    A Survey Of IPv6 Address Usage In The Public Domain Name System

    The IPv6 protocol has been slowly increasing in use on the Internet. The main reason for the development of the protocol is that the address space provided by IPv4 is nearing exhaustion. The pool of addresses provided by IPv6 is 296 times larger than IPv4, and should be sufficient to provide an address for every device for the foreseeable future. Another potential advantage of this significantly large address space is the use of randomly assigned addresses as a security barrier as part of a defence in depth strategy. This research examined the addresses allocated by those implementing IPv6 to determine what method or pattern of allocation was being used by adopters of the protocol. This examination was done through the use of DNS queries of the AAAA IPv6 host record using public DNS servers. It was observed that 55.84% of IPv6 addresses were in the range of 0 to (232 − 1). For those addresses with unique interface identifier (IID) portions, a nearly equal number of sequential and random IIDs were observed. Hong Kong and Germany were found to have the greatest number of IPv6 addresses. These results suggest that adopters are allocating most addresses sequentially, meaning that no security advantage is being obtained. It is unclear as to whether this is through design or the following of accepted practice. Future research will continue to survey the IPv6 address space to determine whether the patterns observed here remain constant

    Activation of Caspases and p53 by Bovine Herpesvirus 1 Infection Results in Programmed Cell Death and Efficient Virus Release

    Programmed cell death (PCD), or apoptosis, is initiated in response to various stimuli, including virus infection. Bovine herpesvirus 1 (BHV-1) induces PCD in peripheral blood mononuclear cells at the G0/G1 phase of the cell cycle (E. Hanon, S. Hoornaert, F. Dequiedt, A. Vanderplasschen, J. Lyaku, L. Willems, and P.-P. Pastoret, Virology 232:351–358, 1997). However, penetration of virus particles is not required for PCD (E. Hanon, G. Meyer, A. Vanderplasschen, C. Dessy-Doize, E. Thiry, and P. P. Pastoret, J. Virol. 72:7638–7641, 1998). The mechanism by which BHV-1 induces PCD in peripheral blood mononuclear cells is not understood, nor is it clear whether nonlymphoid cells undergo PCD following infection. This study demonstrates that infection of bovine kidney (MDBK) cells with BHV-1 leads to PCD, as judged by terminal deoxynucleotidyltransferase- mediated dUTP-biotin nick end labeling, DNA laddering, and chromatin condensation. p53 appears to be important in this process, because p53 levels and promoter activity increased after infection. Expression of proteins that are stimulated by p53 (p21Waf1 and Bax) is also activated after infection. Cleavage of Bcl-xL, a protein that inhibits PCD, occurred after infection, suggesting that caspases (interleukin-1β- converting enzyme-like proteases) were activated. Other caspase substrates [poly(ADP-ribose) polymerase and actin] are also cleaved during the late stages of infection. Inhibition of caspase activity delayed cytotoxic activity and virus release but increased the overall virus yield. Taken together, these results indicate that nonlymphoid cells undergo PCD near the end of productive infection and further suggest that caspases enhance virus release

    Several years of satellite cloud photography were examined to determine to what extent oceanographic phenomena might be revealed through their influence on the local cloud structure. The region of the Peru Current was selected for the study. As anticipated, indications of known oceanographic features including the Peru Current itself and local upwelling centers were noted. Previously unknown features such as an apparent band of warmer water off the Peruvian coast and anomalous patches of calm water in mid-ocean were also found. A seasonal cloud climatology for the Peru Current area was developed. The importance of coordinated studies using satellites, ships and aircraft is emphasized

    L-arginase induces vascular dysfunction in old spontaneously hypertensive rats

    Background: Aging is a major non-modifiable risk factor for hypertension. Changes in aging are similar to those seen in hypertension in the vasculature. Also, aging increases the vascular dysfunction that occurs in hypertension. L-arginase action reduces substrate (L-arginine) availability for the formation of nitric oxide (NO). This reduces the level of NO and leads to reduced vasodilation and ultimately, vascular dysfunction. This study examines the hypothesis that age-dependent vascular dysfunction in SHRs is mediated by arginase.Methods: Young (12-14 weeks) and old (11-12 months) male Wistar and spontaneously hypertensive rats (SHR) were used. Mean arterial pressure (MAP) was measured in the rats. They were then euthanized and mesenteric resistance arteries (MRAs) and thoracic aortae were excised and placed in ice-cold physiological salt solution (PSS). Arterial segments were either snap-frozen in liquid nitrogen and stored for immunoblotting studies or cut into 2mm rings for reactivity studies. Cumulative concentration-response curves to acetylcholine (Ach; 10-9 – 3x10-5M) and sodium nitroprusside (SNP; 10-12 – 3x10-5 M) were performed in the absence or presence (30-minute exposure) of L-arginase, 0.05U/ML (MRA) or 0.5U/ML (aorta). Vessels were pre-contracted with phenylephrine (PE; 3x10-6M)Results: MAP increased during aging in the SHRs p<0.05 but not in the Wistar rats. Arginase impaired the endothelium-dependent relaxation responses of thoracic aortic and MRA arterial rings to Ach in the old Wistars and SHRs (Emax aorta: 29.42±2.19% vs 7.94±1.86%). Arginase also impaired endothelium-independent relaxation response to SNP in the old SHRs only (Emax aorta: 88.62±4.10% vs 31.45±10.61%). We also observed no differences in the serum arginase activity in the four groups of rats. On the contrary, arginase activity in the aortae of young Wistar rats was reduced compared to other groups.Conclusions: Arginase impairs both endothelium-dependent and –independent vasorelaxation responses, through the NO signaling pathway.Keywords: Hypertension, Arginase, aging, vascular dysfunction, endothelium, Nitric oxid

