% after SAVR, p = 0.136). Multivariate analysis determined that aortic regurgitation (AR) was an independent predictor of mortality (OR = 3.623, 95% CI: 1.267–10.358, p = 0.016). Although the rate of AR was higher after TAVI, none of the patients treated with the newest generation devices (10.7%) presented more than a mild degree of AR. Conclusions: TAVI is feasible and shows comparable results to surgery in terms of early, 1-year mortality, as well as cerebrovascular events in patients with severe aortic stenosis and intermediate-low operative risk. Better transvalvular gradients, yet higher rates of AR were found, however, newer devices presented comparable rate of AR.

    Evaluation of 3D/2D Imaging and Image Processing Techniques for the Monitoring of Seed Imbibition

    Seed imbibition is a very important process in plant biology by which, thanks to a simple water income, a dry seed may turn into a developing organism. In natural conditions, this process occurs in the soil, e.g., with difficult access for a direct observation. Monitoring the seed imbibition with non-invasive imaging techniques is therefore an important and possibly challenging task if one tries to perform it in natural conditions. In this report, we describe a set of four different imaging techniques that enable to addressing this task either in 3D or in 2D. For each technique, the following items are proposed. A detailed experimental protocol is provided to acquire images of the imbibition process. With the illustration of real data, the significance of the physical quantities measured in terms of their relation to the income of water in the seed is presented. Complete image analysis pipelines are then proposed to extract dynamic information on the imbibition process from such monitoring experiments. A final discussion compares the advantages and current limitations of each technique in addition to elements concerning the associated throughput and cost.