Herpetological research in the area, as well as frequent visits from amphibian and reptile enthusiasts, have helped to form a more complete understanding of amphibian and reptile distributions across the region. However, gaps in the recognized distribution of many species remain. Efforts to better document species occurrence are confounded by the lack of public land and the fact that most of south Texas is privately owned, making access to suitable habitat difficult (Schmidly et al. 2001

    New insights into anhydrobiosis using cellular dielectrophoresis-based characterization

    Late embryogenesis abundant (LEA) proteins are found in desiccation-tolerant species from all domains of life. Despite several decades of investigation, the molecular mechanisms by which LEA proteins confer desiccation tolerance are still unclear. In this study, dielectrophoresis (DEP) was used to determine the electrical properties of Drosophila melanogaster (Kc167) cells ectopically expressing LEA proteins from the anhydrobiotic brine shrimp, Artemia franciscana. Dielectrophoresis-based characterization data demonstrate that the expression of two different LEA proteins, AfrLEA3m and AfrLEA6, increases cytoplasmic conductivity of Kc167 cells to a similar extent above control values. The impact on cytoplasmic conductivity was surprising, given that the concentration of cytoplasmic ions is much higher than the concentrations of ectopically expressed proteins. The DEP data also supported previously reported data suggesting that AfrLEA3m can interact directly with membranes during water stress. This hypothesis was strengthened using scanning electron microscopy, where cells expressing AfrLEA3m were found to retain more circular morphology during desiccation, while control cells exhibited a larger variety of shapes in the desiccated state. These data demonstrate that DEP can be a powerful tool to investigate the role of LEA proteins in desiccation tolerance and may allow to characterize protein-membrane interactions in vivo, when direct observations are challenging

    An Evaluation of Retinal Imaging Technology for 4-H Beef and Sheep Identification

    The study reported here evaluated retinal imaging technology as a means of permanent identification of 4-H beef and sheep. The OptiReader» Device was used to capture digital images of 491 beef and 220 sheep during 4-H enrollment. A total of 317 beef and 159 sheep were re-imaged. The on-site visual verification rate was 96.2% for beef and 100% for sheep. A visual verification exercise showed that individuals could identify a pair of retinal images as a match 98.6 % of the time for beef. Retinal imaging is a viable method for enrolling beef and sheep

    Premature Expression of the Latency-Related RNA Encoded by Bovine Herpesvirus Type 1 Correlates With Higher Levels of Beta Interferon RNA Expression in Productively Infected Cells

    Bovine herpesvirus type 1 (BHV-1) is an important pathogen that can initiate bovine respiratory disease complex. Like other members of the subfamily Alphaherpesvirinae, BHV-1 establishes latency in sensory neurons. The latency-related (LR) gene expresses a family of alternatively spliced transcripts in infected sensory neurons that have the potential to encode several LR proteins. An LR mutant virus that contains three stop codons near the 5’ terminus of the first open reading frame in the LR gene does not express two LR proteins or reactivate from latency. In addition, the LR mutant virus induces higher levels of apoptosis in trigeminal ganglionic neurons and grows less efficiently in certain tissues of infected calves. In spite of the reduced pathogenesis, the LR mutant virus, wild-type BHV-1, and the LR rescued virus exhibit identical growth properties in cultured bovine cells. In this study, we demonstrated that during early phases of productive infection the LR mutant virus expressed higher levels of LR-RNA relative to the LR rescued virus or wt BHV-1. Bovine kidney cells infected with the LR mutant virus also induced higher levels of beta interferon RNA and interferon response genes. These results suggest that inappropriate expression of LR-RNA, in the absence of LR protein expression, may influence the latency-reactivation cycle and pathogenic potential of BHV-1
